1Transcriptomic and genetic approaches reveal an essential role of the NAC transcription factor SlNAP1 in the growth and defense response of tomato显示文摘With global climate change,plants are frequently being exposed to various stresses,such as pathogen attack,drought,and extreme temperatures.Transcription factors(TFs)play crucial roles in numerous plant biological processes;however,the functions of many tomato(Solanum lycopersicum L.)TFs that regulate plant responses to multiple stresses are largely unknown.Here,using an RNA-seq approach,we identified SlNAP1,a NAC TF-encoding gene,which was strongly induced by various stresses.By generating SlNAP1 transgenic lines and evaluating their responses to biotic and abiotic stresses in tomato,we found that SlNAP1-overexpressing plants showed significantly enhanced defense against two widespread bacterial diseases,leaf speck disease,caused by Pseudomonas syringae pv.tomato(Pst)DC3000,and root-borne bacterial wilt disease,caused by Ralstonia solanacearum.In addition,SlNAP1 overexpression dramatically improved drought tolerance in tomato.Although the SlNAP1-overexpressing plants were shorter than the wild-type plants during the early vegetative stage,eventually,their fruit yield increased by 10.7%.Analysis of different hormone contents revealed a reduced level of physiologically active gibberellins(GAs)and an increased level of salicylic acid(SA)and abscisic acid(ABA)in the SlNAP1-overexpressing plants.Moreover,EMSAs and ChIP-qPCR assays showed that SlNAP1 directly activated the transcription of multiple genes involved in GA deactivation and both SA and ABA biosynthesis.Our findings reveal that SlNAP1 is a positive regulator of the tomato defense response against multiple stresses and thus may be a potential breeding target for improving crop yield and stress resistance.Jiao Wang Chenfei Zheng Xiangqi Shao Zhangjian Hu Jianxin Li Ping Wang Anran Wang Jingquan Yu Kai Shi 2020Horticulture Research2020,7,1:9

3Heterogeneous Network-Based Chronic Disease Progression Mining显示文摘Healthcare insurance fraud has caused billions of dollars in losses in public healthcare funds around the world. In particular, healthcare insurance fraud in chronic diseases is especially rampant. Understanding disease progression can help investigators detect healthcare insurance frauds early on. Existing disease progression methods often ignore complex relations, such as the time-gap and pattern of disease occurrence. They also do not take into account the different medication stages of the same chronic disease, which is of great help when conducting healthcare insurance fraud detection and reducing healthcare costs. In this paper, we propose a heterogeneous network-based chronic disease progression mining method to improve the current understanding on the progression of chronic diseases, including orphan diseases. The method also considers the different medication stages of the same chronic disease. 6Highly branched poly(β-amino ester)s with narrow molecular weight distribution: Fractionation and gene transfection activity显示文摘Highly branched poly(β-amino ester)s(HPAEs) have shown their great promise in gene delivery. However, their broad molecular weight distribution(MWD) poses an additional challenge to the mechanistic understanding of the influence of molecular weight(MW) on their gene transfection activity. Using a stepwise precipitation strategy, HPAEs were fractionated. It is shown that MW has a significant effect on the transfection activity and cytotoxicity of HPAEs. The intermediate MW mediates higher transfection efficiency while maintaining high cell viability. Mechanistic studies show that the intermediate MW confers stronger DNA binding affinity to HPAEs, leading to the formulation of polyplexes with a relatively smaller size and more positive zeta potential.
