| 4 | Antioxidant and Anti-aging Activities of Silybum Marianum Protein Hydrolysate in Mice Treated with D-galactose显示文摘Objective In the present study, we investigated the antioxidant and anti‐aging effects of Silybum marianum protein hydrolysate(SMPH) in D‐galactose‐treated mice. Methods D‐galactose(500 mg/kg body weight) was intraperitoneally injected daily for 7 weeks to accelerate aging, and SMPH(400, 800, 1,200 mg/kg body weight, respectively) was simultaneously administered orally. The antioxidant and anti‐aging effects of SMPH in the liver and brain were measured by biochemical assays. Transmission electron microscopy(TEM) was performed to study the ultrastructure of liver mitochondria. Results SMPH decreased triglyceride and cholesterol levels in the D‐galactose‐treated mice. It significantly elevated the activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH‐Px), and total antioxidant capacity(T‐AOC), which were suppressed by D‐galactose. Monoamine oxidase(MAO) and malondialdehyde(MDA) levels as well as the concentrations of caspase‐3 and 8‐OHd G in the liver and brain were significantly reduced by SMPH. Moreover, it increased Bcl‐2 levels in the liver and brain. Furthermore, SMPH significantly attenuated D‐galactose‐induced liver mitochondrial dysfunction by improving the activities of Na+‐K+‐ATPase and Ca2+‐Mg2+‐ATPase as well as mitochondrial membrane potential(ΔΨm) and fluidity. TEM showed that the degree of liver mitochondrial damage was significantly decreased by SMPH. | 8 | Acupuncture-movement therapy for acute lumbar sprain: a randomized controlled clinical trial显示文摘OBJECTIVE: Several studies have reported that acupuncture is effective for treatment of acute lumbar sprain, but they neglected to consider that acupuncture cannot remarkably improve lumbar activity. We performed a randomized controlled trial to evaluate the effect of acupuncture-movement therapy versus conventional acupuncture in the treatment of acute lumbar sprain.METHODS: Sixty patients were randomized into four groups: the acupuncture-movement(AM)group, sham acupuncture-movement(SAM) group,conventional acupuncture(CA) group, and physical therapy(PT) group. Patients in the AM group were treated with acupuncture at Yintang(EX-HN 3) and exercise of the lumbar region during acupuncture.Patients in the SAM group were treated with sham acupuncture at Yintang(EX-HN 3) and exercise of the lumbar region during sham acupuncture. Conventional acupuncture was performed in the CA group, and physical therapy was applied in the PT group. Each treatment lasted for 20 min. Patients were assessed before and after treatment using a visual analogue scale(VAS) and the Roland Morris Questionnaire(RMQ).RESULTS: The VAS and RMQ scores in the AM group were significantly lower after than before treatment(P < 0.01). The AM group reported lower RMQ scores in after-treatment and 24 h after treatment and lower VAS score in 24 h after treatment in comparison with those of the CA, SAM, and PT groups(P < 0.05).CONCULSION: Acupuncture-movement therapy is effective for treating acute lumbar sprain with a persistent pain-relief and remarkable improvement of lumbar activity. | 10 | Prognostic value of hypoxia-inducible factor-1 alpha and prolyl 4-hydroxylase beta polypeptide overexpression in gastric cancer显示文摘AIM To investigate the relationship between hypoxia-inducible factor-1α(HIF-1α), prolyl 4-hydroxylase beta(P4 HB) expression, and clinicopathologic parameters, as well as the prognostic value of these genes for patients with gastric cancer(Gc).METHODS Hypoxia is a critical factor that shapes the Gc microenvironment. In previous reports, we have demonstrated that P4 HB is a potential target of HIF-1α. In the present study, gene expression profiling interactive analysis(GEPIA) was used to analyze the relationship between P4 HB and hypoxia-associated genes. To this end, 428 Gc tissue samples were used to analyze the expression of HIF-1α and P4 HB via immunohistochemical staining. Patient samples were classified as having weak-expression or over-expression both in terms of HIF-1α and P4 HB. Correlations between biomarkers and clinicopathological factors were analyzed to predict survival. RESULTS P4 HB demonstrated a positive correlation with hypoxiaassociated genes(P < 0.05). HIF-1α and P4 HB overexpression have a significant correlation with TNM staging(χ2 = 23.32, P = 0.00; χ2 = 65.64, P = 0.00) and peritoneum cavity metastasis(χ2 = 12.67, P = 0.00; χ2 = 39.29, P = 0.00). In univariate analysis, patients with a high HIF-1α expression trend had a shorter disease-free survival(DFS: 44.80 mo vs 22.06 mo) and overall survival(OS: 49.58 mo vs 39.92 mo). P4 HB overexpression reflected similar results: patients with over-expression of P4 HB had a shorter survival time than those with weak-expression(DFS: 48.03 mo vs 29.64 mo, OS: 52.48 mo vs 36.87 mo). Furthermore, HIF-1α is also a clinicopathological predictor of dismal prognosis according to multivariate analysis(DFS, 95%c I: 0.52-0.88, P < 0.00; OS, 95%c I: 0.50-0.85, P < 0.00). However, P4 HB was meaningful in DFS(95%c I: 0.58-1.00, P < 0.05) but not in OS(95%c I: 0.72-1.23, P > 0.05).CONCLUSION Overexpression of HIF-1α and P4 HB is associated with poor prognosis in patients with Gc. | 11 | Exploratory clinical study of chidamide,an oral subtype-selective histone deacetylase inhibitor,in combination with exemestane in hormone receptor-positive advanced breast cancer显示文摘Objective: The recurrence or progression under endocrine therapy in hormone receptor-positive(HR+)advanced breast cancer(ABC) remained a critical clinical challenge.Chidamide is an oral subtype-selective histone deacetylase(HDAC) inhibitor with multiple functions in tumor growth inhibition and microenvironment modulation via epigenetic reprogramming.The purpose of this study was to evaluate the safety,pharmacokinetics(PK),and preliminary efficacy of chidamide in combination with exemestane in HR+ ABC patients.Methods: Eligible patients were postmenopausal women with HR+ ABC recurrent or progressed to at least one endocrine therapy.Blood samples were obtained in the run-in period and the first day of combination treatment for PK analysis.In combination treatment,patients were given exemestane 25mg daily and chidamide 30mg twice a week(BIW) until progression of disease or intolerable toxicities.A treatment cycle was defined as 4 weeks.Safety,PK parameters,and preliminary efficacy were evaluated.Results: A total of 20 patients were enrolled between July and December,2015.The median number of treatments cycle was 5.2(20.8 weeks) with 2 patients still on treatment at the data cut-off date of October,2017.The treatment-related adverse events(AE) ≥ grade 3 in more than 2 patients were neutropenia(35%),thrombocytopenia(30%),and leucopenia(20%).The plasma exposure of exemestane was consistent in the presence or absence of chidamide.A slight increase in chidamide exposure was noted in the presence of exemestane,probably due to the inter-and intra-patient variations.The best response in 16 evaluable patients was assessed by Response Evaluation Criteria in Solid Tumors(RECIST),including 4 patients with partial response,10 patients with stable disease.The median progression-free survival(PFS) was 7.6 months.Conclusions: The combination of chidamide with exemestane was generally well tolerated with promising preliminary efficacy in HR+ ABC patients.The overall results from this study encourage further pivotal trial in this patient population. | Qingyuan Zhang Tao Wang Cuizhi Geng Yue Zhang Jinwen Zhang Zhiqiang Ning Zefei Jiang | 2018 | Chinese Journal of Cancer Research2018,30,6: | 11 |

Helminth parasites are the classic inducers of Th2 responses. The Th2-polarized T cell response driven by helminth infection has been linked to the attenuation of some damaging Th1 driven inflammatory responses, preventing some Th1-mediated autoimmune diseases in the host, including experimentally induced colitis. Helminth parasites (the porcine whipworm, Trichuris suis ) have been tested for treating IBD patients, resulting in clinical amelioration of the disease. As a result, there is a great deal of interest in the research community in exploring the therapeutic use of helminth parasites for the control of immune-mediated diseases, including IBD. However, recent studies have provided evidence indicating the exacerbating effects of helminths on bacterial as well as non-infectious colitis in animal models. | 15 | Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients. | Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo | 2021 | Cancer Biology & Medicine2021,18,1: | 10 |

| 17 | Protective effects of ACLF sera on metabolic functions and proliferation of hepatocytes co-cultured with bone marrow MSCs in vitro显示文摘AIM:To investigate whether the function of hepatocytes co-cultured with bone marrow mesenchymal stem cells (MSCs) could be maintained in serum from acute-onchronic liver failure (ACLF) patients.METHODS:Hepatocyte supportive functions and cytotoxicity of sera from 18 patients with viral hepatitis B-induced ACLF and 18 healthy volunteers were evaluated for porcine hepatocytes co-cultured with MSCs and hepatocyte mono-layered culture,respectively.Chemokine profile was also examined for the normal serum and liver failure serum.RESULTS:Hepatocyte growth factor (HGF) and Tumor necrosis factor;tumor necrosis factor (TNF)-α were remarkably elevated in response to ACLF while epidermal growth factor (EGF) and VEGF levels were significantly decreased.Liver failure serum samples induced a higher detachment rate,lower viability and decreased liver support functions in the homo-hepatocyte culture.Hepatocytes co-cultured with MSCs could tolerate the cytotoxicity of the serum from ACLF patients and had similar liver support functions compared with the hepatocytes cultured with healthy human serum in vitro.In addition,cocultured hepatocytes maintained a proliferative capability despite of the insult from liver failure serum.CONCLUSION:ACLF serum does not impair the cell morphology,viability,proliferation and overall metabolic capacities of hepatocyte co-cultured with MSCs in vitro. | Xiao-Lei Shi Jin-Yang Gu Yue Zhang Bing Han Jiang-Qiang xiao Xian-Wen Yuan Ning Zhang Yi-Tao Ding | 2011 | World Journal of Gastroenterology2011,17,19: | 8 |

| 19 | Stability improvement under high efficiency—next stage development of perovskite solar cells显示文摘With efficiency of perovskite solar cells(PSCs) overpassing 23%, to realize their commercialization, the biggest challenge now is to boost the stability to the same level as conventional solar cells. Thus, tremendous effort has been directed over the past few years toward improving the stability of these cells. Various methods were used to improve the stability of bulk perovskites,including compositional engineering, interface adjustment, dimensional manipulation, crystal engineering, and grain boundary decoration. Diverse device configurations, carrier transporting layers, and counter electrodes are investigated. To compare the stability of PSCs and clarify the degradation mechanism, diverse characterization methods were developed. Overall stability of PSCs has become one central topic for the development of PSCs. | 20 | Distributed intrusion detection for mobile ad hoc networks显示文摘Mobile ad hoc networking (MANET) has become an exciting and important technology in recent years,because of the rapid proliferation of wireless devices.Mobile ad hoc networks is highly vulnerable to attacks due to the open medium,dynamically changing network topology,cooperative algorithms,and lack of centralized monitoring and management point.The traditional way of protecting networks with firewalls and encryption software is no longer suffcient and effective for those features.A distributed intrusion detection approach based on timed automata is given.A cluster-based detection scheme is presented,where periodically a node is elected as the monitor node for a cluster.These monitor nodes can not only make local intrusion detection decisions,but also cooperatively take part in global intrusion detection.And then the timed automata is constructed by the way of manually abstracting the correct behaviours of the node according to the routing protocol of dynamic source routing (DSR).The monitor nodes can verify the behaviour of every nodes by timed automata,and validly detect real-time attacks without signatures of intrusion or trained data.Compared with the architecture where each node is its own IDS agent,the approach is much more effcient while maintaining the same level of effectiveness.Finally,the intrusion detection method is evaluated through simulation experiments. | Yi Ping Jiang Xinghao Wu Yue Liu Ning | 2008 | Journal of Systems Engineering and Electronics2008,19,4: | 7 |
