| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 4 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 5 | Correlation between Waist Circumference and Carotid Intima-Media Thickness in Women from Shanghai, China显示文摘Objective To evaluate whether waist circumference (WC) ≥85 cm is related to asymptomatic preclinical atherosclerosis in women from Shanghai, China. Methods A total of 2365 females aged ≥20 years recruited from 4 communities underwent physical examination and carotid artery scanning. Their carotid intima-media thickness (C-IMT) was measured. Results The C-IMT was significantly higher in overweight or obese women with their BMI ≥25.0 kg/m2 (P<0.01) and in those with their WC ≥85 cm than in those with their WC <85 cm (P<0.01). Spearman and partial correlation analysis showed that the C-IMT was significantly correlated with WC which was independent of menopausal status. The C-IMT significantly increased with the increasing WC and reached to a platform in about 85 cm. An increment tendency was found in the subgroup with its WC < 85 cm (P<0.01) while no significant tendency was found in the subgroup with its WC≥85 cm (P=0.07). Multiple stepwise regression analysis showed that the WC was an independent risk factor for C-IMT. In logistic regression model, the odd ratio of WC ≥80 cm, ≥80 cm and <85 cm and ≥85 cm for evaluating the risk of C-IMT elevation was 1.632, 1.501, and 1.878, respectively. | 7 | GM130 regulates pulmonary surfactant protein secretion in alveolar typeⅡcells显示文摘Pulmonary surfactant is a lipid-protein complex secreted by alveolar typeⅡepithelial cells and is essential for the maintenance of the delicate structure of mammalian alveoli to promote efficient gas exchange across the air-liquid barrier.The Golgi apparatus plays an important role in pulmonary surfactant modification and secretory trafficking.However,the physiological function of the Golgi apparatus in the transport of pulmonary surfactants is unclear.In the present study,deletion of GM130,which encodes for a matrix protein of the cis-Golgi cisternae,was shown to induce the disruption of the Golgi structure leading to impaired secretion of lung surfactant proteins and lipids.Specifically,the results of in vitro and in vivo analysis indicated that the loss of GM130 resulted in trapping of Sftpa in the endoplasmic reticulum,Sftpb and Sftpc accumulation in the Golgi apparatus,and an increase in the compensatory secretion of Sftpd.Moreover,global and epithelial-specific GM130 knockout in mice resulted in an enlargement of alveolar airspace and an increase in alveolar epithelial autophagy;however,surfactant repletion partially rescued the enlarged airspace defects in GM130-deficient mice.Therefore,our results demonstrate that GM130 and the mammalian Golgi apparatus play a critical role in the control of surfactant protein secretion in pulmonary epithelial cells. | Qianqian Pang Chunyi Liu Yulong Qiao Jian Zhao Sin Man Lam Mei Mei Guanghou Shui Shilai Bao Qiuling Li | 2022 | Science China(Life Sciences)2022,65,1: | 2 |

| 20 | Experimental Study on Permeability Characteristics of Geotubes for Seepage Analysis on Safety Assessment of Dams显示文摘Geotubes are heterogeneous structures composed of filling sand and bag material,and its pemeability characteristics are different from that of flling sand.The uncertainty of geotubes permeability characteristics results in the failure of seepage analysis of geotube dams,which restricts the safety assessment of the dams.As the basis of the study on the seepage mechanism of the geotubes,the influence of particle grading on pemeability cofficient of flling sand and sand covered with geotextiles were explored by the pemmeability tests of flling sand with different particle grading under the condition of sand covered with or without geotextiles.And the infuence of geotextiles on the permeability coefficient was analyzed by comparing permeability coefficient of sand covered with and without geotextiles.The test results show that the influence of single particle size content on permeability coe ficient is consistent under the condition of sand covered with and without geotextiles.The content of powder,fine,medium and coarse particles is linearly related to their respective permeability cofficients.And the content of powder,fine,medium particles is negatively correlated with their permeability coefficients,while the content of coarse particles is positively correlated with the permeability coefficient.But the pemeability coefficient of sand covered with geotextiles is smaller than that of flling sand under the same conditions.Finally,the parameter d2 50 Cc/Cu was selected as a variable representing the particle grading to fit the empirical formula of permeability coefficient of flling sand and sand covered with geotextiles. | Xiaolei Man Ganggang Sha Shuigen Hu Hui Bao Guangying Liu | 2020 | Structural Durability & Health Monitoring2020,14,4: | 0 |
