| 1 | Time-domain analysis methodology for large-scale RLC circuits and its applications显示文摘With soaring work frequency and decreasing feature sizes, VLSI circuits with RLC parasitic components are more like analog circuits and should be carefully analyzed in physical design. However, the number of extracted RLC components is typically too large to be analyzed efficiently by using present analog circuit simulators like SPICE. In order to speedup the simulations without error penalty, this paper proposes a novel methodology to compress the time-descritized circuits resulted from numerical integration approximation at every time step. The main contribution of the methodology is the efficient structure-level compression of DC circuits containing many current sources, which is an important complement to present circuit analysis theory. The methodology consists of the following parts: 1) An approach is proposed to delete all intermediate nodes of RL branches. 2) An efficient approach is proposed to compress and back-solve parallel and serial branches so that it is error-free and of linear complexity to analyze circuits of tree topology. 3) The Y toπtransformation method is used to error-free reduce and back-solve the intermediate nodes of ladder circuits with the linear complexity. Thus, the whole simulation method is very accurate and of linear complexity to analyze circuits of chain topology. Based on the methodology, we propose several novel algorithms for efficiently solving RLC-model transient power/ground (P/G) networks. Among them, EQU-ADI algorithm of linear-complexity is proposed to solve RLC P/G networks with mesh-tree or mesh-chain topologies. | 2 | Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors. | Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo | 2020 | Cell Research2020,30,3: | 11 |
| 3 | Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images. | Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv | 2018 | Cancer Communications2018,38,1: | 9 |

| 5 | Four Closely-related RING-type E3 Ligases,APD1-4,are Involved in Pollen Mitosis II Regulation in Arabidopsis显示文摘Ubiquitination of proteins is one of the critical regulatory mechanisms in eukaryotes.In higher plants,protein ubiquitination plays an essential role in many biological processes,including hormone signaling,photomorphogenesis,and pathogen defense.However,the roles of protein ubiquitination in the repro-ductive process are not clear.In this study,we identified four plant-specific RING-finger genes designated Aberrant Pollen Development 1 (APD1) to APD4,as regulators of pollen mitosis Ⅱ (PMⅡ) in Arabidopsis thaliana (L.).The apd1 apd2 double mutant showed a significantly increased percentage of bicellular-like pollen at the mature pollen stage.Further downregulation of the APD3 and APD4 transcripts in apd1 apd2 by RNA interference (RNAi) resulted in more severe abnormal bicellular-like pollen phenotypes than in apd1 apd2,suggesting that cell division was defective in male gametogenesis.All of the four genes were expressed in multiple stages at different levels during male gametophyte development.Confocal analysis using green florescence fusion proteins (GFP) GFP-APD1 and GFP-APD2 showed that APDs are associated with intracellular membranes.Furthermore,APD2 had E2-dependent E3 ligase activity in vitro,and five APD2-interacting proteins were identified.Our results suggest that these four genes may be involved,redundantly,in regulating the PMⅡ process during male gametogenesis. | Guo Luo Hongya Gu Jingjing Liu Li-Jia Qu | 2012 | Journal of Integrative Plant Biology2012,54,10: | 8 |
| 6 | Glycolysis reprogramming in cancer-associated fibroblasts promotes the growth of oral cancer through the lncRNA H19/miR-675-5p/PFKFB3 signaling pathway显示文摘As an important component of the tumor microenvironment,cancer-associated fibroblasts(CAFs)secrete energy metabolites to supply energy for tumor progression.Abnormal regulation of long noncoding RNAs(lncRNAs)is thought to contribute to glucose metabolism,but the role of lncRNAs in glycolysis in oral CAFs has not been systematically examined.In the present study,by using RNA sequencing and bioinformatics analysis,we analyzed the lncRNA/mRNA profiles of normal fibroblasts(NFs)derived from normal tissues and CAFs derived from patients with oral squamous cell carcinoma(OSCC).LncRNA H19 was identified as a key lncRNA in oral CAFs and was synchronously upregulated in both oral cancer cell lines and CAFs.Using small interfering RNA(siRNA)strategies,we determined that lncRNA H19 knockdown affected proliferation,migration,and glycolysis in oral CAFs.We found that knockdown of lncRNA H19 by siRNA suppressed the MAPK signaling pathway,6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3)and miR-675-5p.Furthermore,the lncRNA H19/miR-675-5p/PFKFB3 axis was involved in promoting the glycolysis pathway in oral CAFs,as demonstrated by a luciferase reporter system assay and treatment with a miRNA-specific inhibitor.Our study presents a new way to understand glucose metabolism in oral CAFs,theoretically providing a novel biomarker for OSCC molecular diagnosis and a new target for antitumor therapy. | Jin Yang Xueke Shi Miao Yang Jingjing Luo Qinghong Gao Xiangjian Wang Yang Wu Yuan Tian Fanglong Wu Hongmei Zhou | 2021 | International Journal of Oral Science2021,13,1: | 8 |

| 8 | Gene characterization and molecular pathway analysis of reverse thermosensitive genic male sterility in eggplant(Solanum melongena L.)显示文摘The naturally occurring mutant eggplant line 05ms was identified with reverse thermosensitive genic male sterility(rTGMS),but its temperature-responsive fertility mechanisms remain largely unknown.Here,we studied the flower morphology,anther cellular structure,and genome-wide gene expression of this rTGMS line.Candidate genes for thermosensitive male sterility during the microspore development of 05ms and the temperature-insensitive line S63 under low-temperature(LT)and high-temperature(HT)conditions were identified.Under LT,tapetum cells were vacuolated and had delayed disintegration in 05ms.RNA-seq analysis indicated that DEGs were enriched in the KEGG pathways‘plant hormone signal transduction’,‘starch and sucrose metabolism’,and‘phenylpropanoid biosynthesis’.We identified two genes,4CLL1(Sme2.5_00368.1_g00010.1)and CKI1(Sme2.5_10056.1_g00002.1),which could potentially regulate eggplant anther development and may be candidate genes for rTGMS.Finally,we propose a working model of anther abortion for rTGMS in eggplant.CKI1 responds to LT stress and causes expression changes in genes related to anther development,such as 4CLL1,and the cellular structure of the tapetum becomes abnormal,causing male sterility.The findings of this study explain the underlying molecular mechanisms of male sterility in eggplant rTGMS lines. | Bing Li Xueping Chen Yanrong Wu Aixia Gu Jingjing Zhang Shuangxia Luo Xiurui Gao Jianjun Zhao Xiuqing Pan Shuxing Shen | 2019 | Horticulture Research2019,6,1: | 5 |
| 9 | Radiotherapy targeting cancer stem cells “awakens” them to induce tumour relapse and metastasis in oral cancer显示文摘Radiotherapy is one of the most common treatments for oral cancer. However, in the clinic, recurrence and metastasis of oral cancer occur after radiotherapy, and the underlying mechanism remains unclear. Cancer stem cells(CSCs), considered the 'seeds'of cancer, have been confirmed to be in a quiescent state in most established tumours, with their innate radioresistance helping them survive more easily when exposed to radiation than differentiated cancer cells. There is increasing evidence that CSCs play an important role in recurrence and metastasis post-radiotherapy in many cancers. However, little is known about how oral CSCs cause tumour recurrence and metastasis post-radiotherapy. In this review article, we will first summarise methods for the identification of oral CSCs and then focus on the characteristics of a CSC subpopulation induced by radiation, hereafter referred to as 'awakened'CSCs, to highlight their response to radiotherapy and potential role in tumour recurrence and metastasis post-radiotherapy as well as potential therapeutics targeting CSCs. | 12 | High-resolution mapping of brain vasculature and its impairment in the hippocampus of Alzheimer’s disease mice显示文摘Accumulating evidence indicates the critical importance of cerebrovascular dysfunction in the pathogenesis of Alzheimer’s disease(AD).However,systematic comparative studies on the precise brain vasculature of wild-type and AD model mice are still rare.Using an image-optimization method for analysing Micro-Optical Sectioning Tomography(MOST)data,we generated cross-scale whole-brain 3 D atlases that cover the entire vascular system from large vessels down to smallest capillaries at submicron resolution,for both wild-type mice and a transgenic(APP/PS1)mouse model of AD.In addition to distinct vascular patterns in different brain regions,we found that the main vessels of the molecular layer of the hippocampal dentate gyrus(DG-ml)undergo abrupt changes in both diameter and branch angle,spreading a unique comb-like pattern of capillaries.By using a quantitative analysis workflow,we identified in the hippocampus of AD mice an overall reduction of the mean vascular diameter,volume fraction and branch angle,with most significant impairment in the DG-ml.In addition,virtual endoscopy revealed irregular morphological features in the vessel lumen of the AD mice,potentially contributing to the impairment of blood flow.Our results demonstrate the capability of high-resolution cross-scale evaluation of brain vasculature and underscore the importance of studying hippocampal microcirculation for understanding AD pathogenesis. | Xiaochuan Zhang Xianzhen Yin Jingjing Zhang Anan Li Hui Gong Qingming Luo Haiyan Zhang Zhaobing Gao Hualiang Jiang | 2019 | National Science Review2019,6,6: | 4 |

| 17 | Alisol B 23-acetate-induced HepG2 hepatoma cell death through mTOR signaling-initiated G_1 cell cycle arrest and apoptosis: A quantitative proteomic study显示文摘Objective: The present study aimed to investigate the molecular events in alisol B 23-acetate(ABA) cytotoxic activity against a liver cancer cell line.Methods: First, we employed a quantitative proteomics approach based on stable isotope labeling by amino acids in cell culture(SILAC) to identify the different proteins expressed in HepG2 liver cancer cells upon exposure to ABA. Next, bioinformatics analyses through DAVID and STRING on-line tools were used to predict the pathways involved. Finally, we applied functional validation including cell cycle analysis and Western blotting for apoptosis and mTOR pathway-related proteins to confirm the bioinformatics predictions.Results: We identified 330 different proteins with the SILAC-based quantitative proteomics approach. The bioinformatics analysis and the functional validation revealed that the mTOR pathway, ribosome biogenesis, cell cycle, and apoptosis pathways were differentially regulated by ABA. G1 cell cycle arrest, apoptosis and mTOR inhibition were confirmed.Conclusions: ABA, a potential mTOR inhibitor, induces the disruption of ribosomal biogenesis. | 18 | Recruited CD68^(+)CD206^(+)macrophages orchestrate graft immune tolerance to prompt xenogeneic-dentin matrix-based tooth root regeneration显示文摘Successful regenerative medicine strategies of xenogeneic extracellular matrix need a synergistic balance among inflammation,fibrosis,and remodeling process.Adaptive macrophage subsets have been identified to modulate inflammation and orchestrate the repair of neighboring parenchymal tissues.This study fabricated PPARγ-primed CD68+CD206+M2 phenotype(M2γ),and firstly verified their anti-inflammatory and tissue-regenerating roles in xenogeneic bioengineered organ regeneration.Our results showed that Th1-type CD3^(+)CD8^(+)T cell response to xenogeneic-dentin matrix-based bioengineered root complex(xeno-complex)was significantly inhibited by M2γmacrophage in vitro.PPARγactivation also timely recruited CD68^(+)CD206^(+)tissue macrophage polarization to xeno-complex in vivo.These subsets alleviated proinflammatory cytokines(TNF-α,IFN-γ)at the inflammation site and decreased CD3^(+)CD8^(+)T lymphocytes in the periphery system.When translated to an orthotopic nonhuman primate model,PPARγ-primed M2 macrophages immunosuppressed IL-1β,IL-6,TNF-α,MMPs to enable xeno-complex to effectively escape immune-mediated rejection and initiate graft-host synergistic integrity.These collective activities promoted the differentiation of odontoblast-like and periodontal-like cells to guide pulp-dentin and cementum-PDLs-bone regeneration and rescued partially injured odontogenesis such as DSPP and periostin expression.Finally,the regenerated root showed structure-biomechanical and functional equivalency to the native tooth.The timely conversion of M1-to-M2 macrophage mainly orchestrated odontogenesis,fibrogenesis,and osteogenesis,which represents a potential modulator for intact parenchymal-stromal tissue regeneration of targeted organs. | Hui Li Jingjing Sun Hefeng Yang Xue Han Xiangyou Luo LiJun Liao Bo Yang Tian Zhu Fangjun Huo Weihua Guo Weidong Tian | 2021 | Bioactive Materials2021,6,4: | 2 |
