4Loss of BRCA1 expression leads to worse survival in patients with gastric carcinoma显示文摘AIM: To investigate the expression deficiency of key molecular markers in the homologous recombination pathway. METHODS: Expression loss of breast cancer type 1 susceptibility protein (BRCA1), ataxia telangiectasia mutated (ATM), ATM-Rad3-related (ATR), mediator of DNA damage checkpoint protein 1 (MDC1) and meiotic recombination 11 (Mre11) were correlated with their clinicopathological parameters in gastric cancer (GC). One hundred and twenty treatment-naive GC samples were formalin-fixed and paraffin-embedded into tissue blocks. Two representative cores from each block were extracted and constructed into tissue microarrays. Expression levels of BRCA1, ATM, ATR, MDC1 and Mre11 were determined using immunohistochemical analysis, and correlated with clinical parameters, including age, gender, Lauren subtype, tumor grades, clinical stage and overall survival.RESULTS: Expression loss of BRCA1, ATM, ATR, MDC1, and Mre11 was found in 21.4%, 20.2%, 21.0%, 11.1% and 4.6%, respectively, of interpretable cases. BRCA1 loss was significantly associated with patients of diffused subtype (intestinal vs diffused, 8.2% vs 31.7%, P = 0.001), higher tumor grade (Ⅰ/Ⅱ vs Ⅲ, 10.7% vs 20.5;Ⅰ/Ⅱ vs Ⅳ, 10.7% vs 54.5%, P = 0.047) and advanced clinical stage (Ⅰ/Ⅱ vs Ⅲ, 12.9% vs 16.9%;Ⅰ /Ⅱ vs Ⅳ, 12.9% vs 45.5%, P = 0.006). MDC1 loss was significantly associated with patients of diffused subtype (intestinal vs diffused, 0% vs 19.7%, P = 0.001) and higher tumor grade (Ⅰ/Ⅱ vs Ⅲ, 0% vs 12%;Ⅰ/Ⅱ vs Ⅳ, 0% vs 30.8%, P = 0.012). In addition, the survival time of the patients with expression loss of BRCA1 was significantly shorter than those with positive expression of BRCA1 (2-year survival rate, 32.4% vs 62.8%, P = 0.015). No correlations were found between clinicopathological parameters and expression loss of ATM, ATR and Mre11. CONCLUSION: Our results support the hypothesis that homologous recombination deficiency plays an important role in the progression of gastric carcinoma. 5Exogenous Application of Abscisic Acid or Gibberellin Acid Has Different Effects on Starch Granule Size Distribution in Grains of Wheat显示文摘Granule size distribution of wheat starch is an important characteristic that can affect its chemical composition and the functionality of wheat products. Two high-yield winter wheat cultivars were used to evaluate the effects of the application of exogenous ABA or GA during the reproductive phase of the initial grain filling on starch granule size distribution and starch components in grains at maturity. The results indicated that a bimodal curve was found in the volume and surface area distribution of grain starch granules, and a unimodal curve was observed for the number distribution under all treatments. The exogenous ABA resulted in a significant increase in the proportions (both by volume and by surface area) of B-type (<9.9 μm in diameter) starch granules, with a reduction in those of A-type (>9.9 μm) starch granules, while, the exogenous GA_3 led to converse effects on size distribution of those starch granules. The exogenous ABA also increased starch, amylose and amylopectin contents at maturity but significantly reduced the ratio of amylose to amylopectin. Application of GA_3 significantly reduced starch content, amylopectin content but increased the ratio of amylose to amylopectin. 6High-performance silicon−graphene hybrid plasmonic waveguide photodetectors beyond 1.55μm显示文摘Graphene has attracted much attention for the realization of high-speed photodetection for silicon photonics over a wide wavelength range.However,the reported fast graphene photodetectors mainly operate in the 1.55μm wavelength band.In this work,we propose and realize high-performance waveguide photodetectors based on bolometric/photoconductive effects by introducing an ultrathin wide silicon−graphene hybrid plasmonic waveguide,which enables efficient light absorption in graphene at 1.55μm and beyond.When operating at 2μm,the present photodetector has a responsivity of ~70 mA/W and a setup-limited 3 dB bandwidth of >20 GHz.When operating at 1.55μm,the present photodetector also works very well with a broad 3 dB bandwidth of >40 GHz(setup-limited)and a high responsivity of ~0.4 A/W even with a low bias voltage of−0.3 V.This work paves the way for achieving highresponsivity and high-speed silicon-graphene waveguide photodetection in the near/mid-infrared ranges,which has applications in optical communications,nonlinear photonics,and on-chip sensing.Jingshu Guo Jiang Li Chaoyue Liu Yanlong Yin Wenhui Wang Zhenhua Ni Zhilei Fu Hui Yu Yang Xu Yaocheng Shi Yungui Ma Shiming Gao Limin Tong Daoxin Dai 2020Light(Science & Applications)2020,9,1:10

8Rat model of cholelithiasis with human gallstones implanted in cholestasis-induced virtual gallbladder显示文摘AIM:To facilitate translational research on cholelithiasis,we have developed a rat model of human gallstones by exploiting the unique biliopancreatic features of this species.METHODS:Under anesthesia,16 adult rats of equal genders underwent two times of abdominal surgery.First,their common bile duct(CBD)was ligated to cause cholestasis by total biliary obstruction(TBO).On day 0,1,3,7,14,21 and 28 after TBO,magnetic resonance imaging(MRI)was conducted to monitor the dilatation of the CBD,and blood was sampled to analyze total serum bilirubin(TSB).Secondly,on day 30,the abdomen was re-opened and gallstone(s)collected from human patients were implanted in the dilated CBD asa virtual gallbladder(VGB),which was closed by suture ligation.This rat cholelithiasis model was examined by MRI,clinical observation,microcholangiography and histology.RESULTS:All rats survived two laparotomies.After ligation,the CBD was dilated to a stable size of 4 to30 mm in diameter on day 21-28,which became a VGB.The rats initially showed signs of jaundice that diminished over time,which paralleled with the evolving TSB levels from 0.6±0.3 mg/d L before ligation,through a peak of 10.9±1.9 mg/dL on day 14,until a nearly normalized value after day 28.The dilated CBD with thickened wall allowed an incision for implantation of human gallstones of 1-10 mm in diameter.The rat cholelithiasis was proven by in vivo MRI and postmortem microcholangiography and histomorphology.CONCLUSION:A rat model cholelithiasis with human gallstones has been established,which proves feasible,safe,reliable,nontoxic and cost-effective.Given the gallstones of human origin,applications of this model may be of help in translational research such as optical detection and lysis of gallstones by systemic drug administration.Marlein Miranda Cona Yewei Liu Ting Yin Yuanbo Feng Feng Chen Stefaan Mulier Yue Li Jian Zhang Raymond Oyen Yicheng Ni 2016World Journal of Methodology2016,6,2:7
9Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 10KLF4 initiates sustained YAP activation to promote renal fibrosis in mice after ischemia-reperfusion kidney injury显示文摘Acute renal injury(AKI)causes a long-term risk for progressing into chronic kidney disease(CKD)and interstitial fibrosis.Yes-associated protein(YAP),a key transcriptional cofactor in Hippo signaling pathway,shuttles between the cytoplasm and nucleus,which is required for the renal tubular epithelial cells repair in the acute phase of AKI.In this study we investigated the role of YAP during ischemia-reperfusion(IR)-induced AKI to CKD.Mice were subjected to left kidney IR followed by removal of the right kidney on the day before tissue harvests.Mouse shRNA expression adenovirus(Ad-shYAP or Ad-shKLF4)and mouse KLF4 expression adenovirus(Ad-KLF4)were delivered to mice by intrarenal injection on D7 after IR.We showed that the expression and nucleus distribution of YAP were persistently increased until the end of experiment(D21 after IR).The sustained activation of YAP in post-acute phase of AKI was accompanied by renal dysfunction and interstitial fibrosis.Knockdown of YAP significantly attenuated IR-induced renal dysfunction and decreased the expression of fibrogenic factors TGF-βand CTGF in the kidney.We showed that the expression of the transcription factor KLF4,lined on the upstream of YAP,was also persistently increased.Knockdown on KLF4 attenuated YAP increase and nuclear translocation as well as renal functional deterioration and interstitial fibrosis in IR mice,whereas KLF4 overexpression caused opposite effects.KLF4 increased the expression of ITCH,and ITCH facilitated YAP nuclear translocation via degrading LATS1.Furthermore,we demonstrated in primary cultured renal tubular cells that KLF4 bound to the promoter region of YAP and positively regulates YAP expression.In biopsy sample from CKD patients,we also observed increased expression and nuclear distribution of YAP.In conclusion,the activation of YAP in the post-acute phase of AKI is implicated in renal functional deterioration and fibrosis although it exhibits beneficial effect in acute phase.Reprogramming factor KLF4 is responsible for the persistent activation of YAP.Blocking the activation of KLF4-YAP pathway might be a way to prevent the transition of AKI into CKD.Dan Xu Pan-pan Chen Pei-qing Zheng Fan Yin Qian Cheng Zhuan-li Zhou Hong-yan Xie Jing-yao Li Jia-yun Ni Yan-zhe Wang Si-jia Chen Li Zhou Xiao-xia Wang Jun Liu Wei Zhang Li-min Lu 2021Acta Pharmacologica Sinica2021,42,3:6
11PhERF2,an ethylene-responsive element binding factor,plays an essential role in waterlogging tolerance of petunia显示文摘Ethylene-responsive element binding factors(ERFs)are involved in regulation of various stress responses in plants,but their biological functions in waterlogging stress are largely unclear.In this study,we identified a petunia(Petunia×hybrida)ERF gene,PhERF2,that was significantly induced by waterlogging in wild-type(WT).To study the regulatory role of PhERF2 in waterlogging responses,transgenic petunia plants with RNAi silencing and overexpression of PhERF2 were generated.Compared with WT plants,PhERF2 silencing compromised the tolerance of petunia seedlings to waterlogging,shown as 96%mortality after 4 days waterlogging and 14 days recovery,while overexpression of PhERF2 improved the survival of seedlings subjected to waterlogging.PhERF2-RNAi lines exhibited earlier and more severe leaf chlorosis and necrosis than WT,whereas plants overexpressing PhERF2 showed promoted growth vigor under waterlogging.Chlorophyll content was dramatically lower in PhERF2-silenced plants than WT or overexpression plants.Typical characteristics of programmed cell death(PCD),DNA condensation,and moon-shaped nuclei were only observed in PhERF2-overexpressing lines but not in PhERF2-RNAi or control lines.Furthermore,transcript abundances of the alcoholic fermentation-related genes ADH1-1,ADH1-2,ADH1-3,PDC1,and PDC2 were reduced in PhERF2-silenced plants,but increased in PhERF2-overexpressing plants following exposure to 12-h waterlogging.In contrast,expression of the lactate fermentation-related gene LDH was up-regulated in PhERF2-silenced plants,but downregulated in its overexpressing plants.Moreover,PhERF2 was observed to directly bind to the ADH1-2 promoter bearing ATCTA motifs.Our results demonstrate that PhERF2 contributes to petunia waterlogging tolerance through modulation of PCD and alcoholic fermentation system.Dongmei Yin Daoyang Sun Zhuqing Han Dian Ni Ayla Norris Cai-Zhong Jiang 2019Horticulture Research2019,6,1:6
12Caspase-11 promotes NLRP3 inflammasome activation via the cleavage of pannexin1 in acute kidney disease显示文摘Ischemia/reperfusion(I/R)injury is a major cause of acute kidney injury(AKI)in clinic.The activation of NLRP3 inflammasome is associated with inflammation and renal injury in I/R-induced AKI.In the current study we explored the molecular and cellular mechanisms for NLRP3 inflammasome activation following renal I/R.Mice were subjected to I/R renal injury by clamping bilateral renal pedicles.We showed that I/R injury markedly increased caspase-11 expression and the cleavage of pannexin 1(panx1)in the kidneys accompanied by NLRP3 inflammasome activation evidenced by the activation of caspase-1 and interlukin-1β(IL-1β)maturation.In Casp-11^(−/−)mice,I/R-induced panx1 cleavage,NLRP3 inflammasome activation as well as renal functional deterioration and tubular morphological changes were significantly attenuated.In cultured primary tubular cells(PTCs)and NRK-52E cells,hypoxia/reoxygenation(H/R)markedly increased caspase-11 expression,NLRP3 inflammasome activation,IL-1βmaturation and panx1 cleavage.Knockdown of caspase-11 attenuated all those changes;similar effects were observed in PTCs isolated from Casp-11^(−/−)mice.In NRK-52E cells,overexpression of caspase-11 promoted panx1 cleavage;pretreatment with panx1 inhibitor carbenoxolone or knockdown of panx1 significantly attenuated H/R-induced intracellular ATP reduction,extracellular ATP elevation and NLRP3 inflammasome activation without apparent influence on H/R-induced caspase-11 increase;pretreatment with P2X7 receptor inhibitor AZD9056 also attenuated NLRP3 inflammasome activation.The above results demonstrate that the cleavage of panx1 by upregulated caspase-11 is involved in facilitating ATP release and then NLRP3 inflammasome activation in I/R-induced AKI.This study provides new insight into the molecular mechanism of NLRP3 inflammasome activation in AKI.Fan Yin Pei-qing Zheng Liu-qi Zhao Yan-zhe Wang Nai-jun Miao Zhuan-li Zhou Qian Cheng Pan-pan Chen Hong-yan Xie Jing-yao Li Jia-yun Ni Li Zhou Wei Zhang Xiao-xia Wang Jun Liu Li-min Lu 2022Acta Pharmacologica Sinica2022,43,1:5

14Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats.Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang 2022Science China(Life Sciences)2022,65,10:5
15Real-time embedded software testing method based on extended finite state machine显示文摘The reliability of real-time embedded software directly determines the reliability of the whole real-time embedded system,and the effective software testing is an important way to ensure software quality and reliability.Based on the analysis of the characteristics of real-time embedded software,the formal method is introduced into the real-time embedded software testing field and the real-time extended finite state machine (RT-EFSM) model is studied firstly.Then,the time zone division method of real-time embedded system is presented and the definition and description methods of time-constrained transition equivalence class (timeCTEC) are presented.Furthermore,the approaches of the testing sequence and test case generation are put forward.Finally,the proposed method is applied to a typical avionics realtime embedded software testing practice and the examples of the timeCTEC,testing sequences and test cases are given.With the analysis of the testing result,the application verification shows that the proposed method can effectively describe the real-time embedded software state transition characteristics and real-time requirements and play the advantages of the formal methods in accuracy,effectiveness and the automation supporting.Combined with the testing platform,the real-time,closed loop and automated simulation testing for real-time embedded software can be realized effectively.Yongfeng Yin Bin Liu Hongying Ni 2012Journal of Systems Engineering and Electronics2012,23,2:5

17Adsorption of 17β-estradiol from aqueous solution by raw and direct/pre/post-KOH treated lotus seedpod biochar显示文摘Five biochars derived from lotus seedpod(LSP)were applied to examine and compare the adsorption capacity of 17β-estradiol(E2)from aqueous solution.The effect of KOH activation and the order of activation steps on material properties were discussed.The effect of contact time,initial concentration,p H,ionic strength and humic acid on E2 adsorption were investigated in a batch adsorption process.Experimental results demonstrated that the pseudo second-order model fitted the experimental data best and that adsorption equilibrium was reached within 20 hr.The efficiency of E2 removal increased with increasing E2 concentration and decreased with the increase of ionic strength.E2 adsorption on LSP-derived biochar(BCs)was influenced little by humic acid,and slightly affected by the solution p H when its value ranged from 4.0 to 9.0,but considerably affected at p H 10.0.Low environmental temperature is favorable for E2 adsorption.Chemisorption,π–πinteractions,monolayer adsorption and electrostatic interaction are the possible adsorption mechanisms.Comparative studies indicated that KOH activation and the order of activation steps had significant impacts on the material.Post-treated biochar exhibited better adsorption capacity for E2 than direct treated,pretreated,and raw LSP biochar.Pyrolyzed biochar at higher temperature improved E2 removal.The excellent performance of BCs in removing E2 suggested that BCs have potential in E2 treatment and that the biochar directly treated by KOH would be a good choice for the treatment of E2 in aqueous solution,with its advantages of good efficiency and simple technology.Ni Liu Yunguo Liu Guangming Zeng Jilai Gong Xiaofei Tan Jun Wen Shaobo Liu Luhua Jiang Meifang Li Zhihong Yin 2020Journal of Environmental Sciences2020,32,1:4

19Fibroblast growth factor receptor 4 Gly388Arg polymorphism in Chinese gastric cancer patients显示文摘AIM: To investigate the contribution of the fibroblast growth factor receptor 4 (FGFR4) Gly388Arg polymorphism as a genetic risk factor for gastric cancer (GC) and to investigate any associations between this polymorphism and clinicopathological parameters and survival. METHODS: Tumors and matched adjacent non-cancer tissues were collected from 304 GC patients, and 5 mL of venous blood was collected from 62 GC patients and 392 ageand sex-matched healthy controls without cancer history from the same ethnic population. DNA was extracted, and direct sequencing analyses were performed to genotype the FGFR4 Gly388Arg polymorphism in all the samples. Differences in the genotype frequencies of the FGFR4 Gly388Arg polymorphism between GC patients and healthy controls were estimated using the χ 2 test. Binary logistic regression was used for all analysis variables to estimate risk as the ORs with 95%CIs. The relationships between the FGFR4 genotype and clinicopathological parameters were tested with the χ 2 test. The Kaplan-Meier product-limit method, the log-rank test, and the Cox regression model were applied to evaluate the effect of the FGFR4 genotype on the overall survival of patients with GC. RESULTS: In the present GC cohort, 118 patients (38.8%) were homozygous for the Gly388 allele, 124 patients (40.8%) were heterozygous, and 62 patients (20.4%) were homozygous for the Arg388 allele. The frequencies of the Gly/Gly, Gly/Arg, and Arg/Arg genotypes in the healthy controls were 33.6%, 48.0%, and 18.4%, respectively. The distributions of genotypes (χ 2 = 3.589, P = 0.166) and alleles (χ 2 = 0.342, P = 0.559) of the FGFR4 Gly388Arg polymorphism were not different between the GC patients and the healthy controls. Although we observed no correlation between the FGFR4 Gly388Arg polymorphism and clinicopathological parameters or survival in the total cohort of GC patients, the presence of the Arg388 allele was associated with shorter survival time in patients with GC if the tumor was small (log rank χ 2 = 5.449, P = 0.020), well differentiated (log rank χ 2 = 12.798, P = 0.000), T1 or T2 stage (log rank χ 2 = 4.745, P = 0.029), without lymph node involvement (log rank χ 2 = 6.647, P = 0.010), and at an early clinical stage (log rank χ 2 = 4.615, P = 0.032).
