| 1 | Molecular Epidemiological Characteristics of Streptococcus pyogenes Strains Involved in an Outbreak of Scarlet Fever in China,2011显示文摘Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011.Methods Seventy-four Streptococcal pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011.Emm genotyping,pulsed-field gel electrophoresis (PFGE),superantigen (SAg) genes and antimicrobial susceptibility profiling were analyzed for these isolates.Results A total of 4 different emm types were identified.Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles.Emm12 (79.7%) and emml (14.9%) accounted for approximately 94%of all the isolates.The speA gene was all negative in emm12 isolates and positive in emml isolates.All strains were resistant to erythromycin,and 89.4%of them were resistant to erythromycin,tracycline,and clindamycin simultaneously.Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains,though no new emm type was found in this outbreak.The data provide a baseline for further surveillance of scarlet fever,which may contribute to the explanation of the outbreak and development of a GAS vaccine in China. | YOU Yuan Hai SONG Yan Yan YAN Xiao Mei WANG Hai Bin ZHANG Meng Han TAO Xiao Xia Li Lei Lei ZHANG Yu Xin JIANG Xi Hong ZHANG Bing Hua ZHOU Hao XIAO Di JIN Lian Mei FENG Zi Jian LUO Feng Ji ZHANG Jian Zhong | 2013 | Biomedical and Environmental Sciences2013,26,11: | 15 |

| 3 | Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. | 5 | Induction of Endoplasmic Reticulum Stress by Cadmium and Its Regulation on Nrf2 Signaling Pathway in Kidneys of Rats显示文摘Objective This study was conducted to investigate the regulation of endoplasmic reticulum stress on Nrf2 signaling pathway in the kidneys of rats. Methods Rats were divided into twelve groups of six animals each. Some groups were pre-administered with bacitracin or tauroursodeoxycholic acid(TUDCA), and all of them were treated with 5-20 μmol/kg cadmium(Cd) for 48 h. The oxidative stress levels were analyzed using kits. The mRNA and protein expression levels of endoplasmic reticulum stress-related factors and Nrf2 signaling pathway-related factors were determined using RT-PCR and western blot. Results Cd exposure resulted in oxidative stress in the kidneys of rats and upregulated the expression of endoplasmic reticulum stress(ERS)-related factors and Nrf2 signaling pathway-related factors, especially at doses of 10 and 20 μmol/kg Cd, and the expression changes were particularly obvious. Moreover, after pretreatment with bacitracin, Cd upregulated the expression of ERS-related factors to a certain extent and, at higher doses, increased the mRNA expression of Nrf2. After pretreatment with TUDCA, Cd reduced the level of ERS to a certain extent; however, at these doses, there were no significant changes in the expression of Nrf2. Conclusion Cadmium can result in ERS and oxidative stress in the kidneys of rats, activate Nrf2, and upregulate the transcriptional expression of phase II detoxification enzymes under these experimental conditions. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. The inhibition effect increases with the increase of rare earth nitrates concentration. | 15 | A Survey of Urolithiasis in Young Children Fed Infant Formula Contaminated with Melamine in Two Townships of Gansu,China显示文摘Objective To determine the prevalence of urolithiasis in young children fed infant formula(IF) contaminated with melamine,and the association between IF consumption and urolithiasis.Design A total of 2 733 children ≤3 years of age on September 1,2008 in two townships of Gansu Province,China were studied.Cases of urolithiasis were diagnosed by ultrasonography.Milk product consumption was determined by their caregivers.Remaining IF samples were tested for melamine and cyanuric acid.Results Of 2 733 eligible children in the two townships,2 186(80%) were enrolled in our study.Overall,16.6%(362) of 2 186 children had urolithiasis.The prevalence was 24.6% in children exclusively fed Sanlu brand IF,9.7% in those fed other IF,and 8.5% in those fed exclusively on other milk products.For children exclusively breast-fed,no urolithiasis was found(P<0.05).The prevalence of urolithiasis was 11.4% in children fed 400 g of Sanlu IF,rising to 37.5% in children fed over 25 600 g.Of 48 Sanlu IF samples,91.7% contained melamine(median=1 800 ppm;range=45-4 700) and 66.7% contained cyanuric acid(median=1.2 ppm;range=0.4-6.3).Melamine was also detected in 22.2% of 36 other brand IF(median=27.5 ppm,range=4-50).Conclusions Urolithiasis was associated with melamine-contaminated IF.Although one product caused most morbidity,other milk products may have also contributed to the outbreak. | SHI Guo Qing WANG Zi Jun FENG Zi Jian GAO Yong Jun LIU Jian Di SHEN Tao LIMing YANG Jin XU Hai Bing JIANG Xiao Hong WANG Zhao Nan CAI Mei WANG Yu Min ZHU Ye Fan LIU Hui Hui WANG Rui XIONG Wei Yi WANG Zhu Tian YANG Wei Zhong HOU Pei Sen ZENG Guang WANG Yu | 2012 | Biomedical and Environmental Sciences2012,25,2: | 1 |
| 16 | Platinum nanoparticles promote breast cancer cell metastasis by disrupting endothelial barrier and inducing intravasation and extravasation显示文摘Breast cancer is a common malignancy in women with disappointing prognosis especially the triple-negative subtype.Recently,nanomedicine becomes a promising therapeutic strategy for breast cancer,such as platinum nanoparticles(PtNPs).Despite the promising anticancer effects of PtNPs,the safety of PtNPs remains to be fully evaluated.Herein,a series of cell and animal experiments demonstrate that PtNPs facilitate breast cancer metastasis by damaging the vascular endothelial barrier.PtNPs disrupt endothelial cell proliferation,migration and tube-like structure formation,destruct endothelial adhesions junctions and induce endothelial barrier leakiness in vitro most likely by stimulating intracellular reactive oxygen species(ROS)generation and altering the expression and conformation of endothelial junctional proteins,thus promoting intravasation and extravasation of the implanted 4T1 breast cancer cells and leading to cancer metastasis in female BALB/c nude mice in vivo.In addition,smaller PtNPs(5 nm)are more potent than larger PtNPs(70 nm)in exerting the above effects.The study provides the first evidence that PtNPs can promote breast cancer metastasis by damaging endothelial barrier.The unexpected detrimental effects of PtNPs should be considered in future nanomedicine designs for the treatment of breast cancer. | De-Ping Wang Jing Shen Chuan-Yue Qin Yong-Mei Li Li-Juan Gao Jian Zheng Yan-Lin Feng Zi Yan Xin Zhou Ji-Min Cao | 2022 | Nano Research2022,15,8: | 0 |
| 17 | Identification of Dual Receptor-binding Specific Strains of Human H5N1 Viruses in China显示文摘Objective Both the 2,6 linkage and its topology on target cells are critical for the recognition by human influenza virus.The binding preference of avian flu virus H5N1 HA to the 2,3-linked sialylated glycans is considered the major factor limiting its efficient infection and transmission in humans.To monitor potential adaptation of H5N1 virus in human population,the surveillance of receptor-binding specificity was undertaken in China.Methods The binding specificity of 32 human H5N1 virus strains isolated from 2003 to 2009 was tested by 2,3-specific sialidase-treated chicken red blood cell (CRBC) agglutination assay and a solid-phase direct binding assay with synthetic sialylglycopolymers.Results Dual binding preference to 2,3 and 2,6-glycans were found in two strains:A/Guangdong/1/06 (A/GD/1/06) and A/Guangxi/1/08 (A/GX/1/08).Thoughminor effect of short-2,6-binding was detected in A/GX/1/08 at a low virus titer,both showed high affinity to the oligosaccharide at a high load.Notably both are of the long-2,6-recognition,with the same topology as that of human H1N1 and H3N2 viruses.Conclusion The findings suggest that human H5N1 virus in China likely acquired the potential human-adaptation ability.Further research and surveillance on receptor-binding specificity of H5N1 viruses are required. | ZHOU Jian Fang ZOU Shu Mei LI Zi WANG Min DONG Jie GUO Jun Feng WEI He Jiang WEN Le Ying XU Hong SHU Yue Long | 2012 | Biomedical and Environmental Sciences2012,25,1: | 0 |
