1A double-blind,randomized,placebo-and positive-controlled phase III trial of 1% benvitimod cream in mild-to-moderate plaque psoriasis显示文摘Background:Benvitimod cream,a novel synthetic small molecule,was effective in treating mild-to-moderate plaque psoriasis.We conducted a phase III clinical trial to assess the efficacy and safety of benvitimod cream in patients with mild-to-moderate plaque psoriasis.Methods:We randomly assigned 686 patients(2:1:1)to receive 1%benvitimod cream,0.005%calcipotriol ointment or placebo twice a day for 12 weeks.The primary efficacy end points were the percentage of patients with a 75%or greater reduction from baseline in the psoriasis area and severity index(PASI 75)score and with a score of 0 or 1 in static physician’s global assessment(sPGA)at week 12.Results:The results showed that 50.4%of patients in the benvitimod group achieved PASI 75,which was significantly higher than that in the calcipotriol(38.5%,P<0.05)and placebo(13.9%,P<0.05)groups.The proportion of patients achieving an sPGA score 0 or 1 was 66.3%in the benvitimod group and 63.9%in the calcipotriol group,which were both significantly higher than that in the placebo group(34%,P<0.05).In the long-term follow-up study,50.8%of patients experienced recurrence.After retreatment with 1%benvitimod,73.3%of patients achieved an sPGA score of 0 or 1 again at week 52.Adverse events included application site irritation,follicular papules,and contact dermatitis.No systemic adverse reactions were reported.Conclusion:During this 12-week study,benvitimod cream was demonstrated with high effectiveness and safety in patients with mild-to-moderate plaque psoriasis.Lin Cai Gen-Hui Chen Qian-Jin Lu Min Zheng Yu-Zhen Li Jin Chen Jie Zheng Fu-Ren Zhang Jian-Bin Yu Sen Yang Fu-Qiu Li Sheng-Xiang Xiao Qiu-Ning Sun Jin-Hua Xu Xing-Hua Gao Hong Fang Tian-Wen Gao Fei Hao Quan-Zhong Liu Ya-Ting Tu Ruo-Yu Li Bao-Xi Wang Dan-Qi Deng Qing-Shan Zheng Hong-Xia Liu Jian-Zhong Zhang 2020Chinese Medical Journal2020,,24:11

3Comprehensive analysis of NAC transcription factors and their expression during fruit spine development in cucumber (Cucumis sativus L.)显示文摘The cucumber(Cucumis sativus L.)is an important vegetable crop worldwide,and fruit trichomes or spines are an important trait for external fruit quality.The mechanisms underlying spine formation are not well understood,but the plant-specific NAC family of transcription factors may play important roles in fruit spine initiation and development.In this study,we conducted a genome-wide survey and identified 91 NAC gene homologs in the cucumber genome.Clustering analysis classified these genes into six subfamilies;each contained a varying number of NAC family members with a similar intron–exon structure and conserved motifs.Quantitative real-time PCR analysis revealed tissue-specific expression patterns of these genes,including 10 and 12 that exhibited preferential expression in the stem and fruit,respectively.Thirteen of the 91 NAC genes showed higher expression in the wild-type plant than in its near-isogenic trichome mutant,suggesting their important roles in fruit spine development.Exogenous application of four plant hormones promoted spine formation and increased spine density on the cucumber fruits;several NAC genes showed differential expression over time in response to phytohormone treatments on cucumber fruit,implying their essential roles in fruit-trichome development.Among the NAC genes identified,12 were found to be targets of 13 known cucumber micro-RNAs.Collectively,these findings provide a useful resource for further analysis of the interactions between NAC genes and genes underlying trichome organogenesis and development during fruit spine development in cucumber.Xingwang Liu Ting wang Ezra Bartholomew Kezia Black Mingming Dong Yaqi Zhang Sen Yang Yanling Cai Shudan Xue Yiqun Weng Huazhong Ren 2018Horticulture Research2018,5,1:10

5NLRP3 Deficiency Attenuates Secondary Degeneration of Visual Cortical Neurons Following Optic Nerve Injury显示文摘In the visual pathway, optic nerve(ON) injury may cause secondary degeneration of neurons in distal regions, such as the visual cortex. However, the role of the neuroinflammatory response in regulating secondary impairment in the visual cortex after ON injury remains unclear. The NOD-like receptor family pyrin domain containing 3(NLRP3) is an important regulator of neuroinflammation. In this study, we established a mouse model of unilateral ON crush(ONC) and showed that the expression of NLRP3 was significantly increased in the primary visual cortex(V1) as a response to ONC and that the NLRP3 inflammasome was activated in the contralateral V1 1 days–14 days after ONC. Ablation of the NLRP3 gene significantly decreased the trans-neuronal degeneration within 14 days. Visual electrophysiological function was improved in NLRP3-/- mice. 7Role of LAP^+CD4^+ T cells in the tumor microenvironment of colorectal cancer显示文摘AIM To investigate the abundance and potential functions of LAP^+CD4^+ T cells in colorectal cancer(CRC). METHODS Proportions of LAP^+CD4^+ T cells were examined in peripheral blood and tumor/paratumor tissues of CRC patients and healthy controls using flow cytometry. Expression of phenotypic markers such as forkhead box(Fox)p3, cytotoxic T-lymphocyte-associated protein(CTLA)-4, chemokine CC receptor (CCR)4 and CCR5 was measured using flow cytometry. LAP^-CD4^+ and LAP^+CD4^+ T cells were isolated using a magnetic cellsorting system and cell purity was analyzed by flow cytometry. Real-time quantitative polymerase chain reaction was used to measure expression of cytokines interleukin (IL)-10 and transforming growth factor(TGF)-β.RESULTS The proportion of LAP^+CD4^+ T cells was significantly higher in peripheral blood from patients (9.44% ± 3.18%) than healthy controls (1.49% ± 1.00%, P < 0.001). Among patients, the proportion of LAP^+CD4^+ T cells was significantly higher in tumor tissues(11.76% ± 3.74%) compared with paratumor tissues (3.87% ± 1.64%, P < 0.001). We also observed positive correlations between the proportion of LAP^+CD4^+ T cells and TNM stage(P < 0.001), distant metastasis(P < 0.001) and serum level of carcinoembryonic antigen(P < 0.05). Magnetic-activated cell sorting gave an overall enrichment of LAP^+CD4^+ T cells (95.02% ± 2.87%), which was similar for LAP^-CD4^+ T cells(94.75% ± 2.76%). In contrast to LAP^-CD4^+ T cells, LAP^+CD4^+ T cells showed lower Foxp3 expression but significantly higher levels of CTLA-4, CCR4 and CCR5(P < 0.01). 8EGFR/MAPK Signaling Regulates the Proliferation of Drosophila Renal and Nephric Stem Cells显示文摘Tissue homeostasis,accomplished through the self-renewal and differentiation of resident stem cells,is critical for the maintenance of adult tissues throughout an animal's lifetime.Adult Drosoplula Malpighian tubules(MTs or fly kidney) are maintained by renal and nephric stem cells(RNSCs) via self-renewing divisions,however,it is unclear how RNSC proliferation and differentiation are regulated.Here we show that EGFR/MAPK signaling is dispensable for RNSC maintenance,but required for RNSC proliferation in vivo.Inactivation of the EGFR/MAPK pathway blocks or greatly retards RNSC cell cycle progression:conversely,over-activation of EGFR/MAPK signaling results in RNSC over-proliferation and disrupts the normal differentiation of renablasts(RBs),the immediate daughters of RNSC divisions.Our data further suggest that EGFR/MAPK signaling functions independently of JAK/STAT signaling and that dMyc and CycE partially mediate EGFR/MAPK signaling in MTs.Together,our data suggest a principal role of EGFR/MAPK signaling in regulating RNSC proliferation,which may provide important clues for understanding mammalian kidney repair and regeneration following injury.Zhouhua Li Sen Liu Yu Cai 2015Journal of Genetics and Genomics2015,42,1:2

14Nanocrystalline ZrO_2 preparation and kinetics research of phase transition显示文摘The precursor of nanocrystalline ZrO2 was synthesized by solid-state reaction at low heat using ZrOCl2·8H2O,and Na2CO3·10H2O as raw materials.The nanocrystalline ZrO2 was obtained by calcining the precursor.The precursor and its calcined products were characterized using TG/DTA,FT-IR,XRD,and SEM.The results showed that the precursor dried at 353 K was a zirconyl carbonate compound.When the precursor was calcined at 673 K for 150min,highly crystallization ZrO2 with tetragonal structure (space group P42/nmc (137)) was obtained with a crystallite size of 24 nm.However,when the precursor was calcined at 1023 K for 150min,highly crystallization ZrO2 with monoclinic structure (space group P21/c (14)) was obtained with a crystallite size of 20 nm.The mechanism and kinetics of the thermal process of the precursor were studied using DTA and XRD techniques.Based on the Kissinger and Arrhenius equation,the values of the activation energies associated with the thermal process of the precursor were determined to be 26.80 and 566.73 kJ·mol-1 for the first and third steps,respectively.The mechanism of ZrO2 phase transition from tetragonal to monoclinic structure is the random nucleation and growth of nuclei reaction.Wu, Wenwei Cai, Jinchao Wu, Xuehang Li, Yongni Liao, Sen 2012Rare Metals2012,31,1:1

18A Survey of Urolithiasis in Young Children Fed Infant Formula Contaminated with Melamine in Two Townships of Gansu,China显示文摘Objective To determine the prevalence of urolithiasis in young children fed infant formula(IF) contaminated with melamine,and the association between IF consumption and urolithiasis.Design A total of 2 733 children ≤3 years of age on September 1,2008 in two townships of Gansu Province,China were studied.Cases of urolithiasis were diagnosed by ultrasonography.Milk product consumption was determined by their caregivers.Remaining IF samples were tested for melamine and cyanuric acid.Results Of 2 733 eligible children in the two townships,2 186(80%) were enrolled in our study.Overall,16.6%(362) of 2 186 children had urolithiasis.The prevalence was 24.6% in children exclusively fed Sanlu brand IF,9.7% in those fed other IF,and 8.5% in those fed exclusively on other milk products.For children exclusively breast-fed,no urolithiasis was found(P<0.05).The prevalence of urolithiasis was 11.4% in children fed 400 g of Sanlu IF,rising to 37.5% in children fed over 25 600 g.Of 48 Sanlu IF samples,91.7% contained melamine(median=1 800 ppm;range=45-4 700) and 66.7% contained cyanuric acid(median=1.2 ppm;range=0.4-6.3).Melamine was also detected in 22.2% of 36 other brand IF(median=27.5 ppm,range=4-50).Conclusions Urolithiasis was associated with melamine-contaminated IF.Although one product caused most morbidity,other milk products may have also contributed to the outbreak.SHI Guo Qing WANG Zi Jun FENG Zi Jian GAO Yong Jun LIU Jian Di SHEN Tao LIMing YANG Jin XU Hai Bing JIANG Xiao Hong WANG Zhao Nan CAI Mei WANG Yu Min ZHU Ye Fan LIU Hui Hui WANG Rui XIONG Wei Yi WANG Zhu Tian YANG Wei Zhong HOU Pei Sen ZENG Guang WANG Yu 2012Biomedical and Environmental Sciences2012,25,2:1
