| 3 | Prognostic factors of refractory NSCLC patients receiving anlotinib hydrochloride as the third-or further-line treatment显示文摘Objective:Anlotinib hydrochloride is a multitarget tyrosine kinase inhibitor that targets vascular endothelial growth factor receptor,fibroblast growth factor receptor,platelet-derived growth factor receptor,c-Kit,and c-MET;therefore,it exhibits both antitumor and anti-angiogenetic activities.A phase III trial has shown that anlotinib improved progression-free survival(PFS)and overall survival(OS)in patients with advanced non-small cell lung cancer(NSCLC),who presented with progressive disease or intolerance after standard chemotherapy.This study aimed to analyze the characteristics of patients receiving anlotinib treatment to determine the dominant populations who are fit for the treatment.Methods:Data were collected from March 2015 to January 2017 from a randomized,double-blind,placebo-controlled,multicenter,phase III trial of anlotinib(ALTER0303).A total of 437 patients were enrolled and randomly allocated(2:1)to the anlotinib and placebo groups.Kaplan–Meier analysis and log-rank test were performed to compare PFS and OS.Cox proportional hazards model was adopted for multivariate prognostic analysis.Results:Multivariate analysis indicated that high post-therapeutic peripheral blood granulocyte/lymphocyte ratio and elevated alkaline phosphatase levels were independent risk factors for PFS.Meanwhile,elevated thyroid-stimulating hormone,blood glucose,and triglyceride levels;hypertension;and hand–foot syndrome were independent protective factors of PFS.High posttherapeutic peripheral blood granulocyte/lymphocyte ratio,an Eastern Cooperative Oncology Group(ECOG)score≥2,and the sum of the maximal target lesion length at baseline were independent risk factors of OS,and hypertriglyceridemia was an independent protective factor of OS.Conclusions:This study preliminarily explored the possible factors that affected PFS and OS after anlotinib treatment in patients with advanced refractory NSCLC,and the baseline characteristics of the therapeutically dominant populations were then identified. | Jing Wang Yizhuo Zhao Qiming Wang Li Zhang Jianhua Shi Zhehai Wang Ying Cheng Jianxing He Yuankai Shi Hao Yu Yang Zhao Weiqiang Chen Yi Luo Xiuwen Wang Kejun Nan Faguang Jin Jian Dong Baolan Li Zhujun Liu Baohui Han Kai Li | 2018 | Cancer Biology & Medicine2018,15,4: | 46 |

| 9 | Numerical evaluation of the effectiveness of NO_2 and N_2O_5 generation during the NO ozonation process显示文摘Wet scrubbing combined with ozone oxidation has become a promising technology for simultaneous removal of SO_2 and NO_x in exhaust gas.In this paper,a new 20-species,76-step detailed kinetic mechanism was proposed between O_3 and NO_x.The concentration of N_2O_5 was measured using an in-situ IR spectrometer.The numerical evaluation results kept good pace with both the public experiment results and our experiment results.Key reaction parameters for the generation of NO_2 and N_2O_5 during the NO ozonation process were investigated by a numerical simulation method.The effect of temperature on producing NO_2 was found to be negligible.To produce NO_2,the optimal residence time was1.25 sec and the molar ratio of O_3/NO about 1.For the generation of N_2O_5,the residence time should be about 8 sec while the temperature of the exhaust gas should be strictly controlled and the molar ratio of O_3/NO about 1.75.This study provided detailed investigations on the reaction parameters of ozonation of NO_x by a numerical simulation method,and the results obtained should be helpful for the design and optimization of ozone oxidation combined with the wet flue gas desulfurization methods(WFGD) method for the removal of NO_x. | Haiqiang Wang Zhuokai Zhuang Chenglang Sun Nan Zhao Yue Liu Zhongbiao Wu | 2016 | Journal of Environmental Sciences2016,28,3: | 19 |
| 10 | Blockade of Tim-3 Pathway Ameliorates Interferon-γ Production from Hepatic CD8^+ T Cells in a Mouse Model of Hepatitis B Virus Infection显示文摘T cell immunoglobulin-and mucin-domain-containing molecule-3(Tim-3) has been reported to participate in the pathogenesis of inflammatory diseases. However,whether Tim-3 is involved in hepatitis B virus(HBV) infection remains unknown. Here,we studied the expression and function of Tim-3 in a hydrodynamics-based mouse model of HBV infection. A significant increase of Tim-3 expression on hepatic T lymphocytes,especially on CD8+ T cells,was demonstrated in HBV model mice from day 7 to day 18. After Tim-3 knockdown by specific shRNAs,significantly increased IFN-γ production from hepatic CD8+ T cells in HBV model mice was observed. Very interestingly,we found Tim-3 expression on CD8+ T cells was higher in HBV model mice with higher serum anti-HBs production. Moreover,Tim-3 knockdown influenced anti-HBs production in vivo. | 13 | GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://bigd.big.ac.cn/gsa or http://gsa.big.ac.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 15 | The roles of surfactant protein D during Aspergillus fumigatus infection in human corneal epithelial cells显示文摘AIM: To investigate roles of surfactant protein D (SP-D) and relative cytokines in human corneal epithelial(HCE) cells exposed to aspergillus fumigatus (AF) antigens.METHODS: HCE cells cultured in vitro with AF antigens and sampled at 0, 0.5, 1 hour, 2, 4, 6 and 8 hours. The expression of SP-D mRNA was evaluated by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR).The expression of SP-D protein was shown by ELISA and immunocytochemistry SP methods. The expression of NF-κB and relative downstream cytokines such as TNF-α, IL-1β, IL-8 and IL-10 in supernatant fluid were measured by ELISA.RESULTS: SP-D mRNA and protein were detected in untreated HCE cells. The expression of SP-D and the relative downstream cytokines rose after being stimulated with AF antigens. SP-D mRNA began to rise at 0.5 hour and the most significantly peak was in 2 hours. The protein of SP-D in supernatant fluid had the same trend with mRNA. Immunocytochemistry of SP-D showed positive expression and gradually increased to 6 hours, and then the expression began to decline. NF-κB was activated after treated by AF antigens and the changes had correlation with SP-D. TNF-α, IL-1β, IL-8 and IL-10 began to rise after given AF antigens 1 hour and were 1.82, 1.43, 1.12 and 1.28 times higher than the untreated HCE cells separately. The expression of TNF-α and IL-1β reached the peak at 2 hours, separately 2.80 and 2.86 times than the untreated. The expression of IL-8 and IL-10 gradually increased with a time-dependent manner.CONCLUSION: HCE cells exists SP-D and it may play a significant role in pathogenesis of keratomycosis. AF may induce human corneal epithelial cells to express inflammatory cytokines via SP-D and NF-κB pathway. | 16 | Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers. | 17 | Tillage Pedogenesis of Purple Soils in Southwestern China显示文摘Land cultivation and tillage process, and their consequent impacts on soil erosion, have been criticized as the main cause of degradation of land or soil quality. However, purple soils, classified as Regosols in FAO Taxonomy or Entisols in USDA Taxonomy, are formed from purple rocks of the Trias-Cretaceous system, have been developed or at least accelerated the development due to continual tillage operation, especially digging and ridging. The present study took micromorphological investigation on the sedimentary rocks and the soils under different operations of tillage. Results show that the purple rock of Feixiangguan Formation of the Trias system (T1f) is the easiest to physical weathering and the most fertile soil material enriched in nutrients, and it has been, therefore, mostly cultivated and intensively tilled around the year. It has the fastest soil formation rate. Soil formation rate in the cropland with conventional tillage is higher than that in the forestland and the grassland. | 19 | RhMYB108,an R2R3-MYB transcription factor,is involved in ethylene-and JA-induced petal senescence in rose plants显示文摘Rose(Rosa hybrida)plants are major ornamental species worldwide,and their commercial value greatly depends on their open flowers,as both the quality of fully open petals and long vase life are important.Petal senescence can be started and accelerated by various hormone signals,and ethylene is considered an accelerator of petal senescence in rose.To date,however,the underlying mechanism of signaling crosstalk between ethylene and other hormones such as JA in petal senescence remains largely unknown.Here,we isolated RhMYB108,an R2R3-MYB transcription factor,which is highly expressed in senescing petals as well as in petals treated with exogenous ethylene and JA.Applications of exogenous ethylene and JA markedly accelerated petal senescence,while the process was delayed in response to applications of 1-MCP,an ethylene action inhibitor.In addition,silencing of RhMYB108 alter the expression of SAGs such as RhNAC029,RhNAC053,RhNAC092,RhSAG12,and RhSAG113,and finally block ethylene-and JA-induced petal senescence.Furthermore,RhMYB108 was identified to target the promoters of RhNAC053,RhNAC092,and RhSAG113.Our results reveal a model in which RhMYB108 functions as a receptor of ethylene and JA signals to modulate the onset of petal senescence by targeting and enhancing senescence-associated gene expression. | Shuai Zhang Qingcui Zhao Daxing Zeng Jiehua Xu Hougao Zhou Fenglan Wang Nan Ma Yonghong Li | 2019 | Horticulture Research2019,6,1: | 10 |
