Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation显示文摘ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. Since placenta and UCB arehomogeneous, the MSC derived from human placenta can be transplanted combined with hematopoietic stem cells(HSC) from UCB to reduce the potential graft -versus-host disease (GVHD) in recipients.Chang Dong LI Wei Yuan ZHANG He Lian LI Xiao Xia JIANG Yi ZHANG Pei Hsien TANG Ning MAO 2005Cell Research2005,15,7:36

Multimodal treatment for spinal cord injury: a sword of neuroregeneration upon neuromodulation显示文摘Spinal cord injury is linked to the interruption of neural pathways, which results in irreversible neural dysfunction.Neural repair and neuroregeneration are critical goals and issues for rehabilitation in spinal cord injury, which require neural stem cell repair and multimodal neuromodulation techniques involving personalized rehabilitation strategies.Besides the involvement of endogenous stem cells in neurogenesis and neural repair, exogenous neural stem cell transplantation is an emerging effective method for repairing and replacing damaged tissues in central nervous system diseases. However,to ensure that endogenous or exogenous neural stem cells truly participate in neural repair following spinal cord injury, appropriate interventional measures(e.g.,neuromodulation)should be adopted.Neuromodulation techniques, such as noninvasive magnetic stimulation and electrical stimulation, have been safely applied in many neuropsychiatric diseases. There is increasing evidence to suggest that neuromagnetic/electrical modulation promotes neuroregeneration and neural repair by affecting signaling in the nervous system;namely,by exciting, inhibiting,or regulating neuronal and neural network activities to improve motor function and motor learning following spinal cord injury.Several studies have indicated that fine motor skill rehabilitation training makes use of residual nerve fibers for collateral growth, encourages the formation of new synaptic connections to promote neural plasticity, and improves motor function recovery in patients with spinal cord injury. With the development of biomaterial technology and biomechanical engineering, several emerging treatments have been developed, such as robots,brain-computer interfaces, and nanomaterials. These treatments have the potential to help millions of patients suffering from motor dysfunction caused by spinal cord injury.However,large-scale clinical trials need to be conducted to validate their efficacy. This review evaluated the efficacy of neural stem cells and magnetic or electrical stimulation combined with rehabilitation training and intelligent therapies for spinal cord injury according to existing evidence,to build up a multimodal treatment strategy of spinal cord injury to enhance nerve repair and regeneration.Ya Zheng Ye-Ran Mao Ti-Fei Yuan Dong-Sheng Xu Li-Ming Cheng 2020Neural Regeneration Research2020,15,8:29
Efficacy and safety of Xinfeng capsule in patients with rheumatoid arthritis:a multi-center parallel-group double-blind randomized controlled trial显示文摘OBJECTIVE:To evaluate the efficacy and safety of Xinfeng capsule in patients suffering rheumatoid arthritis(RA).METHODS:A multi-center parallel-group designed,double-blind,randomized,controlled trial was conducted.Totally 304 RA patients were assigned to two groups:one group was administered Xinfeng capsule(XFC) plus the placebo of leflunomide and the other given leflunomide(LEF) plus the placebo of XFC for twelve weeks. The clinical and laboratory parameters were compared at baseline and fourth, eighth, and twelfth weeks.RESULTS:After twelve-week treatment, patients in two groups all showed some trend of effectiveness when compared in terms of American Rheumatism Association(ACR) recommended 20%,50%,70%improvement criteria, but it was insignificant. The validity in ameliorate modified disease activity score(DAS28) and laboratory indexes as erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),rheumatoid factor(RF) were also found no difference. The score of health assessment questionnaire(HAQ),self-rating anxiety scale(SAS),self-rating depression scale(SDS) and quality of life questionnaire with rheumatoid arthritis(RAQOL)both lower than the first week and the changes showed no difference. However, the score of SDS dropped more in XFC group than in the other.A total of 147 adverse reaction cases were reported, which shows no difference between the two groups. The most common adverse reactions were hepatic impairment, anemia, leukocytopenia, epigastric discomfort and phalacrosis.CONCLUSION:XFC demonstrated better improvement in the scores of SDS and compared with those of LEF group.Liu Jian Wang Yuan Huang Chuanbing Xu Jianhua Li Zhijun Xu Liang He Liyun Sun Yue Wang Yali Xu Shengqian Zhao Ping Mao Tongjun Tan Bin Zhu Fubing Zhang Pingheng Fang Li 2015Journal of Traditional Chinese Medicine2015,35,5:27
Effects of Short-Term Forest Bathing on Human Health in a Broad-Leaved Evergreen Forest in Zhejiang Province,China显示文摘Objective To investigate the effects of short-term forest bathing on human health. Methods Twenty healthy male university students participated as subjects and were randomly divided into two groups of 10.One group was sent on a two-night trip to a broad-leaved evergreen forest, and the other was sent to a city area.Serum cytokine levels reflecting inflammatory and stress response, indicators reflecting oxidative stress, the distribution of leukocyte subsets, and plasma endothelin-1 (ET-1) concentrations were measured before and after the experiment to evaluate the positive health effects of forest environments.A profile of mood states (POMS) evaluation was used to assess changes in mood states. Results No significant differences in the baseline values of the indicators were observed between the two groups before the experiment.Subjects exposed to the forest environment showed reduced oxidative stress and pro-inflammatory level,as evidenced by decreased malondialdehyde,interleukin-6,and tumor necrosis factor α levels compared with the urban group.Serum cortisol levels were also lower than in the urban group.Notably, the concentration of plasma ET-1 was much lower in subjects exposed to the forest environment. The POMS evaluation showed that after exposure to the forest environment, subjects had lower scores in the negative subscales, and the score for vigor was increased. Conclusion Forest bathing is beneficial to human health, perhaps through preventive effects related to several pathological factors.MAO Gen Xiang LAN Xiao Guang CAO Yong Bao CHEN Zhuo Mei HE Zhi Huat LV Yuan Dong WANG Ya Zhen HU Xi Lian WANG Guo Fu YAN Jing 2012Biomedical and Environmental Sciences2012,25,3:20
Single-cell transcriptomics reveals regulators underlying immune cell diversity and immune subtypes associated with prognosis in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an aggressive malignancy with extremely skewed ethnic and geographic distributions.Increasing evidence indicates that targeting the tumor microenvironment(TME)represents a promising therapeutic approach in NPC,highlighting an urgent need to deepen the understanding of the complex NPC TME.Here,we generated single-cell transcriptome profiles for 7581 malignant cells and 40,285 immune cells from fifteen primary NPC tumors and one normal sample. We revealed malignant signatures capturing intratumoral transcriptional heterogeneity and predicting aggressiveness of malignant cells.Diverse immune cell subtypes were identified, including novel subtypes such as CLEC9A^+dendritic cells(DCs).We further revealed transcriptional regulators underlying immune cell diversity, and cell–cell interaction analyses highlighted promising immunotherapeutic targets in NPC.Moreover,we established the immune subtype-specific signatures, and demonstrated that the signatures of macrophages, plasmacytoid dendritic cells(pDCs),CLEC9A^+DCs,natural killer(NK)cells, and plasma cells were significantly associated with improved survival outcomes in NPC.Taken together, our findings represent a unique resource providing in-depth insights into the cellular heterogeneity of NPC TME and highlight potential biomarkers for anticancer treatment and risk stratification, laying a new foundation for precision therapies in NPC.Yu-Pei Chen Jian-Hua Yin Wen-Fei Li Han-Jie Li Dong-Ping Chen Cui-Juan Zhang Jia-Wei Lv Ya-Qin Wang Xiao-Min Li Jun-Yan Li Pan-Pan Zhang Ying-Qin Li Qing-Mei He Xiao-Jing Yang Yuan Lei Ling-Long Tang Guan-Qun Zhou Yan-Ping Mao Chen Wei Ke-Xu Xiong Hong-Bo Zhang Shi-Da Zhu Yong Hou Ying Sun Michael Dean Ido Amit Kui Wu Dong-Ming Kuang Gui-Bo Li Na Liu Jun Ma 2020Cell Research2020,30,11:19

The Chromosome-Based Rubber Tree Genome Provides New Insights into Spurge Genome Evolution and Rubber Biosynthesis显示文摘The rubber tree,Hevea brasiliensis, produces natural rubber that serves as an essential industrial raw material. Here,we present a high-quality reference genome for a rubber tree cultivar GT1 using single-molecule real-time sequencing(SMRT)and Hi-C technologies to anchor the~1.47-Gb genome assembly into 18 pseudochromosomes. The chromosome-based genome analysis enabled us to establish a model of spurge chromosome evolution, since the common paleopolyploid event occurred before the split of Hevea and Manihot. We show recent and rapid bursts of the three Hevea-specific LTR-retrotransposon families during the last 10 million years, leading to the massive expansion by~65.88%(~970 Mbp)of the whole rubber tree genome since the divergence from Manihot. We identify large-scale expansion of genes associated with whole rubber biosynthesis processes, such as basal metabolic processes, ethylene biosynthesis, and the activation of polysaccharide and glycoprotein lectin, which are important properties for latex production.A map of genomic variation between the cultivated and wild rubber trees was obtained, which contains~15.7 million high-quality single-nucleotide polymorphisms. We identified hundreds of candidate domestication genes with drastically lowered genomic diversity in the cultivated but not wild rubber trees despite a relatively short domestication history of rubber tree, some of which are involved in rubber biosynthesis. This genome assembly represents key resources for future rubber tree research and breeding, providing novel targets for improving plant biotic and abiotic tolerance and rubber production.Jin Liu Cong Shi Cheng-Cheng Shi Wei Li Qun-Jie Zhang Yun Zhang Kui Li Hui-Fang Lu Chao Shi Si-Tao Zhu Zai-Yun Xiao Hong Nan Yao Yue Xun-Ge Zhu Yu Wu Xiao-Ning Hong Guang-Yi Fan Yan Tong Dan Zhang Chang-Li Mao Yun-Long Liu Shi-Jie Hao Wei-Qing Liu Mei-Qi Lv Hai-Bin Zhang Yuan Liu Ge-Ran Hu-tang Jin-Peng Wang Jia-Hao Wang Ying-Huai Sun Shu-Bang Ni Wen-Bin Chen Xing-Cai Zhang Yuan-Nian Jiao Evan E.Eichler Guo-Hua Li Xin Liu Li-Zhi Gao 2020Molecular Plant2020,13,2:15
Significant value of 18F-FDG-PET/CT in diagnosing small cervical lymph node metastases in patients with nasopharyngeal carcinoma treated with intensity-modulated radiotherapy显示文摘Background: Little is known about the nature of metaistasis to small cervical lymph nodes(SCLNS) in the patients with nasopharyngeal carcinoma(NPC)examined by using 18-fluoro-2-deoxy-glucose(^(18)F-FDG) positron emission tomography/computed tomography(PET/CT).The present study aimed to evaluate the diagnostic values of PET/CT in identifying metastasis in SCLNs in NPC patients. Methods: Magnetic resonance images(MRI) and PET/CT scans for 470 patients with newly diagnosed, non-distant metastatic NPC were analyzed. Metastatic rates of SCLNs were defined by the positive number of SCLNs on PET/CT scans and total number of SCLNs on MRI scans. Receiver operating characteristic curve was applied to compare PET/CT-determined stage with MRI-determined stage. Results: In total, 2082 SCLNs were identified, with 808(38.8%) ≥ 5 and < 6 mm in diameter(group A), 526(25.3%)≥ 6 and < 7 mm in diameter(group B),374(18.0%)≥ 7 and < 8 mm in diameter(group C), 237(11.4%) ≥8 and<9 mm in diameter(group D),and 137(6.5%) ≥ 9 and <10 mm in diameter(group E).The overall metastatic rates examined by using PET/CT for groups A, B,C,D, and E were 3.5%, 8.0%, 31.3%, 60.0%, and 83.9%, respectively(P< 0.001). In level IV/Vb, the metastatic rate for nodes ≥ 8 mm was 84.6%. PET/CT examination resulted in modification of N category and overall stage for 135(28.7%) and 46(9.8%) patients, respectively. The areas under curve of MRIdetermined and PET/CT-determined overall stage were 0.659 and 0.704 for predicting overall survival, 0.661 and 0.711 for predicting distant metastasis-free survival, and 0.636 and 0.663 for predicting disease-free survival. Conclusions: PET/CT was more effective than MRI in identifying metastatic SCLNs, and the radiologic diagnostic criteria for metastatic lymph nodes in level IV/Vb should be re-defined.Hao Peng Lei Chen Ling-Long Tang Wen-Fei Li Yan-Ping Mao Rui Guo Yuan Zhang Li-Zhi Liu Li Tian Xu Zhang Xiao-Ping Lin Ying Guo Ying Sun Jun Ma 2017Chinese Journal of Cancer2017,36,12:14

Effect of human umbilical cord-derived mesenchymal stem cells on lung damage in severe COVID-19 patients:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Treatment of severe Coronavirus Disease 2019(COVID-19)is challenging. We performed a phase 2 trial to assess the efficacy andsafety of human umbilical cord-mesenchymal stem cells(UC-MScs)to treat severe coViD-19 patients with lung damage, based onour phase 1 data.In this randomized,double-blind,and placebo-controlled trial,we recruited 101 severe coVID-19 patients withlung damage. They were randomly assigned at a 2:1 ratio to receive either UC-MSCs(4×10^(7)cells per infusion)or placebo on day 0,3,and 6.The primary endpoint was an altered proportion of whole lung lesion volumes from baseline to day 28.Other imagingoutcomes,6-minute walk test(6-MWT),maximum vital capacity, diffusing capacity, and adverse events were recorded and analyzed. In all,100 COVID-19 patients were finally received either UC-MSCs in=65)or placebo(n=35).UC-MSCs administrationexerted numerical improvement in whole lung lesion volume from baseline to day 28 compared with the placebo(the mediandifference was-13.31%,95%Cl-29.14%,2.13%,P=0.08).UC-MSCs significanty reduced the proportions of solid componentlesion volume compared with the placebo(median difference:-15.45%;95%CI-30.82%,-0.39%;P=0.043).The 6-MWT showedan increased distance in patients treated with UC-MSCs(difference:27.00 m;95%CI 0.00,57.00;P=0.057).The incidence of adverseevents was similar in the two groups. These results suggest that UC-MSCs treatment is a safe and potentially effective therapeuticapproach for COVID-19 patients with lung damage.A phase 3 trial is required to evaluate effects on reducing mortality andpreventing long-term pulmonary disability.Lei Shi Hai Huang Xuechun Lu Xiaoyan Yan Xiaojing Jiang Ruonan Xu Siyu Wang Chao Zhang Xin Yuan Zhe Xu Lei Huang Jun-Liang Fu Yuanyuan Li Yu Zhang Wei-Qi Yao Tianyi Liu Jinwen Song Liangliang Sun Fan Yang Xin Zhang Bo Zhang Ming Shi Fanping Meng Yanning Song Yongpei Yu Jiqiu Wen Qi Li Qing Mao Markus Maeurer Alimuddin Zumla Chen Yao Wei-Fen Xie Fu-Sheng Wang 2021Signal Transduction and Targeted Therapy2021,6,3:13
Casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DR5 in human lung cancer cells显示文摘Objective:To assess if casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DR5 in human lung cancer ceils. Methods:Human non-small-cell lung carcinoma cell lines H460,AS49 and H157 were cultured in vitro. The cytotoxic activities were determined using MTT assay. The apoptotic cells death was examined by flow cytometry using PI staining and DMA agarose gel electrophoresis. The activities of caspase-3, -8 and -9 were measured via ELISA.Cellular fractionation was determined by flow cytometry to assess release of cytochrome c and the mitochondrial transmembrane potential.Bcl-2/Bcl-XL/ XIAP/Bid/ DR5 and DR4 proteins were analyzed using western blot. Results:The concentrations required for a 50%decrease in cell growth(IC50) ranged from 1.8 to 3.2 Jt M.Casticin induced rapid apoptosis and triggered a series of effects associated with apoptosis by way of mitochondrial pathway, including the depolarization of the mitochondrial membrane, release of cytochrome c from mitochondria, activation of procaspase-9 and -3,and increase of DNA fragments. Moreover, the pan caspase inhibitor zVAD-FMK and the caspase-3 inhibitor zOEVD-FMK suppressed casticin-induced apoptosis. In addition, casticin induced XIAP and Bcl-XL down-regulation, Bax upregulation and Bid clearage. In H157 cell line, casticin increased expression of DRS at protein levels but not affect the expression of DR4.The prelreatmenl with DR5/Fc chimera protein effectively attenuated casticin-induced apoptosis in H157 cells. No correlation was found between cell sensitivity to casticin and that to p53 status, suggesting that casticin induce a p53- independent apoptosis. Conclusions:Our results demonstrate that casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DRS in human lung cancer cells.Yuan Zhou Yi Peng Qi-Qi Mao Xia Li Ming-Wu Chen Ting Su Li Tian Nai-Quan Mao Ling-Zhi Long Mei-Fang Quan Fei Liu Su-Fang Zhou Yong-Xiang Zhao 2013Asian Pacific Journal of Tropical Medicine2013,6,5:11
First mass measurement of short-lived nuclides at HIRFL-CSR显示文摘Recent commissioning of the HIRFL-CSR has demonstrated its ability to perform direct mass measurement for short-lived nuclides. Projectile fragments produced by the 78Kr ions at 481.88 MeV/u were separated with the new radioactive beam line in Lanzhou (RIBLL2), and injected into and stored in the experimental storage ring (CSRe). By operating the CSRe as an isochronous mass spectrometry, a typical mass resolution around 2.0×10?5 has been achieved. The masses for 63Ge, 65As and 67Se were measured for the first time. The measured masses are compared with theoretical predictions and the location of the proton drip-line for As isotopes is discussed. The implication of the 65As mass in the astrophysical rapid proton capture process has also been addressed.XU HuShan TU XiaoLin YUAN YouJin XIA JiaWen ZANG YongDong WANG Meng MAO RuiShi HU ZhengGuo MEI Bo SHUAI Peng ZHU ZhiChao TANG ShuWen GENG Peng LIU Yong ZHAO TieCheng ZHANG XueYing YANG JianCheng MAO LiJun ZHANG YuHu ZHOU XiaoHong HUANG WenXue XIAO GuoQing ZHAO HongWei ZHAN WenLong 2009Chinese Science Bulletin2009,54,24:11
Characteristics and sedimentary processes of lamina-controlled sand-particle imbricate structure in deposits on Lingshan Island, Qingdao, China显示文摘Imbricate structure is a common sedimentary structure and is well developed in sandy sediments. Here,we report a laminacontrolled fine-sand-particle imbricate structure in a set of very fine grained sedimentary rocks(fine sandstone, siltstone, and mudstone)at the Dockyard and Qianceng Cliff areas of Lingshan Island,Qingdao,Shandong,China.Sets of up to 300 laminae are found in stratigraphic profiles in these areas. The laminae are generally less than 1 mm thick, with most being 0.3–0.4 mm thick and the thinnest being<0.1 mm.The dip angle of the imbricate structure varies widely, between 0°and 90°,with an average dip angle of about 40°–50°,which is higher than that of imbricate riverbed gravels(about 34°).The dip angle is a function of the shape and sorting of the particles,as well as the hydrodynamic conditions under which these fine-grained sediments were deposited.Several profiles show well-developed multiscale,soft-sediment deformation structures.Flute casts, load casts, and groove casts are also common.Fragments of carbon remains occur widely and commonly constitute stringers several millimeters thick and up to 10 cm long, together with fine clasts.Vitrain lenticles are also common.Based on the imbricate structure of the Lingshan Island deposits,it is shown that in addition to paleocurrent analysis, the imbricate structure can be used to infer information about fluid properties, transport characteristics, and sedimentary processes of the depositional environment.A near-bottom underflow, either of authigenic origin or derived from cold river water in winter in a delta interdistributary bay or delta-front environment,is inferred to have provided the hydrodynamic setting in which this imbricate structure formed. The imbricate structure, together with other sedimentary structures and features, shows that the sedimentary rocks on Lingshan Island were deposited in an inland,shallow-water environment, such as a delta, and not in a deep-water or submarine continental-slope environment.SHAO ZhuFu ZHONG JianHua LI Yong MAO Cui LIU ShengXin NI LiangTian TIAN Yuan CUI XinYing LIU YunTian WANG XiaoNan LI WeiHua LIN GangShan 2014Science China Earth Sciences2014,57,5:11
Identification of potential cross-protective epitope between a new type of coronavirus(2019-nCoV)and severe acute respiratory syndrome virus显示文摘Recently,a new type of unknown virus causing severe acute res-piratory infection was reported in Wuhan city,Hubei province,China(WHO,2020b).Infection of this virus was first reported in December 2019,and origin of the virus was traced back to a large seafood/wide animal market in Wuhan city. The serious clinical symptoms of the viral infection, including fever, dry cough,dys-pnea,and pneumonia, may result in progressive respiratory failure and even death. Moreover, the quick spread of the virus has caused an epidemic in China,as well as infection cases worldwide. The whole-genome sequence of Wuhan new virus(WH-Human_.1)was first released on January 10,2020(Zhang,2020),followed by additional ones released in Global Initiative on Sharing All Influ-enza Data(GISAID)(Shu and McCauley,2017).Later,this new virus was determined and announced as a new type of coronavirus(CoV;2019-nCoV)by the World Health Organization(WHO,2020a).Tianyi Qiu Tiantian Mao Yuan Wang Mengdi Zhou Jingxuan Qiu Jianwei Wang Jianqing Xu Zhiwei Cao 2020Journal of Genetics and Genomics2020,47,2:11
Endpoint Prediction of EAF Based on Multiple Support Vector Machines显示文摘The endpoint parameters are very important to the process of EAF steel-making, but their on-line measurement is difficult. The soft sensor technology is widely used for the prediction of endpoint parameters. Based on the analysis of the smelting process of EAF and the advantages of support vector machines, a soft sensor model for predicting the endpoint parameters was built using multiple support vector machines (MSVM). In this model, the input space was divided by subtractive clustering and a sub-model based on LS-SVM was built in each sub-space. To decrease the correlation among the sub-models and to improve the accuracy and robustness of the model, the sub-models were combined by Principal Components Regression. The accuracy of the soft sensor model is perfectly improved. The simulation result demonstrates the practicability and efficiency of the MSVM model for the endpoint prediction of EAF.YUAN Ping MAO Zhi-zhong WANG Fu-li 2007Journal of Iron and Steel Research(International)2007,14,2:10
