Inhibition of cyclooxygenase-2 activity in subchondral bone modifies a subtype of osteoarthritis显示文摘Osteoarthritis(OA) causes the destruction of joints. Its pathogenesis is still under investigation, and there is no effective diseasemodifying therapy. Here, we report that elevated cyclooxygenase-2(COX-2) expression in the osteocytes of subchondral bone causes both spontaneous OA and rheumatoid arthritis(RA). The knockout of COX-2 in osteocytes or treatment with a COX-2 inhibitor effectively rescues the structure of subchondral bone and attenuates cartilage degeneration in spontaneous OA(STR/Ort)mice and tumor necrosis factor-α transgenic RA mice. Thus, elevated COX-2 expression in subchondral bone induces both OAassociated and RA-associated joint cartilage degeneration. The inhibition of COX-2 expression can potentially modify joint destruction in patients with arthritis. | Manli Tu Mi Yang Nanxi Yu Gehua Zhen Mei Wan Wenlong Liu Baochao Ji Hairong Ma Qiaoyue Guo Peijian Tong Li Cao Xianghang Luo Xu Cao | 2019 | Bone Research2019,7,3: | 5
Rapid isolation and immune profiling of SARS-CoV-2 specific memory B cell in convalescent COVID-19 patients via LIBRA-seq显示文摘B cell response plays a critical role against SARS-CoV-2 infection. However, little is known about the diversity and frequency of the paired SARS-CoV-2 antigen-specific BCR repertoire after SARS-CoV-2 infection. Here,we performed single-cell RNA sequencing and VDJ sequencing using the memory and plasma B cells isolated from five convalescent COVID-19 patients, and analyzed the spectrum and transcriptional heterogeneity of antibody immune responses.Via linking BCR to antigen specificity through sequencing(LIBRA-seq),we identified a distinct activated memory B cell subgroup(CD11c^(high) CD95^(high))had a higher proportion of SARS-CoV-2 antigen-labeled cells compared with memory B cells. Our results revealed the diversity of paired BCR repertoire and the non-stochastic pairing of SARS-CoV-2 antigen-specific immunoglobulin heavy and light chains after SARS-CoV-2 infection. The public antibody clonotypes were shared by distinct convalescent individuals. Moreover, several antibodies isolated by LIBRA-seq showed high binding affinity against SARS-CoV-2 receptor-binding domain(RBD)or nucleoprotein(NP)via ELISA assay. Two RBD-reactive antibodies C14646P3S and C2767P3S isolated by LIBRA-seq exhibited high neutralizing activities against both pseudotyped and authentic SARS-CoV-2 viruses in vitro. Our study provides fundamental insights into B cell response following SARS-CoV-2 infection at the single-cell level. | Bing He Shuning Liu Yuanyuan Wang Mengxin Xu Wei Cai Jia Liu Wendi Bai Shupei Ye Yong Ma Hengrui Hu Huicui Meng Tao Sun Yanling Li Huanle Luo Mang Shi Xiangjun Du Wenjing Zhao Shoudeng Chen Jingyi Yang Haipeng Zhu Yusheng Jie Yuedong Yang Deyin Guo Qiao Wang Yuwen Liu Huimin Yan Manli Wang Yao-Qing Chen | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 2
Design and Implementation of Dynamic High-Speed Switches in Super Base Station Architectures显示文摘Novel centralized base station architectures integrating computation and communication functionalities have become important for the development of future mobile communication networks. Therefore, the development of dynamic high-speed interconnections between baseband units(BBUs)and remote radio heads(RRHs)is vital in centralized base station design.Herein, dynamic high-speed switches(HSSs)connecting BBUs and RRHs were designed for a centralized base station architecture. We analyzed the characteristics of actual traffic and introduced a switch traffic model suitable for the super base station architecture. Then,we proposed a data-priority-aware(DPA)scheduling algorithm based on the traffic model.Lastly,we developed the dynamic HSS model based on the OPNET platform and the prototype based on FPGA.Our results show that the DPA achieves close to 100%throughput with lower latency and provides better run-time complexity than iOCF and HE-iSLIP,thereby demonstrating that the proposed switch system can be adopted in centralized base station architectures. | Yingjiao Ma Jinglin Shi Yiqing Zhou Lin Tian Manli Qian | 2020 | China Communications2020,17,3: | 1

Pathological features of COVID-19-associated liver injury-a preliminary proteomics report based on clinical samples显示文摘Dear Editor,Almost one-half of COVID-19 patients have symptoms of liver injury. However, the molecular characteristics in the COVID-19 liver remain unknown. Therefore, the need for long-term medication for recovery make it imperative to study the infectivity and pathogenicity of SARS-CoV-2 in the liver. Here,we have identifed signifcant proteomic changes for the first time in the liver of COVID-19 patients. We detected differential expression levels of several proteins that mediate characteristic cellular host responses elicited by SARS-CoV-2 infection in the liver. | Ling Leng Ruiyuan Cao Jie Ma Luye LV Wei Li Yunping Zhu Zhihong Wu Manli Wang Yiwu Zhou Wu Zhong | 2021 | Signal Transduction and Targeted Therapy2021,6,2: | 1
Analysis of parental genetic diversity and its impact on grain yield and quality of japonica hybrid rice in northern China显示文摘The relationship between parental genetic differences and the quality and yield of japonica hybrid rice strongly influences japonica hybrid rice breeding. In this study,137 parental lines of japonica hybrid rice were genotyped using 8 K rice SNP-Chips to characterize their genetic diversity, population structure, and indica-genotype proportion. The genetic diversity of total parental lines averaged 0.264,with values of0.287 for restorer lines and 0.148 for the sterile lines. The introduction of indica lineage increased the genetic diversity of restorer lines relative to that of sterile lines.By model-based population structure analysis, the 137 lines were divided into 14 groups.According to the grouping results, eight restorer lines and five sterile lines were selected from different groups for cross breeding, yielding 40 japonica hybrid rice combinations(F1).Investigation of the yield and quality of these combinations showed that highyield combinations could be obtained by increasing parental genetic distance to 0.8–0.9,a result accomplished largely by the introduction of indica genomic components of restorer lines. To further improve grain quality, the genetic distance between parents should be reduced to 0.4–0.5,suggesting an indicagenotype proportion of 30%–40%for restorer lines. This study may provide a reference for breeding of japonica hybrid rice. | Hong Gao Zuobin Ma Yuanzheng Wang Manli Zhang Xianju Wang Changhua Wang Zhiqiang Tang Liying Zhang Liang Fu Na He Hui Wang Yongan Yin Yuanjun Bai Guomin Sui Wenjing Zheng | 2022 | The Crop Journal2022,10,3: | 1

Multivariate statistical and bioinformatic analyses for the seasonal variations of actinobacterial community structures in a drinking water reservoir显示文摘Actinobacterial community is a conspicuous part of aquatic ecosystems and displays an important role in the case of biogeochemical cycle, but little is known about the seasonal variation of actinobacterial community in reservoir ecological environment. In this study, the high-throughput techniques were used to investigate the structure of the aquatic actinobacterial community and its inducing water quality parameters in different seasons. The results showed that the highest diversity and abundance of actinobacterial community occurred in winter, with Sporichthya(45.42%)being the most abundant genus and Rhodococcus sp.(29.32%)being the most abundant species.Network analysis and correlation analysis suggested that in autumn the dynamics of actinobacterial community were infuenced by more factors and Nocardioides sp.SX2R5S2 was the potential keystone species which was negatively correlated with temperature(R=-0.72,P<0.05).Changes in environmental factors could significantly affect the changes in actinobacterial community, and the dynamics of temperature, dissolved oxygen(DO),and turbidity are potential conspicuous factors infuencing seasonal actinobacterial community trends. The partial least squares path modeling further elucidated that the combined effects of DO and temperature not only in the diversity of actinobacterial community but also in other water qualities, while the physiochemical parameters(path coefficient=1.571,P<0.05)was strong environmental factors in natural mixture period. These results strengthen our understanding of the dynamics and structures of actinobacterial community in the drinking water reservoirs and provide scientific guidance for further water quality management and protection in water sources. | Haihan Zhang Sixuan Pan BenMa Tinglin Huang Dmitry B.Kosolapov Manli Ma Xiang Liu Huan Liu Xiaoyan Liu | 2024 | Journal of Environmental Sciences2024,,3: | 0
Endoderm contributes to endocardial composition during cardiogenesis显示文摘Heart formation commences from a single heart tube, which fuses from bilateral primordial heart fields. The developing heart tube is composed of outer-layer myocardial cells and inner-layer endocardial cells.Several distinct populations of precardiac cells contribute to cardiac morphogenesis. However,it still remains not very clear about the lineage of endocardium at gastrulation stage.Thereby, this study focused on ascertaining the correlation between the hypoblast in gastrulation and endocardium during cardiogenesis.Firstly, the fusing heart tube morphologically is closed to endoderm-derived pharynx floor, implying the possibility that pharynx floor might be wrapped into the formation of endoderm.Secondly,HNK1 is expressed in hypoblast strongly at gastrula stage and subsequently appeared in endocardium of cardiogenesis. Moreover, fate map data displayed that DiI labeled hypoblast was also present in endocardium later on.One more evidence is chick-quail chimera of hypoblast transplantation,in which quail-hypoblast derivative could be identified inendocardium of cardiogenesis by QCPN antibody. In sum,our current data suggests that endoderm in gastrula contribute at least partly to the formation of endocardium of cardiogenesis. | Yan Li Xiaoyu Wang Zhenglai Ma Manli Chuai Andrea Münsterberg Kenneth KaHo Lee Xuesong Yang | 2014 | Chinese Science Bulletin2014,59,22: | 0

Dehydro-Diels-Alder reaction and diamondization of bowl-shaped clusters C_(18)Te_(3)Br_(4)(Bu-O)_(6)显示文摘Dehydro-Diels-Alder(DDA)reaction is a textbook reaction for preparing six-membered rings in solution but is scarcely seen in solid-state synthesis. In this work, using multiple characterization techniques,we demonstrate that the bowl-shaped clusters C_(18)Te_(3)Br_(4)(Bu-O)_(6) might experience a DDA reaction at room temperature and high pressure between 5.5 and 7.4 GPa.Above 17.0 GPa,it is found that the bonding conversion from the intramolecular sp^(2) to the intermolecular spa occurred,in the form of pressure-induced diamondization. The recovered samples from 20.0 and 36.1 GPa showed incomplete reversibility, while the decompression-induced graphitization of glassy carbon was observed during decompression from 46.5 GPa.The electrochemical impedance spectroscopy results indicated that the transport properties changed from grain boundary dominant to grain dominant due to the DDA reaction and the grain boundary effect disappeared as the intermolecular sp3 bonding building-up and carrier transmission channel formation above 17.0 GPa.The results in this study open a new route to construct the crystalline carbon materials with different transport properties. | Jinbo Zhang Manli Ma Rong Zhou Hongqiang Chu Xue Wang Shaojie Wang Huhu Tian Zhipeng Yan Mingtao Li Zhongyan Wu Bin Li Jiafeng Yan Lan Anh Thi Nguyen Rongxing Cao Guoqing Wu Xianghua Zeng Hao-Li Zhang Jaeyong Kim Lin Wang Yongjun Tian | 2022 | Nano Research2022,15,5: | 0
