1Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection.Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng 2018Cellular & Molecular Immunology2018,15,3:13
2Online adaptive Q-learning method for fully cooperative linear quadratic dynamic games显示文摘A model-based offline policy iteration(PI) algorithm and a model-free online Q-learning algorithm are proposed for solving fully cooperative linear quadratic dynamic games. The PI-based adaptive Q-learning method can learn the feedback Nash equilibrium online using the state samples generated by behavior policies, without sending inquiries to the system model. Unlike the existing Q-learning methods, this novel Q-learning algorithm executes both policy evaluation and policy improvement in an adaptive manner.We prove the convergence of the offline PI algorithm by proving its equivalence to Newton’s method while solving the game algebraic Riccati equation(GARE). Furthermore, we prove that the proposed Q-learning method will converge to the Nash equilibrium under a small learning rate if the method satisfies certain persistence of excitation conditions, which can be easily met by suitable behavior policies. 396-Week Treatment of Tenofovir Amibufenamide and Tenofovir Disoproxil Fumarate in Chronic Hepatitis B Patients显示文摘Background and Aims:Tenofovir amibufenamide(TMF)is a novel phosphoramidated prodrug of tenofovir with nonin-ferior efficacy and better bone and renal safety to tenofovir disoproxil fumarate(TDF)in 48 weeks of treatment.Here,we update 96-week comparison results.Methods:Patients with chronic hepatitis B were assigned(2:1)to receive either 25 mg TMF or 300 mg TDF with matching placebo for 96 weeks.The virological suppression was defined as HBV DNA levels<20 IU/mL at week 96.Safety was evaluated thoroughly with focusing on bone,renal,and metabolic pa-rameters.Results:Virological suppression rates at week 96 were similar between TMF and TDF group in both HBeAg-positive and HBeAg-negative populations.Noninferior efficacy was maintained in the pooled population,while it was first achieved in patients with HBV DNA≥7 or 8 log10 IU/mL at baseline.Non-indexed estimated glomerular filtration rate for renal safety assessment was adopted,while a smaller decline of which was seen in the TMF group than in the TDF group(p=0.01).For bone mineral density,patients receiv-ing TMF displayed significantly lower reduction levels in the densities of spine,hip,and femur neck at week 96 than those receiving TDF.In addition,the lipid parameters were stable after week 48 in all groups while weight change still showed the opposite trend.Conclusions:TMF maintained similar efficacy at week 96 compared with TDF with continued superior bone and renal safety profiles(NCT03903796).Zhihong Liu Qinglong Jin Yuexin Zhang Guozhong Gong Guicheng Wu Lvfeng Yao Xiaofeng Wen Zhiliang Gao Yan Huang Daokun Yang Enqiang Chen Qing Mao Shide Lin Jia Shang Huanyu Gong Lihua Zhong Huafa Yin Fengmei Wang Peng Hu Qiong Wu Chao Pan Wen Jia Chuan Li Chang’an Sun Junqi Niu Jinlin Hou TMF Study Group 2023Journal of Clinical and Translational Hepatology2023,11,3:3

6Construction of hybrid constructed wetlands for phosphorus chemical industry tailwater treatment in the middle Yangtze river basin:Responses of plant growth and root-associated microbial communities显示文摘Constructed wetlands are commonly utilized to treat industrial wastewater due to their effectiveness,affordability,and environmental benefits.Many phosphorus chemical enterprises in the middle Yangtze River basin are facing high pollution load challenges and efforts are needed to improve removal efficiency of pollutants.This work was the first to use constructed wetlands for phosphorus chemical industry tailwater(PCITW)treatment.The new hybrid constructed wetlands(CWs)proposed were the aeration horizontal subsurface flow constructed wetland(AHSCW)and the integrated vertical flow constructed wetland(IVCW),which were constructed on a pilot scale.Here,the effectiveness of pollutant removal along hybrid CWs,wetland plant growth,and rootassociated microbial community responses to the PCITW were investigated.The results showed that there was spatial variation in removal of carbon,nitrogen,and phosphorus in the hybrid CWs,and that the AHSCW and the IVCW played synergistic roles in the removal of pollutants.Compared with influent,the toxic effect of effluent to embryos of a rare minnow(Gobiocypris rarus)was alleviated.Exposure to the effluent resulted in no malformation or death for embryos.Plants and microorganisms in the wetland system of the phosphate chemical tailwater were then compared with the control system of municipal tailwater.Plants in the former system had lower root density,and higher average root diameter,root shoot ratio,specific root length,and specific surface area.Plant roots also had decreased NH_(4)^(+)uptake ability but increased Ca^(2+)uptake to adapt to the high load and complex pollution stress.Unlike the control system,stochastic mechanisms had a more important role than deterministic processes in shaping the microbial community assembly associated with the PCITW.Meanwhile,analysis of microbial network-level topological characteristics demonstrated substantial reduction in network interactions complexity and microbiome stability in the treatment system.Findings from this study suggest wetlands will be helpful for efficient purification of phosphorus chemical industry wastewater in the Yangtze River basin.Qianzheng Li Yao Guo Junqi Yu Lu Yao Shun Liu Yahua Li Disong Chen Fei Peng Dong Xu Zhenbin Wu Qiaohong Zhou 2023Water Biology and Security2023,2,3:1

13EBP50影响HeLa细胞微丝骨架的分布和定位(英文)显示文摘Objective:To explore the influence of EBP50(ezrin-radixin-moesin-binding phospho-protein-50) on microfilament cytoskeleton content and distribution in cultured Hela cells, and to investigate the relationship between the changes in microfilament cytoskeleton localization and EBP50 after PDGF(platelet-derived growth factor) stimulation, and to further clarify the molecular mechanism by which EBP50 suppresses tumor cell proliferation and migration.Methods:pBK-CMV-HAEBP50 wild type recombinant plasmid and pBK-CMV-HA empty vector were transfected into Hela cells.G418 at 350 mg/L was used to screen for cell clones stably expressing EBP50.Western blot was carried out to detect EBP50 expression.Similarities and differences in microfilament cytoskeleton content and distribution in Hela cells transfected with pBK-CMV-HA-EBP50 wild type recombinant plasmid and pBK-CMV-HA empty vector were also treated with PDGF(10 ng/mL and 20 ng/mL, 37 ℃, 15 min) and stained by rhodamine-labeled phalloidin to observe the distribution of microfilament cytoskeleton in the two groups.EBP50 protein distribution in PDGF-stimulated Hela cells was detected by immunofluorescence.Results:Western blot results confirmed that the EBP50 cDNA fragment could express EBP50 in cultured Hela cell lines and that cell lines stably expressing EBP50 were successfully obtained.Western blot and fluorescence results showed that in the cell line transfected with empty vector, the microfilament cytoskeleton was thick, loose, multidirectional and displayed crossing arrangements.The content of microfilament cytoskeleton in the cell line transfected with pBK-CMV-HA-EBP50 was different from that found in the cell line transfected with empty vector.EBP50 expression enhanced microfilament cytoskeleton polymerization into compact thin filaments.Under the stimulation of PDGF, EBP50 migrated to the cell membrane from the cytosol together with microfilament cytoskeleton and co-localized there.Conclusion:EBP50 can change the distribution of microfilament cytoskeleton in cultured Hela cells and can also bind the microfilament cytoskeleton to the cell membrane under the stimulation of PDGF.EBP50 may play a role in the proliferation and migration of tumor cells by influencing the distribution and localization of microfilament cytoskeleton.Kun Liu Junfang Zheng Ying Wang Peng Chen Junqi He 2009The Chinese-German Journal of Clinical Oncology2009,8,5:0
14Stock Price Forecasting: An Echo State Network Approach显示文摘Forecasting stock prices using deep learning models suffers from pro-blems such as low accuracy,slow convergence,and complex network structures.This study developed an echo state network(ESN)model to mitigate such pro-blems.We compared our ESN with a long short-term memory(LSTM)network by forecasting the stock data of Kweichow Moutai,a leading enterprise in China’s liquor industry.By analyzing data for 120,240,and 300 days,we generated fore-cast data for the next 40,80,and 100 days,respectively,using both ESN and LSTM.In terms of accuracy,ESN had the unique advantage of capturing non-linear data.Mean absolute error(MAE)was used to present the accuracy results.The MAEs of the data forecast by ESN were 0.024,0.024,and 0.025,which were,respectively,0.065,0.007,and 0.009 less than those of LSTM.In terms of con-vergence,ESN has a reservoir state-space structure,which makes it perform faster than other models.Root-mean-square error(RMSE)was used to present the con-vergence time.In our experiment,the RMSEs of ESN were 0.22,0.27,and 0.26,which were,respectively,0.08,0.01,and 0.12 less than those of LSTM.In terms of network structure,ESN consists only of input,reservoir,and output spaces,making it a much simpler model than the others.The proposed ESN was found to be an effective model that,compared to others,converges faster,forecasts more accurately,and builds time-series analyses more easily.Guang Sun Jingjing Lin Chen Yang Xiangyang Yin Ziyu Li Peng Guo Junqi Sun Xiaoping Fan Bin Pan 2021Computer Systems Science & Engineering2021,36,3:0
