| 1 | Human umbilical cord Wharton's Jelly-derived mesenchymal stem cells differentiation into nerve-like cells显示文摘Background The two most basic properties of mesenchymal stem cells (MSCs) are the capacities to self-renew indefinitely and differentiate into multiple cells and tissue types. The cells from human umbilical cord Wharton’s Jelly have properties of MSCs and represent a rich source of primitive cells. This study was conducted to explore the possibility of inducing human umbilical cord Wharton’s Jelly-derived MSCs to differentiate into nerve-like cells.Methods MSCs were cultured from the Wharton’s Jelly taken from human umbilical cord of babies delivered after full-term normal labor. Salvia miltiorrhiza and β-mercaptoethanol were used to induce the human umbilical cord-derived MSCs to differentiate. The expression of neural protein markers was shown by immunocytochemistry. The induction process was monitored by phase contrast microscopy, electron microscopy (EM), and laser scanning confocal microscopy (LSCM) .The pleiotrophin and nestin genes were measured by reverse transcription-polymerase chain reaction (RT-PCR). Results MSCs in the Wharton’s Jelly were easily attainable and could be maintained and expanded in culture. They were positive for markers of MSCs, but negative for markers of hematopoietic cells and graft-versus-host disease (GVHD)-related cells. Treatment with Salvia miltiorrhiza caused Wharton’s Jelly cells to undergo profound morphological changes. The induced MSCs developed rounded cell bodies with multiple neurite-like extensions. Eventually they developed processes that formed networks reminiscent of primary cultures of neurons. Salvia miltiorrhiza and β-mercaptoethanol also induced MSCs to express nestin, β-tubulinⅢ, neurofilament (NF) and glial fibrillary acidic protein (GFAP). It was confirmed by RT-PCR that MSCs could express pleiotrophin both before and after induction by Salvia miltiorrhiza. The expression was markedly enhanced after induction and the nestin gene was also expressed.Conclusions MSCs could be isolated from human umbilical cord Wharton’s Jelly. They were capable of differentiating into nerve-like cells using Salvia miltiorrhiza or β-mercaptoethanol. The induced MSCs not only underwent morphologic changes, but also expressed the neuron-related genes and neuronal cell markers. | 3 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 4 | Geologic investigation and mapping of the Sinus Iridum quadrangle from Clementine, SELENE, and Chang’e-1 data显示文摘The objectives of lunar satellite remote sensing are to study lunar surface characteristics, inner structure, and its evolution history. The contents of TiO 2 and FeO are assessed from Clementine UV/VIS data for Sinus Iridum. The geologic stratigraphic units and crates are interpreted visually based on SELENE Terrain Camera (TC) images and the spatial resolution of which is up to 10 m. And the geologic ages of different stratigraphic units are calculated by the crater size-frequency distributions measurements. The gravity anomaly is generated from SELENE gravity model (SGM90d) to show its difference from Mare Imbrium. Furthermore, the thickness of lunar regolith is also derived from microwave radiometer data of Chang’e-1 satellite. Integrating these results, it shows that the Sinus Iridum is different from the Mare Imbrium in inner structure and surface sedimentation. And its history of subsidence, deposition, volcanism, and impact is described. | 5 | Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy. | 8 | In vitro degradation of pure magnesium-the synergetic influences of glucose and albumin显示文摘The biocorrosion of magnesium in the external physiological environment is still difficult to accurately evaluate the degradation behavior in vivo,particularly,in the microenvironment of the patients with hyperglycemia or diabetes.Thus,we explored the synergistic effects of glucose and protein on the biodegradation of pure magnesium,so as to have a deeper understanding the mechanism of the degradation in vivo.The surface morphology and corrosion product composition of pure magnesium were investigated using SEM,EDS,FTIR,XRD and XPS.The effect of glucose and albumin on the degradation rate of pure magnesium was investigated via electrochemical and immersion tests.The adsorption of glucose and albumin on the sample surface was observed using fluorescence microscopy.The results showed that the presence of 2 g/L glucose changed the micromorphology of corrosion products on the magnesium surface by reacting with metal cations,thus inhibiting the corrosion of pure magnesium.Protein formed a barrier layer to protect the magnesium at early stage of immersion.The chelation reaction between protein and magnesium surface might accelerate the degradation at later stage.There may be a critical glucose(albumin)content.Biodegradation of pure magnesium was inhibited at low concentrations and promoted at high concentrations.The synergistic effect of glucose and protein restrained the adsorption of aggressive chloride ions to a certain extent,and thus inhibited the degradation of pure magnesium considerably.Moreover,XPS results indicated that glucose promoted the adsorption of protein on the sample surface. | Wei Yan Yi-Jie Lian Zhi-Yuan Zhang Mei-Qi Zeng Zhao-Qi Zhang Zheng-Zheng Yin Lan-Yue Cui Rong-Chang Zeng | 2020 | Bioactive Materials2020,5,2: | 6 |
| 9 | DropConnect Regularization Method with Sparsity Constraint for Neural Networks显示文摘Drop Connect is a recently introduced algorithm to prevent the co-adaptation of feature detectors.Compared to Dropout, Drop Connect gains state-of-the-art results on several image recognition benchmarks. Motivated by the success of Drop Connect, we extended this algorithm with the ability of sparse feature selection. In Drop Connect algorithm, the dropping masks of weights are generated using Bernoulli gating variables that are independent of the weights and activations. We introduce a new strategy to generate masks depending on the outputs of previous layer. Using this method, neurons which are promising to produce sparser features will be assigned a bigger possibility to keep active in the forward and backward propagations. We then evaluate such sparsity constrained Drop Connect on MNIST and CIFAR datasets in comparison with ordinary Drop Connect and Dropout method. | 10 | Oddi sphincter preserved cholangioplasty with hepaticosubcutaneous stoma for hepatolithiasis显示文摘AIM: To evaluate the long-term outcomes of Oddi sphincter preserved cholangioplasty with hepaticosubcutaneous stoma(OSPCHS) and risk factors for recurrence in hepatolithiasis.METHODS: From March 1993 to December 2012, 202 consecutive patients with hepatolithiasis underwent OSPCHS at our department. The Oddi sphincter preserved procedure consisted of common hepatic duct exploration, stone extraction, hilar bile duct plasty, establishment of subcutaneous stoma to the bile duct. Patients with recurrent stones can undergo stone extraction and/or biliary drainage via the subcutaneous stoma which can be incised under local anesthesia. The long-term results were reviewed. Cox regression model was employed to analyze the risk factors for stone recurrence.RESULTS: Ninety-seven(48.0%) OSPCHS patients underwent hepatic resection concomitantly. The rate of surgical complications was 10.4%. There was no perioperative death. The immediate stone clearance rate was 72.8%. Postoperative cholangioscopic lithotomy raised the clearance rate to 97.0%. With a median follow-up period of 78.5 mo(range: 2-233 mo), 24.8% of patients had recurrent stones, 2.5% had late development of cholangiocarcinoma, and the mortality rate was 5.4%. Removal of recurrent stones and/or drainage of inflammatory bile via subcutaneous stoma were conducted in 44(21.8%) patients. The earthquake occurred no more than half a year after the establishment of the National Strong Motion Observation Network System (NSMONS) of China; what is more, the epicenter was located in the area with dense strong motion observation stations so that a large number of strong motion records of the main shock were obtained. In this paper, 501 strong motion records from 167 observation stations are utilized to establish the ground motion attenuation relations in three directions in the range of fault distance less than 600 km. The result shows the difference of seismic motion attenuation in two horizontal directions is insignificant. | 12 | Therapeutic effects of four strains of probiotics on experimental colitis in mice显示文摘AIM:To investigate the therapeutic effects of four strains of probiotics(E.feacalis,L.acidophilus,C.butyricum and B.adolescentis) on dextran sulphate sodium(DSS)-induced experimental colitis in Balb/c mice.METHODS:Eighty Balb/c mice were randomly divided into 8 groups.Weight-loss,fecal character,fecal occult blood and hematochezia were recorded daily.Disease activity index(DAI) scores were also evaluated everyday.Length of colon was measured and histological scores were evaluated on the 13th day.Myeloperoxidase(MPO) activity was detected.Interleukin-1(IL-1) and IL-4 expression was detected by ELISA and RT-PCR.RESULTS:The four strains of probiotics relieved the inflammatory condition of DSS-induced experimental colitis in mice.Weight loss was slowed down in all probiotics-treated mice.Even weight gain was observed by the end of probiotics treatment.The DAI and histological scores of probiotics-treated mice were lower than those of mice in the control group(1.9 ± 0.2 vs 8.6 ± 0.4,P < 0.05 for E.faecalis).The length of colon of probiotics-treated mice was longer than that of mice in the control group(10.3 ± 0.34 vs 8.65 ± 0.77,P < 0.05 for E.faecalis).The four strains of probiotics decreased the MP activity and the IL-1 expression,but increased the IL-4 expression.E.faecalis had a better effect on DSS-induced experimental colitis in mice than the other three strains.CONCLUSION:The four strains of probiotics have beneficial effects on experimental colitis in mice.E.faecalis has a better effect on DSS-induced experimental colitis in mice than the other three strains.Supplement of probiotics provides a new therapy for UC. | Lin-Lin Chen Xue-Hong Wang Yi Cui Guang-Hui Lian Jie Zhang Chun-Hui Ouyang Fang-Gen Lu | 2009 | World Journal of Gastroenterology2009,15,3: | 5 |
| 13 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 17 | Gentiopicroside ameliorates LKB1/AMPK-dependent alcoholic hepatosteatosis via P2x7R-NLRP3 inflammasome显示文摘OBJECTIVE Regulating P2x7R-NLRP3 inflammasome activation might be a potential therapeutic strategy to treat alcoholic hepatosteatosis.We investigated whether this process would be modulated by gentiopicroside(GPS),which is attributed to the bitterness of gentian root extract.METHODS An in vivo model was established by intragastrically treating mice with ethanol,and an in vitro model was created by treating HepG2 cells with ethanol or treating RAW 264.7 macrophages and murine bone marrow-derived macrophages(BMDMs) with lipopolysaccharides(LPS) plus adenosine triphos.phate(ATP).RESULTS In alcoholic hepatosteatotic mice model,GPS decreased serum aminotrans.ferase and triglyceride accumulation.GPS regulated sterol regulatory element-binding protein-1(Srebp1),peroxisome proliferators-actived receptors α(PPARα) and acetyl CoA carboxylase(ACC) expression via elevating liver kinase B1(LKB1)/AMP-activated Kinase(AMPK).Suppression of nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3),caspase-1 and expression by GPS resulted in the inhibition of interleukin-1β(IL-1β) production.In ethanol-exposed HepG2 cells,GPS reduced lipo.genesis and promoted lipid oxidation via P2x7R-NLRP3 inflammasome activation.P2x7R silencing enhanced AMPK activity,and reduced Srebp1 expression in ethanol-treated hepatocytes.GPS down.regulated P2x7R-mediated inflammatory response to extracellular ATP in LPS-primed RAW 264.7 macro.phages and BMDMs.Additionally,P2x7R deficiency attenuated IL-1β cleavage in RAW 264.7 macro.phages,and GPS further suppressed IL-1β cleavage.CONCLUSION Activation of LKB1/AMPK signaling by GPS might be mediated by P2x7R-NLRP3 inflammasome,suggesting a therapeutic utility of P2x7R blockade in alcoholic hepatosteatosis treatment. | Xia LI Yu ZHANG Kai-li XIA Min JIANG Ben-wen CUI Yan-ling WU Ji-xing NAN Li-hua LIAN | 2018 | 中国药理学与毒理学杂志2018,32,4: | 3 |
| 18 | Porous-hollow nanorods constructed from alternate intercalation of carbon and MoS2 monolayers for lithium and sodium storage显示文摘Weak ion diffusion and electron transport due to limited interlayer spacing and poor electrical conductivity have been identified as critical roadbacks for fast and abundant energy storage of both MoS2-based lithium ion batteries (LIBs) and sodium ion batteries (SIBs). In this work, MoS2 porous-hollow nanorods (MoS2/m-C800) have been designed and synthesized via an annealing-followed chemistry-intercalated strategy to solve the two issues. They are uniformly assembled from ultrathin MoS2 nanosheets, deviated to the rod-axis direction, with expanded interlayer spacing due to alternate intercalation of N-doped carbon monolayers between the adjacent MoS2 monolayers. Electrochemical studies of the MoS2/m-C800 sample, as an anode of LIBs, demonstrate that the superstructure can deliver a reversible discharge capacity of 1,170 mAh·g^-1 after 100 cycles at 0.2 A·g^-1 and maintain a reversible capacity of 951 mAh·g^-1 at 1.25 A·g^-1 after 350 cycles.
