1Mitochondrial tRNA^(Glu) A14693G variant may modulate the phenotypic manifestation of deafness-associated 12S rRNA A1555G mutation in a Han Chinese family显示文摘Mutations in mitochondrial 12S rRNA gene are one of the most important causes of aminoglycoside-induced and nonsyndromic hear- ing loss. Here we report the characterization of one Han Chinese pedigree with aminoglycoside-induced and nonsyndromic hearing loss. This Chinese family carrying the 12S rRNA A1555G mutation exhibited high penetrance and expressivity of hearing impairment. In par- ticular, penetrances of hearing loss in this family pedigree were 43.8% and 25%, respectively, when aminoglycoside-induced hearing loss was included or excluded. Mutational analysis of entire mitochondrial genomes in this family showed the homoplasmic A1555G mutation and a set of variants belonging to haplogroup Y2. Of these, the A14693G variant occurred at the extremely conserved nucleotide (conven- tional position 54) of the TΨC-loop of tRNAGlu and was absent in 156 Chinese controls. Nucleotides at position 54 of tRNAs are often modified, thereby contributing to the structural formation and stabilization of functional tRNAs. Thus, the structural alteration of tRNA by the A14693G variant may lead to a failure in tRNA metabolism and impair mitochondrial protein synthesis, thereby worsening mito- chondrial dysfunctions altered by the A1555G mutation. 2Formation Mechanism of Ground Fissures Originated from the Hanging Wall of Normal Fault:A Case in Fen-Wei Basin,China显示文摘This paper takes Fen-Wei Basin(FWB)as a case to study the ground fissures controlled by normal fault.Based on the field investigation,geophysical exploration,drilling,GNSS data and numerical calculation,the characteristics and mechanism of ground fissures originated from the hanging wall of normal faults are revealed.The results show that the distribution of ground fissures in the hanging wall and heading wall of the active faults is not uniform.Ground fissures are mostly distributed in the hanging wall of active faults and show a linear distribution on the surface,their strike is consistent with the fault,mainly characterized by vertical offset and horizontal tension.Ground fissures destroy the farmland and building foundation through which they pass and cause the rupture or displacement.In profile section,the ground fissure shows the characteristics of normal faults and dislocates the strata,and is connected with the underlying faults.Numerical analysis shows that the vertical displacement of normal fault activity in hanging wall is much larger than that in heading wall,which is the reason that tectonic ground fissures mainly originate from hanging wall.The range of dangerous area of ground fissures is controlled by the depth of fault,the strength of the ground fissures disaster is mainly controlled by the activity of fault.The formation of the ground fissures originated from the hanging wall of the fault experienced three stages:the main fault activity stage,the secondary fault activity stage and the fissure formation stage.Zhijie Jia Jianbing Peng Quanzhong Lu Jianwei Qiao Feiyong Wang Mingdong Zang Yang Liu Junyan Zhao Fengji Zhu 2022Journal of Earth Science2022,33,2:2
3Effects of fish meal replacement with Chlorella meal on growth performance,pigmentation,and liver health of largemouth bass(Micropterus salmoides)显示文摘Chlorella meal is a potential protein source for aquafeeds.However,the physiological response of carnivorous fish fed Chlorella meal remains elusive.This study evaluated the effects of replacing dietary fish meal with Chlorella meal on growth performance,pigmentation,and liver health in largemouth bass.Five diets were formulated to replace dietary fish meal of 0%(C0,control),25%(C25),50%(C50),75%(C75),and 100%(C100)with Chlorella meal,respectively.Total 300 fish(17.6±0.03 g)were randomly assigned to 15 tanks(3 tanks/group).Fish were fed the experimental diet twice daily for 8 weeks.The increased dietary Chlorella meal quadratically influenced the final body weight(FBW),weight gain rate(WGR),specific growth rate(SGR),and feed intake(FI),which were significantly lower in the C100 group than in the other groups(P<0.05).The feed conversion ratio(FCR)increased linearly or quadratically with dietary Chlorella meal.Dietary Chlorella meal linearly or quadratically increased the lutein content of plasma,liver,and dorsal muscle of largemouth bass(P<0.05).Compared to the C0 group,all supplemented Chlorella meal groups significantly improved the yellowness(b*)of the dorsal body(1.5 to 2.0 fold),abdominal body(1.5 to 1.8 fold),and dorsal muscle(3.8 to 5.4 fold)of largemouth bass(P<0.05).In addition,compared to the C0 group,the liver vacuolation area of fish was significantly increased in the C75 and C100 groups(P<0.05).Transcriptional levels of apoptosis-related genes of b-cell lymphoma-2(bcl2),caspase-9-like(casp9),and caspase-3a(casp3)were markedly upregulated(0.9 to 1.6 fold)in the C100 group compared to the C0 group(P<0.05).Based on the quadratic regression analysis between FBW,WGR,or SGR and dietary Chlorella meal level,largemouth bass had the best growth when replacing 31.7%to 32.6%of fish meal with 15.03%to 15.43%dietary Chlorella meal.The present results indicated that dietary supplementation with Chlorella meal(11.85%to 47.45%)significantly enhanced the pigmentation;however,total replacement of fish meal(40%)with Chlorella meal(47.45%)caused growth retardation,apoptosis,and liver damage in largemouth bass.Longwei Xi Qisheng Lu Yulong Liu Jingzhi Su Wen Chen Yulong Gong Dong Han Yunxia Yang Zhimin Zhang Junyan Jin Haokun Liu Xiaoming Zhu Shouqi Xie 2022Animal Nutrition2022,,3:2
4Duplication and transcriptional divergence of three Kunitz protease inhibitor genes that modulate insect and pathogen defenses in tea plant(Camellia sinensis)显示文摘Kunitz protease inhibitors(KPIs)are ubiquitous in plants and act as crucial compounds in defense responses against insect attack and pathogen infection.However,the influence of gene duplication on the postdivergence of the CsKPI genes involved in biotic stresses in tea plant is not well known.Here,we identified three CsKPI genes from tea plant(Camellia sinensis)and characterized their expression and evolutionary patterns among plant species.We found that CsKPI1,CsKPI2,and CsKPI3 diverged from their common ancestor 72.94 million years ago(MYA),and the tandem duplication of CsKPI2 and CsKPI3 occurred 26.78 MYA.An in vitro protein assay showed that the three CsKPI proteins were functional and inhibited the production of p-nitroanilide(PNA)from an artificial substrate.The three CsKPI-GFP fusion proteins localized to the cytoplasm.We showed that salicylic acid(SA)and transcripts of CsKPI2 and CsKPI3 significantly accumulated after infection with Glomerella cingulata.The application of exogenous SA stimulated the high expression of both CsKPI2 and CsKPI3 by activating cis-elements within their promoters.Under Ectropis oblique attack,CsKPI1 expression and jasmonic acid(JA)levels were more abundant in both insect-damaged leaf tissues and undamaged neighboring leaves.The application of jasmonic acid methyl ester elicited high expression levels of CsKPI1,suggesting that CsKPI1 accumulation requires JA production in tea plant.The overall findings suggest that the transcriptional divergence of KPI genes after duplication led to the specialized role of CsKPI1 in the physiological response to insect stress;the functional conservation between CsKPI2 and CsKPI3 confers resistance to pathogen infection in tea plant.Junyan Zhu Yaxian He Xiaomei Yan Lu Liu Rui Guo Xiaobo Xia Daojie Cheng Xiaozeng Mi Lidiia Samarina Shenrui Liu Enhua Xia Chaoling Wei 2019Horticulture Research2019,6,1:2

8Critical Amino Acid Residues for Nicotine 5'-Hydroxylation in Human CYP2A Enzymes显示文摘Objective: We have continued previous work in which we demonstrated that #117 and #372 amino acids contrib- uted to the high activities of human CYP2A13 in catalyzing 4-methylnitrosamino-1-(3-pyridyl)-1-butanone(NNK) and aflatoxin B1(AFB1) carcinogenic activation. The present study was designed to identify other potential amino acid residues that contribute to the different catalytic characteristics of two CYP2A enzymes, CYP2A6 and CYP2A13, in nicotine metabolism and provide insights of the substrate and related amino acid residues interactions. Methods: A series of reciprocally substituted mutants of CYP2A6Ile300 → Phe, CYP2A6Gly301Ala, CYP2A6Ser369 → Gly, CYP2A13Phe300 → Ile, CYP2A13Ala301 → Gly and CYP2A13Gly369 → Ser were generated by site-directed mutagenesis/baculovirus-Sf9 insect cells expression. Comparative kinetic analysis of nicotine 5'hydroxylatin by wild type and mutant CYP2A proteins was performed. Results:All amino acid residue substitutions at 300, 301 and 369 caused significant kinetic property changes in nicotine metabolism. While CYP2A6Ile300 → Phe and CYP2A6Gly301→Ala mutations had notable catalytic efficiency increases compared to that for the wild type CYP2A6, CYP2A13Phe300→Ile and CYP2A13Ala301→Gly replacement introduced remarkable catalytic efficiency decreases. 12Tracing the formation of oxygen vacancies at the conductive LaAlO3/SrTiO3 interface via photoemission显示文摘The two-dimensional electron gas(2DEG)generated at the LaAlO3/SrTiO3 interface has been in the focus of oxides re-search since its first discovery.Although oxygen vacancies play an important role in the generation of the insulator-to-metal transition of the SrTiO3 bare surface,their contribution at the LaAlO3/SrTiO3 interface remains unclear.In this work,we investigated a LaAlO3/SrTiO3 heterostructure with regional distribution of defect-based localized polar sites at the interface.Using static and time-resolved threshold photoemission electron microscopy,we prove that oxygen vacan-cies are induced near those polar sites,resulting in the increase of carrier density of the 2DEG states.In addition,oxy-gen-related surface states were uncovered,which we attributed to the release of lattice oxygen during the formation of oxygen vacancies.Such effects are mainly found spatially located around the defect sites at the buried interface,while other regions remain unaffected.Our results confirm that the itinerant electrons induced by oxygen vacancies can coex-ist with the charge transfer mechanism in the LaAlO3/SrTiO3 heterostructure,together leading to the formation of the metallic interface.These observations provide fundamental insights into the nature of LaAlO3/SrTiO3 interface based 2DEG and unique perspectives for potential applications.Junyan Chen Tobias Eul Lu Lyu Yaolong Li Xiaoyong Hu Xingkun Ning Shufang Wang Martin Aeschlimann Qihuang Gong 2022Opto-Electronic Science2022,1,7:1

14A Comprehensive Method for the Risk Assessment of Ground Fissures:Case Study of the Eastern Weihe Basin显示文摘Ground fissures are influenced by the coupling of geological factors and human activi-ties.They threaten the safety of infrastructure and restrict town planning in many areas in China.One of the area most severely affected by ground fissures is the Weihe Basin in North China,which has the most extensive distribution of ground fissures and all the elements that control their occurrence,pro-viding an ideal study area for the study of ground fissures.In this study,we took the eastern Weihe Ba-sin as the study area and determined the distribution and hazard characteristics of ground fissures via field investigation.Based on the analytic hierarchy process,we propose a multi-level,comprehensive method for evaluating the hazards of ground fissures.This method considers the geological back-ground,development status,and triggering factors of ground fissures,including all nine currently known assessment indices of ground fissures.We used judgment matrices to rank the constructed ground fissure assessment index system at the single and total levels and quantified the nine assessment indices of ground fissures according to the field survey data.Finally,we plotted the ground fissure risk zoning map and evaluated the ground fissure risk at the study area.Our findings indicated that the proposed method could facilitate ground fissure hazard assessment and prediction and provide support for hazard prevention and urban/rural planning.Zhijie Jia Jianbing Peng Quanzhong Lu Penghui Ma Jianwei Qiao Feiyong Wang Mingdong Zang Yang Liu Junyan Zhao Fengji Zhu 2023Journal of Earth Science2023,34,6:0

18Effects of dietary oil sources and fat extraction methods on apparent and standardized ileal digestibility of fat and fatty acids in growing pigs显示文摘Background:There is a lack of data for the standardized ileal digestibility(SID) of fat and fatty acids in national feed databases.In addition,it is important to specify the procedures used for fat analyses.Therefore,an experiment was conducted to 1) determine the apparent ileal digestibility(AID) and SID of fat and fatty acids in ten different oil sources for growing pigs and to develop prediction equations for SID of fat based on fatty acid composition;and 2) compare the effect of the fat extraction methods on the calculated values for endogenous loss and digestibility of fat.Methods:Twenty-two barrows(initial body weight:32.1 ± 2.3 kg) were surgically fitted with a T-cannula in the distal ileum,and allotted to 1 of 11 experimental diets in a 4-period Youden Square design.A fat-free diet was formulated using cornstarch,soy protein isolate and sucrose.Ten oil-added diets were formulated by adding 6% of dietary oil sources to the fat-free diet at the expense of cornstarch.All diets contained 26% sugar beet pulp and 0.40% chromic oxide.Results:The endogenous loss of ether extract(EE) was lower than that of acid-hydrolyzed fat(AEE;P < 0.01).There were significant differences in the AID and SID of fat and saturated fatty acids across the dietary oil sources(P < 0.05).The SID of AEE for palm oil was lower than that of sunflower oil,corn oil,canola oil,rice oil and flaxseed oil(P < 0.01).The AID and SID of fat ranged from 79.65% to 86.97% and from 91.14% to 99.18%.Although the AID of EE was greater than that of AEE(P < 0.01),there was no significant difference in SID of EE and AEE except for palm oil.The ratio of unsaturated to saturated fatty acids(U/S) had a positive correlation with SID of fat(P < 0.05),whereas C16:0 and long chain saturated fatty acids(LSFA) were significant negatively correlated with SID of fat(P < 0.01).The best-fit equation to predict SID of fat was SID AEE = 102.75-0.15 × LSFA-0.74 × C18:0-0.03 × C18:1(Adjusted coefficient of determination = 0.88,P < 0.01).Conclusions:When calculating the SID of fat,the EE content of the samples can be analyzed using the direct extraction method,whereas the acid hydrolysis procedure should be used to determine the AID of fat.Fat digestibility of dietary oils was affected by their fatty acid composition,especially by the contents of C16:0,LSFA and U/S.Lu Wang Wenjun Gao Junyan Zhou Huangwei Shi Tenghao Wang Changhua Lai 2023Journal of Animal Science and Biotechnology2023,14,2:0
19Different dietary starch patterns in lowprotein diets:effect on nitrogen efficiency,nutrient metabolism,and intestinal flora in growing pigs显示文摘Background:Protein releases amino acids faster than starch releases glucose in digestive tract of pigs fed lowprotein(LP)diets.Poor synchronization of dietary glucose and amino acids supply leads to compromised nitrogen efficiency.Dietary starch patterns modulation may improve this situation.Methods:Growing barrows(29.7±2.0 kg)were randomly allotted into 5 dietary treatments with LP diets consisting of different purified starches.Treatments included:waxy corn starch(W LP),corn starch+waxy corn starch(C+W LP),corn starch(C LP),pea starch+waxy corn starch(P+W LP)and pea starch(P LP).In the experiment,growth performance,protein deposition,nutrient metabolism,and fecal microbial community of pigs were investigated.In vitro starch digestion was used for predicting the in vivo glucose response.Results:Dietary starch in vitro glucose release profile was determined by starch source and the ratio of amylopectin and amylose.C+W LP treatment showed decreased total nitrogen excretion and plasma citrulline concentration and improved plasma leptin concentration among treatments(P<0.05).Besides,the highest nitrogen apparent biological value,whole-body protein deposition and growth performance and lowest urinary nitrogen excretion were also observed in C+W LP treatment.Compared with the other groups,C+W LP and C LP showed increased plasma pyruvate,IGF-1,and lipase concentrations(P<0.05).The W LP group presented dramatically increased plasma alanine and urea nitrogen concentration and decreased aldolase and leptin concentrations(P<0.05).Dietary starch patterns did not make an impact on bacterial richness and diversity,but changed the taxonomic and functional structures of the microbial communities.Microbial protein fermentation product(isobutyrate and isovalerate)presented increased in P LP treatments compared with the other treatments(P<0.05).Conclusions:Dietary starch patterns modulation can regulate dietary glucose release profile,nutrient metabolism,protein turnover,and fecal microbial fermentation in pigs.The optimal dietary glucose release profile effectively strengthened whole-body protein deposition and improve nitrogen efficiency and growth performance in growing pigs fed LP diets.Junyan Zhou Lu Wang Lijie Yang Guangxin Yang Xiangfang Zeng Shiyan Qiao 2022Journal of Animal Science and Biotechnology2022,13,5:0
