| 1 | Alginate oligosaccharide-induced intestinal morphology, barrier function and epithelium apoptosis modifications have beneficial effects on the growth performance of weaned pigs显示文摘Background: Alginate oligosaccharide(AOS), produced from alginate by alginate lyase-mediated depolymerisation, is a potential substitute for antibiotics and possesses growth-enhancing effects. Nevertheless, the mechanisms by which AOS regulates porcine growth remain to be elucidated. Therefore, we investigated the AOS-mediated changes in the growth performance of weaned pigs by determining the intestinal morphology, barrier function,as well as epithelium apoptosis.Methods: Twenty-four weaned pigs were distributed into two groups(n = 12) and received either a basal diet(control group) or the same diet supplemented with 100 mg/kg AOS. On d 15, D-xylose(0.1 g/kg body weight)was orally administrated to eight randomly selected pigs per treatment, and their serum and intestinal mucosa samples were collected 1 h later.Results: Our results showed that inclusion of AOS in the diet for 2 wk increased(P < 0.05) the average daily body weight gain in weaned pigs. Notably, AOS supplementation ameliorated the intestinal morphology and barrier function, as suggested by the enhanced(P < 0.05) intestinal villus height, secretory immunoglobulin A content and goblet cell counts. Compared to the control group, AOS ingestion both decreased(P < 0.05) the total apoptotic percentage and increased(P < 0.05) the proportion of S phase in the intestinal epithelial cells. | 3 | Arabidopsis VQ10 interacts with WRKY8 to modulate basal defense against Botrytis cinerea显示文摘Recent studies in Arabidopsis have revealed that some VQ motif-containing proteins physically interact with WRKY transcription factors;however,their specific biological functions are still poorly understood.In this study,we confirmed the interaction between VQ10and WRKY8,and show that VQ10 and WRKY8 formed a complex in the plant cell nucleus.Yeast two-hybrid analysis showed that the middle region of WRKY8 and the VQ motif of VQ10 are critical for their interaction,and that this interaction promotes the DNA-binding activity of WRKY8.Further investigation revealed that the VQ10protein was exclusively localized in the nucleus,and VQ10was predominantly expressed in siliques.VQ10 expression was strongly responsive to the necrotrophic fungal pathogen,Botrytis cinerea and defense-related hormones.Phenotypic analysis showed that disruption of VQ10 increased mutant plants susceptibility to the fungal pathogen B.cinerea,whereas constitutive-expression of VQ10 enhanced resistance to B.cinerea.Consistent with these findings,expression of the defenserelated PLANT DEFENSIN1.2(PDF1.2)gene was decreased in vq10 mutant plants,after B.cinerea infection,but increased in VQ10-overexpressing transgenic plants.Taken together,our findings provide evidence that VQ10 physically interacts with WRKY8 and positively regulates plant basal resistance against the necrotrophic fungal pathogen B.cinerea. | Junqiu Chen Houping Wang Yang Li Jinjing Pan Yanru Hu Diqiu Yu | 2018 | Journal of Integrative Plant Biology2018,60,10: | 8 |

| 5 | Tannic acid extracted from gallnut prevents post-weaning diarrhea and improves intestinal health of weaned piglets显示文摘This experiment was conducted to evaluate the effects of different levels of tannic acid(TA)on growth performance,diarrhea rate,nutrient digestibility and intestinal health in weaned piglets.A total of 180 weaned piglets(Duroc×Landrace×Yorkshire,24 d of age,initial average BW=7.77±0.17 kg)were allotted to 5 groups(6 pigs/pen and 6 replicates/group)in a randomized complete block design according to their gender and body weight.Piglets were fed a basal diet,or the basal diet supplemented with 0.05%,0.1%,0.2%or 0.4%TA for 28 d.The supplementary levels of TA in the diets were obtained by adding tannalbin containing 51%TA and 40.17%protein.The results showed that,compared with the CON group,dietary TA did not affect ADFI,ADG or F:G,and linearly reduced(P<0.01)the diarrhea rate and diarrhea index of piglets.There were no significant effects on apparent total tract digestibility(ATTD)in the 0.05%,0.1%and 0.2%TA groups,while negative effects(P<0.05)on apparent digestibility of crude protein and gross energy were observed in the 0.4%TA group.In addition,the nutrient digestibility of dry matter,crude protein and gross energy linearly decreased(P<0.01)with the increase of TA dosage.Supplementation of TA increased(P<0.05)the villus height of the duodenum and jejunum,as well as increased(P<0.05)catalase(CAT)activity in serum.Dietary TA improved(P<0.05)the Bacillus counts in cecal digesta.Further,TA significantly improved(P<0.05)Bacillus counts and reduced(P<0.05)the Escherichia coli counts in colonic digesta.The concentration of acetic acid,pro pionic acid,butyric acid and isovaleric acid in cecal digesta were significantly increased(P<0.05).The mRNA expression level of zonula occludens-1(Z0-1),zonula occludens-2(ZI-2),and claudin-2(CLDN-2)in the jejunum were greater(P<0.05)in TA supplemented groups.The study showed that,compared to the control,TA prevented post-weaning diarrhea and improved intestinal health of weaned piglets,and the appropriate level of TA supplementation would be from 0.1%to 0.2%. | Yanyan Song Yong Luo Bing Yu Jun He Ping Zheng Xiangbing Mao Zhiqing Huang Junqiu Luo Yuheng Luo Hui Yan Quyuan Wang Huifen Wang Daiwen Chen Jie Yu | 2021 | Animal Nutrition2021,,4: | 5 |
| 6 | Differential expression, molecular cloning,and characterization of porcine beta defensin 114显示文摘Background:β-defensins have attracted considerable research interest because of their roles in protecting hosts from various pathogens.This study was conducted to investigate the expression profiles of the porcineβ-defensin114(PBD114)in different breeds and in response to infections.Moreover,the function of PBD114 protein was partially investigated.Methods:Six Tibetan pigs(TP)and six DLY(Duroc×Landrace×Yorkshire)pigs were slaughtered to explore the expression profiles of PBD114 in different breeds and tissues.For infection models,sixteen DLY pigs were divided into two groups and challenged either with sterile saline or E.coli K88.The recombinant protein PBD114(r PBD114)was obtained by using a heterologous expression system in E.coli.Results:PBD114 gene was highly expressed in tissues such as the intestine,liver,spleen,and thymus.Interestingly,the expression level of PBD114 gene was higher in the TP pigs than in the DLY pigs(P<0.05),and was significantly elevated upon E.coli K88 challenge(P<0.05).The nucleotide sequences of PBD114 from Tibetan and DLY pigs was identical,and both showed a 210-bp open reading frame encoding a 69-amino acid mature peptide.To explaore the function of PBD114 protein,PBD114 gene was successfully expressed in E.coli Origami B(DE3)and the molecular weight of the r PBD114 was estimated by SDS-PAGE to be 25 k Da.The r PBD114 was purified and mass spectrometry verified the protein as PBD114.Importantly,r PBD114 showed antimicrobial activities against E.coli DH5αand E.coli K88,and the minimal inhibitory concentrations(MICs)were 64 and 128μg/m L,respectively.Hemolytic and cytotoxicity assays showed that r PBD114 did not affect cell viability under physiological concentrations.Conclusions:PBD114 is an infection response gene that is differentially-expressed between different porcine breeds and tissues.The antimicrobial activity of PBD114 protein,against pathogens such as the E.coli K88,suggested that it may serve as a candidate for the substitution of conventionally used antibiotics. | Guoqi Su Kunhong Xie Daiwen Chen Bing Yu Zhiqing Huang Yuheng Luo Xiangbing Mao Ping Zheng Jie Yu Junqiu Luo Jun He | 2019 | Journal of Animal Science and Biotechnology2019,10,4: | 4 |
| 7 | Supplementing daidzein in diets improves the reproductive performance, endocrine hormones and antioxidant capacity of multiparous sows显示文摘Certain hormones play important roles in modulating mammalian reproductive behaviour.Daidzein is a well-known isoflavonic phytoestrogen that possesses oestrogenic activity.This study was conducted to probe the effects of daidzein supplementation in gestation diets on the reproductive performance in sows.A total of 120 multiparous sows(Landrace x Yorkshire)were randomly assigned to 2 groups(n=60)and fed either a base diet(control)or one containing 200 mg/kg daidzein during gestation.We discovered that daidzein supplementation significantly increased the total number of piglets born per litter and number of piglets born alive per litter(P<0.05),decreased the farrowing time(P<0.05)and increased the serum oestrogen and progesterone concentrations(P<0.05)at 35 d of gestation.Moreover,serum immunoglobulin G(IgG)concentration and superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities were higher in the daidzein-treated group than in the control group at35 d of gestation(P<0.05).Daidzein increased the serum SOD activity and total anti-oxidative capacity(T-AOC)at 85 d of gestation(P<0.05).Interestingly,daidzein elevated the expression levels of the sodium-coupled neutral amino acid transporter 1(SLC38 A1)and insulin-like growth factor 1(IGF-1)genes in the placenta(P<0.05).These results suggest that daidzein ingestion could improve sow reproductive performance by changing serum hormones,elevating anti-oxidative capacity and upregulating critical functional genes in the placenta. | Yan Li Guoru He Daiwen Chen Bing Yu Jie Yu Ping Zheng Zhiqing Huang Yuheng Luo Junqiu Luo Xiangbing Mao Hui Yan Jun He | 2021 | Animal Nutrition2021,,4: | 4 |
| 8 | Tannic acid prevents post-weaning diarrhea by improving intestinal barrier integrity and function in weaned piglets显示文摘Background:Tannic acid(TA)is potential to reduce diarrhea in weaning pigs,but knowledge about the influence of TA on intestinal barrier integrity and function is still scarce.This experiment was conducted to investigate the effects of dietary TA supplementation on growth performance,diarrhea rate,intestinal barrier integrity and function of weaned pigs.Methods:A total of 108 crossbred(Duroc×Landrace×Yorkshire)piglets,with an initial average body weight of 6.60±0.27 kg,were allotted to 3 groups(6 pigs/pen and 6 replicates/group)in a randomized complete block design according to their gender and body weight.Piglets were fed the basal diet with 0(control,CON),0.2%and 1.0%TA,respectively.The trial lasted for 28 d.Results:Compared with the CON group,dietary 0.2%and 1.0%TA supplementation didn’t affect ADFI,ADG and F:G(P>0.05),but reduced diarrhea rate,diarrhea index and diarrhea score of piglets(P<0.05),reduced diamine oxidase(DAO)activity and D-lactic acid concentration in serum(P<0.01).The higher occludin expression and localization were observed in the duodenum,jejunum and ileum after supplementation with 0.2%or 1.0%TA(P<0.05).Adding 0.2%TA to diet significantly decreased crypt depth,increased villus height/crypt depth ratio in the duodenum(P<0.05),and dietary 1.0%TA tended to decrease crypt depth(P<0.10)and significantly decreased villus height(P<0.05)of the ileum.Moreover,lower malondialdehyde content in the ileum was detected in the pigs fed 1.0%TA(P<0.05).In the duodenum,both 0.2%and 1.0%TA groups had higher occludin(OCLN)mRNA and 0.2%TA group had higher zonula occludens-2(ZO-2)level(P<0.05).Meanwhile,dietary 1.0%TA supplementation tended to up-regulate OCLN mRNA levels in the jejunum(P<0.10)and 0.2%TA supplementation tended to up-regulate zonula occludens-1(ZO-1)mRNA levels in the ileum(P<0.10).Conclusion:In conclusion,dietary supplementation of 0.2%or 1.0%TA could effectively alleviate postweaning diarrhea without altering growth performance in weaned piglets,which might be achieved by improving intestinal barrier integrity and function. | Jie Yu Yanyan Song Bing Yu Jun He Ping Zheng Xiangbing Mao Zhiqing Huang Yuheng Luo Junqiu Luo Hui Yan Quyuan Wang Huifen Wang Daiwen Chen | 2021 | Journal of Animal Science and Biotechnology2021,12,2: | 3 |
| 9 | Atmospheric pollution of agriculture-oriented cities in Northeast China: A case in Suihua显示文摘Agriculture-oriented cities in Northeastern China have experienced frequent atmospheric pollution events.Deeper understandings of the pollution characteristics,haze causes and effects of management on local air quality are crucial for conducting integrated management approaches for the sustainable development of agriculture-oriented cities.Taking a typical agriculture-dominant city(i.e.,Suihua)in Northeast China,we analyzed in detail the characteristics and causes of atmospheric pollution and evaluated the straw-burning prohibition using multisource data.The results showed a clear temporal pattern of air quality index(AQI)on an annual scale(i.e.,2015-April 2019),with two typical pollution periods occurring in late autumn and early spring.The large areas of concentrated straw burning at local and regional scales accounted for the first period(i.e.,October and November),while dust emissions and farming disturbances comprised the second period.The interannual variation in pollution periods among these years was large,showing similar trends from 2015 to 2017 and the postponed late-autumn pollution period in 2018.Our evaluation has shown that the prohibition effect of straw burning significantly improved air quality in 2018,with a reduction of 59%±88%in the PM2.5 concentrations in October and November compared to 2015–2017.However,From October to April of the following year,the improvement effect was not significant due to postponement of straw burning to February or March.Our analysis also highlighted the roles of meteorological conditions,Therefore,combined with the promotion of straw utilization,scientifically prescribed burning considering the burning amount and location,meteorological conditions and regional transportation should be implemented. | Lili Li Kun Wang Weiwei Chen Qingliang Zhao Lijuan Liu Wei Liu Yang Liu Junqiu Jiang Jiumeng Liu Mengduo Zhang | 2020 | Journal of Environmental Sciences2020,32,11: | 3 |
| 10 | Leucine modulates the IPEC-J2 cell proteome associated with cell proliferation, metabolism and phagocytosis显示文摘Leucine can affect intestinal protein expressions, and improve mucosal immune function. However, little study has been conducted to determine the change of protein component by leucine treatment in intestine epithelial cells. The present study was to cover the key proteins and cell pathways that could be regulated by leucine treatment in porcine intestinal epithelial cell line(IPEC-J2) cells with the approach of proteome analysis. A total number of 3,211 proteins were identified in our approach by searching the database of Uniprot sus scrofa. Among identified proteins, there were 101 proteins expressed differently between control group and leucine group. Compared with the control group, there were 50 up-regulated proteins and 51 down-regulated proteins in leucine group. In these proteins, leucine treatment decreased the expression of some proteins including pyruvate kinase, glyceraldehyde-3-phosphate dehydrogenase,E3 ubiquitin ligase, cathepsin D, caspase 3 and caspase 6, and increased the levels of some proteins, such as some eukaryotic translation initiation factors, ubiquitin carboxyl-terminal hydrolase, DNA-related RNA polymerase II, urokinase plasminogen activator, cyclin-dependent kinase inhibitor 2 b, Mut L homolog 1,5-methylcytosine binding domain 4, polymerase d, a-tubulin, syntaxin 18, Ras homolog D, actin related protein 2/3 complex and cofilin. Via the analysis of Gene Ontology and pathways, these proteins in IPECJ2 cells were related with some physiological functions, such as protein metabolism, glycolysis, cell proliferation, apoptosis and phagocytosis. | 12 | A high-quality sponge gourd(Luffa cylindrica)genome显示文摘Sponge gourd(Luffa cylindrica)is an important cultivated vegetable and medicinal plant in the family Cucurbitaceae.In this study,a draft genome sequence of the sponge gourd inbred line P93075 was analyzed.Using Illumina,PacBio,and 10×Genomics sequencing techniques as well as new assembly techniques such as FALCON and chromatin interaction mapping(Hi-C),a chromosome-scale genome of approximately 656.19 Mb,with an N50 scaffold length of 48.76 Mb,was generated.From this assembly,25,508 protein-coding gene loci were identified,and 63.81%of the whole-genome consisted of transposable elements,which are major contributors to the expansion of the sponge gourd genome.According to a phylogenetic analysis of conserved genes,the sponge gourd lineage diverged from the bitter gourd lineage approximately 41.6 million years ago.Additionally,many genes that respond to biotic and abiotic stresses were found to be lineage specific or expanded in the sponge gourd genome,as demonstrated by the presence of 462 NBS-LRR genes,a much greater number than are found in the genomes of other cucurbit species;these results are consistent with the high stress resistance of sponge gourd.Collectively,our study provides insights into genome evolution and serves as a valuable reference for the genetic improvement of sponge gourd. | Haibin Wu Gangjun Zhao Hao Gong Junxing Li Caixia Luo Xiaoli He Shaobo Luo Xiaoming Zheng Xiaoxi Liu Jinju Guo Junqiu Chen Jianning Luo | 2020 | Horticulture Research2020,7,1: | 2 |

| 17 | Micro/nano-scale Characterization and Fatigue Fracture Resistance of Mechanoreceptor with Crack-shaped Slit Arrays in Scorpion显示文摘The nocturnal scorpion Heterometrus petersii uses Basitarsal Compound Slit Sensilla (BCSS) as mechanoreceptor to detect mechanical signal (e.g. substrate vibration, cyclic loads caused by walking) without fatigue failure such as initiation of fatigue crack and further propagation of crack-shaped slit. The outstanding perceptive function has been discovered for over half a century. However, it is not yet clear about the microstructure, material composition and micromechanical property which are all important factors that determine the fatigue fracture resistance of the BCSS. Here, the microscopic characteristics of the BCSS were thoroughly studied. The results dicate that anti-fatigue resilin and stiff chitinous cuticle form multilayered composite as the main body of the BCSS. Meanwhile, the pre-existing slit as mechanosensory structure is covered by cuticular membrane which has different mechanical property with the epicuticle. | 18 | Soliton formation and spectral translation into visible on CMOS-compatible 4H-silicon-carbide-oninsulator platform显示文摘Recent advancements in integrated soliton microcombs open the route to a wide range of chip-based communication,sensing,and metrology applications.The technology translation from laboratory demonstrations to real-world applications requires the fabrication process of photonics chips to be fully CMOS-compatible,such that the manufacturing can take advantage of the ongoing evolution of semiconductor technology at reduced cost and with high volume.Silicon nitride has become the leading CMOS platform for integrated soliton devices,however,it is an insulator and lacks intrinsic second-order nonlinearity for electro-optic modulation.Other materials have emerged such as AlN,LiNbO_(3),AlGaAs and GaP that exhibit simultaneous second-and third-order nonlinearities.Here,we show that silicon carbide(SiC)--already commercially deployed in nearly ubiquitous electrical power devices such as RF electronics,MOSFET,and MEMS due to its wide bandgap properties,excellent mechanical properties,piezoelectricity and chemical inertia--is a new competitive CMOS-compatible platform for nonlinear photonics.High-quality-factor microresonators(Q=4×10^(6))are fabricated on 4H-SiC-on-insulator thin films,where a single soliton microcomb is generated.In addition,we observe wide spectral translation of chaotic microcombs from near-infrared to visible due to the second-order nonlinearity of SiC.Our work highlights the prospects of SiC for future low-loss integrated nonlinear and quantum photonics that could harness electro-opto-mechanical interactions on a monolithic platform. | Chengli Wang Jin Li Ailun Yi Zhiwei Fang Liping Zhou Zhe Wang Rui Niu Yang Chen Jiaxiang Zhang Ya Cheng Junqiu Liu Chun-Hua Dong Xin Ou | 2022 | Light(Science & Applications)2022,11,12: | 1 |
| 19 | Bioresponsive cisplatin crosslinked albumin hydrogel served for efficient cancer combination therapy显示文摘Combination therapy is one of the potential strategies for tackling complicated tumor treatments like drug resistance.In this work,we have generated a therapeutic cisplatin-crosslinked albumin hydrogel(BC-Gel)that allows the local release of L-Buthionine-sulfoximine(BSO),cisplatin,and glucose oxidase(GOx)with distinct release kinetics.The BC-Gel with favorable biostimuli degradability and injectability could release therapeutic agents in a programmed manner within the tumor microenvironment(TME).The preferentially released BSO significantly suppressed the glutathione(GSH)-related cisplatin resistance and sensitized the tumor cells to cisplatin by inhibiting theγ-glutamylcysteine synthetase.Meanwhile,cisplatin achieved a sequential release and long-term treatment following the bioresponsive gel degradation under the combined action of chloride ions(Cl−)and proteinase in the body.In addition,the overproduced H_(2)O_(2)of GOx-catalyzed glucose oxidation accelerated the depletion of existed GSH within cells and further weakened the cisplatin resistance,achieving enhanced tumor treatment together with a strong cell-killing effect.The above sequential drug release strategy based on the dual GSH depletion effect breaks the balance of the GSH-mediated redox TME and enhances the sensitivity of A549 cells to cisplatin forcefully,and provides a promising way for temporal control of drug release as well as efficient cancer combination therapy. | An Yan Zherui Zhang Jiamei Gu Xiaoran Ding Yongchen Chen Jingjing Du Shu Wei Hongcheng Sun Jiayun Xu Shuangjiang Yu Junqiu Liu | 2023 | Nano Research2023,16,2: | 1 |
| 20 | Foundry manufacturing of tight-confinement,dispersion-engineered,ultralow-loss silicon nitride photonic integrated circuits显示文摘The foundry development of integrated photonics has revolutionized today’s optical interconnect and datacenters.Over the last decade,we have witnessed the rising of silicon nitride(Si_(3)N_(4)) integrated photonics,which is currently transferring from laboratory research to foundry manufacturing.The development and transition are triggered by the ultimate need for low optical loss offered by Si_(3)N_(4),which is beyond the reach of silicon and III-V semiconductors.Combined with modest Kerr nonlinearity,tight optical confinement,and dispersion engineering,Si_(3)N_(4) has today become the leading platform for linear and Kerr nonlinear photonics,and it has enabled chip-scale lasers featuring ultralow noise on par with table-top fiber lasers.However,so far all the reported fabrication processes of tight-confinement,dispersion-engineered Si_(3)N_(4) photonic integrated circuits(PICs)with optical loss down to few dB/m have only been developed on 4-inch(100 mm diameter)or smaller wafers.Yet,to transfer these processes to established CMOS foundries that typically operate 6-inch or even larger wafers,challenges remain.In this work,we demonstrate the first foundry-standard fabrication process of Si_(3)N_(4) PICs with only 2.6 dB/m loss,thickness above 800 nm,and near 100%fabrication yield on 6-inch(150 mm diameter)wafers.Such thick and ultralow-loss Si_(3)N_(4) PIC enables low-threshold generation of soliton frequency combs.Merging with advanced heterogeneous integration,active ultralow-loss Si_(3)N_(4) integrated photonics could pave an avenue to addressing future demands in our increasingly information-driven society. | ZHICHAO YE HAIYAN JIA ZHANGJUN HUANG CHEN SHEN JINBAO LONG BAOQI SHI YI-HAN LUO LAN GAO WEI SUN HAIRUN GUO JIJUN HE JUNQIU LIU | 2023 | Photonics Research2023,11,4: | 1 |
