1Natural Variation in CCD4 Promoter Underpins Species-Specific Evolution of Red Coloration in Citrus Peel显示文摘Carotenoids and apocarotenoids act as phytohormones and volatile precursors that influence plant development and confer aesthetic and nutritional value critical to consumer preference.Citrus fruits display considerable natural variation in carotenoid and apocarotenoid pigments.In this study,using an integrated genetic approach we revealed that a 5;c/s-regulatory change at CCD4b encoding CAROTENOID CLEAVAGE DIOXYGENASE 4b is a major genetic determinant of natural variation in C3 0 apocarotenoids responsible for red coloration of citrus peel.Functional analyses demonstrated that in addition the known role in synthesizing 3-citraurin,CCD4b is also responsible for the production of another important C3 0 apocarotenoid pigment,p-citraurinene.Furthermore,analyses of the CCD4b promoter and transcripts from various citrus germplasm accessions established a tight correlation between the presence of a putative 5'c/s-regulatory enhancer within an MITE transposon and the enhanced allelic expression of CCD4b in C3 0 apocarotenoid-rich red-peeled accessions.Phylogenetic analysis provided further evidence that functional diversification of CCD4b and naturally occurring variation of the CCD4b promoter resulted in the stepwise evolution of red peels in mandarins and their hybrids.Taken together,our findings provide new insights into the genetic and evolutionary basis of apocarotenoid diversity in plants,and would facilitate breeding efforts that aim to improve the nutritional and aesthetic value of citrus and perhaps other fruit crops.Xiongjie Zheng Kaijie Zhu Quan Sun Weiyi Zhang Xia Wang Hongbo Cao Meilian Tan Zongzhou Xie Yunliu Zeng Junli Ye Lijun Chai Qiang Xu Zhiyong Pan Shunyuan Xiao Paul D.Fraser Xiuxin Deng 2019Molecular Plant2019,12,9:18
2Yangtze Three Gorges Reservoir, China: A holistic assessment of organic pollution, mutagenic effects of sediments and genotoxic impacts on fish显示文摘Besides obvious benefits,the Three Gorges Dam's construction resulted in new pollution scenarios with the potentials to threaten the Three Gorges Reservoir(TGR) ecosystem.In order to record organic contamination,to find links to ecotoxicological impacts and to serve as reference for ensuing monitoring,several sites in the TGR area were screened applying the triad approach with additional lines-of-evidence as a holistic assessment method.Sediments and the benthic fish species Pelteobagrus vachellii were sampled in 2011 and 2012 to determine organic pollution levels,mutagenic potentials and genotoxic impacts.Two regional hot-spots near the cities of Chongqing and Kaixian were identified and further investigated in 2013.Only polycyclic aromatic hydrocarbons(PAHs) could be detected in sediments in 2011(165–1653 ng/g),emphasizing their roles as key pollutants of the area.Their ubiquity was confirmed at Chongqing(150–433 ng/g) and Kaixian(127–590 ng/g) in2013.Concentrations were comparable to other major Chinese and German rivers.However,the immense sediment influx suggested a deposition of 216–636 kg PAH/day(0.2–0.6 mg PAH/(m2·day)),indicating an ecotoxicological risk.PAH source analysis highlighted primary impacts of combustion sources on the more industrialized upper TGR section,whereas petrogenic sources dominated the mid-low section.Furthermore,sediment extracts from several sites exhibited significant activities of frameshift promutagens in the Ames fluctuation assay.Additionally,significant genotoxic impairments in erythrocytes of P.vachellii were detected(Chongqing/Kaixian),demonstrating the relevance of genotoxicity as animportant mode of action in the TGR's fish.PAHs,their derivatives and non-target compounds are considered as main causative agents.Tilman Floehr Bj?rn Scholz-Starke Hongxia Xiao Josef Koch Lingling Wu Junli Hou Anja Wolf Axel Bergmann Kerstin Bluhm Xingzhong Yuan Martina Ro?-Nickoll Andreas Sch?ffer Henner Hollert 2015Journal of Environmental Sciences2015,27,12:9
3Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients.Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu 2021Hepatobiliary Surgery and Nutrition2021,10,1:4

6Beyond direct neighbourhood effects:higher-order interactions improve modelling and predicting tree survival and growth显示文摘It is known that biotic interactions are the key to species coexistence and maintenance of species diversity.Traditional studies focus overwhelmingly on pairwise interactions between organisms,ignoring complex higher-order interactions(HOIs).In this study,we present a novel method of calculating individual-level HOIs for trees,and use this method to test the importance of size-and distance-dependent individual-level HOIs to tree performance in a 25-ha temperate forest dynamic plot.We found that full HOI-inclusive models improved our ability to model and predict the survival and growth of trees,providing empirical evidence that HOIs strongly influence tree performance in this temperate forest.Specifically,assessed HOIs mitigate the competitive direct effects of neighbours on survival and growth of focal trees.Our study lays a foundation for future investigations of the prevalence and relative importance of HOIs in global forests and their impact on species diversity.Yuanzhi Li Margaret M.Mayfield Bin Wang Junli Xiao Kamil Krai David Janik Jan Holik Chengjin Chu 2021National Science Review2021,8,5:2

10Synergistic-engineered van der Waals photodiodes with high efficiency显示文摘Van der Waals(vdW)heterostructures based on two-dimensional transitionmetal dichalcogenides have provided unprecedented opportunities for photovoltaic detectors owing to their strong light-matter interaction and ultrafast interfacial charge transfer.Despite continued advancement,insufficient control of photocarrier behaviors still limits the external quantum efficiency(EQE)and operation speed of such detectors.Here,we propose a synergistic strategy of contact-configuration design and thickness-modulation to construct high-performance vdW photodiodes based on the typical type II heterostructure(MoS2/WSe2).Through integrating three contact architectures into one device to exclude other factors,we solid the superiority of designed 1L-MoS2/WSe2/graphene heterostructures incorporating efficient photocarrier collection and gate modulation.Together with leveraging the layer-numberdependent properties of WSe2,we observe the critical thickness of WSe2(11 layers)for the highest EQE,which verifies the thickness-dependent competition between photocarrier generation,dissociation,and collection.Finally,we demonstrate the synergistic-engineered vdW heterostructure can trigger record-high EQE(61%)and manifest ultrafast photoresponse(4.1μs)at the atomically thin limit(8 nm).The proposed strategy enables architecture-design and thickness-engineering to unlock the potential to realize high-performance optoelectronic devices.Baishan Liu Xiankun Zhang Junli Du Jiankun Xiao Huihui Yu Mengyu Hong Li Gao Yang Ou Zhuo Kang Qingliang Liao Zheng Zhang Yue Zhang 2022InfoMat2022,4,3:1

12The ubiquitin ligase Pelil inhibits ICOS and thereby Tfh-mediated immunity显示文摘T follicular helper(Tfh)cells are crucial for regulating autoimmune inflammation and protective immunity against viral infection.However,the molecular mechanism controlling Tfh cell differentiation is poorly understood.Here,through two mixed bone marrow chimeric experiments,we identified Peli1,a T cell-enriched E3 ubiquitin ligase,as an intrinsic regulator that inhibits Tfh cell differentiation.Peli1 deficiency significantly promoted c-Rel-mediated inducible T-cell costimulator(ICOS)expression,and PELI1 mRNA expression was negatively associated with ICOS expression on human CD4^(+)T cells.Mechanistically,increased ICOS expression on Peli1-KO CD4^(+)T cells enhanced the activation of PI3K-AKT signaling and thus suppressed the expression of Klf2,a transcription factor that inhibits Tfh differentiation.Therefore,reconstitution of Klf2 abolished the differences in Tfh differentiation and germinal center reaction between WT and Peli1-KO cells.As a consequence,Peli1-deficient CD4^(+)T cells promoted lupus-like autoimmunity but protected against H1N1 influenza virus infection in mouse models.Collectively,our findings established Peli1 as a critical negative regulator of Tfh differentiation and indicated that targeting Peli1 may have beneficial therapeutic effects in Tfhrelated autoimmunity or infectious diseases.Xinfang Huang Shumeng Hao Junli Liu Yuanyuan Huang Manman Liu Chunyuan Xiao Yan Wang Siyu Pei Tao Yu Jing Xu Haikun Wang Dongfang Dai Xiao Su Yichuan Xiao 2021Cellular & Molecular Immunology2021,18,4:1

18Triethylamine-catalyzed Isomerization of Glucose to Fructose under Low Temperature Conditions in Aqueous Phase显示文摘Isomerization of glucose derived from lignocellulosic biomass is an important step in biorefinery.Fructose isomerized from glucose,is used as a highly attractive sweetener in the food and beverages industries.However,the prevalence of side reactions at high glucose concentrations is a serious issue,leading to a significant reduction in the fructose yield,especially in the aqueous phase.In this study,an efficient method for the conversion of highly concentrated glucose into fructose under low temperature conditions using triethylamine as the catalyst was developed.It was demonstrated that high fructose yield could be maintained at high glucose concentration.At 60℃,fructose yield of 38.7%and fructose selectivity of 80.6%were achieved in 1 mol/L(approximately 17 wt%)glucose.When glucose concentration was increased to 2 mol/L(approximately 31 wt%),the fructose yield and selectivity were maintained at 34.7%and 77.4%,respectively.13C nuclear magnetic resonance(NMR)spectrometer was used to examine the glucose isomerization reaction.Compared to the NaOH catalytic system,triethylamine acted as a buffer to provide a stable alkaline environment for the catalytic system,further maintaining a high level of catalytic efficiency for the isomerization of glucose to fructose.Xiao Zhang Banggui Cheng Qixuan Lin Xiaohui Wang Rui Li Junli Ren 2020Paper And Biomaterials2020,5,4:0
