| 1 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 4 | Effects of water salinity and N application rate on water- and N-use efficiency of cotton under drip irrigation显示文摘In arid and semi-arid regions, freshwater scarcity and high water salinity are serious and chronic problems for crop production and sustainable agriculture development. We conducted a field experiment to evaluate the effect of irrigation water salinity and nitrogen(N) application rate on soil salinity and cotton yield under drip irrigation during the 2011 and 2012 growing seasons. The experimental design was a 3×4 factorial with three irrigation water salinity levels(0.35, 4.61 and 8.04 dS/m) and four N application rates(0, 240, 360 and 480 kg N/hm2). Results showed that soil water content increased as the salinity of the irrigation water increased, but decreased as the N application rate increased. Soil salinity increased as the salinity of the irrigation water increased. Specifically, soil salinity measured in 1:5 soil:water extracts was 218% higher in the 4.61 dS/m treatment and 347% higher in the 8.04 dS/m treatment than in the 0.35 dS/m treatment. Nitrogen fertilizer application had relatively little effect on soil salinity, increasing salinity by only 3%–9% compared with the unfertilized treatment. Cotton biomass, cotton yield and evapotranspiration(ET) decreased significantly in both years as the salinity of irrigation water increased, and increased as the N application rate increased regardless of irrigation water salinity; however, the positive effects of N application were reduced when the salinity of the irrigation water was 8.04 dS/m. Water use efficiency(WUE) was significantly higher by 11% in the 0.35 dS/m treatment than in the 8.04 dS/m treatment. There was no significant difference in WUE between the 0.35 dS/m treatment and the 4.61 dS/m treatment. The WUE was also significantly affected by the N application rate. The WUE was highest in the 480 kg N/hm2 treatment, being 31% higher than that in the 0 kg N/hm2 treatment and 12% higher than that in the 240 kg N/hm2 treatment. There was no significant difference between the 360 and 480 kg N/hm2 treatments. The N use efficiency(NUE) was significantly lower in the 8.04 dS/m treatment than in either the 4.61 dS/m or the 0.35 dS/m treatment. There was no significant difference in NUE between the latter two treatments. These results suggest that irrigation water with salinity <4.61 dS/m does not have an obvious negative effect on cotton production, WUE or NUE under the experimental conditions. | 6 | Protective effects of a novel drug RC28-E blocking both VEGF and FGF2 on early diabetic rat retina显示文摘AIM: To investigate protective effects of a novel recombinant decoy receptor drug RC28-E on retinal damage in early diabetic rats.METHODS: The streptozotocin(STZ)-induced diabetic rats were randomly divided into 6 groups: diabetes mellitus(DM) group(saline, 3 μL/eye); RC28-E at low(0.33 μg/μL, 3 μL), medium(1 μg/μL, 3 μL), and high(3 μg/μL,3 μL) dose groups; vascular endothelial growth factor(VEGF) Trap group(1 μg/μL, 3 μL); fibroblast growth factor(FGF) Trap group(1 μg/μL, 3 μL). Normal control group was included. At week 1 and 4 following diabetic induction,the rats were intravitreally injected with the corresponding solutions. At week 6 following the induction, apoptosis in retinal vessels was detected by TUNEL staining.Glial fibrillary acidic protein(GFAP) expression was examined by immunofluorescence. Blood-retinal barrier(BRB) breakdown was assessed by Evans blue assay.Ultrastructural changes in choroidal and retinal vessels were analyzed by transmission electron microscopy(TEM).Content of VEGF and FGF proteins in retina was measured by enzyme linked immunosorbent assay(ELISA). The retinal expression of intercellular cell adhesion molecule-1(ICAM-1), tumor necrosis factor-α(TNF-α), VEGF and FGF genes was examined by quantitative polymerase chain reaction(qPCR).RESULTS: TUNEL staining showed that the aberrantly increased apoptotic cells death in diabetic retinal vascular network was significantly reduced by treatments of medium and high dose RC28-E, VEGF Trap, and FGF Trap(all P<0.05), the effects of medium and high dose RC28-E or FGF Trap were greater than VEGF Trap(P<0.01). GFAP staining suggested that reactive gliosis was substantially inhibited in all RC28-E and VEGF Trap groups, but the inhibition in FGF Trap group was not as prominent.Evans blue assay demonstrated that only high dose RC28-E could significantly reduce vascular leakage in early diabetic retina(P<0.01). TEM revealed that the ultrastructures in choroidal and retinal vessels were damaged in early diabetic retina, which was ameliorated to differential extents by each drug. The expression of VEGF and FGF2 proteins was significantly upregulated in early diabetic retina, and normalized by RC28-E at all dosages and by the corresponding Traps. | 7 | Simultaneous quantification of ginsenoside Rg1 and its metabolites by HPLC–MS/MS:Rg1 excretion in rat bile, urine and feces显示文摘Ginsenoside Rg1(Rg1), the major effective component of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography–tandem mass spectrometry(LC–MS/MS) method, which could be used to validate and quantify the concentrations of Rg1 and its metabolites in Sprague-Dawley rat bile,urine, and feces after oral administration(25 mg/kg). Calibration curves offered satisfactory linearity(r40.995)within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80–120%. The excretion recoveries of Rg1, ginsenoside Rh1(Rh1), and protopanaxatriol(Ppt) in bile,urine, and feces combined were all greater than 70%. The fecal excretion recoveries of Rg1, Rh1, and Ppt were40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in bile.Urinary excretion accounted for only 0.04% of Rg1. | 15 | Optimal Waist Circumference Cut-off values for Identifying Metabolic Risk Factors in Middle-aged and Elderly Subjects in Shandong Province of China显示文摘Objective To study the optimal waist circumference(WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China. Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome(MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men. | 20 | Role of carbon in modifying solidification and microstructure of a Ni?based superalloy with high Al and Ti contents显示文摘The effect of carbon ranging from 0.014 to 0.071 wt.%on the solidification and microstructure of a Ni-based superalloy with high Al and Ti contents was studied.The results show that the increase in carbon addition significantly increases the size and volume fraction of MC carbides and promotes the change of their morphology from blocky to elongated shape.However,the carbon addition obviously decreases the size and volume fraction of eutectic(γ+γ′)and reducesηphase and borides formation.The change in carbide characteristics is mainly because of the increasing carbide-forming element and carbides precipitation temperature with the increase in carbon which favors the growth of them along the interdendritic liquid film.MC carbides are formed at an earlier solidification stage than the eutectic(γ+γ′).The increased carbide formation consumes more Ti,which delays and reduces the eutectic(γ+γ′)precipitation.The delay of eutectic(γ+γ′)precipitation leads to a deeper undercooling,which significantly decreases the critical Ti concentration for its precipitation.This,in turn,lowers Ti/Al ratio in residual liquids ahead of the eutectic(γ+γ′)and hence reducesηformation subsequently.B and Zr are slightly enriched in the carbides,which are considered during discussing how carbon influences the eutectic(γ+γ′)precipitation. | Guang‑di Zhao Xi‑min Zang Wen‑ru Sun | 2021 | Journal of Iron and Steel Research(International)2021，28，1： | 2 |
