| 7 | Metastatic tumor to the iris and ciliary body as an initial sign of lung cancer: a case report显示文摘The most common sites of lung cancer metastases are pleura, bone, brain, pericardium and liver. 1 Tumor metastasis to the eye is a rare complication of lung cancer. Metastatic cancer to the ocular region most often involves the uveal tract, however, most uveal metastases occur in the posterior uvea, and iris metastases are relatively rare. We describe a patient with adenocarcinoma of the lung metastatic to the iris as the first clinical sign without symptoms of lung cancer.CASE REPORTIn September 2003, a 54-year-old man was presented at outpatient department of ophthalmology because of progressive blurred vision and discomfort in his left eye during the previous week. Ocular examination of the left showed the best-corrected visual acuity 0.4 and intraocular pressure of 34 mmHg. Slit lamp examination of the left eye showed a saddle-shaped yellowish tumor with prominent blood vessels involving the iris in the temporal quadrant and a dome-shaped yellow-brown mass at the inferior-temporal pupil margin (Fig. 1). Tyndall sign was ++ and moderate cells in the left anterior chamber. Brownish material and blood vessels were found in the inferior and temporal chamber angle gonioscopically. Visual acuity and slit lamp examination of the right eye were not significant. Ophthalmoscopy revealed the cup/disc ratio was 0.3 and 0.5 for the right and left eye respectively. The retinae of both eyes were otherwise within normal limits. Ultrasound biomicroscopy showed mass lesion in the iris of left eye. Ciliary body was also involved. He had no other systemic symptom and no history of systemic cancer. His father died of lung cancer; one of his brothers died of colon cancer; his son was diagnosed as brain tumor. However, it is unclear whether different mouse models with mitochondrialrelated diseases have similar changes in mitochondrial morphology of the same tissues. Moreover, whether similarities in mitochondrial morphology can be a suitable marker for screening and/or discovering mitochondrial-protective substances remains unknown. Mitochondria morphology in different tissues of a novel mitochondrial outer membrane protein Slc25a46 knockout mouse and a traditional APP_(SWE)/PS1ΔE9 transgenic mouse were examined using transmission electron microscope(TEM). Both young Slc25a46 knockout mice and aged APP_(SWE)/PS1ΔE9 mice models showed similar mitochondrial damage in cerebellum tissues. The results indicated that different mitochondrial-related diseases shared similar alteration and defects in mitochondrial morphology. Furthermore, Lycium ruthenicum Murr. extract, a bioactive food substance with cognition-improving property, could effectively improve muscle strength and increase body weight in the Slc25a46 knockout mice. These findings suggest that mitochondrial morphology defects in mice models, particularly in the mitochondrial compartment, represent a unified and effective marker for screening and validating natural product-derived functional substances with mitochondrial protective properties. | 20 | Identification of the core promoter of STK11 gene and its transcriptional regulation by p53显示文摘Peutz-Jeghers syndrome (PJS) is an autosomal dominant disease characterized by mucocutaneous pigmentation and hamartomatous polyps. Most cases of PJS involve the inactivation of germline mutations in the serine/threonine kinase gene STK11 which is also known as LKB1. The function of STK11 was previously linked to the tumor suppressor p53 and was shown to activate the p53 target p21/ WAF1. Recently, STK11 was reported to be interacting with p53 physically in the nucleus and it can directly or indirectly phosphorylate p53. Here we characterized the 50-flanking region of human STK11 gene and identified a 161-bp fragment with promoter activity. Sequence analysis, mutagenesis and gel shift studies revealed a binding site of Sp1 and p53, which affects the promoter activity. Mutation analyses showed that this fragment was required for p53-mediated transcriptional activation. This transcriptional activation was further confirmed by real-time quantitative RT-PCR and Western blot analysis. Transient transfection of p53 expression plasmid into fetal liver cell lines increased STK11 mRNA and protein levels.
