| 1 | Three-dimensional ATUM-SEM reconstruction and analysis of hepatic endoplasmic reticulum-organelle interactions显示文摘The endoplasmic reticulum(ER)is a contiguous and complicated membrane network in eukaryotic cells,and membrane contact sites(MCSs)between the ER and other organelles perform vital cellular functions,including lipid homeostasis,metabolite exchange,calcium level regulation,and organelle division.Here,we establish a whole pipeline to reconstruct all ER,mitochondria,lipid droplets,lysosomes,peroxisomes,and nuclei by automated tape-collecting ultramicrotome scanning electron microscopy and deep learning techniques,which generates an unprecedented 3D model for mapping liver samples.Furthermore,the morphology of various organelles and the MCSs between the ER and other organelles are systematically analyzed.We found that the ER presents with predominantly flat cisternae and is knitted tightly all throughout the intracellular space and around other organelles.In addition,the ER has a smaller volume-to-membrane surface area ratio than other organelles,which suggests that the ER could be more suited for functions that require a large membrane surface area.Our data also indicate that ER-mitochondria contacts are particularly abundant,especially for branched mitochondria.Our study provides 3D reconstructions of various organelles in liver samples together with important fundamental information for biochemical and functional studies in the liver. | Yi Jiang Linlin Li Xi Chen Jiazheng Liu Jingbin Yuan Qiwei Xie Hua Han | 2021 | Journal of Molecular Cell Biology2021,13,9: | 2 |

| 8 | AHA1 upregulates IDH1 and metabolic activity to promote growth and metastasis and predicts prognosis in osteosarcoma显示文摘Osteosarcoma(OS)is the most common primary malignant bone tumor in children and adolescents.Although activator of HSP90 ATPase activity 1(AHA1)is reported to be a potential oncogene,its role in osteosarcoma progression remains largely unclear.Since metabolism reprogramming is involved in tumorigenesis and cancer metastasis,the relationship between AHA1 and cancer metabolism is unknown.In this study,we found that AHA1 is significantly overexpressed in osteosarcoma and related to the prognosis of osteosarcoma patients.AHA1 promotes the growth and metastasis of osteosarcoma both in vitro and in vivo.Mechanistically,AHA1 upregulates the metabolic activity to meet cellular bioenergetic needs in osteosarcoma.Notably,we identifed that isocitrate dehydrogenase 1(IDH1)is a novel client protein of Hsp90 AHA1.Furthermore,the IDH1 protein level was positively correlated with AHA1 in osteosarcoma.And IDH1 overexpression could partially reverse the effect of AHA1 knockdown on cell growth and migration of osteosarcoma.Moreover,high IDH1 level was also associated with poor prognosis of osteosarcoma patients.This study demonstrates that AHA1 positively regulates IDH1 and metabolic activity to promote osteosarcoma growth and metastasis,which provides novel prognostic biomarkers and promising therapeutic targets for osteosarcoma patients. | Diwei Zheng Weihai Liu Wenlin Xie Guanyu Huang Qiwei Jiang Yang Yang Jiarong Huang Zihao Xing Mengling Yuan Mengning Wei Yao Li Junqiang Yin Jingnan Shen Zhi Shi | 2021 | Signal Transduction and Targeted Therapy2021,6,2: | 1 |

| 11 | Application of AHP and DTOPSIS in Sugarcane Breeding显示文摘The weights of ten important traits of sugarcane,including the number of hair groups,plant height,defoliation capability,stalk diameter,ratooning property,stalk number of disease resistance,cane yield,sucrose content and sugar yield,were distributed with the Analytic Hierarchy Process( AHP) method. Ten Yuetang series of sugarcane lines were evaluated by Dynamic Technique for Order Preference by Similarity to Ideal Solution( DTOPSIS). AHP results indicated that the weights of ten traits were 0. 013 4,0. 022 7,0. 030 2,0. 040 8,0. 065 3,0. 088 1,0. 116 2,0. 160 6,0. 198 4 and 0. 264 5,respectively,with random consistency ratio C. Comprehensive evaluation of 10 sugarcane lines by DTOPSIS method combining with the conclusion of AHP showed that the comprehensive performances of Yuetang 03-393 and Yuetang 03-373 were much better than other sugarcane lines,with relative approach degree C i ranking the first and second,respectively,which was basically consistent with the practical survey,suggesting that Yuetang 03-393 and Yuetang 03-373 were suitable for large-scale popularization. | 12 | Highly efficient and anti-poisoning single-atom cobalt catalyst for selective hydrogenation of nitroarenes显示文摘Developing non-precious metal catalysts to selectively reduce functionalized nitroarenes with high efficiency is urgently desirable for the production of value-added amines.Herein,we report a novel,efficient,anti-poisoning single-atom cobalt catalyst(Co-NAC)for the highly selective hydrogenation of the nitro to amino group for nitroarenes baring various functional groups,including vinyl,cyano,and halogen.Using a combination of structure characterization techniques,we have confirmed that the cobalt species are predominantly present in the form of four-coordinated Co single sites anchored on nitrogen-assembly carbon(NAC)as the ordered mesoporous support.Co-NAC catalysts enable the full conversion and>99%selectivity with molecular H2 as a green reductant under mild conditions(80℃,2 MPa H2).As for the selective hydrogenation of 3-nitrostyrene,Co-NAC catalyst affords high catalytic productivity(19.7 h-1),which is superior to the cobalt nanoparticles(NPs)catalysts and most of the recently reported Co-based catalysts.This is attributed to the highly accessible atomically-dispersed Co active sites,the high surface area with ordered-mesoporous morphology and the prominent high content of nitrogen dopants.Notably,Co-NAC catalyst displays resistance towards sulfur-containing poisons(20 equivalents)and strong non-oxidizing acid(8 M),showing great potential for continuous application in the chemical industry. | Yuemin Lin Renfeng Nie Yuting Li Xun Wu Jiaqi Yu Shaohua Xie Yajing Shen Shanjun Mao Yuzhuo Chen Dan Lu Zongbi Bao Qiwei Yang Qilong Ren Yiwen Yang Fudong Liu Long Qi Wenyu Huang Zhiguo Zhang | 2022 | Nano Research2022,15,12: | 0 |
