| 1 | DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR. | Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen | 2020 | Acta Biochimica et Biophysica Sinica2020,52,11: | 5 |

| 4 | Nickel oxide for inverted structure perovskite solar cells显示文摘The emergence of inverted perovskite solar cells(PSCs) has attached great attention derived from the potential in improving stability. Charge transporting layer, especially hole transporting layer is crucial for efficient inverted PSCs. Organic materials were used as hole transporting layer previously. Recently, more and more inorganic hole transporting materials have been deployed for further improving the device stability. Nickel oxide(NiOx) as p-type metal oxide, owning high charge mobility and intrinsic stability,has been widely adopted in inverted PSCs. High performance over 20% efficiency has been achieved on NiOx base inverted PSCs. Herein, we have summarized recent progresses and strategies on the NiOx based PSCs, including the synthesis or deposition methods of NiOx, doping and surface modification of NiOx for efficient and stable PSCs. | 8 | Structure-guided manipulation of the regioselectivity of the cyclosporine A hydroxylase CYP-sb21 from Sebekia benihana显示文摘The cytochrome P450 enzyme CYP-sb21 from the rare actinomycete Sebekia benihana is capable of hydroxylating the immunosuppressive drug molecule cyclosporine A(CsA)primarily at the 4th N-methyl leucine(MeLeu4),giving rise toγ-hydroxy-N-methyl-L-Leu4-CsA(CsA-4-OH).This oxidative modification of CsA leads to dramatically reduced immunosuppressive activity while retaining the hair growth-promoting side-effect,thus demonstrating great application potential in both pharmaceutical and cosmetic industries.However,this P450 enzyme also hydroxylates CsA at the unwanted position of the 9th N-methyl leucine(MeLeu9),indicating that the regioselectivity needs to be improved for the development of CsA-4-OH into a commercial hair growth stimulator.Herein,we report the crystal structure of CYP-sb21 in its substrate-free form at 1.85Å.Together with sequence and 3D structure comparisons,Autodock-based substrate docking,molecular dynamics(MD)simulation,and site-directed mutagenesis,we identified a number of key residues including R294,E264,and M179 that can improve catalytic efficiency or change the regioselectivity of CYP-sb21 towards CsA,setting the stage for better enzymatic preparation of CsA-4-OH.This study also provides new insights into the substrate recognition and binding mechanism of P450 enzymes that accommodate bulky substrates. | Fengwei Li Li Ma Xingwang Zhang Jingfei Chen Feifei Qi Yinyue Huang Zepeng Qu Lishan Yao Wei Zhang Eung-Soo Kim Shengying Li | 2020 | Synthetic and Systems Biotechnology2020,5,3: | 1 |

| 17 | Pyridoxal 5'-phosphate alleviates prenatal pyridaben exposureinduced anxiety-like behaviors in offspring显示文摘Pyridaben(PY)is a widely used organochlorine acaricide,which can be detected in the peripheral blood of pregnant women.Available evidence suggests that PY has reproductive toxicity.However,it remains uncertain whether prenatal PY exposure impacts neurobehavioral development in offspring.Here,we administered PY to pregnant mice at a dose of 0.5 and 5 mg kg^(-1)day^(-1)via gavage and observed anxietylike behaviors in PY offspring aged five weeks.We then integrated the metabolome and transcriptome of the offspring's brain to explore the underlying mechanism.Metabolome data indicated that the vitamin B6 metabolism pathway was significantly affected,and the pyridoxal 50-phosphate(PLP)concentration and the active form of vitamin B6 was significantly reduced.Moreover,the transcriptome data showed that both PLP generation-related Pdxk and anxiety-related Gad1 were significantly down-regulated.Meanwhile,there was a decreasing trend in the concentration of GABA in the hippocampal DG region.Next,we supplemented PLP at a dose of 20 mg kg^(-1)day^(-1)to the PY offspring via intraperitoneal injection at three weeks.We found up-regulated expression of Pdxk and Gad1 and restored anxiety-like behaviors.This study suggests that prenatal exposure to PY can disrupt vitamin B6 metabolism,reduce the concentration of PLP,down-regulate the expression levels of Pdxk and Gad1,inhibit the production of GABA,and ultimately lead to anxiety-like behaviors in offspring. | Xingwang Ding Ya Wen Xuan Ma Yuepei Zhang Yuting Cheng Zhaofeng Liu Weiyue Hu Yankai Xia | 2023 | Environmental Science and Ecotechnology2023,,1: | 0 |
| 18 | Association between SLC17A7 gene polymorphisms and venlafaxine for major depressive disorder in a Chinese Han population:a prospective pharmacogenetic case-control study显示文摘Objective: Venlafaxine is a common antidepressant and its therapeutic effect varies among people with different genetic backgrounds. The aim of this study was to investigate whether single nucleotide polymorphisms (SNPs) in theSLC17A7 gene are associated with the treatment outcome of venlafaxine in a Chinese Han population with major depressive disorder.Methods: This prospective pharmacogenetic case-control study that involved genotyping of four SNPs ofSLC17A7 was conducted on 175 major depressive disorder patients of Chinese Han origin, aged 18 to 65 years, participated in the study from April 2005 to September 2006. Comparisons of allele and genotype frequencies of all SNPs were performed between the responder/remission group and the nonresponder/nonremission group. This study was approved by the Institutional Ethics Committee of Sichuan University (approval No. 20151112-265).Results: The allele and genotype frequencies of the four candidate SNPs inSCL17A7 showed no significant difference between responders and nonresponders. Meanwhile, no significant difference was detected in the four investigatedSLC17A7 SNPs between patients who did and did not exhibit remission. | 20 | Selenized liposomes with ameliorative stability that achieve sustained release of emodin but fail in bioavailability显示文摘Stability of liposomes plays a crucial role in drug delivery,especially in oral aspect.The structural modification of liposomes has been the orientation of efforts to improve their stability and enable the controllability of payload release.This study reported a selenylation strategy to optimize the liposomal structure in an attempt to enhance the nanocarrier’s stability,hence the bioavailability of emodin(EM),an active compound with poor water-solubility.EM-loaded selenized liposomes(EM-Se@LPs)were prepared by thin film dispersion followed by in situ reduction technique.The results showed that EM-Se@LPs were provided with enhancive gastrointestinal stability and exhibited sustained release of drug compared with EM-loaded liposomes(EM-LPs).However,the modified liposomes with Se depositing onto the interior and exterior bilayers did not substantially facilitate absorption of EM.The reinforced structure of liposomes irrelevant to absorption was affirmed to be due to good stability and absorbability of EM itself.Nevertheless,the present work provides an alternative option for stabilization of liposomes instead of conventional methods,which may be promising for oral delivery of physiologically unstable and/or poorly absorbed drugs and systemic drug delivery. | Mujuan Zhu Shiping Zhu Qiubo Liu Yuehong Ren Zhiguo Ma Xingwang Zhang | 2023 | Chinese Chemical Letters2023,34,1: | 0 |
