1Gene cloning and function analysis of ABP9 protein which specifically binds to ABRE2 motif of maize Cat1 gene显示文摘A cDNA library was constructed using mRNA extracted from 17 days post-pollination (dpp) maize embryos and was screened by employing a yeast one-hybrid system for proteins specifically interacting with ABRE2 motif of maize Call gene. Three truncated overlapping positive clones designated ABP9 were obtained and the full-length cDNA was isolated by 5’ RACE. Searching the database revealed that ABP9 protein belongs to a bZIP-type transcription factor family. ABP9 protein speciflcally binds to ABRE2 motif and activates the expression of downstream reporter gene in yeast cells. 2Isolation and Characterization of a C-repeat Binding Transcription Factor from Maize显示文摘C-repeat binding proteins (CBFs) are a group of transcription factors that have been proven to be important for stress tolerance in plants. Many of these transcription factors transactivate the promoters of cold-regulated genes via binding to low temperature-or dehydration-responsive cis-elements, thus conferring plants cold acclimation. In the present study, we isolated a C-repeat binding transcription factor from maize using the yeast one-hybrid system with the C-repeat motif from the promoter of the Arabidopsis COR15a gene as bait. The isolated transcription factor is highly similar to the Arabidopsis CBF3 in their predicted amino acid sequences, and is therefore designated ZmCBF3. Point mutation analyses of the ZmCBF3-binding cis-element revealed (A/G)(C/T)CGAC as the core binding sequence. 6Isolation and functional analysis of transcription factor GmWRKY57B from soybean显示文摘WRKY proteins are a large family of transcriptional regulators in higher plants involved in many biological processes, such as responses to biotic and abiotic stress, plant development and metabolism. In this study, a soybean cDNA library was constructed and screened by yeast one-hybrid system using the 3×W-box element as a bait, and consequently a transcription factor, named GmWRKY57B, a member of group 3 of WRKY family, was obtained. GmWRKY57B specifically binds to the W-box of the Arabidopsis NPR1 (AtNPR1) promoter in yeast cells. This specific binding was further confirmed with an in vitro electrophoretic mobility shift assay. RT-PCR analysis showed that GmWRKY57B was expressed at a similar level in root, stem and leaf of the soybean plant. Overexpression of the GmWRKY57B gene in tobacco conferred the transgenics tolerance to drought stress. 12Expression of tachyplesin gene in yeast Pichia pastoris显示文摘Tachyplesin gene was designed and chemically synthesized with codon usage bias. The stop codon TAG and some restriction sites convenient for further cloning were added to this gene. The synthesized gene cloned into yeast expression vector pPIC9 (with α_secretion signal) was transformed into host strain GS115 by electroporation. The tachyplesin expressed from recombinants Y PIC27 and Y PIC42 showed an inhibition activity against the germination of the spores of \%Magnaporthe grisea\%.
