2CXCL5/CXCR2 axis in tumor microenvironment as potential diagnostic biomarker and therapeutic target显示文摘The components of the tumor microenvironment(TME)in solid tumors,especially chemokines,are currently attracting much attention from scientists.C-X-C motif chemokine ligand 5(CXCL5)is one of the important chemokines in TME.Over-expression of CXCL5 is closely related to the survival time,recurrence and metasta-sis of cancer patients.In TME,CXCL5 binds to its receptors,such as C-X-C motif chemokine receptor 2(CXCR2),to participate in the recruitment of immune cells and promote angiogenesis,tumor growth,and metastasis.The CXCL5/CXCR2 axis can act as a bridge between tumor cells and host cells in TME.Blocking the trans-mission of CXCL5/CXCR2 signals can increase the sensitivity and effectiveness of immunotherapy and slow down tumor progression.CXCL5 and CXCR2 are also regarded as biomarkers for predicting prognosis and molecular targets for customiz-ing the treatment.In this review,we summarized the current literature regarding the biological functions and clinical significance of CXCL5/CXCR2 axis in TME.The possibility to use CXCL5 and CXCR2 as potential prognostic biomarkers and thera-peutic targets in cancer is also discussed.Wen Zhang Huishan Wang Mingyang Sun Xueting Deng Xueru Wu Yilan Ma Mengjing Li Said Maisam Shuoa Qiang You Lin Miao 2020Cancer Communications2020,40,2:19

4Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 6On a New Genus of Basal Neoceratopsian Dinosaur from the Early Cretaceous of Gansu Province, China显示文摘A new genus and species of basal neoceratopsian dinosaur, Auroraceratops rugosus, is reported based on material from the Early Cretaceous Xinminpu Group in the Gongpoquan Basin of Gansu Province, China. Auroraceratops is represented by a nearly complete skull and low jaws, and different greatly from all other neoceratopsians by its considerable breadth of the nasals, fungiform expansion of the dorsal end of the lacrimal, highly developed rugosity of the jugal, dentary and surangular, and inflated, striated premaxillary teeth. 12An Early Cretaceous Avialian Bird, Shenzhouraptor sinensis from Western Liaoning, China显示文摘This paper describes an avialian bird from the Early Cretaceous Jiufotang Formation in Yixian county,western Liaoning, China, which is named as Shenzhouraptor sinensis by Ji et al. on July 15, 2002. Shenzhouraptor sinensis is characterized by no teeth in its mouth, the forelimbs longer than the hindlimbs, a long tail with more than 23 caudal vertebrae, U-shaped wishbone, and remiges longer than the total length of ulna and manus. 13Effect of arsenic trioxide on human hepatoma cell line BEL-7402 cultured in vitro显示文摘AIM To study the effect of a varyingconcentrations of arsenic trioxide on humanhepatoma cell line BEL-?402 cultured in vitro andits mechanism of action.METHODS The BEL-7402 cells were treatedwith arsenic trioxide(at the concentrations of0.5,1,2 μmol/L,respectively)for 4 successivedays.The cell growth and proliferation wereobserved by cell counting and cell-growth curve.Morphologic changes were studied withelectronmicroscopy.Flow cytometry was usedto assay celI-DNA distribution and the proteinexpression of Bcl-2 and Bax detected byimmunocytochemical method.RESULTS The cell growth was significantlyinhibited by varying concentrations of arsenictrioxide as revealed by cell counting and cell-growth curve,which was dose- and time-dependent.Arsenic trioxide treatment at 0.5,1and 2 μmol/L resulted in a sub-G1 cell peak,theapoptosis rate of the control group was 9.31%and that of 0.5 μmol/L arsenic trioxide 15.53%,no significant difference was seen between thetwo.The apoptosis rates of 1,2 μmol/L arsenictrioxide were 19.10% and 21.87% respectively,which were much higher(both P<0.05).Decrease of G0/G1 phase cells and increase of Sphase cells were observed by flow cytometry,suggesting the inhibition effect of 0.5,1,2 μmol/L arsenic trioxide on BEL-7402 cell lay in the G0/G1 phase.Morphologic changes such asintact cell membrane,nucleic condensation,apoptotic body formation were seen undertransmission electronmicrescopy,whereas the0.5 mol/L arsenic trioxide-treated BEL-7402cells showed decrease of nucleocytoplasmicratio,round nucleus,well-differentiatedorganelles in the cytoplasm.The processes andmicrovilli on the cell surface of the experimentalgroups under scanning electron microscopy weresignificantly decreased.High expressions ofBcl-2 and Bax were detected in 1 and 2 μmol/Larsenic trioxide-treated cells,these were 46%,87.33% and 83.08%,95.83% respectively,among which that of Bax was more significant.Arsenic trioxide treatment at 0.5 μmol/Lresulted in a higher expression level of Bcl-2 andlower expression level of Bax,which were8.81% and 3.83% respectively,as comparedwith that of the control group(15.33%)(P1<0.01,P2<0.01).CONCLUSION Arsenic trioxide not onlyinhibited proliferation but also induced apoptosisof human hepatoma cell line BEL-7402.Theinduced-apoptosis effect of 1,2 μmol/L arsenictrioxide was related to the expression level ofBcl-2 and Bax.Hong Yu Xu You Lin Yang Yuan Yuan Gao Qiao Li Wu Guang Qiang Gao 2000World Journal of Gastroenterology2000,6,5:8
14CuFe2O4@GO nanocomposite as an effective and recoverable catalyst of peroxymonosulfate activation for degradation of aqueous dye pollutants显示文摘Recently,heterogeneous activation of peroxymonosulfate(PMS) to oxidatively degrade organic pollutants has been a hotspot.In the present work,copper ferrite-graphite oxide hybrid(CuFe2 O4@GO)was prepared and used as catalyst to activate PMS for degradation of methylene blue(MB) in aqueous solution.A high degradation efficiency(93.3%) was achieved at the experimental conditions of20 mg/L MB,200 mg/L CuFe2 O4@GO,0.8 mmol/L PMS,and 25℃temperature.Moreover,CuFe2 O4@GO showed an excellent reusability and stability.The effects of various operational parameters including pollutant type,solution pH,catalyst dosage,PMS dosage,pollutant concentration,temperature,natural organic matter(NOM),and inorganic anions on the catalytic degradation process were comprehensively investigated and elucidated.The further mechanistic study revealed the Cu(Ⅱ)/Cu(Ⅰ) redox couple on CuFe2 O4@GO played the dominant role in PMS activation,where both hydroxyl and sulfate radicals were generated and proceeded the degradation of pollutants.In general,CuFe2 O4@GO is a promising heterocatalyst for PMS-based advanced oxidation processes(AOPs) in wastewater treatment.Xiaoman Lei Menghan You Fei Pan Min Liu Peng Yang Dongsheng Xia Qiang Li Yanting Wang Jie Fu 2019Chinese Chemical Letters2019,30,12:8
15Chitinase-3 like-protein-1 function and its role in diseases显示文摘Non-enzymatic chitinase-3 like-protein-1(CHI3L1)belongs to glycoside hydrolase family 18.It binds to chitin,heparin,and hyaluronic acid,and is regulated by extracellular matrix changes,cytokines,growth factors,drugs,and stress.CHI3L1 is synthesized and secreted by a multitude of cells including macrophages,neutrophils,synoviocytes,chondrocytes,fibroblast-like cells,smooth muscle cells,and tumor cells.It plays a major role in tissue injury,inflammation,tissue repair,and remodeling responses.CHI3L1 has been strongly associated with diseases including asthma,arthritis,sepsis,diabetes,liver fibrosis,and coronary artery disease.Moreover,following its initial identification in the culture supernatant of the MG63 osteosarcoma cell line,CHI3L1 has been shown to be overexpressed in a wealth of both human cancers and animal tumor models.To date,interleukin-13 receptor subunit alpha-2,transmembrane protein 219,galectin-3,chemo-attractant receptor-homologous 2,and CD44 have been identified as CHI3L1 receptors.CHI3L1 signaling plays a critical role in cancer cell growth,proliferation,invasion,metastasis,angiogenesis,activation of tumor-associated macrophages,and Th2 polarization of CD4+T cells.Interestingly,CHI3L1-based targeted therapy has been increasingly applied to the treatment of tumors including glioma and colon cancer as well as rheumatoid arthritis.This review summarizes the potential roles and mechanisms of CHI3L1 in oncogenesis and disease pathogenesis,then posits investigational strategies for targeted therapies.Ting Zhao Zhongping Su Yingchang Li Xiaoren Zhang Qiang You 2020Signal Transduction and Targeted Therapy2020,5,1:8
16THE RADIATION AND DIFFRACTION OF WATER WAVES BY A BOTTOM-MOUNTED CIRCULAR CYLINDER IN A TWO-LAYER FLUID显示文摘An analytical method was proposed to analyze the radiation and diffraction of water waves by a bottom-mounted circular cylinder in a two-layer fluid. Analytical expressions for added mass and damping coefficients, as well as the wave excitation forces of the circular cylinder were obtained by an eigenfunction expansion method. The hydrodynamic forces on the bottom-mounted circular cylinder in a two-layer fluid include not only the added mass and damping coefficients, but also the wave forces of the surface and internal-wave modes. This is different from the case of a homogenous fluid. 17The Chinese Hα Solar Explorer(CHASE) mission: An overview显示文摘The Chinese Hα Solar Explorer(CHASE), dubbed “Xihe”—Goddess of the Sun, was launched on October 14, 2021 as the first solar space mission of China National Space Administration(CNSA). The CHASE mission is designed to test a newly developed satellite platform and to acquire the spectroscopic observations in the Hα waveband. The Hα Imaging Spectrograph(HIS)is the scientific payload of the CHASE satellite. It consists of two observational modes: raster scanning mode and continuum imaging mode. The raster scanning mode obtains full-Sun or region-of-interest spectral images from 6559.7 to 6565.9 ? and from 6567.8 to 6570.6 ? with 0.024 ? pixel spectral resolution and 1 min temporal resolution. The continuum imaging mode obtains photospheric images in continuum around 6689 ? with the full width at half maximum of 13.4 ?. The CHASE mission will advance our understanding of the dynamics of solar activity in the photosphere and chromosphere. 18Effect of metoprolol on myocardial apoptosis after coronary microembolization in rats显示文摘BACKGROUND:Coronary microembolization(CME) is a serious complication following percutaneous coronary intervention(PCI) in patients with acute coronary syndromes.The use of metoprolol before PCI can significantly protect ischemic myocardium from myocardial damage,but the function of metoprolol in the treatment of CME is not entirely clear.This study was to explore the effect and significance of metoprolol on myocardial apoptosis and caspase-3 activation after CME in rats.METHODS:Thirty rats were randomly divided into three groups including sham-operation(control group),CME plus saline(CME group),CME plus metoprolol(metoprolol group),10 rats for each group.The CME group was induced by injecting 3 000 polyethylene microspheres(42 urn)into the left ventricle during a 10-second occlusion of the ascending aorta;the control group was injected with physiological saline instead of microembolization ball;the metoprolol or saline group was given three intravenous bolus injections before CME.Echocardiography,TUNEL staining,and Western blotting were used to evaluate cardiac function,proportion of apoptotic cells and activation of caspase-3 respectively at 6 hours after operation.RESULTS:Echocardiography parameters displayed that the metoprolol group improved cardiac function significantly compared with the CME group(PO.05).The myocardial apoptotic rate of the CME group as well as the contents of activated caspase-3 increased significantly(P<0.05),both of which were ameliorated significantly by metoprolol treatment(P<0.05).CONCLUSIONS:This study demonstrates that metoprolol can protect the myocardium during CME in rats by inhibiting apoptosis and improving cardiac function.These results suggest that the inhibition of apoptosis can be a potential therapeutic strategy for the treatment of CME.Qiang Su Lang Li Yang-chun Liu You Zhou Wei-ming Wen 2013World Journal of Emergency Medicine2013,4,2:7

20Cornuside alleviates experimental autoimmune encephalomyelitis by inhibiting Th17 cell infiltration into the central nervous system显示文摘The present study was conducted to clarify the therapeutic effect of cornuside on experimental autoimmune encephalomyelitis(EAE)and its influence on T helper 17(Th17)cell and regulatory T(Treg)cell infiltration into the central nervous system.Rats were randomly placed into four treatment groups:control,EAE,EAE+cornuside,and EAE+prednisolone.The neurological function scores of rats were assessed daily.On the second day after EAE rats began to show neurological deficit symptoms,the four groups were treated with normal saline,normal saline,cornuside(150 mg/kg),and prednisolone(5 mg/kg),respectively.The treatment was discontinued after two weeks,and the spinal cord was obtained for hematoxylin and eosin(H&E)and luxol fast blue staining,as well as retinoic acid receptor-related orphan receptorγ(RORγ)and forkhead box protein P3(Foxp3)immunohistochemical staining.Blood was collected for Th17 and Treg cell flow cytometry testing,and the serum levels of interleukin(IL)-17A,IL-10,transforming growth factor-β(TGF-β),IL-6,IL-23,and IL-2 were measured via enzymelinked immunosorbent assay(ELISA).Compared with rats in the EAE group,rats in the EAE+cornuside and EAE+prednisolone groups began to recover from neurological deficits earlier,and had a greater degree of improvement of symptoms.Focal inflammation,demyelination,and RORγ-positive cell infiltration were reduced by cornuside or prednisolone treatment,whereas the Foxp3-positive cell numbers were not significantly different.Meanwhile,the number of Th17 cells and the IL-17A,IL-6,and IL-23 levels were lower in the blood after cornuside or prednisolone treatment,whereas the number of Treg cells or the levels of IL-10,TGF-β,and IL-2 were not markedly different.Cornuside can alleviate symptoms of EAE neurological deficits through its anti-inflammatory and immunosuppressive effects,and Th17 cells may be one of its therapeutic targets.Rongbo ZHANG Jin LIU Bin XU You WU Shunli LIANG Qiang YUAN 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,5:5
