1Effects of environmental contaminants on fertility and reproductive health显示文摘Recent research indicates that the human infertility rate is increasing. Although various reasons have been hypothesized for the growing infertility rate, environmental contaminants are potentially important causal agents associated with this change.Chemical contaminants are widespread throughout our environment and human exposure is virtually unavoidable. The overall contribution of environmental exposure to infertility is unknown, but studies involving occupational exposure, together with results from animal experiments, suggest that environmental contaminants may adversely affect fertility. We reviewed the adverse effects of environmental exposure on fertility and related reproductive outcomes. Environmental contaminants covered in this review include heavy metals, organic solvents, pesticides and endocrine disrupting chemicals. 2Mechanisms of Traditional Chinese Medicine Bushenantai granules(补肾安胎颗粒) in promoting angiogenesis at the maternal-fetal interface of recurrent spontaneous abortion mice显示文摘OBJECTIVE:To assess the effects of Bushenantai(BSAT)granule(补肾安胎颗粒)on angiogenesis-related factors[E2,P,and vascular endothelial growth factor(VEGF)]at the maternal-fetal interface of recurrent spontaneous abortion(RSA)mice,and to evaluate the role of BSAT in promoting angiogenesis at the maternal-fetal interface by influencing the expression of sex hormones,and VEGF.METHODS:A mouse model with normal pregnancy and another with Clark’s classic RSA were established.The RSA mice were randomly assigned to six groups:normal,model,progesterone,high-doseBSAT granule(BSAT-H),medium-dose-BSAT granule(BSAT-M),and low-dose-BSAT granule(BSAT-L)(n=10 for each group).The embryo loss rate and the histopathological changes in the decidual tissues were measured.Serum levels of estrogen(E2),progesterone(P),and VEGF were detected by enzyme-linked immunosorbent assay.The m RNA and protein expressions of estradiol receptor(ER),progesterone receptor(PR),VEGF,and vascular endothelial growth factor receptor 2(VEGFR2)in the decidual tissues were identified by immunohistochemistry,Western blotting,and quantitative reverse transcription polymerase chain reaction.RESULTS:The embryo loss rate in all groups that received BSAT treatment was reduced,while the number of blood vessels at decidual tissues was increased.The serum levels of E2,P and VEGF were elevated,and the m RNA and protein expressions of ER,PR,VEGF,and VEGFR2 in the decidual tissues were enhanced.CONCLUSION:BSAT can improve angiogenesis at the maternal-fetal interface and reduce the embryo loss rate,which may be associated with its ability to increase the serum levels of estrogen,progesterone,and VEGF,in addition to up-regulation of m RNA and protein expression of ER,PR,VEGF,and VEGFR2 in the decidual tissues.LI Yuanqi LI Weili YU Xinhui WU Hua WANG Yuanzhong JIN Ya HAO Lele LIU Mingmin SONG Xiaoge 2021Journal of Traditional Chinese Medicine2021,41,4:3
3Inducible expression of Wnt7b promotes bone formation in aged mice and enhances fracture healing显示文摘There remain unmet clinical needs for safe and effective bone anabolic therapies to treat aging-related osteoporosis and to improve fracture healing in cases of nonunion or delayed union. Wnt signaling has emerged as a promising target pathway for developing novel bone anabolic drugs. Although neutralizing antibodies against the Wnt antagonist sclerostin have been tested,Wnt ligands themselves have not been fully explored as a potential therapy. Previous work has demonstrated Wnt7b as an endogenous ligand upregulated during osteoblast differentiation, and that Wnt7b overexpression potently stimulates bone accrual in the mouse. The earlier studies however did not address whether Wnt7b could promote bone formation when specifically applied to aged or fractured bones. Here we have developed a doxycycline-inducible strategy where Wnt7b is temporally induced in the bones of aged mice or during fracture healing. We report that forced expression of Wnt7b for 1 month starting at 15 months of age greatly stimulated trabecular and endosteal bone formation, resulting in a marked increase in bone mass. We further tested the effect of Wnt7b on bone healing in a murine closed femur fracture model. Induced expression of Wnt7b at the onset of fracture did not affect the initial cartilage formation but promoted mineralization of the subsequent bone callus. 4Sensing carboxylesterase 1 in living systems by a practical and isoformspecific fluorescent probe显示文摘Carboxylesterase 1(CES1), one of the most abundant serine hydrolases in mammals, has drawn much attentions in recent years, owing to this enzyme involves in many physiological processes via hydrolysis of both endogenous esters and xenobiotic esters. Herein, to real-time monitor the activities of CES1 in various biological systems, a practical and iso form-specific fluorescent probe was developed on the basis of the substrate preference of CES1, as well as the structural and optical properties of BODIPY dyes. After screening of a panel of BODIPY ester derivatives, probe 1 displayed the best combination of specificity,sensitivity, enzymatic kinetics and applicability for monitoring CES1 activities in real samples. This probe was successfully used to detect CESl activities in several biological systems including tissue preparations,living cells, tissue slices and zebrafish. Furthermore, the biomedical applications of probe 1 for screening of CES1 inhibitors were also demonstrated using tissue preparations or living cells as enzyme sources. 6Comprehensive characterization of CRC with germline mutations reveals a distinct somatic mutational landscape and elevated cancer risk in the Chinese population显示文摘Objective:Hereditary colorectal cancer(CRC)accounts for approximately 5%–10%of all CRC cases.The full profile of CRC-related germline mutations and the corresponding somatic mutational profile have not been fully determined in the Chinese population.Methods:We performed the first population study investigating the germline mutation status in more than 1,000(n=1,923)Chinese patients with CRC and examined their relationship with the somatic mutational landscape.Germline alterations were examined with a 58-gene next-generation sequencing panel,and somatic alterations were examined with a 605-gene panel.Results:A total of 92 pathogenic(P)mutations were identified in 85 patients,and 81 likely pathogenic(LP)germline mutations were identified in 62 patients,accounting for 7.6%(147/1,923)of all patients.MSH2 and APC was the most mutated gene in the Lynch syndrome and non-Lynch syndrome groups,respectively.Patients with P/LP mutations had a significantly higher ratio of microsatellite instability,highly deficient mismatch repair,family history of CRC,and lower age.The somatic mutational landscape revealed a significantly higher mutational frequency in the P group and a trend toward higher copy number variations in the non-P group.The Lynch syndrome group had a significantly higher mutational frequency and tumor mutational burden than the nonLynch syndrome group.Clustering analysis revealed that the Notch signaling pathway was uniquely clustered in the Lynch syndrome group,and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Population risk analysis indicated that the overall odds ratio was 11.13(95%CI:8.289–15.44)for the P group and 20.68(95%CI:12.89–33.18)for the LP group.Conclusions:Distinct features were revealed in Chinese patients with CRC with germline mutations.The Notch signaling pathway was uniquely clustered in the Lynch syndrome group,and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Patients with P/LP germline mutations exhibited higher CRC risk.Jianfei Yao Yunhuan Zhen Jing Fan Yuan Gong Yumeng Ye Shaohua Guo Hongyi Liu Xiaoyun Li Guosheng Li Pan Yang Xiaohui Wang Danni Liu Tanxiao Huang Huiya Cao Peisu Suo Yuemin Li Jingbo Yu Lele Song 2022Cancer Biology & Medicine2022,19,5:1

9The Jidong Eye Cohort Study:objectives,design,and baseline characteristics显示文摘Background:To describe the objective,design and baseline characteristics of the Jidong Eye Cohort Study(JECS),a community-based cohort in China based on etiology,imaging and biomarkers.The JECS will clarify the pathogenesis of visual impairment and status of ocular indicators in the occurrence and progression of cardiocerebrovascular and neurological diseases.Methods:Between August 2019 and January 2020,the JECS recruited consecutive participants aged 18 years and older from the Jidong communities in China.The demographic and clinical characteristics were collected by trained site personnel via face-to-face interviews.The relevant biological samples were also collected.The participants underwent comprehensive ophthalmic examination,such as retinal photography and optical coherence tomography(OCT)angiography.The following outcomes were measured annually:ocular vascular abnormality,optic nerve degeneration,cardiovascular diseases(CVD)and neurological diseases.The study will be performed until 2024.Results:Among 3377 participants,the average age was 45.0±12.5 years and 1809(53.6%)were women.Hypertension occurred in 825 individuals(25.0%),diabetes in 258(7.7%),hyperglycemia in 474(14.2%),and a CVD history in 100(3.0%).The mean best-corrected visual acuity was 0.1 logMAR in the recruited subjects.The average OCT signal index was 8.2±1.2.Additionally,the mean vessel densities for the entire measured area were 46.4%and 50.8%for the superficial and deep vascular complex,respectively.Mean area and perimeter of foveal avascular zone was 0.3 mm2 and 2.3 mm.Conclusions:The JECS is a large community-based prospective cohort in North China.Rich data collected from this study will provide the opportunity to identify risk factors,imaging,and biomarkers of visual impairment(either ocular vascular anomalies or optic nerve degeneration)and to evaluate their associations with CVD and neurological diseases.Kai Yang Lele Cui Guoyun Zhang Xianwei Wang Xiaoxuan Zhu Yunfan Xiao Binbin Su Daiyu Song Xinyao Zhang Yang Zheng Fan Lu Jia Qu Ming Li 2020Eye and Vision2020,7,1:0
10Magnetic properties,crystalline and magnetic microstructures of dual-main-phase(Nd,Ce)-Fe-B sintered magnets显示文摘The(Nd,Ce)-Fe-B sintered magnets were prepared by a dual-main-phase(DMP)process under the same conditions using different combinations of Nd-Fe-B with Ce-Fe-B and(Nd,Ce)-Fe-B strips.The crystalline and magnetic microstructures of DMP(Nd,Ce)-Fe-B magnets with the remanence of 11.92-12.68 kGs,the intrinsic coercivity of 3.97-5.31 kOe,and the maximum energy product of 23.08-32.99 MGOe have been investigated.Magnetic force microscope(MFM)investigations reveal that the DMP(Nd,Ce)-Fe-B magnets show maze-like patterns,which are like that of standard anisotropy Nd-Fe-B sintered magnets by and large.However,much finer domain structures mixing with coarse ones can be observed obviously in DMP(Nd,Ce)-Fe-B sintered magnets.The size distribution of the domain width of the DMP(Nd,Ce)-Fe-B magnet is not uniform obviously.The average domain width is W=0.912μm,and the fine domain width has only 0.216μm.The smaller domain width and more branch domain patterns exist in poorer coercivity DMP magnets.This is caused by the non-uniform Ce composition of poorer property DMP magnets and the grain boundary microstructure that is not conducive to improving the coercivity.Furthermore,it is found that some domains of rare-earth-rich grain boundary phases exhibit the characteristics of plate-like patterns rather than no-contrast by using MFM,indicating their ferromagnetism.Obvious correlations between the crystalline microstructure,chemical composition of phases,and magnetic structure were demonstrated for the DMP magnets.Yifei Xiao Lele Zhang Tao Liu Xiaolong Song Qisong Sun Yikun Fang Minggang Zhu Wei Li 2023Journal of Materials Science & Technology2023,,31:0
11Simultaneous Desorption of Polycyclic Aromatic Hydrocarbons and Heavy Metals from Contaminated Soils by Rhamnolipid Biosurfactants显示文摘Hydrophobic organic compounds(HOCs)and heavy metals(HMs)are toxins that usually coexist in natural environments.Due to the differences in their properties,it remains challenging to simultaneously remove HMs and HOCs.In this study,the removal of phenanthrene(Phe)and lead(Pb)from co-contaminated soils by single rhamnolipid(RL)and mixed RL-sophorolipid(SL)biosurfactants were evaluated via soil column experiments.Biosurfactant micelle sizes were determined by dynamic light scattering,and the mechanisms of micelle solubilization were studied.The effects of biosurfactant concentrations,pH,washing agent salinity and the ageing time of polluted soils on Phe and Pb desorption efficiencies were also assessed.The substantial removal of Phe and Pb using mixed RL-SL systems,when molar fractions of RLs were 0.7,was attributed to large mixed micelle formation and lower sorption losses of these systems.The optimal pH value was 6.0,while Phe desorption was favoured at high RLs and low ionic strengths.However,the RLs concentration and ionic strength had no obvious influence on Pb removal.In addition,both Phe and Pb desorption decreased with increased ageing of the polluted soils.Combined RL-SL biosurfactants can be effective for simultaneously removing HOCs and HMs from polluted soils.WU Liang SONG Dandan YAN Lele LIANG Shengkang YANG Yanqun PENG Changsheng SHANG Yujun WANG Xiuli DONG Xueliang 2020Journal of Ocean University of China2020,19,4:0
12Enhanced oil recovery by low-salinity water spontaneous imbibition (LSW-SI) in a typical tight sandstone formation of mahu sag from core scale to field scale显示文摘Accelerating mass exchange between matrix and fractures is the essence of enhanced oil recovery(EOR)in tight formations after natural depletion.Low salinity water(LSW)injection has been commerciallyproven in conventional reservoirs EOR,with scale projects in progress worldwide.There is,however,a lack of understanding of the EOR effect in tight formations.Therefore,in this work,we introduced LSWEOR to a target tight formation using huff-puff mode.Spontaneous imbibition(SI)tests were firstly performed on homogenous Berea sandstone cores with decreasing salinity brine to observe the production response.The results indicated that the oil recovery of the tight rock was boosted by tuning brine salinity.Of all the used brines with salinity ranging from 0.021%to 2.1%TDS(total dissolved salinity),the 0.21%TDS brine showed a rapid increase in oil production over imbibing time,which finally led to an incremental oil recovery of 4.5%OOIP(original oil in place).Core-scale modeling was conducted by history-matching the oil recovery dynamics of the SI results through modifying capillary pressure and relative permeability.A full-scale reservoir model was constructed using micro-seismic data to model fracture geometry combing fracturing results and scaling parameters obtained from core scale historymatching.It is proven that LSW huff-n-puff stimulated the oil production after natural depletion and improved MEE(mass exchange efficiency)of the target formation,but the EOR benefit was not comparable to CO2 and surfactant-assisted water huff-puff methods.Bing Wei Lele Wang Tao Song Mengying Zhong Mikhail A.Varfolomeev 2021Petroleum2021,7,3:0
13New insights into the dispersion history and adaptive evolution of taxon Aegilops tauschii in China显示文摘Aegilops tauschii,the wild progenitor of wheat D-genome and a valuable germplasm for wheat improvement,has a wide natural distribution from eastern Turkey to China.However,the phylogenetic relationship and dispersion history of Ae.tauschii in China has not been scientifically clarified.In this study,we genotyped 208 accessions(with 104 in China)using dd RAD sequencing and 55K SNP array,and classified the population into six sublineages.Three possible spreading routes or events were identified,resulting in specific distribution patterns,with four sublineages found in Xinjiang,one in Qinghai,two in Shaanxi and one in Henan.We also established the correlation of SNP-based,karyotypebased and spike-morphology-based techniques to demonstrate the internal classification of Ae.tauschii,and developed consensus dataset with 1245 putative accessions by merging data previously published.Our analysis suggested that eight inter-lineage accessions could be assigned to the putative Lineage 3and these accessions would help to conserve the genetic diversity of the species.By developing the consensus phylogenetic relationships of Ae.tauschii,our work validated the hypothesis on the dispersal history of Ae.tauschii in China,and contributed to the efficient and comprehensive germplasm-mining of the species.Hao Li Fang Nie Lele Zhu Menghua Mu Ruixiao Fan Jingyao Li Aaqib Shaheen Yifan Liu Can Li Wenjuan Liu Huihui Liang Xinpeng Zhao Shenglong Bai Guanghui Guo Zheng Li Yiheng Hu Yuannian Jiao Jonathan Adams Assaf Distelfeld Guiling Sun Suoping Li Yun Zhou Chun-Peng Song 2022Journal of Genetics and Genomics2022,49,3:0
14Interactions between ShPP2-1, an F-box family gene, and ACR11A regulate cold toleranee of tomato显示文摘There is a critical need to identify germplasm resources and genes that promote cold tolerance of tomato because global tomato production is threatened by cold stress.We found that the expression of an F-box gene family member named ShPP2-1 from Solanum habrochaites is cold inducible and studied its contribution to cold tolerance.Overexpression of ShPP2-1 in cultivated tomato(AC)reduced cold tolerance by intensifying damage to cell membranes.To explore the underlying molecular mechanism,we conducted a yeast two-hybrid library screen and found that a protein containing ACT domain repeats named ACR11A interacts with PP2-1.Overexpression of SIACR11A in AC enhanced the cold tolerance of seedlings and germinating seeds.Cold tolerance decreased in tomato plants that overexpressed both of these genes.Additionally,we performed seed germination experiments in the cold with 177 tomato accessions and identified two alleles of SlACR11A that differ in one single-nucleotide polymorphism.We found that one of these alleles,SlACR11A G,is significantly enriched in cold-tolerant tomato plants.Taken together,our fi ndings indicate that the combination of low expression levels of PP2-1 and high expression levels of ACR11A can promote cold tolerance.These genes may therefore serve as direct targets for both genetic engineering and improvement projects that aim to enhance the cold tolerance of tomato.Jianwen Song Lele Shang Shiwei Chen Yongen Lu Yuyang Zhang Bo Ouyang Zhibiao Ye Junhong Zhang 2021Horticulture Research2021,8,1:0
15Rare earth metals modified Ni-S2O8^2-/ZrO2-Al2O3 catalysts for n-pentane isomerization显示文摘The effects of adding rare earth(RE) metals,such as Ce,Yb and Pr to Ni-S_2O_8^(2-)/ZrO_2-Al_2O_3(Ni-SZA) on the structure of catalysts as well as their isomerization performance were studied.The prepared catalysts were characterized by XRD,BET,FT-IR,Py-IR,and H_2-TPR,The results showed that the addition of RE metals can increase the strength and amounts of the acid sites,improve the redox properties of catalysts.The Yb-Ni-SZA catalyst showed the best redox properties,which could provide enough metallic sites.In addition,it provided the largest amounts of weak and moderately strong acid sites.Among RE metals modified Ni-SZA catalyst,Yb-Ni-SZA exhibited the highest isopentane yield of 61.7%at 160 °C.The optimum isomerization catalytic performance of the catalysts decreased in the order of Yb-Ni-SZA > Pr-Ni-SZA > Ni-SZA > Ce-Ni-SZA.Hua Song Lele Zhao Na Wang 2017Chinese Journal of Chemical Engineering2017,25,1:0
16VG,encoding a thylakoid formation protein,regulates the formation of variegated leaves in tomato显示文摘Leaf-color mutations have been studied extensively in plants.However,to better understand the complex mechanisms underlying the formation of leaf color,it is essential to continue discover novel genes involved in the process of leaf color development.In this study,we identified a variegated-leaf(vg)mutant in tomato that exhibited defective phenotypes in thylakoids and photosynthesis.To clone the vg locus,an F2population was constructed from the cross between the vg mutant(Solanum lycopersicum)and the wild tomato LA1589(S.pimpinellifolium).Using the map-based cloning approach,the vg locus was mapped on chromosome 7 and narrowed down to a 128 kb region that contained 21 open reading frames(ORFs).The expression levels of ORF9,ORF10,and ORF13 were significantly lower in vg than in the wild-type plants,while the ORF11 transcript level was elevated in vg.We then mutagenized ORF9,ORF10,and ORF13 by the CRISPR/Cas9 system in the wild-type tomato background and found that only the ORF10 mutation reproduced the phenotype of variegated leaves,indicating that ORF10 represents VG and its down-regulated expression was responsible for the variegated leaf phenotype.ORF10 encodes a thylakoid formation protein and its mutant lines showed reduced levels of chlorophyll synthesis and photosynthesis.Taken together,these results suggest that VG is necessary for chloroplast development,chlorophyll synthesis,and photosynthesis in tomato.Jianwen Song Lijie Guo Lele Shang Wenqian Wang Chuying Yu Zhibiao Ye Junhong Zhang 2023Horticultural Plant Journal2023,9,1:0
17Introgression of QTL from Aegilops tauschii enhances yield-related traits in common wheat显示文摘To break the narrow diversity bottleneck of the wheat D genome, a set of Aegilops tauschii-wheat introgression(A-WI) lines was developed by crossing Ae. tauschii accession T015 with common wheat elite cultivar Zhoumai 18(Zhou18). A high-density genetic map was constructed based on Single Nucleotide Polymorphism(SNP) markers and 15 yield-related traits were evaluated in 11 environments for detecting quantitative trait loci(QTL). A total of 27 environmentally stable QTL were identified in at least five environments, 20 of which were derived from Ae. tauschii T015, explaining up to 24.27% of the phenotypic variations. The major QTL for kernel length(KL), QKl-2D.5, was delimited to a physical interval of approximately 2.6 Mb harboring 52 candidate genes. Three Kompetitive Allele Specific PCR(KASP)markers were successfully developed based on nonsynonymous nucleotide mutations of candidate gene AetT093_2Dv1G100900.1 and showed that A-WI lines with the T015 haplotype had significantly longer KL than the Zhou18 haplotype across all 11 environments. Four primary valuable A-WIs with good trait performance and carrying yield-related QTL were selected for breeding improvement.
