1Characteristics of volatile organic compounds,NO_2,and effects on ozone formation at a site with high ozone level in Chengdu显示文摘Chengdu is a megacity in the southwest of China with high ozone(O_3) mixing ratio.Observation of volatile organic compounds(VOCs), NO_2 and O_3 with high temporal resolution was conducted in Chengdu to investigate the chemical processes and causes of high O_3 levels. The hourly mixing ratios of VOCs, NO2, and O_3 were monitored by an online system from 28 August to 7 October, 2016. According to meteorological conditions,Chengdu, with relative warm weather and low wind speed, is favorable to O_3 formation.Part of the O_3 in Chengdu may be transported from the downtown area. In O_3 episodes,the average mixing ratios of NO_2 and O_3 were 20.20 ppbv and 47.95 ppbv, respectively. In non-O_3 episodes, the average mixing ratios of NO_2 and O_3 were 16.38 ppbv and 35.15 ppbv,respectively. The average mixing ratio of total VOCs(TVOCs) was 40.29 ppbv in non-O_3 episodes, which was lower than that in O_3 episodes(53.19 ppbv). Alkenes comprised51.7% of the total O_3 formation potential(OFP) in Chengdu, followed by aromatics which accounted for 24.2%. Ethylene, trans-pentene, propene, and BTEX(benzene, ethylbenzene,toluene, m/p-xylene, o-xylene) were also major contributors to the OFP in Chengdu. In O_3 episodes, intensive secondary formations were observed during the campaign. Oxygenated VOCs(OVOCs), such as acetone, Methylethylketone(MEK), and Methylvinylketone(MVK)were abundant. Isoprene rapidly converted to MVK and Methacrolein(MACR) during O_3 episodes. 3Multifunctional dextran micelles as drug delivery carriers and magnetic resonance imaging probes显示文摘Multifunctional nanoparticles combining diagnostic and therapeutic agents into a single platform make cancer theranostics possible and have attracted wide interests in the field. In this study, a multifunctional nanocomposite based on dextran and superparamagnetic iron oxide nanoparticles(SPIO) was prepared for drug delivery and magnetic resonance imaging(MRI). Amphiphilic dextran was synthesized by grafting stearyl acid onto the carbohydrate backbone, and micelle was formed by the resulted amphiphilic dextran with low critical micelle concentration at 1.8 mg L-1. Doxorubicin(DOX) and a cluster of the manganese-doped iron oxide nanoparticles(Mn-SPIO)nanocrystals were then coencapsulated successfully inside the core of dextran micelles, resulting in nanocomposites with diameter at about 100 nm. Cell culture experiments demonstrated the potential of these Mn-SPIO/DOX nanocomposites as an effective multifunctional nanoplatform for the delivery of anticancer drug DOX with a loading content(DLC) of 16 %. Confocal laser scanning microscopyreveals that the Mn-SPIO/DOX had excellent internalization ability against MCF-7/Adr cells after 2-h labeling compared with free DOXáHCl. Under a 3.0-T MRI scanner, Mn-SPIO/DOX nanocomposite-labeled cells in gelatin phantom show much darker images than the control. Their transverse relaxation(T2) rate is also significantly higher than that of the control cells(33.9 versus 2.3 s-1). 4Reduction of polyethylenimine-coated iron oxide nanoparticles induced autophagy and cytotoxicity by lactosylation显示文摘Superparamagnetic iron oxide(SPIO)nanoparticles are excellent magnetic resonance contrast agents and surface engineering can expand their applications.When covered with amphiphilic alkyl-polyethyleneimine(PEI),the modified SPIO nanoparticles can be used as MRI visible gene/drug delivery carriers and cell tracking probes.However,the positively charged amines of PEI can also cause cytotoxicity and restricts their further applications.In this study,we used lactose to modify amphiphilic low molecular weight polyethylenimine(C_(12-)PEI_(2K))at different lactosylation degree.It was found that the N-alkyl-PEI-lactobionic acid wrapped SPIO nanocomposites show better cell viability without compromising their labelling efficacy as well as MR imaging capability in RAW 264.7 cells,comparing to the unsubstituted ones.Besides,we found the PEI induced cell autophagy can be reduced via lactose modification,indicating the increased cell viability might rely on down-regulating autophagy.Thus,our findings provide a new approach to overcome the toxicity of PEI wrapped SPIO nanocomposites by lactose modification.Jiuju Du Wencheng Zhu Li Yang Changqiang Wu Bingbing Lin Jun Wu Rongrong Jin Taipeng Shen Hua Ai 2016Regenerative Biomaterials2016,3,4:5
5Nuclear m^(6)A reader YTHDC1 regulates the scaffold function of LINE1 RNA in mouse ESCs and early embryos显示文摘N^(6)-methyladenosine(m^(6)A)on chromosome-associated regulatory RNAs(carRNAs),including repeat RNAs,plays important roles in tuning the chromatin state and transcription,but the intrinsic mechanism remains unclear.Here,we report that YTHDC1 plays indispensable roles in the self-renewal and differentiation potency of mouse embryonic stem cells(ESCs),which highly depends on the m^(6)A-binding ability.Ythdcl is required for sufficient rRNA synthesis and repression of the 2-cell(2C)transcriptional program in ESCs,which recapitulates the transcriptome regulation by the LINE1 scaffold.Detailed analyses revealed that YTHDC1 recognizes m^(6)A on LINE1 RNAs in the nucleus and regulates the formation of the LINE1-NCL partnership and the chromatin recruitment of KAP1.Moreover,the establishment of H3K9me3 on 2C-related retrotrans-posons is interrupted in Ythdcl-depleted ESCs and inner cell mass(ICM)cells,which consequently increases the transcriptional activities.Our study reveals a role of m^(6)A in regulating the RNA scaffold,providing a new model for the RNA-chromatin cross-talk.Chuan Chen Wenqiang Liu Jiayin Guo Yuanyuan Liu Xuelian Liu Jun Liu Xiaoyang Dou Rongrong Le Yixin Huang Chong Li Lingyue Yang Xiaochen Kou Yanhong Zhao You Wu Jiayu Chen Hong Wang Bin Shen Yawei Gao Shaorong Gao 2021Protein & Cell2021,12,6:4
6Temporal and spatial cellular and molecular pathological alterations with single-cell resolution in the adult spinal cord after injury显示文摘Spinal cord injury(SCI)involves diverse injury responses in different cell types in a temporally and spatially specific manner.Here,using single-cell transcriptomic analyses combined with classic anatomical,behavioral,electrophysiological analyses,we report,with single-cell resolution,temporal molecular and cellular changes in crush-injured adult mouse spinal cord.Data revealed pathological changes of 12 different major cell types,three of which infiltrated into the spinal cord at distinct times post-injury.We discovered novel microglia and astrocyte subtypes in the uninjured spinal cord,and their dynamic conversions into additional stage-specific subtypes/states.Most dynamic changes occur at 3-days post-injury and by day-14 the second wave of microglial activation emerged,accompanied with changes in various cell types including neurons,indicative of the second round of attacks.By day-38,major cell types are still substantially deviated from uninjured states,demonstrating prolonged alterations.This study provides a comprehensive mapping of cellular/molecular pathological changes along the temporal axis after SCI,which may facilitate the development of novel therapeutic strategies,including those targeting microglia.Chen Li Zhourui Wu Liqiang Zhou Jingliang Shao Xiao Hu Wei Xu Yilong Ren Xingfei Zhu Weihong Ge Kunshan Zhang Jiping Liu Runzhi Huang Jing Yu Dandan Luo Xuejiao Yang Wenmin Zhu Rongrong Zhu Changhong Zheng Yi Eve Sun Liming Cheng 2022Signal Transduction and Targeted Therapy2022,7,4:4
7Application of next-generation sequencing to screen for pathogenic mutations in 123 unrelated Chinese patients with Marfan syndrome or a related disease显示文摘Marfan syndrome(MFS) is a systemic connective tissue disease principally affecting the ocular, skeletal and cardiovascular systems. This autosomal dominant disorder carries a prevalence of 1:3,000 to 1:5,000. This study aims to define the mutational spectrum of MFS related genes in Chinese patients and to establish genotype-phenotype correlations in MFS. Panel-based targeted next-generation sequencing was used to analyze the FBN1, TGFBR1 and TGFBR2 genes in 123 unrelated Chinese individuals with MFS or a related disease. Genotype-phenotype correlation analyses were performed in mutation-positive patients. The results showed that 97 cases/families(78.9%;97/123) harbor at least one(likely) pathogenic mutation, most of which were in FBN1;four patients had TGFBR1/2 mutations;and one patient harbored a SMAD3 mutation. Three patients had two FBN1 mutations, and all patients showed classical MFS phenotypes. Patients with a dominant negative-FBN1 mutation had a higher prevalence of ectopia lentis(EL). Patients carrying a haploinsufficiency-FBN1 mutation tended to have aortic dissection without EL. 8A study of the acoustical radiation force considering attenuation显示文摘Acoustical tweezer is a primary application of the radiation force of a sound field. When an ultrasound focused beam passes through a micro-particle, like a cell or living biological specimens, the particle will be manipulated accurately without physical contact and invasion, due to the three-dimensional acoustical trapping force. Based on the Ray acoustics approach in the Mie regime, this work discusses the effects on the particle caused by Gaussian focused ultrasound, studies the acoustical trapping force of spherical Mie particles by ultrasound in any position, and analyzes the numerical calculation on the two-dimensional acoustical radiation force. This article also analyzes the conditions for the acoustical trapping phenomenon, and discusses the impact of the initial position and size of the particle on the magnitude of the acoustical radiation force. 9Flexible cation-based threshold selector for resistive switching memory integration显示文摘Emerging resistive switching random access memory(RRAM), considered as the most promising candidate of flash memory, is favorable for in flexible electronic system. However, in high density flexible crossbar RRAM array, crosstalk issue that currents from the neighboring unselected cell lead to failure of write and read operations, still keeps a main bottleneck. Therefore, flexible selector compatible with the flexibility of the RRAM array should be focused on to configure one selector-one resistor(1 S1 R) system,which is immune to crosstalk issue. In this paper, flexible cation-based threshold switching(TS) selectors(Pt/Ag/Hf O_2/Pt/Ti/parylene) are fabricated and the compressive performance is studied systematically.The device shows excellent bidirectional volatile TS characteristics, including high selectivity ratio(10~9), low operating voltages(|VTH|<1 V), ultra-low leakage current(~10^(-13) A) and good flexibility. 15Overexpression of Lrp5 enhanced the anti-breast cancer effects of osteocytes in bone显示文摘Osteocytes are the most abundant cells in bone,which is a frequent site of breast cancer metastasis.Here,we focused on Wnt signaling and evaluated tumor-osteocyte interactions.In animal experiments,mammary tumor cells were inoculated into the mammary fat pad and tibia.The role of Lrp5-mediated Wnt signaling was examined by overexpressing and silencing Lrp5 in osteocytes and establishing a conditional knockout mouse model.The results revealed that administration of osteocytes or their conditioned medium(CM)inhibited tumor progression and osteolysis.Osteocytes overexpressing Lrp5 or β-catenin displayed strikingly elevated tumor-suppressive activity,accompanied by downregulation of tumor-promoting chemokines and upregulation of apoptosis-inducing and tumor-suppressing proteins such as p53.The antitumor effect was also observed with osteocyte-derived CM that was pretreated with a Wnt-activating compound.Notably,silencing Lrp5 in tumors inhibited tumor progression,while silencing Lrp5 in osteocytes in conditional knockout mice promoted tumor progression.Osteocytes exhibited elevated Lrp5 expression in response to tumor cells,implying that osteocytes protect bone through canonical Wnt signaling.Thus,our results suggest that the Lrp5/β-catenin axis activates tumor-promoting signaling in tumor cells but tumor-suppressive signaling in osteocytes.We envision that osteocytes with Wnt activation potentially offer a novel cell-based therapy for breast cancer and osteolytic bone metastasis.Shengzhi Liu Di Wu Xun Sun Yao Fan Rongrong Zha Aydin Jalali Yan Feng Kexin Li Tomohiko Sano Nicole Vike Fangjia Li Joseph Rispoli Akihiro Sudo Jing Liu Alexander Robling Harikrishna Nakshatri Bai-Yan Li Hiroki Yokota 2021Bone Research2021,9,4:1

17Genome-wide association analysis reveals genetic loci and candidate genes associated with intramuscular fat in Duroc pigs显示文摘Intramuscular fat(IMF) is a major meat-quality trait in pigs. The content of IMF is directly associated with the taste and flavor of pork. As a complex trait, there could be multiple genes affecting IMF content in pork. Genomewide association study is a powerful tool to detect genomic regions associated with phenotypic variations. The objectives of the present study were to identify or refine the positions of genomic regions affecting IMF, and to characterize candidate genes and pathways that may influence this trait. Of note, we identified a significant region in longissium dorsi muscle in a Duroc pig population for IMF content with Porcine SNP60 v2 Bead Chip. This region spans 1.24 Mb on chromosome 8 and had been identified as a quantitative trait locus for IMF in Pietrain, Large White, Landrace, and Leicoma pigs. In this region, eight SNPs were significantly associated with IMF content. Three genes proximal to these significant SNPs were considered candidate genes, including ZDHHC16, LOC102162218 and PCDH7.
