| 4 | Differentiation of isolated human umbilical cord mesenchymal stem cells into neural stem cells显示文摘AIM:To investigate whether umbilical cord human mesenchymal stem cell(UC-MSC)was able to differentiate into neural stem cell and neuron in vitro.·METHODS:The umbilical cords were obtained from pregnant women with their written consent and the approval of the Clinic Ethnics Committee.UC-MSC were isolated by adherent culture in the medium contains 20%fetal bovine serum(FBS),then they were maintained in the medium contain 10%FBS and induced to neural cells in neural differentiation medium.We investigated whether UC-MSC was able to differentiate into neural stem cell and neuron in vitro by using flow cytometry,reverse transcriptase-polymerase chain reaction(RT-PCR)and immunofluorescence(IF)analyzes.·RESULTS:A substantial number of UC-MSC was harvested using the tissue explants adherent method at about 2wk.Flow cytometric study revealed that these cells expressed common markers of MSCs,such as CD105(SH2),CD73(SH3)and CD90.After induction of differentiation of neural stem cells,the cells began to form clusters;RT-PCR and IF showed that the neuron specific enolase(NSE)and neurogenic differentiation 1-positive cells reached 87.3%±14.7%and 72.6%±11.8%,respectively.Cells showed neuronal cell differentiation after induced,including neuron-like protrusions,plump cell body,obviously and stronger refraction.RT-PCR and IF analysis showed that microtubule-associated protein 2(MAP2)and nuclear factor-M-positive cells reached 43.1%±10.3%and 69.4%±19.5%,respectively.·CONCLUSION:Human umbilical cord derived MSCscan be cultured and proliferated in vitro and differentiate into neural stem cells,which may be a valuable source for cell therapy of neurodegenerative eye diseases. | Song Chen Wei Zhang Ji-Ming Wang Hong-Tao Duan Jia-Hui Kong Yue-Xin Wang Meng Dong Xue Bi Jian Song | 2016 | International Journal of Ophthalmology(English edition)2016,9,1: | 22 |

RF ablation has the following advantages in the treatment of hepatic hemangiomas: minimal invasiveness, definite efficacy, high safety, fast recovery, relatively simple operation, and wide applicability. | 10 | Study on the compatibility of cement-superplasticizer system based on the amount of free solution显示文摘The changes of free solution amount,fluidity and the time-depended fluidity loss of cement paste were examined by varying the water-cement ratio and the dosages of superplasticizer. The distribution of solution and flocculation microstructure in fresh cement paste was observed with optical microscope. The change of free solution amount and its effect on the fluidity and bleeding of cement paste was studied. The results show that the adsorbed solution amount has a great influence on the compatibility of cement-superplasticizer system,including the bleeding degree,the fluidity and the time-depended fluidity loss of cement paste. Superplasticizer increases the fluidity of cement paste by destroying the flocculated cement particle structure and increasing the amount of adsorbed solution. Polycarboxylate superplasticizer shows higher ability of adsorption than naphthalene superplasticizer. Over dosage of superplasticizer is not the primary cause of bleeding. | 11 | 3D Shapeable,Superior Electrically Conductive Cellulose Nanofibers/Ti_(3)C_(2)T_(x) MXene Aerogels/Epoxy Nanocomposites for Promising EMI Shielding显示文摘In this work,3D highly electrically conductive cellulose nanofibers(CNF)/Ti_(3)C_(2)T_(x) MXene aerogels(CTA)with aligned porous structures are fabricated by directional freezing followed by freeze-drying technique,and the thermally annealed CTA(TCTA)/epoxy nanocomposites are then fabricated by thermal annealing of CTA,subsequent vacuum-assisted impregnation and curing method.Results show that TCTA/epoxy nanocomposites possess 3D highly conductive networks with ultralow percolation threshold of 0.20 vol%Ti_(3)C_(2)T_(x).When the volume fraction of Ti_(3)C_(2)T_(x) is 1.38 vol%,the electrical conductivity(σ),electromagnetic interference shielding effectiveness(EMI SE),and SE divided by thickness(SE/d)values of the TCTA/epoxy nanocomposites reach 1672 Sm-1,74 dB,and 37 dBmm-1,respectively,which are almost the highest values compared to those of polymer nanocomposites reported previously at the same filler content.In addition,compared to those of the samples without Ti_(3)C_(2)T_(x),the storage modulus and heat-resistance index of TCTA/epoxy nanocomposites are enhanced to 9792.5 MPa and 310.7℃,increased by 62%and 6.9℃,respectively,presenting outstanding mechanical properties and thermal stabilities.The fabricated lightweight,easy-to-process,and shapeable TCTA/epoxy nanocomposites with superior EMI SE values,excellent mechanical properties,and thermal stabilities greatly broaden the applications of MXene-based polymer composites in the field of EMI shielding. | Lei Wang Ping Song Cheng-Te Lin Jie Kong Junwei Gu | 2020 | Research2020,,1: | 13 |

| 13 | Parameter identification and global sensitivity analysis of Xin'anjiang model using meta-modeling approach显示文摘Parameter identification, model calibration, and uncertainty quantification are important steps in the model-building process, and are necessary for obtaining credible results and valuable information. Sensitivity analysis of hydrological model is a key step in model uncertainty quantification, which can identify the dominant parameters, reduce the model calibration uncertainty, and enhance the model optimization efficiency. There are, however, some shortcomings in classical approaches, including the long duration of time and high computation cost required to quantitatively assess the sensitivity of a multiple-parameter hydrological model. For this reason, a two-step statistical evaluation framework using global techniques is presented. It is based on (1) a screening method (Morris) for qualitative ranking of parameters, and (2) a variance-based method integrated with a meta-model for quantitative sensitivity analysis, i.e., the Sobol method integrated with the response surface model (RSMSobol). First, the Morris screening method was used to qualitatively identify the parameters' sensitivity, and then ten parameters were selected to quantify the sensitivity indices. Subsequently, the RSMSobol method was used to quantify the sensitivity, i.e., the first-order and total sensitivity indices based on the response surface model (RSM) were calculated. The RSMSobol method can not only quantify the sensitivity, but also reduce the computational cost, with good accuracy compared to the classical approaches. | 15 | Promoter hypermethylation of death-associated protein kinase gene in cholangiocarcinoma显示文摘BACKGROUND:Death-associated protein kinase(DAPK) is a Ca2+/calmodulin-regulated Ser/Thr kinase which is involved in apoptosis. The aberrant methylation of its promoter region CpG islands may be one of the important mechanisms of carcinogenesis. We studied the relationship of methylation status and expression of the DAPK gene with the clinical findings in cholangiocarcinoma. METHODS:Target DNA was modified by sodium bisulfite,coverting all unmethylated,but not methylated,cytosines to uracil,and subsequently detected by methylation-specific PCR. Moreover,mRNA expression of the DAPK gene was assessed by RT-PCR. RESULTS:Aberrant methylation of the DAPK gene was detected in 11(30.6%) of 36 tissue specimens of cholangiocarcinoma,and in 2(5.6%) of 36 specimens of adjacent normal tissues. DAPK mRNA was not expressed in tumor and adjacent tissues with hypermethylation of the DAPK promoter. There were no statistical differences in the extent of differentiation and invasion,lymph node metastasis or pathologic type between the methylated and unmethylated tissues. CONCLUSIONS:The frequency of DAPK gene methylation in cholangiocarcinoma is high and it may offer an effective means for earlier auxiliary diagnosis of the malignancy. The DAPK gene is probably suppressed by methylation,and it could become resistant to apoptosis and immunologicalsurveillance. | 16 | Spatial-Temporal Changes in Grain Production, Consumption and Driving Mechanism in China显示文摘The spatial-temporal patterns of grain production and consumption have an important influence on the effective national grain supply on condition of tight balance in the total grain amount in China. In this paper, we analyze the spatial-temporal patterns of grain production, consumption and the driving mechanism for their evolution processes in China. The results indicate that both gravity centers of grain production and consumption in China moved toward the northern and eastern regions, almost in the same direction. The coordination of grain production and consumption increased slightly from 1995 to 2007 but decreased from 2000 to 2007. There is a spatial difference between the major districts of output increase and the strong growth potential in grain consumption, which indicates an increasing difficulty in improving the regional coordination of grain production and consumption. The movement of the gravity center of grain production is significantly correlated with regional differences in grain production policy, different economic development models, and spatial disparity of land and water resource use. | 17 | A comparative study on the transplantation of different concentrations of human umbilical mesenchymal cells into diabetic rats显示文摘AIM: To observe the effects of intravitreal injections of different concentrations of human umbilical mesenchymal stem cells on retinopathy in rats with diabetes mellitus.METHODS: Healthy and adult male Sprague-Dawley(SD) rats were randomly assigned to a normal control group(group A), a diabetic retinopathy(DR) blank control group(group B), a high-concentration transplantation group(group C), a low-concentration transplantation group(group D) and a placebo transplantation group(group E). The expression of nerve growth factor(NGF)protein in the retinal layers was detected by immunohistochemical staining at 2, 4, 6 and 8wk.RESULTS: The expression of NGF was positive in group A and most positive in the retinal ganglion cell layer. In groups B and E, the expression of NGF was positive 2wk after transplantation and showed an increase in all layers. However, the level of expression had decreased in all layers at 4wk and was significantly reduced at 8wk. In groups C and D, the expression of NGF had increased at 2wk and continued to increase up to 8wk. | 19 | Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. | 20 | Genome-wide identification and analysis of the DREB genes and their expression profiles under abiotic stresses in Chinese jujube(Ziziphus jujuba Mill.)显示文摘CBF/DREB proteins play a critical role in abiotic stress-mediated gene expression and represent attractive regulons for plant breeding programs.However,no study has been conducted for CBF/DREB protein-related genes in jujube(Ziziphus jujuba Mill.).In this study,twenty-five ZjDREB genes were identified and annotated from the jujube(Z.jujuba‘Dongzao’)genome.Detailed analysis,including gene classification,annotation,phylogenetic evaluation,conserved motif determination and expression profiling were performed on all genes.Phylogenetic analysis showed that ZjDREB proteins were divided into five subgroups(A1–A5),but lacking a subgroup A6 corresponding to AtDREBs.The ZjDREB genes were distributed in nine of twelve chromosomes in the genome.Additionally,the expression patterns of the DREB genes under different abiotic stresses were investigated using q RT-PCR.Nineteen ZjDREB genes were down-regulated under low temperature,in contrast six ZjDREB genes(01,03,05,11,23 and 24)were up-regulated.Under drought,salinity and high temperature conditions,expression of ZjDREB03,09,10,14,15,17 and 20 genes were induced and showed similar expression patterns,suggesting that various stress conditions share common elements in the signaling pathway.The results suggest that the family of DREB genes play an important role in abiotic stresses in jujube,and provide a foundation for further functional studies of this important class of transcriptional regulators. | Heying Zhou Jiping Jia Decang Kong Zhendong Zhang Shuang Song Yingyue Li Xiaoming Pang | 2019 | Journal of Forestry Research2019,30,4: | 8 |
