| 1 | Effects of Sc, Zr and Tion the microstructure and properties of Al alloys with high Mg content显示文摘The effects of trace Sc, Zr, and Ti on the microstructure and hardness of Al alloys with high Mg content (Al-6Mg, Al-8Mg, and Al-10Mg) were studied by optical microscope, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and Brinell hardness. The grain size of the ascast alloys was refined by the addition of Sc and Zr, and it was further refined by the addition of Ti. With the same contents of Sc, Zr, and Ti, an increase in Mg content was beneficial to the refinement due to the solution of Mg into α-Al. The refined microstructures of the as-cast alloys were favorable for Brinell hardness. | 2 | Syntheses and characterization of anti-thrombotic and anti-oxidative Gastrodin-modified polyurethane for vascular tissue engineering显示文摘Vascular grafts must avoid negative inflammatory responses and thrombogenesis to prohibit fibrotic deposition immediately upon implantation and promote the regeneration of small diameter blood vessels(<6 mm inner diameter).Here,polyurethane(PU)elastomers incorporating anti-coagulative and anti-inflammatory Gastrodin were fabricated.The films had inter-connected pores with porosities equal to or greater than 86%and pore sizes ranging from 250 to 400μm.Incorporation of Gastrodin into PU films resulted in desirable mechanical properties,hydrophilicity,swelling ratios and degradation rates without collapse.The released Gastrodin maintained bioactivity over 21 days as assessed by its anti-oxidative capability.The Gastrodin/PU had better anti-coagulation response(less observable BSA,fibrinogen and platelet adhesion/activation and suppressed clotting in whole blood).Red blood cell compatibility,measured by hemolysis,was greatly improved with 2Gastrodin/PU compared to other Gastrodin/PU groups.Notably,Gastrodin/PU upregulated anti-oxidant factors Nrf2 and HO-1 expression in H2O2 treated HUVECs,correlated with decreasing pro-inflammatory cytokines TNF-αand IL-1β in RAW 264.7 cells.Upon implantation in a subcutaneous pocket,PU was encapsulated by an obvious fibrous capsule,concurrent with a large amount of inflammatory cell infiltration,while Gastrodin/PU induced a thinner fibrous capsule,especially 2Gastrodin/PU.Further,enhanced adhesion and proliferation of HUVECs seeded onto films in vitro demonstrated that 2Gastrodin/PU could help cell recruitment,as evidenced by rapid host cell infiltration and substantial blood vessel formation in vivo.These results indicate that 2Gastrodin/PU has the potential to facilitate blood vessel regeneration,thus providing new insight into the development of clinically effective vascular grafts. | Meng Zheng Jiazhi Guo Qing Li Jian Yang Yi Han Hongcai Yang Mali Yu Lianmei Zhong Di Lu Limei Li Lin Sun | 2021 | Bioactive Materials2021,6,2: | 7 |
| 3 | Synthesis of nitrogen-doped carbon nanotubes-FePO_4 composite from phosphate residue and its application as effective Fenton-like catalyst for dye degradation显示文摘Phosphate residue is regarded as a hazardous waste, which could potentially create significant environmental and health problems if it is not properly treated and disposed of. In this study, nitrogen-doped carbon nanotubes-FePO_4(NCNTs-FePO_4) composite was successfully synthesized from phosphate residue, and its application as an effective catalyst was explored. Firstly, an effective method was developed to recover FePO_4 from phosphate residue, achieving an impressive FePO_4 mass recovery rate of 98.14%. Then, the NCNTsFePO_4 catalyst was synthesized from the recovered FePO_4 by two main reactions, including surface modification and chemical vapor deposition. Finally, the synthesized NCNTs-FePO_4 was applied to photo-degrade 15 mg/L Rhodamine B(RhB) in a Fenton-like system. The results showed that 98.9% of RhB could be degraded in 60 min, closely following the pseudofirst-order kinetics model. It was found that even after six consecutive cycles, NCNTs-FePO_4 still retained a high catalytic capacity(>50%). Moreover, ·OH radicals participating in the RhB degradation process were evidenced using quenching experiments and electron paramagnetic resonance analysis, and a rational mechanism was proposed. | 4 | Inhibition of the lncRNA DANCR attenuates cardiomyocyte injury induced by oxygen-glucose deprivation via the miR-19a-3p/MAPK1 axis显示文摘Long noncoding RNAs(lncRNAs)have been considered as crucial regulators of acute myocardial infarction(AMI).In this study,to analyze the effect of differentiation antagonizing nonprotein coding RNA(DANCR)of lncRNA on cardiomyocyte damage in AMI,cardiomyocyte injury was induced by oxygen-glucose deprivation(OGD).Cell counting kit-8(CCK-8)assay and flow cytometry were used to assess cell viability and apoptosis,respectively.Quantitative real-time PCR was used to measure the expression levels of DANCR and miR-19a-3p.Bioinformatics analysis and luciferase gene reporter assay were utilized to explore the relationship among DANCR,miR-19a-3p,and mitogen-activated protein kinase 1(MAPK1).CCK-8 and TUNEL assays were used to explore the effects of DANCR alone or plus miR-19a-3p on the viability and apoptosis of OGD/R-exposed HL-1 cells.Western blot analysis was used to detect changes in the MAPK1/ERK1/2 pathway in HL-1 cells.We found that DANCR expression and miR-19a-3p level are negatively correlated as DANCR expression is increased,while miR-19a-3p level is decreased in AMI patients’serum and OGD/R-exposed HL-1 cells.DANCR knockdown increased miR-19a-3p level,and miR-19a-3p inhibition increased DANCR expression.Moreover,DANCR directly binds to miR-19a-3p.DANCR knockdown reduced viability but induced apoptosis in OGD/R-exposed HL-1 cells,while miR-19a-3p inhibition weakens these effects.Furthermore,MAPK1 is a target of miR-19a-3p.miR-19a-3p overexpression decreases MAPK1 and ERK1/2 in HL-1 cells,while miR-19a-3p inhibition increases MAPK1 and ERK1/2 in HL-1 cells.Moreover,DANCR knockdown reduces myocardium apoptosis in mice with the left anterior descending artery ligated.DANCR knockdown effectively restores myocardial cell apoptosis by regulating the miR-19a-3p/MAPK1/ERK1/2 axis. | Yang Ruan Hong Li Xiaojing Cao Shuai Meng Ruofei Jia Lianmei Pu Hao Fu Zening Jin | 2021 | Acta Biochimica et Biophysica Sinica2021,53,10: | 4 |
| 5 | Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors. | Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang | 2022 | Animal Models and Experimental Medicine2022,5,1: | 3 |
| 6 | Flotillin1 promotes EMT of human small cell lung cancer via TGF-β signaling pathway显示文摘Objective:Small cell lung carcinoma(SCLC)is considered one of the most aggressive types of lung cancer due to its rapid growth and early metastasis.No tumor markers or therapeutic targets have been demonstrated to be specific or effective in SCLC to date.This study aims to evaluate the potential of Flotillin1(Flot1)as a target of SCLC treatment.Methods:Flot1 expression level in the tissue of SCLC and other tissue of lung disease was detected using immunohistochemical staining.Transwell and Matrigel assays were employed to examine migration and invasion of cancer cells.Flow cytometry and xCELLigence system were used to evaluate cell apoptosis and cell viability,respectively.Expression levels of Flot1,epithelialmesenchymal transition(EMT)marker E-cadherin,vimentin,cyclinD1,TGF-β-Smad2/3,and p-AKT were examined using Western blot.Furthermore,xenograft tumor in nude mice was used to evaluate the growth and metastasis of NCI-H446 cells in vivo.Results:Our results demonstrated that Flot1 is highly expressed in SCLC samples and that its expression correlates strongly with clinical stage,distant metastasis,and poor survival.The knockdown of Flot1 decreased the growth,migration,and invasiveness of SCLC cells and reversed EMT phenotype in vitro and in vivo,while enhanced Flot1 expression exhibited the opposite behavior.Gene expression profile analysis demonstrated that Flot1-regulated genes frequently mapped to the AKT and TGF-β-Smad2/3pathways.Our results further revealed that Flot1 affected the progression of SCLC via regulation of EMT progression.Conclusions:These findings indicated an oncogenic role of Flot1 via promoting EMT in SCLC and suggested its potential as a tumor marker and prognostic indicator. | Lianmei Zhao Jie Li Yueping Liu Wei Zhou Yanan Shan Xinyi Fan Xinliang Zhou Baoen Shan Yongmei Song Qimin Zhan | 2018 | Cancer Biology & Medicine2018,15,4: | 3 |
| 7 | Potential chronic liver toxicity in rats orally administered an ethanol extract of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE: To investigate the potential chronic liver toxicity of oral administration of ethanol extract of Huangqin(Radix Scutellariae Baicalensis)(SBE) in Wistar rats.METHODS: SBE was administered to rats by gavage for 26 weeks, at doses of 300, 1250, or 2500 mg·kg-1·d-1 respectively. The rats were euthanized at the end of 13 and 26 weeks daily oral dosing and following 4 weeks of recovery time. The changes of hematology, urinary, blood biochemistry and histomorphology were examined at each time point and focus on liver function and histological changes.RESULTS: When SBE at a dose of up to 2500 mgkg-1 d-1 was fed to male and female rats for 2··6 weeks, the liver tissue showed some inflammatory change that predominated by leukocyte infiltrationbut returned to normal after withdrawal. In addition, high-dose SBE treatment of 26 weeks in rats,glucose, electrolyte and lipid levels also have some changes. In addition, there are no other functional or organic lesions related to SBE treatment.CONCLUSIONS: Long-term and high-dose SBE may cause liver damage, however, the structural damage of the liver can be restored after the ethanol extract stopping. | 14 | Gastrodin modified polyurethane conduit promotes nerve repair via optimizing Schwann cells function显示文摘Peripheral nerve regeneration and functional recovery remain a major clinical challenge.Nerve guidance conduit(NGC)that can regulate biological behavior of Schwann cells(SCs)and facilitate axonal regeneration through microenvironmental remodeling is beneficial for nerve regeneration and functional recovery.Gastrodin,a main constituent of a Chinese traditional herbal medicine,has been known to display several biological and pharmacological properties,especially antioxidative,anti-inflammatory and nerve regeneration.Herein,polyurethane(PU)NGCs modified by different weight ratio of Gastrodin(0,1 and 5 wt%)were designed for sequential and sustainable drug release,that created a favorable microenvironment for nerve regeneration.The scaffold showed suitable pore structure and biocompatibility in vitro,and evidently promoted morphological and functional recovery of regenerated sciatic nerves in vivo.Compared to the PU and 1% Gastrodin/PU scaffolds,the 5% Gastrodin/PU significantly enhanced the proliferation,migration and myelination of SCs and up-regulated expression of neurotrophic factors,as well as induction of the differentiation of PC12 cells.Interestingly,the obvious anti-inflammatory response was observed in 5% Gastrodin/PU by reduced expression of TNF-α and iNOS,which also evidenced by the few fibrous capsule formation in the subcutaneous implantation.Such a construct presented a similarity to autograft in vivo repairing a 10 mm sciatic nerve defects.It was able to not only boost the regenerated area of nerve and microvascular network,but also facilitate functional axons growth and remyelination,leading to highly improved functional restoration.These findings demonstrate that the 5% Gastrodin/PU NGC efficiently promotes nerve regeneration,indicating their potential for use in peripheral nerve regeneration applications. | Hongcai Yang Qing Li Limei Li Shaochun Chen Yu Zhao Yingrui Hu Lu wang Xiaoqian Lan Lianmei Zhong Di Lu | 2022 | Bioactive Materials2022,7,2: | 1 |
