| 2 | Effect of Lingguizhugan decoction on myocardial Nuclear factor kappa B protein expression in rats with chronic heart failure显示文摘OBJECTIVE: To investigate the effect of Lingguizhugan decoction (LGZGD) on changes of cardiac structure and function, and its putative mechanism of action, by investigating mRNA and protein expression of myocardial nuclear factor kappa B(NF-κB), and the plasma content of NF-κB in rats with chronic heart failure.METHODS: The chronic heart failure (CHF) model in rats was induced by coronary artery ligation.Sham operation was performed in control rats. Six weeks after the procedure, rats were randomly classified into the various treatment groups: model CHF, Captopril (4.4 mg/kg), low LGZGD dose (2.1 g/kg), medium LGZGD dose (4.2 g/kg), and high LG-ZGD dose (8.4 g/kg). Treatments continued for 4 consecutive weeks. Changes of hemodynamic indices were observed by the PowerLab data acquisition and analysis system. Morphological changes of myocardium were observed by hematoxylin and eosin staining, and Masson staining. The mRNA and protein expression of myocardial NF-κB were detected by reverse transcription-polymerase chain reaction and western blotting, respectively. The plasma content of NF-κB was detected by enzyme-linked immuno-sorbent assay.RESULTS: CHF rats showed significant dysfunction in hemodynamic indices and in cardiac structure.Compared with the sham operation group, mRNA expression of myocardial NF-κB and plasma content of NF-κB of the model group was significantly increased. All three doses of LGZGD, and Captopril,improved the hemodynamic dysfunction, and inhibited the change of cardiac structure while significantly improving the survival rate. | 4 | Climate indexes of phytoliths from Homo erectus' cave deposits in Nanjing显示文摘A study on phytoliths and their climate indexes is carried out from Homo erectus?cave deposits in HuluCave, Nanjing. Evidence shows that phytolith assemblages of the cave deposits are dominated by the cold resistant types with a lower warm index, reflecting an overall cold inclined climate. This possibly connects the cave deposits with glacial climate to a great extent, which is in accordance with thenorthern fauna revealed by fossil mammals and temperateclimate indicated by pollen assemblages. | 6 | Prognosis of microscopic polyangiitis with renal involvement:report of 60 Chinese patients显示文摘With the widespread clinical application of renal biopsy and serum antineutrophil cytoplasmic antibody (ANCA) testing, the percentage of renal injuries caused by systemic small vessel vasculitis (SVV) including microscopic polyangiitis (MPA) and Wegener’ s granulomatosis (WG) is on the rise in China. According to our previous report, SVV constituted 1.09% of diagnoses from all renal biopsies (5. 8% in secondary nephritis) and 14. 4% of that from renal biopsies from patients with chronic renal,failure, among which MPA is the most common type. Many studies reported that SVV with rapid progressive glomerulonephritis had poor prognosis with a 5-year renal survival less than 75%. However, there is a lack of large-sample statistical analysis about the long-term prognosis of SVV in China. Indeed, the clinical manifestations of SVV in China are quite different from those reported in other countries. | 7 | Effect of Wuziyanzong Pill on sperm quality and calcium ion content in oligoasthenospermia rats显示文摘OBJECTIVE:To study the effect of Wuziyanzong treatment on the sperm quality and content of calcium ions(Ca 2+) in oligoasthenospermia rats.METHODS:A model of oligoasthenospermia was induced in 50 Sprague Dawley rats by treatment with tripterygium glycosides at 30 mg/kg per day for 8 weeks.They were divided randomly into a model group,a positive group(Huangjingzanyu capsule,3.01 g/kg),and low,medium and high dose Wuziyanzong treatment groups(2.30,4.60,9.20 g/kg crude drug respectively) with 10 in each group.Another 10 rats were used as a control group.The rats in the control and model groups were administered distilled water,while the rats in the remaining groups were administered Wuziyanzong for 30 d.The epididymides were removed,spermatozoa recovered and the sperm density and viability were measured.The spermatozoa were purified and the contents of Ca 2+ in the cytoplasm and mitochondria were detected by flow cytometry and atomic absorption spectrometry,respectively.RESULTS:After 8 weeks of treatment with tripterygium glycosides,the sperm density,sperm activity and the Ca 2 + content of spermatozoa in the model rats were all significantly decreased compared with the control group(all P<0.05).After 30 d treatment,the sperm density and activity improved and the Ca 2 + content of sperm were increased significantly in the medium and high dose Wuziyanzong treatment groups in comparison with the model group(all P<0.05).CONCLUSION:The Wuziyanzong treatment increased sperm density,improved sperm viability and enhanced the content of Ca 2+ in the sperm cytoplasm and mitochondria in this rat model of oligoasthenospermia. | Tongsheng Wang Jinling Huang Deling Wu Qing Li Xiangguo Liu Han Chen Lei Qiao Dongdong Li Jun Li | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 11 |
| 8 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. | 9 | The effects of the formula of amino acids enriched BCAA on nutritional support in traumatic patients显示文摘AIM: To investigate the formula of amino acid enriched BCAA on nutritional support in traumatic patients after operation.METHODS: 40 adult patients after moderate or large abdominal operations were enrolled in a prospective,randomly and single-blind-controlled study, and total parenteral nutrition (TPN) with either formula of amino acid (AA group, 20 cases) or formula of amino acid enriched BCAA (BCAA group, 20 cases). From the second day after operation, total parenteral nutrition was infused to the patients in both groups with equal calorie and equal nitrogen by central or peripheral vein during more than 12 hours per day for 6 days. Meanwhile, nitrogen balance was assayed by collecting 24 hours urine for 6 days. The markers of protein metabolism were investigated such as amino acid patterns, levels of total protein, albumin, prealbumin,transferrin and fibronectin in serum.RESULTS: The positive nitrogen balance in BCAA group occurred two days earlier than that in AA group. The serum levels oftotal protein and albumin in BCAA group were increased more obviously than that in AA group. | 11 | Enrichment and hydrogen production by marine anaerobic hydrogen-producing microflora显示文摘Acid, alkali, heat-shock, KNO3 and control pretreatment methods applied to anaerobic sludge were evaluated for their ability to selectively enrich the marine hydrogen-producing mixed microflora. Sea-water culture medium was used as the substrate. The hydrogen yield of pretreated microflora was higher than that of the un-pretreated control (P < 0.05). Among the pretreatment methods studied, heat-shock pretreatment yielded the greatest hydrogen production, which was 14.6 times that of the control. When the effect of initial pH on hydrogen production of heat-shock pretreated samples was studied, hydrogen was produced over the entire pH range (pH 4 - 10). The hydrogen yield peaked at initial pH 8 (79 mL/g sucrose) and then steadily decreased as the initial pH increased. Sucrose con-sumption was high at neutral initial pH. | 13 | Molecular evolution of the exon 2 of CHS genes and the possibility of its application to plant phylogenetic analysis显示文摘The exon 2 of chalcone synthase (CHS) gene is relatively conserved during evolution. In this study, three exon 2 fragments from two species in gymnosperm (Cycas panzhihuaensis, Ginkgo biloba ) and seven from four species in angiosperm (Magnolia denudata, Salix babylonica, Nymphaea tetragona, Camellia japonica) have been amplified by PCR from genomic DNA and sequenced. Together with other 73 sequences of CHS collected from EMBL database and literature, these sequences, which embrace 19 families of gymnosperm and angiosperm, have been analyzed for their phylogenetic relations by parsimony method. The result indicated that sequences from the same systematic family usually grouped together except those from Theaceae, Magnoliaceae and Nymphaeaceae. The relative rate test revealed the rate heterogeneity of CHS genes among the families. | 14 | Vulnerabilities and integrity of precise point positioning for intelligent transport systems:overview and analysis显示文摘The implementation of Intelligent Transport System (ITS) technology is expected to significantly improve road safety and traffic efficiency. One of the key components of ITS is precise vehicle positioning. Positioning with decimetre to sub-metre accuracy is a fundamental capability for self-driving, and other automated applications. Global Navigation Satellite System (GNSS) Precise Point Positioning (PPP) is an attractive positioning approach for ITS due to its relatively low-cost and flexibility. However, GNSS PPP is vulnerable to several effects, especially those caused by the challenging urban environments, where the ITS technology is most likely needed. To meet the high integrity requirements of ITS applications, it is necessary to carefully analyse potential faults and failures of PPP and to study relevant integrity monitoring methods. In this paper an overview of vulnerabilities of GNSS PPP is presented to identify the faults that need to be monitored when developing PPP integrity monitoring methods. These vulnerabilities are categorised into different groups according to their impact and error sources to assist integrity fault analysis, which is demonstrated with Failure Modes and Effects Analysis (FMEA) and Fault Tree Analysis (FTA) methods. The main vulnerabilities are discussed in detail, along with their causes, characteristics, impact on users, and related mitigation methods. In addition, research on integrity monitoring methods used for accounting for the threats and faults in PPP for ITS applications is briefly reviewed. | 15 | Pinning controllability of autonomous Boolean control networks显示文摘Autonomous Boolean networks(ABNs), which are developed to model the Boolean networks(BNs)with regulatory delays, are well known for their advantages of characterizing the intrinsic evolution rules of biological systems such as the gene regulatory networks. As a special type of ABNs with binary inputs, the autonomous Boolean control networks(ABCNs) are introduced for designing and analyzing the therapeutic intervention strategies where the binary inputs represent whether a certain medicine is dominated or not. An important problem in the therapeutic intervention is to design a control sequence steering an ABCN from an undesirable location(implying a diseased state) to a desirable one(corresponding to a healthy state). Motivated by such background, this paper aims to investigate the reachability and controllability of ABCNs with pinning controllers. Several necessary and sufficient criteria are provided by resorting to the semi-tensor product techniques of matrices. | 20 | Use of endoscopic naso-pancreatic drainage in the treatment of severe acute pancreatitis显示文摘AIM: To review the experience on the use of endoscopic nasopancreatic drainage (ENPD) in the treatment of severe acute pancreatitis (SAP).METHODS: Since March 1998, under the regular management of SAP with non-operative method, ENPD has been randomly used in 14 patients. The average age of the patients was 41.3±15.9 (years), with 8 males and 6 females.The time from onset to admission was 32.9±22.8 (hours). 8cases were found to have gallbladder stone. The daily output of pancreatic fluid was measured. The body temperature,heart rate, WBC count, blood glucose, blood calcium, PaO2,blood and urine levels of amylase were detected on the fifth day and compared with their respective data on the first day.Therapeutic results and hospitalization times were recorded.RESULTS: The time of drainage was 7.3±4.0 days. The daily drainage outputs of the first five days were 236.4±176.6,287.1±164.7, 284.6±216.4, 435.0±357.8 and 377.8±223.8 ml,respectively. The decreases in body temperature, heart rate,WBC counts, blood and urine levels of amylase and the increase in PaO2 were significant on the fifth day when compared with those on the first day. Infection of pancreatic necrosis was found in one patient and controlled by antiinfectives. 6 out of 8 patients with gallbladder stone were operated during hospital stay. All patients were cured and diischarged and the average hospital stay was 28.1±11.6 days.CONCLUSION: ENPD is an effective method for the drainage of pancreatic fluid and might have an important role in the treatment of SAP.
