1A new bone repair scaffold combined with chitosan/ hydroxyapatite and sustained releasing icariin显示文摘Icariin, a plant-derived flavonol glycoside, has been proved as an osteoinductive agent for bone tissue engineering. A new bone repair scaffold was generated by thorough mixing of icariin and chitosan/ hydroxyapatite (icariin-CS/HA) using freeze-drying technigue. Characteristics of morphology, mechanical properties, biocompatibility, drug release behavior and bone repair abilities in vivo were evaluated. The results show that drug loading process of icariin did not affect physical structure of CS/HA composite significantly but decreased mechanical properies of CS/HA composite, which happened with a high dosage; icariin-CS/HA had favorable cell compatibility and promoted osteogenic differentiation of hBMSCs; the controlled release of icariin was satisfactory and the release retained after 90 d in vitro. In addition, icariin-CS/HA scaffolds had favorable osteoconduction and osteoinduction in vivo, and could fill bone defect sites and stimulate newborn bone tissues formation at early stage. 2Inhibition of gasdermin D-dependent pyroptosis attenuates the progression of silica-induced pulmonary inflammation and fibrosis显示文摘Silicosis is a leading cause of occupational disease-related morbidity and mortality worldwide,but the molecular basis underlying its development remains unclear.An accumulating body of evidence supports gasdermin D(GSDMD)-mediated pyroptosis as a key component in the development of various pulmonary diseases.However,there is little experimental evidence connecting silicosis and GSDMD-driven pyroptosis.In this work,we investigated the role of GSDMD-mediated pyroptosis in silicosis.Single-cell RNA sequencing of healthy and silicosis human and murine lung tissues indicated that GSDMD-induced pyroptosis in macrophages was relevant to silicosis progression.Through microscopy we then observed morphological alterations of pyroptosis in macrophages treated with silica.Measurement of interleukin-1βrelease,lactic dehydrogenase activity,and real-time propidium iodide staining further revealed that silica induced pyroptosis of macrophages.Additionally,we verified that both canonical(caspase-1-mediated)and non-canonical(caspase-4/5/11-mediated)signaling pathways mediated silica-induced pyroptosis activation,in vivo and in vitro.Notably,Gsdmd knockout mice exhibited dramatically alleviated silicosis phenotypes,which highlighted the pivotal role of pyroptosis in this disease.Taken together,our results demonstrated that macrophages underwent GSDMD-dependent pyroptosis in silicosis and inhibition of this process could serve as a viable clinical strategy for mitigating silicosis.Meiyue Song Jiaxin Wang Youliang Sun Junling Pang Xiaona Li Yuan Liu Yitian Zhou Peiran Yang Tianhui Fan Ying Liu Zhaoguo Li Xianmei Qi Baicun Li Xinri Zhang Jing Wang Chen Wang 2022Acta Pharmaceutica Sinica B2022,12,3:5
3Klotho in Osx+-mesenchymal progenitors exerts pro-osteogenic and anti-inflammatory effects during mandibular alveolar bone formation and repair显示文摘Maxillofacial bone defects are commonly seen in clinical practice.A clearer understanding of the regulatory network directing maxillofacial bone formation will promote the development of novel therapeutic approaches for bone regeneration.The fibroblast growth factor(FGF)signalling pathway is critical for the development of maxillofacial bone.Klotho,a type I transmembrane protein,is an important components of FGF receptor complexes.Recent studies have reported the presence of Klotho expression in bone.However,the role of Klotho in cranioskeletal development and repair remains unknown.Here,we use a genetic strategy to report that deletion of Klotho in Osx-positive mesenchymal progenitors leads to a significant reduction in osteogenesis under physiological and pathological conditions.Klotho-deficient mensenchymal progenitors also suppress osteoclastogenesis in vitro and in vivo.Under conditions of inflammation and trauma-induced bone loss,we find that Klotho exerts an inhibitory function on inflammation-induced TNFR signaling by attenuating Rankl expression.More importantly,we show for the first time that Klotho is present in human alveolar bone,with a distinct expression pattern under both normal and pathological conditions.In summary,our results identify the mechanism whereby Klotho expressed in Osx+-mensenchymal progenitors controls osteoblast differentiation and osteoclastogenesis during mandibular alveolar bone formation and repair.Klotho-mediated signaling is an important component of alveolar bone remodeling and regeneration.It may also be a target for future therapeutics.Yi Fan Chen Cui Clifford JRosen Tadatoshi Sato Ruoshi Xu Peiran Li Xi Wei Ruiye Bi Quan Yuan Chenchen Zhou 2022Signal Transduction and Targeted Therapy2022,7,6:3
4Drinking water treatment using a submerged internal-circulation membrane coagulation reactor coupled with permanganate oxidation显示文摘A submerged internal circulating membrane coagulation reactor(MCR) was used to treat surface water to produce drinking water. Polyaluminum chloride(PACl) was used as coagulant,and a hydrophilic polyvinylidene fluoride(PVDF) submerged hollow fiber microfiltration membrane was employed. The influences of trans-membrane pressure(TMP), zeta potential(ZP) of the suspended particles in raw water, and KMnO_4 dosing on water flux and the removal of turbidity and organic matter were systematically investigated. Continuous bench-scale experiments showed that the permeate quality of the MCR satisfied the requirement for a centralized water supply, according to the Standards for Drinking Water Quality of China(GB 5749-2006), as evaluated by turbidity(<1 NTU) and total organic carbon(TOC)(<5 mg/L)measurements. Besides water flux, the removal of turbidity, TOC and dissolved organic carbon(DOC) in the raw water also increased with increasing TMP in the range of 0.01–0.05 MPa. High ZP induced by PACl, such as 5–9 mV, led to an increase in the number of fine and total particles in the MCR, and consequently caused serious membrane fouling and high permeate turbidity.However, the removal of TOC and DOC increased with increasing ZP. A slightly positive ZP, such as 1–2 mV, corresponding to charge neutralization coagulation, was favorable for membrane fouling control. Moreover, dosing with KMnO_4 could further improve the removal of turbidity and DOC, thereby mitigating membrane fouling. 5Recognition of high-specificity hERG K+ channel inhibitor-induced arrhythmia in cardiomyocytes by automated template matching显示文摘Cardiovascular disease(CVD)is the number one cause of death in humans.Arrhythmia induced by gene mutations,heart disease,or hERG K+channel inhibitors is a serious CVD that can lead to sudden death or heart failure.Conventional cardiomyocyte-based biosensors can record extracellular potentials and mechanical beating signals.However,parameter extraction and examination by the naked eye are the traditional methods for analyzing arrhythmic beats,and it is difficult to achieve automated and efficient arrhythmic recognition with these methods.In this work,we developed a unique automated template matching(ATM)cardiomyocyte beating model to achieve arrhythmic recognition at the single beat level with an interdigitated electrode impedance detection system.The ATM model was established based on a rhythmic template with a data length that was dynamically adjusted to match the data length of the target beat by spline interpolation.The performance of the ATM model under long-term astemizole,droperidol,and sertindole treatment at different doses was determined.The results indicated that the ATM model based on a random rhythmic template of a signal segment obtained after astemizole treatment presented a higher recognition accuracy(100%for astemizole treatment and 99.14%for droperidol and sertindole treatment)than the ATM model based on arrhythmic multitemplates.We believe this highly specific ATM method based on a cardiomyocyte beating model has the potential to be used for arrhythmia screening in the fields of cardiology and pharmacology.Hao Wang Hongbo Li Xinwei Wei Tao Zhang Yuting Xiang Jiaru Fang Peiran Wu Xi Xie Ping Wang Ning Hu 2021Microsystems & Nanoengineering2021,7,2:2

7Floating Escherichia coli by Expressing Cyanobacterial Gas Vesicle Genes显示文摘Gas vesicles are hollow, air-filled polyprotein structures that provide the buoyancy to cells. They are found in a variety of prokaryotes. In this study, we isolated a partial gas vesicle protein gene cluster containing gvp A and gvp C20Ψ from Planktothrix rubescens, and inserted it into an expression vector and expressed it in E. coli. The gas vesicle was developed in bacterial cells, which made bacterial cells to float on medium surface. We also amplified gvp A and gvp C20Ψ separately and synthesized an artificial operon by fusing these two genes with the standardized gene expression controlling elements of E. coli. The artificial operon was expressed in E. coli, forming gas vesicles and floating bacteria cells. Our findings verified that the whole set of genes and the overall structure of gas vesicle gene cluster are not necessary for developing gas vesicles in bacteria cells. Two genes, gvp A and gvp C20Ψ, of the gas vesicle gene cluster are sufficient for synthesizing an artificial operon that can develop gas vesicles in bacteria cells. 8Extracellular and intracellular infection of Botryosphaeria dothidea and resistance mechanism in apple cells显示文摘Apple ring rot,which is caused by Botryosphaeria dothidea,severely affects apple production.The mechanisms employed in apple cells against B.dothidea remain unknown.In this research,the pathogen infection mode and the relationship between cell death and disease resistance in‘Fuji’/B.dothidea interaction pathosystem were investigated.By using transmission electron microscopy(TEM),our research showed that the pathogen infects apple cells both intracellularly and extracellularly.However,compared with that in immature fruit,the incidence of hyphae in the interior of mature apple fruit cells increased dramatically,suggesting that cell wall-mediated penetration resistance could be important in apple resistance against B.dothidea.TEM ultrastructural characterization identified the nuclear morphology of programmed cell death induction in both apple fruit and callus cells under B.dothidea infection.Overexpression of MdVDAC2(MDP0000271281),which encodes an outer-membrane localized anion channel protein in mitochondria,significantly promoted cell death under B.dothidea infection and simultaneously inhibited pathogen infection,suggesting that cell death represents a disease resistance mechanism in apple against B.dothidea infection.Furthermore,BdCatalase(KAF4307763),a cytochromeP450 family protein BdCYP52A4(KAF4300696),and subtilisin-domain containing proteinswere identified fromB.dothidea-secreted proteins,which suggested the potential involvement of active oxygen species and phytoalexins in combating B.dothidea infection and triggering or dampening apple resistance.Collectively,our research suggested that cell wall-mediated penetration resistance,programmed cell death machinery and microbial effector-interrelated signaling were among strategies recruited in apple to combat B.dothidea.The current research laid the foundation for further investigations into resistance mechanisms in apple.Li Xin Rui Zhang Xianpu Wang Xiuxia Liu Yicheng Wang Peiran Qi Lishuang Wang Shujing Wu Xuesen Chen 2023Horticultural Plant Journal2023,9,2:1
9Gefitinib and fostamatinib target EGFR and SYK to attenuate silicosis:a multi-omics study with drug exploration显示文摘Silicosis is the most prevalent and fatal occupational disease with no effective therapeutics,and currently used drugs cannot reverse the disease progress.Worse still,there are still challenges to be addressed to fully decipher the intricated pathogenesis.Thus,specifying the essential mechanisms and targets in silicosis progression then exploring anti-silicosis pharmacuticals are desperately needed.In this work,multi-omics atlas was constructed to depict the pivotal abnormalities of silicosis and develop targeted agents.By utilizing an unbiased and time-resolved analysis of the transcriptome,proteome and phosphoproteome of a silicosis mouse model,we have verified the significant differences in transcript,protein,kinase activity and signaling pathway level during silicosis progression,in which the importance of essential biological processes such as macrophage activation,chemotaxis,immune cell recruitment and chronic inflammation were emphasized.Notably,the phosphorylation of EGFR(p-EGFR)and SYK(pSYK)were identified as potential therapeutic targets in the progression of silicosis.To inhibit and validate these targets,we tested fostamatinib(targeting SYK)and Gefitinib(targeting EGFR),and both drugs effectively ameliorated pulmonary dysfunction and inhibited the progression of inflammation and fibrosis.Overall,our drug discovery with multi-omics approach provides novel and viable therapeutic strategies for the treatment of silicosis.Mingyao Wang Zhe Zhang Jiangfeng Liu Meiyue Song Tiantian Zhang Yiling Chen Huiyuan Hu Peiran Yang Bolun Li Xiaomin Song Junling Pang Yanjiang Xing Zhujie Cao Wenjun Guo Hao Yang Jing Wang Juntao Yang Chen Wang 2022Signal Transduction and Targeted Therapy2022,7,6:1

11Self-assembly of phosphorylated peptide driven by Dy^(3+)显示文摘Nature chooses phosphorylation as a key modification to modulate and program the functions of proteins.Various phosphorylated peptides(PPs)have been widely identified and investigated by biologists,but the possibility that PPs could become a building unit for artificial materials is neglected.Here we report for the first time a supramolecular assembly of PPs with the assistance of dysprosium ions(Dy^(3+)).Dy^(3+)bridges multiple phosphate groups in double-phosphorylated peptides(di-PPs),and braid these peptide chains into nanofibers.The assembly occurs inside nanochannels and blocks the channels,leading to prominent“ON–OFF”switching in transmembrane ionic current.The di-PPs’assembling process could be dynamically regulated by the addition or deletion of phosphate groups under the control of kinases or phosphatases.This study proves the huge potential of PPs being utilized as materials via self-assembling,which will promote the design of novel bio-inspired artificial materials and devices.Hang Yang Yuting Xiong Minmin Li Zhiying Yang Peiran Meng Guangyan Qing 2023Chinese Chemical Letters2023,34,8:0
12Temporal Trends of Maternal Mortality Due to Obstetric Hemorrhage in Chinese Mainland: Evidence from the Population-Based Surveillance Data Between 2000 and 2019显示文摘Objective:To analyze the temporal trends of maternal mortality ratio(MMR)due to obstetric hemorrhage and its specific causes in Chinese mainland from 2000 to 2019,to identify whether the rate of change has accelerated or slowed down during this period,and to find the prior cause of obstetric hemorrhage that needs to be intervened in the future.Methods:Individual information on maternal deaths and total number of live births from 336 surveillance sites across 31 provinces in Chinese mainland was collected from the National Maternal and Child Health Surveillance System between 2000 and 2019.Maternal death was defined according to the World Health Organization’s criterion.The final underlying cause of death was confirmed by the national review and was coded according to International Classification of Diseases-10.Linear trends for changes in characteristics of maternal deaths were assessed using linear or logistic models with the year treated as a continuous variable.The MMR and 95%confidence intervals(CI)for regions or causes were estimated by Poisson’s distribution.Joinpoint regression was used to assess the accurate temporal patterns.Results:The national MMR due to obstetric hemorrhage was 18.4 per 100,000 live births(95%CI:15.0–22.2)in 2000.It peaked in 2001(22.1 per 100,000 live births,95%CI:18.3–26.4)and was lowest in 2019(1.6 per 100,000 live births,95%CI:1.0–2.3).For specific regions,the MMR due to obstetric hemorrhage in rural areas and western regions both experienced a slight rise,followed by a rapid decline,and then a slow decline.For specific causes,no change point was found in joinpoint analysis of the national MMR caused by placenta previa,postpartum uterine atony,and retained placenta(the annual percent change was12.0%,10.5%,and21.0%,respectively).The MMR caused by postpartum hemorrhages(PPH)significantly declined by 8.0%(95%CI:1.9–13.6)per year from 2000 to 2007.The annual percent change of MMR caused by PPH accelerated further to25.0%between 2007 and 2011,and then decreased to7.8%between 2011 and 2019.The proportion of maternal deaths due to antepartum hemorrhages increased from 7.6%(8/105)in 2000 to 14.3%(4/28)in 2019.The changes in the proportion of causes were different for maternal deaths due to PPH.The proportion of postpartum uterine atony increased from 39.0%(41/105)in 2000 to 60.7%(17/28)in 2019,and the proportion of uterine rupture also increased from 12.3%(13/105)in 2000 to 14.3%(4/28)in 2019.However,the proportion of retained placenta decreased from 37.1%(39/105)in 2000 to 7.1%(2/28)in 2019.Conclusion:Over the last 20 years,the intervention practice in China has proved that targeted interventions are beneficial in reducing the MMR due to obstetric hemorrhage.However,the MMR has reached a plateau and is likely to increase for some specific causes such as uterine rupture.China needs to develop more effective interventions to reduce maternal deaths due to obstetric hemorrhage,especially for postpartum uterine atony and uterine rupture.Yi Mu Jun Zhu Yanping Wang Jiani Zhang Mingrong Li Peiran Chen Yanxia Xie Juan Liang Xiaodong Wang 2022Maternal-Fetal Medicine2022,4,3:0
13Spatially controlled lithium deposition on Li_(x)Cu_(y)P_(z) arrays enabling highly stable lithium metal batteries显示文摘Lithium(Li)metal is a potential anode for high-energy-density batteries because of its low potential and ultrahigh capacity.Nevertheless,the Li dendrites formation,the ununiform Li deposition,and the growth of Li dendrites hamper its application,especially under high deposition capacity/high rate.Here,a spatially controlled Li deposition mode with array-oriented morphology is achieved based on the novel mixed ion/electron-conducting Li_(x)Cu_(y)P_(z) arrays constructed on Cu foil,which can be facile fabricated via an in-situ transformation of metal phosphide.Theoretic calculations indicate the excellent lithiophilicity and low Li diffusion barrier of the arrays,especially for the Li_(2)CuP phase,which are conducive to ho-mogenizing the Li nucleation/deposition of Li.Moreover,such mixed conducting arrays promote fast Li+diffusion via the continuous Li+pathways as well as modulate the Li+flux/electric field.Furthermore,the arrays with enlarged specific surface area and open spaces reduce the local current density and alle-viate the volume fluctuation of Li.Consequently,a dendrite-free Li anode is obtained under a high rate(20 mA cm^(–2))or a high deposition capacity(10 mAh cm^(–2)).In addition,even if the negative/positive ratio reduces to only 1.1,the full cells still perform outstanding stability for over 200 cycles.This work empha-sizes the importance of the design of the framework in terms of the intrinsic properties and structure and reveals a pathway for developing Li metal batteries.Ying Zhou Xiangru Sun Peiran Tan Yueming Wang Hongyang Dong Dejun Li 2023Journal of Materials Science & Technology2023,,21:0

16Automated optical inspection of FAST’s reflector surface using drones and computer vision显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST)is the world’s largest single-dish radio telescope.Its large reflecting surface achieves unprecedented sensitivity but is prone to damage,such as dents and holes,caused by naturally-occurring falling objects.Hence,the timely and accurate detection of surface defects is crucial for FAST’s stable operation.Conventional manual inspection involves human inspectors climbing up and examining the large surface visually,a time-consuming and potentially unreliable process.To accelerate the inspection process and increase its accuracy,this work makes the first step towards automating the inspection of FAST by integrating deep-learning techniques with drone technology.First,a drone flies over the surface along a predetermined route.Since surface defects significantly vary in scale and show high inter-class similarity,directly applying existing deep detectors to detect defects on the drone imagery is highly prone to missing and misidentifying defects.As a remedy,we introduce cross-fusion,a dedicated plug-in operation for deep detectors that enables the adaptive fusion of multi-level features in a point-wise selective fashion,depending on local defect patterns.Consequently,strong semantics and fine-grained details are dynamically fused at different positions to support the accurate detection of defects of various scales and types.Our AI-powered drone-based automated inspection is time-efficient,reliable,and has good accessibility,which guarantees the long-term and stable operation of FAST.Jianan Li Shenwang Jiang Liqiang Song Peiran Peng Feng Mu Hui Li Peng Jiang Tingfa Xu 2023Light(Advanced Manufacturing)2023,4,1:0
17Self-Awakened Particle Swarm Optimization BN Structure Learning Algorithm Based on Search Space Constraint显示文摘To obtain the optimal Bayesian network(BN)structure,researchers often use the hybrid learning algorithm that combines the constraint-based(CB)method and the score-and-search(SS)method.This hybrid method has the problemthat the search efficiency could be improved due to the ample search space.The search process quickly falls into the local optimal solution,unable to obtain the global optimal.Based on this,the Particle SwarmOptimization(PSO)algorithm based on the search space constraint process is proposed.In the first stage,the method uses dynamic adjustment factors to constrain the structure search space and enrich the diversity of the initial particles.In the second stage,the update mechanism is redefined,so that each step of the update process is consistent with the current structure which forms a one-to-one correspondence.At the same time,the“self-awakened”mechanism is added to prevent precocious particles frombeing part of the best.After the fitness value of the particle converges prematurely,the activation operation makes the particles jump out of the local optimal values to prevent the algorithmfromconverging too quickly into the local optimum.Finally,the standard network dataset was compared with other algorithms.The experimental results showed that the algorithmcould find the optimal solution at a small number of iterations and a more accurate network structure to verify the algorithm’s effectiveness.Kun Liu Peiran Li Yu Zhang Jia Ren Xianyu Wang Uzair Aslam Bhatti 2023Computers, Materials & Continua2023,76,9:0
18Novel empirical model for predicting residual flexural capacity of corroded steel reinforced concrete beam显示文摘In this study,a total of 177 flexural experimental tests of corroded reinforced concrete(CRC)beams were collected from the published literature.The database of flexural capacity of CRC beam was established by using unified and standardized experimental data.Through this database,the effects of various parameters on the flexural capacity of CRC beams were discussed,including beam width,the effective height of beam section,ratio of strength between longitudinal reinforcement and concrete,concrete compressive strength,and longitudinal reinforcement corrosion ratio.The results indicate that the corrosion of longitudinal reinforcement has the greatest effect on the residual flexural capacity of CRC beams,while other parameters have much less effect.In addition,six available empirical models for calculating the residual flexural strength of CRC beams were also collected and compared with each other based on the established database.It indicates that though five of six existing empirical models underestimate the flexural capacity of CRC beams,there is one model overestimating the flexural capacity.Finally,a newly developed empirical model is proposed to provide accurate and effective predictions in a large range of corrosion ratio for safety assessment of flexural failure of CRC beams confirmed by the comparisons.Zhao-Hui LU Hong-Jun WANG Fulin QU Yan-Gang ZHAO Peiran LI Wengui LI 2020Frontiers of Structural and Civil Engineering2020,14,4:0
19On the study of aerosol composition at the Great Wall Station, South Pole显示文摘The South pole aera offers one of the best locations on the Earth for studying the composition of the background aerosol. For this purpose, aerosol samples were collected in particle size fractions by 8 stages cascade impactors. Aerosol sampling was begun in Jan. and finished in Dec. 1986 at the St. Great Wall, South Pole. The samples were analysed by Proton Induced X-ray Emission (PIXE) method. In order to learn the enrichment factors of various elements all the results were separated into two groups: coarse (> 10μm) and fine (< 10μm) particles. The correlation coefficients of elements of both coarse and fine particles were discussed. The aerosol levels measured at St. Great Wall were compared to those recently measured at some other locations. 20Optimal beamforming with aliasing cancellation in the spherical harmonics domain显示文摘The existing beamforming with aliasing elimination in the spherical harmonics domain usually adopts a single design goal,making it challenging to adapt to diverse application scenarios.Therefore,we propose optimal beamforming with aliasing cancellation in the spherical harmonics domain and establish a corresponding multiply constrained convex optimization model with a second-order cone programming form.Benefiting from this model,we further propose a maximum directivity beamformer with sidelobe constraint and aliasing cancellation,a robust maximum directivity beamformer with aliasing cancellation,a minimum variance distortionless response beamformer with aliasing elimination,and a maximum directivity beamformer with null constraint and aliasing elimination.The proposed method not only inherits the aliasing-cancellation capability of the existing aliasing cancellation beamforming,but also achieves a proper trade-off among multiple conflicting performance measures such as directivity factor,white noise gain,and sidelobe level.Hence,the proposed optimal beamforming provides users with the flexibility to adjust the constraint parameters according to actual application requirements and thus obtain a beamformer with the best all-around performance in a specific scenario.JIANG Peihong CHU Zhigang LI Peiran YANG Yang 2023Chinese Journal of Acoustics2023,42,3:0
