6Genetically Modified Pig Models for Human Diseases显示文摘Genetically modified animal models are important for understanding the pathogenesis of human disease and developing therapeutic strategies.Although genetically modified mice have been widely used to model human diseases,some of these mouse models do not replicate important disease symptoms or pathology.Pigs are more similar to humans than mice in anatomy,physiology,and genome. 7Toward an improved data stewardship and service for environmental and ecological science data in West China显示文摘Sharing of scientific data can help scientific research to flourish and facilitate more widespread use of scientific data for the benefit of society.The Environmental and Ecological Science Data Center for West China(WestDC),sponsored by the National Natural Science Foundation of China(NSFC),aims to collect,manage,integrate,and disseminate environmental and ecological data from western China.It also aims to provide a long-term data service for multidisciplinary research within NSFC’s‘‘Environment and Ecology of West China Research Plan’’(NSFC West Plan).An integrated platform has been developed by the WestDC,and this has the function of data sharing,acting as a knowledge repository.Major data sets developed by the WestDC include basic geographic data,the regionalization of global data set for China,scientific data for cold and arid regions in China,scientific data for the cryosphere in countries that neighbor China,data relating to the inland river basins in northwestern China,and data submitted by the NSFC West Plan projects.In compliance with the‘‘full and open’’data sharing policy,most data in the WestDC can be accessed online.Highlights include detailed data documentation,the integration of data with bibliographic knowledge,data publishing,and data reference.Xin Li Zhuotong Nan Guodong Cheng Yongjian Ding Lizong Wu Liangxu Wang Jian Wang Youhua Ran Hongxing Li Xiaoduo Pan Zhongming Zhu 2011International Journal of Digital Earth2011,4,4:11

10Comprehensive Marine Observing Experiment Based on High-Altitude Large Unmanned Aerial Vehicle(South China Sea Experiment2020 of the “Petrel Project”)显示文摘In collaboration with 12 other institutions, the Meteorological Observation Center of the China Meteorological Administration undertook a comprehensive marine observation experiment in the South China Sea using the Yilong-10 high-altitude large unmanned aerial vehicle(UAV). The Yilong-10 UAV carried a self-developed dropsonde system and a millimeter-wave cloud radar system. In addition, a solar-powered unmanned surface vessel and two drifting buoys were used. The experiment was further supported by an intelligent, reciprocating horizontal drifting radiosonde system that was deployed from the Sansha Meteorological Observing Station, with the intent of producing a stereoscopic observation over the South China Sea. Comprehensive three-dimensional observations were collected using the system from 31 July to2 August, 2020. This information was used to investigate the formation and development processes of Typhoon Sinlaku(2020). The data contain measurements of 21 oceanic and meteorological parameters acquired by the five devices, along with video footage from the UAV. The data proved very helpful in determining the actual location and intensity of Typhoon Sinlaku(2020). The experiment demonstrates the feasibility of using a high-altitude, large UAV to fill in the gaps between operational meteorological observations of marine areas and typhoons near China, and marks a milestone for the use of such data for analyzing the structure and impact of a typhoon in the South China Sea. 11Multi-satellite observations on the storm-time enhancements of energetic outer zone electron fluxes driven by chorus waves显示文摘The evolution of energetic outer zone electron fluxes during the strong magnetic storm on September 28, 2002 is investigated based on the observations of SAMPEX and GOES-10 satellites. The observations of both satellites showed that energetic electron fluxes increased significantly during the storm recovery phase, and reached the maximum on October 6. The 1.5–14 MeV and 2.5–14 MeV electron fluxes observed by SAMPEX peaked around L=3.5 with values of 6×10 2 cm -2 s -1 sr -1 keV -1 and 5×10 3 cm -2 s -1 sr -1 keV -1 , which were about 10 and 8 times the pre-storm values. At the geostationary orbit, the >0.6 MeV and >2 MeV electron fluxes observed by GOES-10 showed enhancement up to 50 and 30 times. The plasma parameters and whistler-mode chorus waves in the outer radiation belt are also analyzed based on the data from Cluster C3 satellite. Cluster C3 satellite went through the outer radiation belt twice from 1 October to 4 October, and observed whistler-mode chorus waves with high intensity (10 -5 –10 -4 nT 2 Hz -1 ). Numerical calculations indicated that the observed chorus waves were in gyro-resonance with the radiation belt electrons. 12Climatology of Cloud-base Height from Long-term Radiosonde Measurements in China显示文摘Clouds are critical to the global radiation budget and hydrological cycle, but knowledge is still poor concerning the observed climatology of cloud-base height(CBH) in China. Based on fine-resolution sounding observations from the China Radiosonde Network(CRN), the method used to estimate CBH was modified, and uncertainty analyses indicated that the CBH is good enough. The accuracy of CBH estimation is verified by the comparison between the sounding-derived CBHs and those estimated from the micro-pulse lidar and millimeter-wave cloud radar. As such, the CBH climatology was compiled for the period 2006–16. Overall, the CBH exhibits large geographic variability across China, at both 0800 Local Standard Time(LST) and 2000 LST, irrespective of season. In addition, the summertime cloud base tends to be elevated to higher altitudes in dry regions [i.e., Inner Mongolia and the North China Plain(NCP)]. By comparison, the Tibetan Plateau(TP),Pearl River Delta(PRD) and Sichuan Basin(SCB) have relatively low CBHs(< 2.4 km above ground level). In terms of seasonality, the CBH reaches its maximum in summer and minimum in winter. A low cloud base tends to occur frequently(> 70%) over the TP, PRD and SCB. In contrast, at most sites over the Yangtze River Delta(YRD) and the NCP, about half the cloud belongs to the high-cloud category. The CBH does not exhibit marked diurnal variation in summer, throughout all CRN sites, probably due to the persistent cloud coverage caused by the East Asia Summer Monsson. 13Generation of rat blood vasculature and hematopoietic cells in rat-mouse chimeras by blastocyst complementation显示文摘Interspecies chimera through blastocyst complementation could be an alternative approach to create human organs in animals by using human pluripotent stem cells.A mismatch of the major histocompatibility complex of vascular endothelial cells between the human and host animal will cause graft rejection in the transplanted organs.Therefore,to achieve a transplantable organ in animals without rejection,creation of vascular endothelial cells derived from humans within the organ is necessary.In this study,to explore whether donor xeno-pluripotent stem cells can compensate for blood vasculature in host animals,we generated rat-mouse chimeras by injection of rat embryonic stem cells(rESCs)into mouse blastocysts with deficiency of Flk-1 protein,which is associated with endothelial and hematopoietic cell development.We found that rESCs could differentiate into vascular endothelial and hematopoietic cells in the rat-mouse chimeras.The whole yolk sac(YS)of Flk-1^EGFP/ECFP rat-mouse chimera was full of rat blood vasculature.Rat genes related to vascular endothelial cells,arteries,and veins,blood vessels formation process,as well as hematopoietic cells,were highly expressed in the YS.Our results suggested that rat vascular endothelial cells could undergo proliferation,migration,and self-assembly to form blood vasculature and that hematopoietic cells could differentiate into B cells,T cells,and myeloid cells in rat-mouse chimeras,which was able to rescue early embryonic lethality caused by Flk-1 deficiency in mouse.Xiaomin Wang Hui Shi Juanjuan Zhou Qingjian Zou Quanjun Zhang Shixue Gou Pengfei Chen Lisha Mou Nana Fan Yangyang Suo Zhen Ouyang Chengdan Lai Quanmei Yan Liangxue Lai 2020Journal of Genetics and Genomics2020,47,5:2

15Production of transgenic pigs over-expressing theantiviral gene Mx1显示文摘The myxovirus resistance gene (Mx1) has a broad spectrum of antiviral activities. It is therefore an interestingcandidate gene to improve disease resistance in farm animals. In this study, we report the use of somatic cellnuclear transfer (SCNT) to produce transgenic pigs over-expressing the Mx1 gene. These transgenic pigs expressapproximately 15–25 times more Mx1 mRNA than non-transgenic pigs, and the protein level of Mx1 was alsomarkedly enhanced. We challenged fibroblast cells isolated from the ear skin of transgenic and control pigs withinfluenza A virus and classical swine fever virus (CFSV). Indirect immunofluorescence assay (IFA) revealed a profounddecrease of influenza A proliferation in Mx1 transgenic cells. Growth kinetics showed an approximately 10-foldreduction of viral copies in the transgenic cells compared to non-transgenic controls. Additionally, we found thatthe Mx1 transgenic cells were more resistant to CSFV infection in comparison to non-transgenic cells. 16Efficient and high-fidelity base editor with expanded PAM compatibility for cytidine dinucleotide显示文摘Cytidine base editor(CBE),which is composed of a cytidine deaminase fused to Cas9 nickase,has been widely used to induce C-to-T conversions in a wide range of organisms.However,the targeting scope of current CBEs is largely restricted to protospacer adjacent motif(PAM)sequences containing G,T,or A bases.In this study,we developed a new base editor termed“nNme2-CBE”with excellent PAM compatibility for cytidine dinucleotide,significantly expanding the genome-targeting scope of CBEs.Using nNme2-CBE,targeted editing efficiencies of 29.0%-55.0%and 17.3%—52.5%were generated in human cells and rabbit embryos,respectively.In contrast to conventional nSp-CBE,the nNme2-CBE is a natural high-fidelity base editing platform with minimal DNA off-targeting detected in vivo.Significantly increased efficiency in GC context and precision were determined by combining nNme2Cas9 with rationally engineered cytidine deaminases.In addition,the Founder rabbits with accurate single-base substitutions at Fgf5 gene loci were successfully generated by using the nNme2-CBE system.These novel nNme2-CBEs with expanded PAM compatibility and high fidelity will expand the base editing toolset for efficient gene modification and therapeutic applications.Zhiquan Liu Siyu Chen Yingqi Jia Huanhuan Shan Mao Chen Yuning Song Liangxue Lai Zhanjun Li 2021Science China(Life Sciences)2021,64,8:2
17Fully Organic Self-Powered Electronic Skin with Multifunctional and Highly Robust Sensing Capability显示文摘Electronic skin(e-skin)with skin-like flexibility and tactile sensation will promote the great advancements in the fields of wearable equipment.Thus,the multifunction and high robustness are two important requirements for sensing capability of the e-skin.Here,a fully organic self-powered e-skin(FOSE-skin)based on the triboelectric nanogenerator(TENG)is developed.FOSE-skin based on TENG can be fully self-healed within 10 hours after being sheared by employing the self-healing polymer as a triboelectric layer and ionic liquid with the temperature sensitivity as an electrode.FOSE-skin based on TENG has the multifunctional and highly robust sensing capability and can sense the pressure and temperature simultaneously.The sensing capability of the FOSE-skin based on TENG can be highly robust with no changes after self-healing.FOSE-skin based on TENG can be employed to detect the arm swing,the temperature change of flowing water,and the motion trajectory.This work provides a new idea for solving the issues of monofunctional and low robust sensing capability for FOSE-skin based on TENG,which can further promote the application of wearable electronics in soft robotics and bionic prosthetics.Lijuan Song Zheng Zhang Xiaochen Xun Liangxu Xu Fangfang Gao Xuan Zhao Zhuo Kang Qingliang Liao Yue Zhang 2021Research2021,,1:2
