2Analytical validation of GMEX rapid point- of care CYP2C19 genotyping system for the CHANCE-2 trial显示文摘Background and purpose Rapid genotyping is useful for guiding early antiplatelet therapy in patients with high-risk nondisabling ischaemic cerebrovascular events(HR-NICE).Conventional genetic testing methods used in CYP2C19 genotype-guided antiplatelet therapy for patients with HR-NICE did not satisfy the needs of the Clopidogrel in High-Risk Patients with Acute Nondisabling Cerebrovascular Events(CHANCE)-2 trial.Therefore,we developed the rapid-genotyping GMEX(point-of care)system to meet the needs of the CHANCE-2 trial.Methods Healthy individuals and patients with history of cardiovascular diseases(n=408)were enrolled from six centres of the CHANCE-2 trial.We compared the laboratory-based genomic test results with Sanger sequencing test results for accuracy verification.Next,we demonstrated the accuracy,timeliness and clinical operability of the GMEX system compared with laboratory-based technology(YZY Kit)to verify whether the GMEX system satisfies the needs of the CHANCE-2 trial.Results Genotypes reported by the GMEX system showed 100%agreement with those determined by using the YZY Kit and Sanger sequencing for all three CYP2C19 alleles(*2,*3 and*17)tested.The average result’s turnaround times for the GMEX and YZY Kit methods were 85.0(IQR:85.0-86.0)and 1630.0(IQR:354.0-7594.0)min(p<0.001),respectively.Conclusions Our data suggest that the GMEX system is a reliable and feasible point-of care system for rapid CYP2C19 genotyping for the CHANCE-2 trial or related clinical and research applications.Xia Meng Anxin Wang Guojun Zhang Siying Niu Wei Li Sifei Han Fang Fang Xingquan Zhao Kehui Dong Zening Jin Huaguang Zheng Kelin Chen Hao Li Chengyuan Yang Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:7
3Gastrointestinal bleeding during acute ischaemic stroke hospitalisation increases the risk of stroke recurrence显示文摘Objective Gastrointestinal(GI)bleeding in patients who had a stroke is strongly associated with a higher risk of death and loss of independence.However,it is unknown whether GI bleeding increases risk for recurrence of stroke.In this study,we assess the potential relationship between GI bleeding and stroke recurrence in patients within 12 months of an acute ischaemic stroke(AIS),using the China National Stroke Registry(CNSR).Methods This study included 22216 patients who had an ischaemic stroke included in the CNSR from 2007 to 2008.We analysed baseline patient characteristics,GI bleeding and outcomes of patients who had an AIS,specifically stroke recurrence at 3,6 and 12 months.We used multivariable logistic regression to evaluate a possible association between GI bleeding and stroke recurrence.results Of the 12415 patients included in our study,12.3%,15.5%and 17.7%had a stroke recurrence at 3,6 and 12 months,respectively.GI bleeding was an independent stroke recurrence risk factor in patients after ischaemic stroke at 3 months(adjusted OR 1.481,95%CI 1.118 to 1.962),6 months(adjusted OR 1.448,95%CI 1.106 to 1.896)and 12 months(adjusted OR 1.350;95%CI 1.034 to 1.763).Conclusion GI bleeding was associated with the increased risk of stroke recurrence after an AIS.Wanliang Du Xingquan Zhao Yilong Wang Yuesong Pan Gaifen Liu Anxin Wang Ruijun Ji Liping Liu Hongqiu Gu Kehui Dong Penglian Wang Yongjun Wang 2020Stroke & Vascular Neurology2020,5,2:7
4The efficacy and safety of nimodipine in acute ischemic stroke patients with mild cognitive impairment: a double-blind, randomized,placebo-controlled trial显示文摘Nimodipine might be effective in subcortical vascular dementia(VaD). Its benefit in preventing further cognitive decline in patients with acute ischemic stroke(AIS) and vascular mild cognitive impairment(VaMCI) remains to be established. In this multicenter, double-blind trial, we randomly assigned 654 eligible patients to nimodipine 30 mg three times a day or placebo. The primary outcome was any cognitive decline defined by the changes on the Mini-Mental State Examination(DMMSE à3) or vascular AD assessment scale cognitive subscale(DADAS-cog ! 4) at 6 months. Secondary outcomes included any distribution shift of DADAS-cog, DMMSE or cognitive improvement defined by DADAS-cog à2, or DMMSE ! 0. The primary outcome in the nimodipine group and placebo group were similar for DMMSE à3(4.18% and 7.22%, respectively, P = 0.15) and DADAS-cog ! 4(8.36% and 8.93% respectively,P = 0.88). The distribution shift of DADAS-cog and DMMSE differed significantly between the two groups(P = 0.03 and P = 0.05 respectively). Cognitive improvement occurred in 55.4% in the nimodipine group and 43.6% in the placebo group measured by DADAS-cog à2(Odds Ratio, 1.54; 95% confidence interval[CI] 1.10–2.14, P < 0.01) or 84.0% and 74.6% respectively by DMMSE ! 0(Odds Ratio, 1.79; 95% CI 1.18–2.70, P < 0.01). Nimodipine was associated with better cognitive function in the memory domain. The adverse events rate was similar in two groups. This study is registered with ClinicalTrials.gov,NCT01220622. 5Enclosed stigma contributes to higher spikelet fertility for rice(Oryza sativa L.) subjected to heat stress显示文摘With global warming, rice plants may be subjected to heat stress more regularly during the heatsensitive flowering stage, causing spikelet sterility and grain yield loss.Stigma exsertion is considered to increase pollen reception and promote female reproductive success.The aim of this study was to investigate the role of stigma exsertion on spikelet fertility at high temperatures.Five rice cultivars(Liangyoupeijiu, Shanyou 63, Huanghuazhan, Nagina 22, and IR64) with differing degrees of stigma exsertion were cultivated and exposed to high temperature at anthesis.Heat-tolerant cultivars did not always show a high percentage of spikelets with exserted stigmas, and vice versa.Irrespective of the presence of more pollen grains on exserted stigmas, spikelets with exserted stigmas did not show greater spikelet fertility than spikelets with fewer exserted stigmas or hidden stigmas under heat stress.GA3 application augmented the percentage of spikelets with exserted stigmas;however, it did not increase spikelet fertility under heat stress.Spikelet fertility of whole panicles was negatively correlated with the percentage of spikelets with exserted stigmas, but positively with that with hidden stigmas.Viability of the hidden stigmas was less reduced than that of exserted stigmas under heat stress, suggesting that hidden stigmas have an advantage in maintaining viability.Heat stress delayed anther dehiscence and reduced the viabilities of both exserted stigmas and pollens, thereby causing low spikelet fertility.Together, these results suggest that high spikelet fertility does not depend on stigma exsertion and that enclosed stigma generally contributes to higher spikelet fertility and heat tolerance under high-temperature conditions during flowering in rice.Chao Wu Kehui Cui Qiuqian Hu Wencheng Wang Lixiao Nie Jianliang Huang Shaobing Peng 2019The Crop Journal2019,7,3:4

7Hydrolysis of polyaluminum chloride prior to coagulation:Effects on coagulation behavior and implications for improving coagulation performance显示文摘The effects of polyaluminum chloride(PACl) hydrolysis prior to coagulation on both the coagulation zone and coagulation performance of a kaolin suspension were investigated by a novel jar test named the 'reversed coagulation test'.The tests showed that PACl hydrolysis prior to coagulation decreased the performance of charge neutralization coagulation in the case of short-time slow mixing(10 min;G = 15 sec-1) and increased the optimal dosage for charge neutralization and sweep coagulation.Moreover,the hydrolysis time had insignificant effects on the size and zeta potential of PACl precipitates and the residual turbidity of the raw water.However,PACl hydrolysis prior to coagulation and the size of PACl precipitates had a negligible effect on the performance of sweep coagulation.The results imply that,in practice,preparing a PACl solution with deionized water,rather than tap water or the outlet water from a wastewater treatment unit,can significantly save PACl consumption and improve the performance of charge neutralization coagulation,while preparing the PACl solution with tap or outlet water would not affect the performance of sweep coagulation.In addition,the optimal rapid mixing intensity appears to be determined by a balance between the degree of coagulant hydrolysis before contacting the primary particles and the average size of flocs in the rapid mixing period.These results provide new insights into the role of PACl hydrolysis and will be useful for improving coagulation efficiency.Zhongguo Zhang Jun Wang Dan Liu Jiuyi Li Xiaolin Wang Boyu Song Bing Yue Kehui Zhao Yun Song 2017Journal of Environmental Sciences2017,29,7:4
8Double Position Servo Synchronous Drive System Based on Cross-Coupling Integrated Feedforward Control for Broacher显示文摘Synchronization errors directly deteriorate the machining accuracy of metal parts and the existed method cannot keep high synchronization precision because of external disturbances. A new double position servo synchronous driving scheme based on semi-closed-loop crosscoupling integrated feedforward control is proposed. The scheme comprises a position error cross-coupling feedforward control and a load torque identification with feedforward control. A digital integrated simulation system for the dual servo synchronous drive system is established.Using a 20 t servo broacher, performance analysis of the scheme is conducted based on this simulation system and the simulation results show that systems with traditional parallel or single control have problems when the worktable works with an unbalanced load. However, the system with proposed scheme shows good synchronous performance and positional accuracy. Broaching tests are performed and the experimental results show that the maximum dual axis synchronization error of the system is only 8 μm during acceleration and deceleration processes and the error between the actual running position and the given position is almost zero. 13Exploring a particle-size-reduction strategy of YAG:Ce phosphor via a chemical breakdown method显示文摘Particle size reduction of Y_(3)Al_(5)O_(12):Ce(YAG:Ce) phosphor is highly needed for micro-LED display applications.In this work,size control of YAG:Ce phosphor particles is achieved via carbon coating and further heat treatment.A thin layer of carbon is deposited on the surface of YAG:Ce by chemical vapor deposition.During the heat treatment,carbon reacts with oxygen element in the phosphor and escapes from phosphor particles.The reaction results in the phosphor breaking into smaller particles.The phosphors were characterized by laser light scattering for particle size distribution,scanning electron microscopy(SEM),transmission electron microscopy for morphologies,X-ray diffraction refinements for crystal structure and electron energy-loss spectroscopy for elemental analysis.It is demonstrated that the median diameter(D_(50)) of the phosphor particle size is significantly reduced from 19.1 to 0.96 μm yet the photoluminescent properties have little changes.The carbon coating and further heat treatment method show potential application in size control of phosphors.Hongren Chen Licheng Ju Li Zhang Xin Wang Linbo Zhang Xin Xu Lin Gao Kehui Qiu Liangjun Yin 2021Journal of Rare Earths2021,39,8:1

15Novel Natural Allelic Variations at the Rht-1 Loci in Wheat显示文摘Plant height is an important agronomic trait.Dramatic increase in wheat yield during the'green revolution'is mainly due to the widespread utilization of the Reduced height(Rht)-1 gene.We analyzed the natural allelic variations of three homoeologous loci Rht-A1,Rht-B1,and Rht-D1 in Chinese wheat(Triticum aestivum L.)micro-core collections and the Rht-B1/D1 genotypes in over 1,500 bred cultivars and germplasms using a modifed EcoTILLING.We identifed six new Rht-A1 allelic variations(Rht-A1b–g),eight new Rht-B1 allelic variations(Rht-B1h–o),and six new Rht-D1 allelic variations(Rht-D1e–j).These allelic variations contain single nucleotide polymorphisms(SNPs)or small insertions and deletions in the coding or uncoding regions,involving two frame-shift mutations and 15 missenses.Of which,Rht-D1e and Rht-D1h resulted in the loss of interactions of GID1-DELLA-GID2,Rht-B1i could increase plant height.We found that the Rht-B1h contains the same SNPs and 197 bp fragment insertion as reported in Rht-B1c.Further detection of Rht-B1h in Tibet wheat germplasms and wheat relatives indicated that Rht-B1c may originate from Rht-B1h.These results suggest rich genetic diversity at the Rht-1 loci and provide new resources for wheat breeding.Aixia Li Wenlong Yang Xueyuan Lou Dongcheng Liu Jiazhu Sun Xiaoli Guo Jing Wang Yiwen Li Kehui Zhan Hong-Qing Ling Aimin Zhang 2013Journal of Integrative Plant Biology2013,55,11:1

17Response of spikelet water status to high temperature and its relationship with heat tolerance in rice显示文摘In rice, high-temperature stress(HT) during flowering results in decreased grain yield via a reduction in spikelet fertility;however, the effect of plant water status on spikelet fertility under HT remains unknown. To investigate the relationship between spikelet water status and spikelet fertility under HT, two experiments were performed under temperature-controlled conditions using four genotypes with varying tolerance to HT. Rice plants were exposed to HT for seven consecutive days during the flowering stage under three soil water treatments(soil water potential 0, -20, and -40 kPa), as well as under hydroponic conditions in a separate experiment. HT significantly decreased spikelet fertility, pollen fertility, and anther dehiscence under each of the three water treatments. HT significantly increased the spikelet transpiration rate, and this change was accompanied by a significant decrease in the internal temperature of the spikelets. HT decreased pollen grain diameter in heat-sensitive genotypes. HT had varying effects on the water potential of panicles and anthers but increased anther soluble-sugar concentration. Different aquaporin genes showed different expression profiles under HT, and the expression levels of PIPs for plasma membrane intrinsic proteins and TIPs for tonoplast intrinsic proteins increased in anthers but decreased in glumes.Correlation analyses showed that anther dehiscence and pollen(spikelet) fertility were tightly associated with anther water status, and the expression levels of almost all anther aquaporin genes were significantly correlated with anther dehiscence under HT. In summary, an increased spikelet transpiration rate and decreased internal spikelet temperature were associated with alleviation of the effects of HT in rice genotypes with varying degrees of heat tolerance, and the response of spikelet water status to HT, involving increased total expression of aquaporins and soluble sugar content, thereby improved pollen fertility,anther dehiscence, and spikelet fertility, especially in heat-resistant genotypes. 19Chimonanthus nitens Oliv. leaves flavonoids alleviate hyperuricemia by regulating uric acid metabolism and intestinal homeostasis in mice显示文摘This study aimed to evaluate the alleviating effect of Chimonanthus nitens Oliv.leaves flavonoids(CLF)on hyperuricemia induced by potassium oxonate in mice.The results showed that CLF lowered the serum levels of uric acid(UA),creatinine and blood urea nitrogen,downregulated hepatic mRNA expressions of xanthine oxidase(XO),phosphate ribose pyrophosphate synthetase(PRPS)and adenosine deaminase(ADA)in hyperuricemia mice.In addition,CLF repaired renal injury by significantly down-regulating mRNA and protein expressions of renal UA reabsorption-related proteins and up-regulating the mRNA and protein expressions of UA secretory-related proteins.Finally,CLF inhibited UA synthesis and promoted UA excretion to alleviate hyperuricemia.Besides,CLF supplementation repaired the intestinal barrier function as demonstrated by significant increased mRNA levels of intestinal zonula occludens-1(ZO-1),Occludin,mucin 2(MUC2)and mucin 4(MUC4),as well as decreased mRNA levels of toll-like receptor 4(TLR4)and myeloid differentiation factor 88(MyD88)in mice.Further research showed that CLF treatment restored intestinal homeostasis mediated by improving the composition of gut microbiota and elevating the abundance of beneficial bacteria like Lactobacillus,Alistipes,Prevotellaceae_UCG-001 and Parasutterella.Overall,our findings revealed a novel function of CLF as a promising therapeutic candidate for the treatment of hyperuricemia.Wenya Meng Lingli Chen Kehui Ouyang Suyun Lin Yang Zhang Jing He Wenjun Wang 2023Food Science and Human Wellness2023,12,6:0
20Construction of Tetrathiafulvalene-based Covalent Organic Frameworks for Superior Iodine Capture显示文摘The effective capture of radioiodine species during nuclear fuel reprocessing and nuclear accidents is of primary importance but remains challenging for the sustainable development of nuclear energy.Herein,we report two newly designed two-dimensional(2D)and three-dimensional(3D)covalent organic frameworks by introducing tetrathiafulvalene functional groups into the building units for the simultaneous physisorption and chemisorption capture of iodine molecules.Remarkably,the obtained 3D TTF-TAPT-COF material exhibited a superior iodine vapor adsorption capacity of up to 5.02 g/g at 348 K and under ambient pressure and an adsorption kinetics of 0.515 g/(g∙h),surpassing most of other materials reported so far.The strong physiochemical interactions between iodine molecules and the frameworks of the obtained COFs were revealed by a set of experimental techniques.This study provides a feasible approach for the rational design and the construction of novel and effective COF-based adsorbents for iodine enrichment and related environmental remediation.WANG Guangbo XIE Kehui ZHU Fucheng KAN Jinglan LI Sha GENG Yan DONG Yubin 2022Chemical Research in Chinese Universities2022,38,2:0
