| 1 | The mutational pattern of homologous recombination(HR)-associated genes and its relevance to the immunotherapeutic response in gastric cancer显示文摘Objective:Currently,there is an urgent need to identify immunotherapeutic biomarkers to increase the benefit of immune checkpoint inhibitors(ICIs)for patients with gastric cancer(GC).Homologous recombination deficiency(HRD)can modify the tumor immune microenvironment by increasing the presence of tumor-infiltrating lymphocytes and therefore might serve as a biomarker of immunotherapeutic response.We aimed to analyze the mutational pattern of HR-associated genes in Chinese patients with GC and its relevance to the tumor immune profile and clinical immunotherapeutic response.Methods:A panel of 543 cancer-associated genes was used to analyze genomic profiles in a cohort comprising 484 Chinese patients with GC.Correlations between HR gene mutations and tumor immunity or clinical outcomes were identified via bioinformatic analysis using 2 GC genomic datasets(TCGA and MSK-IMPACT).Results:Fifty-one of the 484(10.54%)patients carried at least one somatic mutation in an HR gene;ATM(16/484,3.31%)was among the most frequently mutated HR genes in the Chinese cohort.Mutations in HR genes were associated with elevated tumor mutational burden,enhanced immune activity,and microsatellite instability status.In the MSK-IMPACT cohort comprising 49 patients with stomach adenocarcinoma or gastroesophageal junction adenocarcinoma treated with ICIs,patients with HR-mut GC(n=12)had significantly better overall survival than those with HR-wt GC(n=37)(log-rank test,P<0.05).Conclusions:Our data suggest that detection of somatic mutations in HR genes might aid in identifying patients who might benefit from immune checkpoint blockade therapy. | Yue Fan Haifeng Ying Xueying Wu Huan Chen Ying Hu Henghui Zhang Lijia Wu Ying Yang Beibei Mao Lan Zheng | 2020 | Cancer Biology & Medicine2020,17,4: | 6 |
| 2 | Filamin B:The next hotspot in skeletal research?显示文摘Filamin B(FLNB)is a large dimeric actin-binding protein which crosslinks actin cytoskeleton filaments into a dynamic structure.Up to present,pathogenic mutations in FLNB are solely found to cause skeleta deformities,indicating the important role of FLNB in skeletal development.FLNB-related disorders are classifiedasspondylocarpotarsalsynostosis(SCT),Larsensyndrome(LS),atelosteogenesis(AO)boomerang dysplasia(BD),and isolated congenital talipes equinovarus,presenting with scoliosis,shortlimbed dwarfism,clubfoot,joint dislocation and other unique skeletal abnormalities.Several mecha?nisms of FLNB mutations causing skeletal malformations have been proposed,including delay of ossification in long bone growth plate,reduction of bone mineral density(BMD),dysregulation of muscle differentiation,ossification of intervertebral disc(IVD),disturbance of proliferation,differentiation and apoptosis in chondrocytes,impairment of angiogenesis,and hypomotility of osteoblast,chondrocyte and fibroblast.Interventions on FLNB-related diseases require prenatal surveillance by sonography,gene testing in high-risk carriers,andproper orthosis or orthopedic surgeries to correct malformations including scoliosis,cervical spine instability,large joint dislocation,and clubfoot.Gene and cell therapies for FLNB-related diseases are also promising but require further studies. | Qiming xu Nan Wu Lijia Cui Zhihong Wu Guixing Qiu | 2017 | Journal of Genetics and Genomics2017,44,7: | 5 |

| 4 | Low cycle fatigue behavior of die cast Mg-Al-Mn-Ce magnesium alloy显示文摘Fatigue failure is a main failure mode for magnesium and other alloys. It is beneficial for fatigue design and fatigue life improvement to investigate the low cycle fatigue behavior of magnesium alloys. In order to investigate the low cycle fatigue behavior of die cast Mg-Al-Mn-Ce magnesium alloy, the strain controlled fatigue experiments were performed at room temperature and fatigue fracture surfaces of specimens were observed with scanning election microscopy for the alloys under die-cast and aged states. Cyclic stress response curves, strain amplitude versus reversals to failure curve, total strain amplitude versus fatigue life curves and cyclic stress-strain curves of Mg-Al-Mn-Ce alloys were analyzed. The results show that the Mg-Al-Mn-Ce alloys under die-cast(F) and aged(T5) states exhibit cyclic strain hardening under the applied total strain amplitudes, and aging treatment could greatly increase the cyclic stress amplitudes of die cast Mg-Al-Mn-Ce alloys. The relationships between the plastic strain amplitude, the elastic strain amplitude and reversals to failure of Mg-Al-Mn-Ce magnesium alloy under different treatment states could be described by Coffin-Manson and Basquin equations, respectively. | 10 | Reduced pigmentation and thyroid hormone disruption in zebrafish embryos caused by industrial sludge near Bohai Bay,China显示文摘In recent years,pollution caused by the discharge of industrial wastewater into Bohai Bay has posed a potential threat to the health of surrounding residents.Sludge was collected from the outlet of a factory that discharges effluent into Bohai Bay,and alcohol extracts of sludge(SE)were prepared.We confirmed by UPLC-MS/MS analysis that the SE contained PAHs,including fluorene,pyrene,and phenanthrene.Zebrafish embryos as animal models were exposed to 0.1,0.3,0.5,1 and 5 mg/mL SE from 2 to 4 h post-fertilization(hpf)until 120 hpf.The results showed that SE caused a concentration-dependent increase in mortality and a decrease in hatchability.We found that SE significantly reduced eye pigmentation and decreased the movement of embryos and larvae.In addition,SE decreased triiodothyronine(T3)content and down-regulated the mRNA expression of some thyroid hormone-related genes including TPO and Thrβ,and caused the up-regulation of Dio2 and Dio3 at 120 hpf.Exposure to three individual PAHs found in SE,namely fluorene,pyrene,and phenanthrene,caused morphological and transcriptional changes that were similar to those caused by SE exposure.These findings indicate that PAHs in SE can reduce the pigmentation of zebrafish,which may be related to the genetic changes associated with thyroid hormones,and that zebrafish eye pigmentation can be used as an indicator of PAHs exposure. | Wenjing Dong Xiaoyu Yin Chelimuge Qi Tingting Wei Lijia Wei Jingfeng Yang Jingli Mu Hiroki Teraoka Wu Dong | 2023 | Water Biology and Security2023,2,4: | 0 |
| 11 | Low-temperature superplasticity of ZK40 magnesium alloy processed using equal channel angular pressing显示文摘Microstructure evolution and superplastic behaviors of ZK40 magnesium alloy were investigated in the temperature range of 473~623 K. Transmission electron microscopy (TEM) was used to study the microstructure changes, twinning occurred significantly after being processed by equal channel angular pressing (ECAP) for one pass through the die, the mean grain size was 5.6μm. Finer grains can be obtained after further processing through ECAP, the average grain size of the alloy processed by ECAP for three passes was as low as 0.8 μm; this alloy exhibited low temperature superplasticity at 473~523 K, elongations obtained at the same initial strain rate of 1×10-3 s-1 were 260% at 473 K and 612% at 523 K, respectively. Corresponding values for the ZK40 alloy processed by ECAP for only one pass were 124% at 473 K and 212% at 523 K, respectively; poor superplastic behavior of this material was related to the long-range stresses associated with the non-equilibrium grain boundaries within the coarse grains. | 14 | Enhanced design of hourglass truss sandwich structures for compressive resistance显示文摘In this article,the design of the hourglass truss sandwich structure is improved by optimizing the number of layers to enhance the compressive strength of both the core and the face sheet and then its mechanical performance. The hourglass truss structures characterized by three different numbers of layers are manufactured using an interlocking and vacuum brazing method. The effect of the layer number of the hourglass core panels on their out-of-plane compression and in-plane compression performance is investigated,and the results from calculations and experiments are in reasonable agreement. The results show that as the layer number of the hourglass core increases,the out-of-plane compressive strengths show little change,but their energy absorption properties are effectively increased. The in-plane compressive failure mechanism maps are constructed,and the specimens are designed to examine the local elastic and inelastic buckling failure modes of the face sheets. The results suggest that as the number of layers of the hourglass core increases,its maximum in-plane compressive load increases. The maximum in-plane compressive loads of the two-layer hourglass truss panels are 57%–70% higher than those of the single-layer panels. It can also be concluded that the out-of-plane and in-plane compression mechanical properties of the multilayer hourglass truss outperform those of the pyramidal truss. | 16 | Metallogenic model of the Shuangjianzishan Ag-Pb-Zn district,Northeast China:Revealed from integrated geophysical investigation显示文摘The Shuangjianzishan deposit in Inner Mongolia is a typical Ag-Pb-Zn deposit of the southern Great Xing’an Range.Proven reserves of Ag,Pb,and Zn in this deposit have reached the scale of super-large deposits,with favorable metallogenic conditions,strong prospecting signs,and high metallogenic potential.This paper reports a study involving integrated geophysical methods,including controlled-source audio-frequency magnetotelluric,gravity,magnetic,and shallow-seismic-reflection methods,to determine the spatial distribution of ore-controlling structures and subsurface intrusive rock for a depth range of<2000 m in the Shuangjianzishan ore district.The objective of this study is to construct a metallogenic model of the ore district and provide a scientific basis for the exploration of similar deposits in the deep and surrounding regions.We used three-dimensional inversion for controlled-source audio-frequency magnetotelluric data based on the limited memory quasi-Newton algorithm,and three-dimensional physical-property inversion for the gravity and magnetic data to obtain information about the subsurface distribution of ore-controlling structures and intrusive rocks.Under seismic reflection results,regional geology,petrophysical properties,and borehole information,the geophysical investigation shows that the Dashizhai group,which contains the main ore-bearing strata in the ore district,is distributed within a depth range of<1239 m,and is thick in the Xinglongshan ore block and the eastern part of the Shuangjianzishan ore block.The mineralization is spatially associated with a fault system characterized by NE-,NW-,and N-trending faults.The magnetic and electrical models identify large,deep bodies of intrusive rock that are inferred to have been involved in mineralization,with local shallow emplacement of some intrusions.Combining the subsurface spatial distributions of ore-bearing strata,ore-controlling faults,and intrusive rock,we propose two different metallogenic models for the Shuangjianzishan ore district,which provide a scientific basis for further prospecting in the deep regions of the ore district and surrounding areas. | Yihao Wu Jiangtao Han Yunhe Liu Guoqing Ma Fuxing Han Yanchen Yang Lijia Liu Lei Guo Ye Guan Yinghui Zhang Zhenyu Guo Qian Mu | 2022 | Geoscience Frontiers2022,13,2: | 0 |
| 17 | Study the relationship between the expression of MRP and the radiosensitivity of primary lesions of nasopharyngeal carcinoma显示文摘Objective: To study the relationship between the expression of multidrug resistance-associated protein (MRP) gene in nasopharyngeal carcinoma (NPC) and the radiosensitivity of primary lesions of NPC. Methods: Total 51 cases of NPC were analyzed retrospectively, who were treated by radiotherapy between January 1, 1999 and December 31, 2000 in our department, including the cases of pure radiotherapy and chemotherapy after radiotherapy. Using immunohistochemiscal S-P method, the expression of MRP in NPC was detected, and the relationship with the radiosensitivity of primary lesions of NPC was analyzed. Results: The rate of positive expression was 68.63% (35/51) in NPC, and there were significant difference between T stages (P<0.05), but no significant difference with radiosensitivity of primary lesions of NPC (P>0.05). Conclusion: There is a higher expression of MRP in NPC, but it has no relationship with radiosensitivity of primary lesions of NPC.
