2Transplantation of X-box-binding protein-1 gene-modified neural stem cells in the lateral ventricle of brain ischemia rats显示文摘X-box-binding protein-1 (XBP-1) is an essential transcription factor in endoplasmic reticulum stress. In this study, XBP-1 gene-transfected neural stem cells (NSCs) were transplanted into lesion sites to ensure stability and persistent expression of XBP-1, resulting in the exertion of anti-apoptotic effects. Simultaneously, XBP-1 gene transfection promotes the survival and differentiation of transplanted NSCs. Results from this study demonstrated that survival, proliferation and differentiation of XBP-1 gene-modified NSCs were enhanced when compared to unmodified NSCs at 28 days post-transplantation (P < 0.05). A diminished number of apoptotic neural cells increased Bcl-2 expression and reduced Bax expression, and were observed in the ischemic region of the XBP-1-NSCs group (P < 0.05). 3Key indicators affecting maize stalk lodging resistance of different growth periods under different sowing dates显示文摘The accurate evaluation of maize stalk lodging resistance in different growth periods enables timely management of lodging risks and ensures stable and high maize yields.Here,we established five diferent sowing dates to create diferent conditions for maize growth.We evaluated the effects of the different growth conditions on lodging resistance by determining stalk morphology,moisture content,mechanical strength and dry matter,and the relationship between stalk breaking force and these indicators during the silking stage(R1),milk stage(R3),physiological maturity stage(R6),and 20 days after R6.Plant height at R1 positively affected stalk breaking force.At R3,the cofficient of ear height and the dry weight per unit length of basal internodes were key indicators of stalk lodging resistance.At R6,the key indicators were the coefficient of the center of gravity height and plant fresh weight.After R6,the key indicator was the coefficient of the center of gravity height.The crushing strength of the fourth internode correlated significanty and positively with the stalk breaking force from R1 to R6,which indicates that crushing strength is a reliable indicator of stalk mechanical strength.These results suggest that high stalk strength and low ear height beneft lodging resistance prior to R6.During and after R6,the cofficient of the center of gravity height and the mechanical strength of basal internodes can be used to evaluate plant lodging resistance and the appropriate time for harvesting in fields with a high lodging risk.WANG Qun XUE Jun CHEN Jiang-lu FAN Ying-hu ZHANG Guo-qiang XIE Rui-zhi MING Bo HOU Peng WANG Ke-ru LI Shao-kun 2020Journal of Integrative Agriculture2020,19,10:13

5Exploratory study on new pulse detonation engines显示文摘The worldwide progress in studies on pulse detonation engines (PDE) is briefly reviewed and some results of our exploratory study on PDE are presented. Analysis of thermodynamic cycle is made and the specific impulse formula is improved. A proof-of-principle experiment of a two-phase PDE is successfully carried out, using poor-detonable liquid C8H16/air mixture with a low-energy system (total spark energy of 50 mJ) and a newly developed one-step detonation initiation method. The measured detonation wave pressure ratio is very close to that of C-J detonation. The effects of length, diameter and detonation frequency on PDE performance are experimentally investigated. For liquid hydrocarbon fuel/air mixture, the PDE operation is successfully realized with an engine length of 1000 mm and detonation frequency up to 36 Hz, which has made an important step toward practical PDE. The developed code can be used for simulating PDE operation processes including deflagration to detonation transition (DDT) phenomenon. 7Prognostic values of the integrated model incorporating the volume of metastatic regional cervical lymph node and pretreatment serum Epstein-Barr virus DNA copy number in predicting distant metastasis in patients with N1 nasopharyngeal carcinoma显示文摘Background: According to the 7 th edition of the American Joint Committee on Cancer(AJCC) staging system, over50% of patients with nasopharyngeal carcinoma(NPC) have N1 disease at initial diagnosis. However, patients with N1 NPC are relatively under-researched, and the metastasis risk of this group is not well-stratified. This study aimed to evaluate the prognostic values of gross tumor volume of metastatic regional lymph node(GTVnd) and pretreatment serum copy number of Epstein-Barr virus(EBV) DNA in predicting distant metastasis of patients with N1 NPC, and to develop an integrated prognostic model that incorporates GTVnd and EBV DNA copy number for this group of patients.Methods: The medical records of 787 newly diagnosed patients with nonmetastatic, histologically proven N1 NPC who were treated at Sun Yat-sen University Cancer Center between November 2009 and February 2012 were analyzed. Computed tomography-derived GTVnd was measured using the summation-of-area technique. Blood samples were collected before treatment to quantify plasma EBV DNA. The receiver operating characteristic(ROC) curve analysis was used to evaluate the cut-off point for GTVnd, and the area under the ROC curve was used to assess the predicted validity of GTVnd. The survival rates were assessed by Kaplan-Meier analysis, and the survival curves were compared using a log-rank test. Multivariate analysis was conducted using the Cox proportional hazard regression model.Results: The 5-year distant metastasis-free survival(DMFS) rates for patients with GTVnd > 18.9 vs.≤ 18.9 mL were82.2% vs. 93.2%(P < 0.001), and for patients with EBV DNA copy number > 4000 vs. < 4000 copies/mL were 83.5% vs.93.9%(P < 0.001). After adjusting for GTVnd, EBV DNA copy number, and T category in the Cox regression model, both GTVnd > 18.9 mL and EBV DNA copy number > 4000 copies/mL were significantly associated with poor prognosis(both P < 0.05). According to combination of GTVnd and EBV DNA copy number, all patients were divided into low-,moderate-, and high-risk groups, with the 5-year DMFS rates of 96.1,87.4, and 73.8%, respectively(P < 0.001). Multivariate analysis confirmed the prognostic value of this model for distant metastatic risk stratification(hazard ratio [HR],4.17; 95% confidence interval [CI] 2.34-7.59; P < 0.001).Conclusions: GTVnd and serum EBV DNA copy number are independent prognostic factors for predicting distant metastasis in NPC patients with N1 disease. 8Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 9Effects of Glutamine Supplementation on Patients Undergoing Abdominal Surgery显示文摘Objective To evaluate the effects of supplementation of glutamine (GLN) on maintaining glutathione (GSH) level, immune system function, liver function, and clinical outcome of patients receiving abdominal operation. Methods Forty patients undergoing elective abdominal surgical treatment were randomly divided into 2 groups: study group (n=20) and control group (n=20). All patients received total parenteral nutrition (TPN) for up to 7 days during perioperative period. The study group received TPN supplemented with GLN dipeptide while the control group received TPN without GLN dipeptide. Patients in both groups received equivalent nitrogen and caloric intake. Blood sample was taken on preoperative day, and the 1st,3rd,6th postoperative day to measure GSH level, immune indexes, and liver function indexes. Results The decrease of GSH level in plasma and red blood cell (RBC) in study group was less than that in control group during postoperative period. Ratio of GSH/glutathione disulfide (GSSG) in plasma in study group was higher than that in control group on the 3rd postoperative day (52.53±11.46 vs. 31.43±7.27, P=0.001). Albumin level in study group was higher than that in control group on the 3rd postoperative day (37.7±3.8g/L vs. 33.8±4.2g/P=0.02). There was no significant difference in the levels of immunoglobin (IgG,IgM,IgA) or T lymphocyte subgroup (CD4, CD8, CD4/CD8) in both groups during postoperative period. There was one case with infectious complication in control group, while none in study group. A trend of shortened hospital stay was observed in study group compared with control group (22.3± 2.1d vs.24.9±1.7d,P=0.32). Conclusions Supplementation of GLN-enriched TPN has beneficial effects on maintaining GSH levels in plasma and RBC, sustaining GSH/GSSG ratio and albumin level, and keeping antioxidant abilities during postoperative period in patients with abdominal operation, with the trends of decreasing incidence of infectious complication and shortening hospital stay.Yue-Dina Fan Jian-chun Yu Wei-ming Kang Qun Zhang 2009Chinese Medical Sciences Journal2009,24,1:7
10Mussel-inspired nanozyme catalyzed conductive and self-setting hydrogel for adhesive and antibacterial bioelectronics显示文摘Adhesive hydrogels have broad applications ranging from tissue engineering to bioelectronics;however,fabricating adhesive hydrogels with multiple functions remains a challenge.In this study,a mussel-inspired tannic acid chelated-Ag(TA-Ag)nanozyme with peroxidase(POD)-like activity was designed by the in situ reduction of ultrasmall Ag nanoparticles(NPs)with TA.The ultrasmall TA-Ag nanozyme exhibited high catalytic activity to induce hydrogel self-setting without external aid.The nanozyme retained abundant phenolic hydroxyl groups and maintained the dynamic redox balance of phenol-quinone,providing the hydrogels with long-term and repeatable adhesiveness,similar to the adhesion of mussels.The phenolic hydroxyl groups also afforded uniform distribution of the nanozyme in the hydrogel network,thereby improving its mechanical properties and conductivity.Furthermore,the nanozyme endowed the hydrogel with antibacterial activity through synergistic effects of the reactive oxygen species generated via POD-like catalytic reactions and the intrinsic bactericidal activity of Ag.Owing to these advantages,the ultrasmall TA-Ag nanozyme-catalyzed hydrogel could be effectively used as an adhesive,antibacterial,and implantable bioelectrode to detect bio-signals,and as a wound dressing to accelerate tissue regeneration while preventing infection.Therefore,this study provides a promising approach for the fabrication of adhesive hydrogel bioelectronics with multiple functions via mussel-inspired nanozyme catalysis.Zhanrong Jia Xuanhan Lv Yue Hou Kefeng Wang Fuzeng Ren Dingguo Xu Qun Wang Kelong Fan Chaoming Xie Xiong Lu 2021Bioactive Materials2021,6,9:7
11Gibberellin repression of axillary bud formation in Arabidopsis by modulation of DELLA-SPL9 complex activity显示文摘The formation of lateral branches has an important and fundamental contribution to the remarkable developmental plasticity of plants,which allows plants to alter their architecture to adapt to the challenging environment conditions.The Gibberellin(GA)phytohormones have been known to regulate the outgrowth of axillary meristems(AMs),but the specific molecular mechanisms remain unclear.Here we show that DELLA proteins regulate axillary bud formation by interacting and regulating the DNA-binding ability of SQUAMOSA-PROMOTER BINDING PROTEIN LIKE 9(SPL9),a micro RNA156-targeted squamosa promoter binding protein-like transcription factor.SPL9 participates in the initial regulation of axillary buds by repressing the expression of LATERAL SUPPRESSOR(LAS),a key regulator inthe initiation of AMs,and LAS contributes to the specific expression pattern of the GA deactivation enzyme GA2ox4,which is specifically expressed in the axils of leaves to form a low-GA cell niche in this anatomical region.Nevertheless,increasing GA levels in leaf axils by ectopically expressing the GA-biosynthesis enzyme GA20ox2 significantly impaired axillary meristem initiation.Our study demonstrates that DELLA-SPL9-LAS-GA2ox4 defines a core feedback regulatory module that spatially pattern GA content in the leaf axil and precisely control the axillary bud formation in different spatial and temporal.Qi-Qi Zhang Jia-Gang Wang Ling-Yan Wang Jun-Fang Wang Qun Wang Ping Yu Ming-Yi Bai Min Fan 2020Journal of Integrative Plant Biology2020,62,4:6
12Effect of microenvironmental quantitative regulation on growth of Korean pine trees planted under secondary forest显示文摘Korean pine(Pinus koraiensis) and broadleaved mixed forest in Northeast China has been changed regressively into secondary forest with almost no conifers.Planting Korean pine trees under the canopy of secondary forest is a feasible approach for recovering Korean pine and broadleaved mixed forest.For establishing an effective growth promotion method for under-canopy planted young Korean pine trees,two stands were selected as the experiment plots,Stand A(planted in 1989) and Stand B(planted in 1982),and an experiment of microenvironment regulation was conducted relying mainly on Opening degree(K=1,K=1.5,K=2,CK) in 2004.The results were shown that the adjustment had promoted growth of diameter and height of Korean pine planted in Stand A and Stand B,and had a significant influence on the growth rate of basal diameter,diameter at breast height and height in the two growth stands.The four years periodic increment of mean diameter and height of Korean pine planted in 1989 and in 1982 after regulation in K=1 level were 63.4%(D0) and 82.7%(H),64.8%(D1.3) and 69.7%(H) higher than that of control respectively.Quantitative regulation had significant influence on specific leaf area of Korean pine planted in 1989,and the current year specific leaf area(SLA) was lager than perennial year SLA.Quality indexes of natural pruning capacity,normal form quotient and crown size was not significantly changed but shown a positive tendency.The regulation scheme of Opening degree K=1 might be proper for adjusting the microenvironment of Korean pine trees planted under the canopy of secondary forest when the Korean pine trees were in the growth period of 15 to 26 years old in the experiment region.CONG Jian Shen Hai-Long YANG Wen-Hua FAN Shao-Hui ZHANG Qun 2011Journal of Forestry Research2011,22,2:5

14Distribution control and formation mechanism of gas inclusions in directionally solidified Al2O3-Er3Al5O12-ZrO2 ternary eutectic ceramic by laser floating zone melting显示文摘Distribution control and formation mechanism of gas inclusions formed in directionally solidified Al2O3-Er3Al5O12-ZrO2 eutectic ceramic rods are explored during laser floating zone melting. In atmospheric environment, highly-dense bubble-free eutectic rods are well fabricated at low solidification rate(<25μm/s). Gas inclusions form intermittently when the solidification rate is in the range of 25-50 μm/s,but produce continuously at higher solidification rates(100-200 μm/s). The gas inclusions exhibit an elongated finger-like pattern along the growth direction, which of the maximum value of diameter first increases and then decreases with increasing the solidification rate. Meanwhile, the volume fraction of gas inclusions increased gradually with the solidification rate. 16Whole-genome sequencing of monozygotic twins discordant for schizophrenia indicates multiple genetic risk factors for schizophrenia显示文摘Schizophrenia is a common disorder with a high heritability, but its genetic architecture is still elusive.We implemented whole-genome sequencing(WGS) analysis of 8 families with monozygotic(MZ) twin pairs discordant for schizophrenia to assess potential association of de novo mutations(DNMs) or inherited variants with susceptibility to schizophrenia. Eight non-synonymous DNMs(including one splicing site) were identified and shared by twins, which were either located in previously reported schizophrenia risk genes(p.V24689 I mutation in TTN, p.S2506 T mutation in GCN1L1, IVS3+1G > T in DOCK1) or had a benign to damaging effect according to in silico prediction analysis. By searching the inherited rare damaging or loss-of-function(LOF) variants and common susceptible alleles from three classes of schizophrenia candidate genes, we were able to distill genetic alterations in several schizophrenia risk genes, including GAD1, PLXNA2, RELN and FEZ1. Four inherited copy number variations(CNVs; including a large deletion at 16p13.11) implicated for schizophrenia were identified in four families, respectively. Most of families carried both missense DNMs and inherited risk variants, which might suggest that DNMs, inherited rare damaging variants and common risk alleles together conferred to schizophrenia susceptibility. 17Effects of transfected adenovirus-mediated transcription factor X-box binding protein 1 on hippocampal-derived neural stem cell proliferation and apoptosis under hypoxia显示文摘BACKGROUND:Neural stem cell(NSC) survival is closely associated with cell apoptosis in ischemic-hypoxic regions following transplantation.Numerous studies have revealed that X-box binding protein 1(XBP1) is a transcription factor during endoplasmic reticulum unfolded protein response and is essential for cell survival,differentiation,and anti-apoptotic effects.OBJECTIVE:To determine the effects of the XBP1 gene on NSC proliferation and apoptosis under hypoxic conditions following XBP1 gene transfection into rat embryonic hippocampal NSCs using recombinant adenovirus vector.DESIGN,TIME AND SETTING:In vitro experiments were performed at the Laboratory of Cell Biology of Jilin University and Laboratory of Proteomics,Department of Neurology,Jilin University,China from September 2008 to November 2009.MATERIALS:Recombinant adenovirus package XBP1 gene and Ad-XBP1-enhanced green fluorescent protein plasmid(Guangzhou Easywin BioMed Technology,China),rabbit anti-XBP1 and its target gene estrogen receptor degradation-enhancing α-mannosidase-like protein(EDEM),glucose-regulated protein 78(GRP78),anti-apoptotic molecule Bcl-2 and proapoptotic molecule Bax polyclonal antibody(Santa Cruz Biotechnology,Inc.,Santa Cruz,CA,USA),and CoCl2(Sigma,St.Louis,MO,USA) were used in the present study.METHODS:Hippocampi from embryonic,Sprague Dawley rats on gestational day 16 were harvested for NSC isolation and cloning,followed by immunofluorescence for Nestin and sub-culturing.The recombinant adenovirus Ad-XBP1-enhanced green fluorescent protein plasmid was transfected into rat embryonic hippocampal NSCs,and then CoCl2 was applied to induce hypoxia.MAIN OUTCOME MEASURES:Cell quantification and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colorimetric assay were utilized to detect proliferation in XBP1-transfected NSCs for 7 consecutive days.Western blot assay was utilized to quantify XBP1,GRP78,EDEM,Bcl-2,and Bax expression.Flow cytometry was used to measure apoptosis.RESULTS:NSC proliferation was significantly enhanced following XBP1 gene transfection(P < 0.05).Under hypoxic conditions,GRP78,EDEM,and Bcl-2 levels increased,but Bax levels decreased.In addition,NSC apoptosis decreased following transfection(P < 0.05).CONCLUSION:The XBP1 gene was successfully transfected into rat embryonic hippocampal NSCs using a recombinant adenovirus vector.NSC proliferation following transfection,as well as anti-apoptotic effects under hypoxia,was significantly increased.Ying Sha Baohua Liu Qun Liu Lei Song Jia Fan Yong Liu 2010Neural Regeneration Research2010,5,13:4
18Experimental Investigation of Nozzle Effects on Thrust and Inlet Pressure of an Air-breathing Pulse Detonation Engine显示文摘Nozzle effects on thrust and inlet pressure of a multi-cycle air-breathing pulse detonation engine (APDE) are investigated experimentally. An APDE with 68 mm in diameter and 2 050 mm in length is operated using gasoline/air mixture. Straight nozzle, converging nozzle, converging-diverging nozzle and diverging nozzle are tested. The results show that thrust augmentation of converging-diverging nozzle, diverging nozzle or straight nozzle is better than that of converging nozzle on the whole. Thrust augmentation of straight nozzle is worse than those of converging-diverging nozzle and diverging nozzle. Thrust augmentations of diverging nozzle with larger expansion ratio and converging-diverging nozzle with larger throat area range from 20% to 40% on tested frequencies and are better than those of congeneric other nozzles respectively. Nozzle effects on inlet pressure are also researched. At each frequency it is indicated that filling pressures and average peak pressures of inlet with diverging nozzle and converging-diverging nozzle with large throat cross section area are higher than those with straight nozzle and converging nozzle. 20Zinc finger protein 139 expression in gastric cancer and its clinical significance显示文摘AIM:To investigate the expression of zinc finger protein 139(ZNF139)in gastric cancer(GC),and to analyze its clinical significance.METHODS:A total of 108 patients who were diagnosed with GC and underwent surgery between January 2005and March 2007 were enrolled in this study.Gastric tumor specimens and paired tumor-adjacent tissues were collected and paraffin-embedded,and the clinicopathologic characteristics and prognosis were recorded.The expression of ZNF139,Bcl-2,Bax,and caspase-3 were determined by immunohistochemistry,and apoptosis was assessed by terminal deoxynucleotidyl transferasemediated d UTP-biotin nick end labeling.SPSS 13.0software was used for data processing and analyses,and significance was determined at P<0.05.RESULTS:The expression of ZNF139 was stronger in tumors than in tumor-adjacent tissues(66.67%vs 44.44%;P<0.01).Overexpression of ZNF139correlated with tumor differentiation,invasion depth,clinical stage,lymphatic metastasis,and blood vessel invasion(all P s<0.05).Patients with overexpression of ZNF139 had a poorer prognosis(P<0.01),and overexpression of ZNF139 was an independent factor for the prognosis of GC patients by a Cox survival analysis(P=0.02).A negative relationship between ZNF139 and the apoptosis index was observed(r=-0.686;P<0.01).The expression of Bcl-2 in GC was stronger than in tumor-adjacent tissues(66.67%vs41.67%),whereas the expression levels of Bax and caspase-3 were lower in primary tumors(54.63%and47.22%,respectively)than in tumor-adjacent tissues(73.15%and 73.15%,respectively)(all Ps<0.05).The expression of ZNF139 negatively correlated with caspase-3(r=-0.370;P<0.01).The expressions of Bcl-2 and Bax were also negatively correlated(r=-0.231;P=0.02).The expressions of caspase-3 and Bax protein were positively correlated(r=0.217;P=0.024).CONCLUSION:ZNF139 is related to clinicopathologic characteristics and prognosis of GC.Furthermore,it is overexpressed and involved in apoptosis in GC tissues by regulating caspase-3.Yong Li Qun Zhao Li-Qiao Fan Li-Li Wang Bi-Bo Tan Yan-Li Leng Yu Liu Dong Wang 2014World Journal of Gastroenterology2014,20,48:2
