| 3 | Polysaccharides from Angelica sinensis alleviate neuronal cell injury caused by oxidative stress显示文摘Angelica sinensis has antioxidative and neuroprotective effects. In the present study, we aimed to determine the neuroprotective effect of polysaccharides isolated from Angelica sinensis. In a preliminary experiment, Angelica sinensis polysaccharides not only protected PC12 neuronal cells from H 2 O 2-induced cytotoxicity, but also reduced apoptosis and intracellular reactive oxygen species levels, and increased the mitochondrial membrane potential induced by H 2 O 2 treatment. In a rat model of local cerebral ischemia, we further demonstrated that Angelica sinensis polysaccharides enhanced the antioxidant activity in cerebral cortical neurons, increased the number of microvessels, and improved blood flow after ischemia. | 4 | Immunotherapy using IL-2 and GM-CSF is a potential treatment for multidrug-resistant Mycobacterium tuberculosis显示文摘This study investigated the therapeutic effects of interleukin (IL)-2 and granulocyte-macrophage colony-stimulating factor (GM-CSF) coadministrated with antibacterial agents isoniazid (INH) and rifampin (RIF) to treat a mouse model of tuberculosis (TB) infection. A drug-susceptible TB strain, H37Rv was used to infect mice and the effectiveness of IL-2 and GM-CSF was initially evaluated based on survival rate, bacterial counts in lungs and spleens and the pathological condition of the lungs. Next, the therapeutic effect of the immunotherapy regimen was assessed in multidrug-resistant strain OB35-infected mice. In the H37Rv infection model, IL-2 and GM-CSF monotherapies reduced bacterial numbers in the lungs by 0.82 (P<0.01) and 0.58 (P<0.05) lg colony-forming units (CFU), respectively, and in the spleens by 1.42 (P<0.01) and 1.22 (P<0.01) lg CFU, respectively, compared with the untreated group. Mice receiving immunotherapy developed fewer lesions in the lungs compared with mice receiving antibacterial therapy alone. In the OB35 infection model, immunotherapy with either cytokine resulted in a significant reduction of bacterial load in the lungs and spleens and less severe lesions in the lungs compared with the untreated or antibacterial therapy treated mice. Notably, mice receiving immunotherapy with both cytokines had a 30% survival rate which was higher than that in other treated groups, and had significantly less CFUs in the lungs and spleens (1.02 and 1.34 lg CFU) compared with antibacterial therapy alone (P<0.01). | 5 | Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. | 7 | Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis. | Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu | 2021 | Pediatric Investigation2021,5,3: | 12 |
| 8 | Preliminary analysis on the source properties and seismogenic structure of the 2017 M_s7.0 Jiuzhaigou earthquake显示文摘At GMT time 13:19, August 8, 2017, an Ms7.0 earthquake struck the Jiuzhaigou region in Sichuan Province, China,causing severe damages and casualties. To investigate the source properties, seismogenic structures, and seismic hazards, we systematically analyzed the tectonic environment, crustal velocity structure in the source region, source parameters and rupture process, Coulomb failure stress changes, and 3-D features of the rupture plane of the Jiuzhaigou earthquake. Our results indicate the following:(1) The Jiuzhaigou earthquake occurred on an unmarked fault belonging to the transition zone of the east Kunlun fault system and is located northwest of the Huya fault.(2) Both the mainshock and aftershock rupture zones are located in a region where crustal seismic velocity changes dramatically. Southeast to the source region, shear wave velocity at the middle to lower crust is significantly low, but it rapidly increases northeastward and lies close to the background velocity across the rupture fault.(3) The aftershock zone is narrow and distributes along the northwest-southeast trend, and most aftershocks occur within a depth range of 5–20 km.(4) The focal mechanism of the Jiuzhaigou earthquake indicates a left-lateral strike-slip fault, with strike, dip, and rake angles of 152°, 74° and 8°, respectively. The hypocenter depth measures 20 km, whereas the centroid depth is about 6 km. The co-seismic rupture mainly concentrates at depths of 3–13 km, with a moment magnitude(M_w) of 6.5.(5) The co-seismic rupture also strengthens the Coulomb failure stress at the two ends of the rupture fault and the east segment of the Tazang fault. Aftershocks relocation results together with geological surveys indicate that the causative fault is a near vertical fault with notable spatial variations: dip angle varies within 66°–89° from northwest to southeast and the average dip angle measures ~84°. | 9 | Functional analysis of the ABA-responsive protein family in ABA and stress signal transduction in Arabidopsis显示文摘The phytohormone abscisic acid(ABA)plays an important role in plant growth and development,for example in seed dormancy and germination,as well as in plant responses to environmental stresses,such as drought and high salinity.Previous studies have shown that ABA regulates the expression of genes with an ABA-responsive element(ABRE)and their corresponding physiological responses.Bioinformatics analysis identified a GRAM domain-containing gene family that has a multiple ABRE cis-element,which was termed the ABA-responsive protein(ABR)family.To analyze the function of the ABR family,we identified homozygous T-DNA insertion mutants and constructed abr1,2,3 double mutants and triple mutant.The abr1,abr2 and abr3single mutants showed a normal phenotype;however,the germination of seeds of the double mutants and triple mutant were insensitive to ABA,NaCl,mannitol and glucose.ABR1-GFP was distributed as a punctate structure in the cytosol and may be localized in the endomembrane system.The ABR2-GFP and ABR3-GFP proteins localized in the cytoplasm.In addition,ABR1,ABR2 and ABR3 were expressed in various tissues,and could be induced by several abiotic stresses,especially by ABA.The expressions of these genes were significantly suppressed in aba2,abi1 and abi2 null mutants.These results suggested that the ABR family may act downstream of ABI1 and ABI2 in the ABA signal transduction process in plants. | LIU LingYun LI Na YAO ChunPeng MENG SaSa SONG ChunPeng | 2013 | Chinese Science Bulletin2013,58,31: | 7 |

| 11 | Proteasome inhibition suppresses Th17 cell generation and ameliorates autoimmune development in experimental Sjögren’s syndrome显示文摘Immunoproteasome activation in immune cells is involved in the modulation of immune responses.Increasing evidence indicates that proteasome inhibitors show beneficial effects in treating autoimmune diseases,but it remains unclear whether proteasome inhibition is an effective approach for suppressing autoimmune development in Sjögren’s syndrome(SS).Our previous work has demonstrated a critical role for Th17 cells in the development of experimental SS(ESS)in mice.In this study,we detected high levels of low-molecular-weight protein 7(LMP7),a key subunit of the immunoproteasome,in Th17 cells from ESS mice.Moreover,treatment with bortezomib(BTZ),a proteasome inhibitor,markedly suppressed Th17 differentiation in both murine and human naive T cells in culture.Furthermore,ESS mice treated with BTZ displayed significantly higher saliva flow rates and a reduction in tissue destruction in the salivary glands compared with vehicle-treated ESS mice.Notably,BTZ-treated ESS mice showed markedly decreased Th17 cells,germinal center B cells and plasma cells in the peripheral lymphoid organs.In addition,adoptively transferred wild type naive CD4+T cells rapidly differentiated into Th17 cells and induced salivary dysfunction in IL-17-deficient mice immunized for ESS induction.However,BTZ treatment profoundly suppressed the donor T-cell-derived Th17 response and ameliorated the reduction in salivary secretion in IL-17-deficient recipient mice.Taken together,our findings demonstrate that proteasome inhibition can effectively ameliorate ESS by suppressing the Th17 response,which may contribute to the development of a novel therapeutic strategy for the treatment of SS. | Fan Xiao Xiang Lin Jie Tian Xiaohui Wang Qian Chen Ke Rui Jie Ma Shengjun Wang Qingwen Wang Xiaoqi Wang Dongzhou Liu Lingyun Sun Liwei Lu | 2017 | Cellular & Molecular Immunology2017,14,11: | 6 |
| 12 | In vivo retention of poloxamer-based in situhydrogels for vaginal application in mouse and rat models显示文摘The purpose of this study is to evaluate the in vivo retention capabilities of poloxamer-based in situ hydrogels for vaginal application with nonoxinol-9 as the model drug. Two in situ hydrogel formulations, which contained 18% poloxamer 407 plus 1% poloxamer 188(GEL1, relative hydrophobic)or 6% poloxamer 188(GEL2, relative hydrophilic), were compared with respect to the rheological properties, in vitro hydrogel erosion and drug release. The vaginal retention capabilities of these hydrogel formulations were further determined in two small animal models, including drug quantitation of vaginal rinsing fluid in mice and isotope tracing with99 m Tc in rats. The two formulations exhibited similar phase transition temperatures ranging from 27 to 32 1C. Increasing the content of poloxamer 188 resulted in higher rheological moduli under body temperature, but slightly accelerated hydrogel erosion and drug release. When compared in vivo, GEL1 was eliminated significantly slower in rat vagina than GEL2,while the vaginal retention of these two hydrogel formulations behaved similarly in mice. In conclusion,increases in the hydrophilic content of formulations led to faster hydrogel erosion, drug release andintravaginal elimination. | 18 | A viral vector expressing hypoxia-inducible factor 1 alpha inhibits hippocampal neuronal apoptosis显示文摘Hypoxia-inducible factor 1(HIF-1) attenuates amyloid-beta protein neurotoxicity and decreases apoptosis induced by oxidative stress or hypoxia in cortical neurons. In this study, we constructed a recombinant adeno-associated virus(rAAV) vector expressing the human HIF-1α gene(rAAV-HIF-1α), and tested the assumption that rAAV-HIF-1α represses hippocampal neuronal apoptosis induced by amyloid-beta protein. Our results confirmed that rAAV-HIF-1α significantly reduces apoptosis induced by amyloid-beta protein in primary cultured hippocampal neurons. Direct intracerebral rAAV-HIF-1α administration also induced robust and prolonged HIF-1α production in rat hippocampus. Single rAAV-HIF-1α administration resulted in decreased apoptosis of hippocampal neurons in an Alzheimer's disease rat model established by intracerebroventricular injection of aggregated amyloid-beta protein(25–35). | 19 | TiO_2-doped Fe_3O_4 nanoparticles as high-performance Fenton-like catalyst for dye decoloration显示文摘Fenton reaction based on Fe2+-H2O2 system has been widely applied in water remediation, but the obvious drawbacks largely hinder its practical uses. Alternatively, heterogeneous nanomaterials with proper surface modification could be used as Fenton-like catalysts. Surface doping of Ti O2 could concentrate the pollutants surrounding the Fe3O4 catalyst, which might benefit the catalytic performance of Fe3O4. Herein, we reported that Ti O2-doped Fe3O4 nanoparticles(NPs) could be used as high-performance Fenton-like catalyst for dye decoloration in near neutral environment, where the doping of Ti O2 on Fe3O4 surface dramatically improved the catalytic activity of Fe3O4 in Fenton-like reaction. Ti O2-doped Fe3O4 NPs catalyzed the decomposition of H2O2 to oxidize methylene blue without external energy supply, resulting in effective decoloration. Ti O2-doped Fe3O4 NPs showed high catalytic activity under various p H values and even in the presence of radical scavenger. More catalysts and H2O2 would facilitate the decoloration. At higher temperature, the decoloration became faster and more effective. | 20 | Non-dominated sorting culture differential evolution algorithm for multi-objective optimal operation of Wind-Solar-Hydro complementary power generation system显示文摘Due to the intermittency and instability of Wind-Solar energy and easy compensation of hydropower, this study proposes a Wind-Solar-Hydro power optimal scheduling model. This model is aimed at maximizing the total system power generation and the minimum ten-day joint output. To effectively optimize the multi-objective model, a new algorithm named non-dominated sorting culture differential evolution algorithm(NSCDE) is proposed. The feasibility of NSCDE was verified through several well-known benchmark problems. It was then applied to the Jinping Wind-Solar-Hydro complementary power generation system.
