3Variation in allelic frequencies at loci associated with kernel weight and their effects on kernel weight-related traits in winter wheat显示文摘Knowledge of allelic frequencies at loci associated with kernel weight and effects on kernel weight-related traits is crucial for yield improvement in wheat. Kernel weight-related traits were evaluated in 200 Chinese winter wheat cultivars(lines) grown at the Xinxiang Experimental Station, Chinese Academy of Agricultural Sciences, Xinxiang in Henan Province, for three consecutive years from 2014 to 2016. Alleles associated with kernel weight at nine loci, TaCKX6-D1, TaCwi-A1, TaCWI-4A, TaGS1a, TaGS5-A1, TaGS3-3A, TaGW2-6A, TaSus2-2B, and TaTGW6-A1, were determined for all cultivars(lines). ANOVA showed that genotypes, years and their interactions had significant effects on thousand-kernel weight(TKW), kernel length(KL) and kernel width(KW). The overall mean frequencies of alleles conferring high and low TKW at the nine loci were 65.9% and 33.4%, with the ranges of 37.0%–85.0% and 13.5%–63.0% for single loci. The frequencies of high-TKW alleles were over 50.0% at eight of the loci. Genotypes at each locus with the high-TKW allele had higher TKW than those with the low-TKW allele. The high-TKW allele Hap-H at the TaSus2-2B locus can be preferably used to increase grain yield due to its high TKW(49.32 g). A total of 18 main allelic combinations(ACs) at nine loci were detected. Three ACs(AC1–AC3) had significantly higher TKW than AC6 with high-TKW alleles at all nine loci even though they contained some low-TKW alleles. This indicated that other loci controlling kernel weight were present in the high-TKW cultivars. 6Deciphering structural and functional roles of disulfide bonds in decorsin显示文摘Decorsin,an antagonist of integrin glycoprotein IIb/IIIa,contains Arg-Gly-Asp(RGD)sequence and three disulfide bridges.The function of RGD sequence has already been well defined,but the roles of conserved disulfide bonds in antihemostatic proteins still remain unclear.Herein we use the fusion expression and characterization of mutant decorsin to study the functions of disulfide bonds in protein structure,stability and biological activity.The purified protein shows an apparent inhibition of activity to platelet aggregation induced by ADP with IC50 of 500 nM.The removal of cys7-cys15(from cysteine to serine)at the N-terminal causes a thirty-fold decrease of the inhibition activity with IC50 of 15 M,whereas the mutation of cys22-cys38 at the C-terminal completely impairs the biological activity of decorsin.The overall secondary and tertiary structures of decorsin are disrupted inevitably without disulfide bonds.Using a domain insertion mutation,the retaining of RGD loop and the adjacent disulfide bond produces a week antihemostatic activity of decorsin.This reveals that the overall structure of decorsin stabilized by the three conserved disulfide bridges is cooperative for antihemostatic function.Our study on the effect of disulfide bonds together with RGD-sequence on the protein function is helpful for structure-based drug design of antithrombotic research.WU LingZhi LI Ying YANG Yang QIN Meng 2013Science China Chemistry2013,56,10:1

9Thermodynamical properties of protein kinase with adenine inhibitors显示文摘The protein-based molecular recognition of the adenine ring is essential to understand protein function and drug design as well.In this paper,a variety of the adenine-based inhibitors with modified groups of amino groups,nitrogen and oxygen atoms in the aromatic ring are designed,and the binding capability of these adenosine analogues with an aminoglycoside antibiotic kinase [APH(3')-IIIa] are investigated with activity assays and isothermal titration calorimetry(ITC) experiments.1-aminoisoquinoline is one of the weakest substrates bound to APH(3')-IIIa with the lowest affinity(high ki and high kd) and the smallest negative value of free energy change(G) among the inhibitors tested.The binding process of adenine and 5-nitroisoquinoline to APH(3')-IIIa is an enthalpy-driven event with unfavorable entropy,which is consistent with the energy change induced by the binding of ATP to the enzyme.However,the reverse is true for 1-aminoisoquinoline,3-amino-5-nitrobenzisothiazole,5-aminoisoquinoline binding to the enzyme because the entropy is more favorable and the enthalpy makes a lower contribution to the binding process.These results suggest that small changes of the adenine ring can lead to significant influence on the ability of these analogues to occupy the adenine-binding region of the enzyme,which can be the potential inhibitors as drug candidates against the bacterial resistance.WU LingZhi HU Dong TANG LiHua HU XiaoFei QIN Meng 2013Chinese Science Bulletin2013,58,1:1

11In-Fiber Thermally Diffused Coupler and Fiber Bragg Grating Inscribed in Twin-Core Fiber for Sensitivity-Enhanced Vector Bending Sensing显示文摘A vector bending fiber sensor based on core-by-core inscribed fiber Bragg gratings in a twin-core fiber has been proposed and experimentally demonstrated.An in-fiber integrated vector bending sensor is realized by using the thermal diffusion technique to fabricate the coupler.The characteristics of the coupler fabricated by thermal diffusion are simulated and experimented.By inscribing fiber Bragg gratings with different reflection wavelengths in the two cores of a symmetrical twin-core fiber,the curvature sensitivity can be enhanced by tracking the wavelength difference between the fiber Bragg gratings of the two cores.The measured bending sensitivity of the fiber Bragg grating ranges from–161.6 pm/m^(−1) to+165.5 pm/m^(−1).The differential sensitivity of the two cores is twice that of a conventional single grating,and the temperature-induced crosstalk is also reduced.The bending sensor proposed in this paper has the advantages of high integration,enhancing the sensitivity and two-dimensional orientation recognizability,and reducing temperature crosstalk,which can be a promising candidate for structural health monitoring or wearable artificial electronics applications.Lingzhi MENG Hongye WANG Qi XIA Tingting YUAN Xiaotong ZHANG Libo YUAN 2023Photonic Sensors2023,13,3:0

13Induced temperature-dependent DNA degradation by C_(60) without photoactivation显示文摘This study shows that C_(60) can degrade pBR322 plasmid DNA at room temperature without photoactivation. The degradation was enhanced by increasing incubation temperature, reaction time or C_(60) concentration. We also found that superoxide radical anions (O 2 · ) were formed in the C_(60) solution. Superoxide dismutase significantly inhibited DNA cleavage and O 2 · generation induced by C_(60) . These results suggest that DNA cleavage was caused by the formation of reactive oxygen species induced by C_(60) at room temperature. Furthermore, we demonstrate that the DNA degradation was significantly inhibited by acid amide chemicals such as formamide, and by increased ionic strength of the reaction solution. These results indicate that the DNA conformation stability and the surface properties of C_(60) are important factors regulating DNA degradation. We propose that C_(60) can bind DNA, decrease DNA conformation stability, and induce the formation of reactive oxygen species and DNA cleavage at room temperature.
