1Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment.Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang 2022Cancer Communications2022,42,4:10
2Dissimilar Al/Mg alloys friction stir lap welding with Zn foil assisted by ultrasonic显示文摘The Zn-added ultrasonic assisted friction stir lap welding(UaFSLW) was carried out to improve the quality of dissimilar Al/Mg alloys joint. The effects of ultrasonic power on the joint quality were also investigated.The results indicated that the larger effective lap width and mixing region between Mg and Al(Mg/Al MR) were attained by Zn foil addition and external ultrasonic assistance. Compared with the conventional joint, the finer and better-distributed Mg-Zn IMCs placing the continuous Al-Mg IMCs were formed in the Mg/Al MR of the Zn-added UaFSLW joint. The Zn foil addition and external ultrasonic assistance significantly improved the tensile shear load of the joint, and the load was increased with the increase of the ultrasonic power. 3Plasma spray of biofunctional(Mg, Sr)-substituted hydroxyapatite coatings for titanium alloy implants显示文摘Plasma-sprayed hydroxyapatite (HA) coatings have been widely utilized in load-bearing titanium alloy implants. In this study, Mg, Sr co-substituted HA ((Mg,Sr)-HA) nano-scale powders have been synthesized, which are further used to prepare (Mg,Sr)-HA coatings on Ti-6A1-4V alloys in order to improve the biological functions. The average size of (Mg,Sr)-HA nano particles is ~75nm. The average bonding strength for (Mg,Sr)-HA coating and samples after heat treatment at 500℃ or 600℃ for 3h are 26.17±2.11 MPa, 36.07±4.48 MPa and 37.07 ±2.95 MPa, respectively. There is a significantly increase of bonding strength likely due to low residual stress after heated treatment.MC3T3-E1 cells show a high proliferation rate when cultured with (Mg,Sr)-HA coating extract compared to the normal culture medium, which also exhibit large extension and depositi on of extracellular matrices when adhered on the coating surfaces. 4Improving Joint Morphologies and Tensile Strength of Al/Mg Dissimilar Alloys Friction Stir Lap Welding by Changing Zn Interlayer Thickness显示文摘The pure Zn foils with different thicknesses(0.02, 0.05, 0.1, 0.2 and 0.3 mm) were selected as interlayers to improve the quality of friction stir lap welding joint of 7075-T6 Al and AZ31 B Mg dissimilar alloys. The effects of the interlayer thickness on joint formation, microstructure and tensile strength were analyzed. The results displayed that the maximum length of the boundary between stir zone(SZ) and thermo-mechanically affected zone in lower plate was obtained by the addition of the Zn interlayer with 0.05 mm thickness. The Mg–Zn intermetallic compounds(IMCs) were discontinuously distributed in the SZ, replacing the continuous Al–Mg IMCs. The size of Mg–Zn IMCs increased with the increase in the thickness of the Zn interlayer. 6Ambient air pollution and low temperature associated with case fatality of COVID-19: A nationwide retrospective cohort study in China显示文摘The evidence for the effects of environmental factors on COVID-19 case fatality remains controversial,and it is crucial to understand the role of preventable environmental factors in driving COVID-19 fatality.We thus conducted a nationwide cohort study to estimate the effects of environmental factors(temperature,particulate matter[PM2.5,PM10],sulfur dioxide[SO2],nitrogen dioxide[NO2],and ozone[O3])on COVID-19 case fatality.A total of 71,808 confirmed COVID-19 cases were identified and followed up for their vital status through April 25,2020.Exposures to ambient air pollution and temperature were estimated by linking the city-and county-level monitoring data to the residential community of each participant.For each participant,two windows were defined:the period from symptom onset to diagnosis(exposure window I)and the period from diagnosis date to date of death/recovery or end of the study period(exposure window II).Cox proportional hazards models were used to estimate the associations between these environmental factors and COVID-19 case fatality.COVID-19 case fatality increased in association with environmental factors for the two exposure windows.For example,each 10 mg/m^(3) increase in PM2.5,PM10,O3,and NO2 in window I was associated with a hazard ratio of 1.11(95%CI 1.09,1.13),1.10(95%CI 1.08,1.13),1.09(95 CI 1.03,1.14),and 1.27(95%CI 1.19,1.35)for COVID-19 fatality,respectively.A significant effect was also observed for low temperature,with a hazard ratio of 1.03(95%CI 1.01,1.04)for COVID-19 case fatality per 1C decrease.Subgroup analysis indicated that these effects were stronger in the elderly,as well as in those with mild symptoms and living in Wuhan or Hubei.Overall,the sensitivity analyses also yielded consistent estimates.Short-term exposure to ambient air pollution and low temperature during the illness would play a nonnegligible part in causing case fatality due to COVID-19.Reduced exposures to high concentrations of PM2.5,PM10,O3,SO2,and NO2 and low temperature would help improve the prognosis and reduce public health burden.Fei Tian Xiaobo Liu Qingchen Chao Zhengmin(Min)Qian Siqi Zhang Li Qi Yanlin Niu Lauren DArnold Shiyu Zhang Huan Li Hualiang Lin Qiyong Liu 2021The Innovation2021,2,3:2
7Effects of chronic dexamethasone administration on hyperglycemia and insulin release in goats显示文摘Background: Dexamethasone(Dex), a synthetic glucocorticoid, is among the most commonly used drugs worldwide in animals and humans as an anti-inflammatory and immunosuppressive agent. GC has profound effects on plasma glucose level and other metabolic conditions. However, the effect of prolonged use of Dex on glucose metabolism in ruminants is still unclear.Results: Ten goats were randomly assigned to two groups: the control goats were injected with saline, and the Dex-treated goats were intramuscularly injected daily for 21 d with 0.2 mg/kg Dex. The results showed that plasma glucose and insulin concentrations were significantly increased after Dex administration(P < 0.05). Additionally, the content of hepatic glycogen was also markedly increased in Dex-treated goats(P < 0.01), while the content of glycogen in dorsal longissimus was unchanged by Dex(P > 0.05). The expression of several key genes, involved in blood glucose regulation, was detected by real-time PCR in the small intestine, skeletal muscle and liver. The expression of glucose transporter type 2(GLUT2), sodium-glucose transporter 1(SGLT1) and sodium-potassium ATPase(Na-K/ATPase) in the small intestine were generally increased by Dex, and GLUT2 m RNA expression was significantly up-regulated(P < 0.05). In liver, the expression of genes involved in gluconeogenesis including glucose-6-phosphatase catalytic subunit(G6 PC), cytosolic form of phosphoenolpyruvate carboxykinase(PCK1) and pyruvate carboxylase(PC), were significantly down-regulated by Dex. However, the protein expression levels of PCK1 & PCK2 were significantly increased by Dex, suggesting a post-transcriptional regulation. 9Precise phase adjustment and antibacterial property of copper sulfide particles in confined mesoporous silica nanoreactor显示文摘Bacteria-caused wound infection greatly threatens human health,thus developing an efficient and safe antibacterial agent without drug resistance is still a great challenge.Herein,a confined vulcanization strategy is proposed to construct copper sulfides-loaded dual-mesoporous silica nanospheres(Cux-Sy@DMSNs)with various crystal phases for reactive oxygen species(ROS)-mediated and photothermal antibacterial application.With the pore confinement of DMSNs,the crystal phases of copper sulfides including CuS,Cu_(9)S_(5)and Cui.96S can be easily controlled by changing the vulcanization temperature.The relationships between the crystal phases and photothermal properties as well as peroxidase-like activity of copper sulfides were systematically investigated.Results show that the obtained CuS@DMSNs exhibited higher photothermal ability with remarkable photothermal conversion efficiency of 36.86%in the second near-infrared region(NIR-II)and better peroxidase-like activity,compared to those of Cu_(9)S_(5)@DMSNs and Cu_(1.96)S@DMSNs.As a result,the in vitro experiments showed the good antibacterial effect against both gram-negative E.coli and gram-positive S.aureus under 1064 nm laser irradiation and the presence of H_(2)O_(2).Besides,the CCK-8 assay indicated that CuS@p-DMSNs have minimal cytotoxicity against normal human umbilical vein endothelial cells at the ranged concentrations.Therefore,the resultant CuS@p-DMSNs could act as a promising antibacterial agent for deep wound bacterial infection treatment.Shiyu Xu Jinyong Wu Zhongzhen Ren Dechao Niu 2024Particuology2024,85,2:0
10Coulometric Response of H-Selective Solid-Contact Ion-Selective Electrodes and Its Application in Flexible Sensors+显示文摘The analytical performance of H+-selective solid-contact ion-selective electrodes(SCISEs)based on solid contact polyaniline doped with chloride(PANI(Cl))and poly(3,4-ethylenedioxythiophene)doped with poly(styrene sulfonate)(PEDOT(PSS))was characterized by a developed coulometric signal transduction method.PEDOT(PSS)solid contact is covered by PVC based H+-selective membrane.The obtained coulometric signal demonstrates that the cumulated charge can be amplified by increasing the capacitance of solid contact.SCISEs covered with spin-coated membrane behave faster amperometric response than electrodes with drop-cast mem-brane.In contrast to earlier works,the amperometric response and impedance spectrum demonstrates H+transfer through SCISEs is independent from the thickness of membrane.The exceptional behavior of PANI(Cl)H+-SCISEs shows that the capacitance estimated from impedance spectrum at low frequency 10 mHz and coulometric signal of PANI(Cl)based SCISEs is influenced by the applied po-tentials,whereas PEDOT(PSS)solid contact is independent from the chosen applied potentials.Furthermore,preliminary investiga-tions of coulometric signal transduction on flexible pH sensor implies its potential applications in wearable sensors for sweat ion concentration detection.Tingting Han Tao Song Shiyu Gan Dongxue Han Johan Bobacka Li Niu Ari lvaska 2023Chinese Journal of Chemistry2023,41,2:0
11Phosphorous and selenium tuning Co-based non-precious catalysts for electrosynthesis of H_(2)O_(2)in acidic media显示文摘Electrosynthesis of hydrogen peroxide(H2O2)is an on-site method that enables independent distribution applications in many fields due to its small-scale and sustainable features.The crucial point remains developing highly active,selective and cost-effective electrocatalysts.The electrosynthesis of H2O2 in acidic media is more practical owing to its stability and no need for further purification.We herein report a phosphorus and selenium tuning Co-based non-precious catalyst(CoPSe)toward two-electron oxygen reduction reaction(2e–ORR)to produce H2O2 in acidic media.The starting point of using both P and Se is finding a balance between strong ORR activity of CoSe and weak activity of CoP.The results demonstrated that the CoPSe catalyst exhibited the optimized 2e–ORR activity compared with CoP and CoSe.It disclosed an onset potential of 0.68 V and the H2O2 selectivity 76%-85%in a wide potential range(0–0.5 V).Notably,the CoPSe catalyst overcomes a significant challenge of a narrow-range selectivity for transitionmetal based 2e–ORR catalysts.Finally,combining with electro-Fenton reaction,an on-site system was constructed for efficient degradation of organic pollutants.This work provides a promising non-precious Co-based electrocatalyst for the electrosynthesis of H2O2 in acidic media.Jingxin Xie Lijie Zhong Xin Yang Dequan He Kanglong Lin Xiaoxia Chen Huan Wang Shiyu Gan Li Niu 2024Chinese Chemical Letters2024,35,1:0
12Real-time monitoring system for quality monitoring of jujube slice during drying process显示文摘The real-time monitoring and prediction system for quality attributes of jujube slices during the drying process was designed to solve the problem of destructive and inconvenient of the traditional quality detection method and realize quality online monitoring.Firstly,machine vision and automatic weighing were employed to monitor the color and moisture content changes of jujube slices in real-time.Secondly,correlation models between color parameter(a^(*)value)and nutritional quality attributes(vitamin C,reducing sugar)were established to predict vitamin C and reducing sugar content of jujube slices during the drying process.Finally,the upper computer monitoring software was integrated and designed based on LABVIEW virtual instrument,and the real-time monitoring system was tested and validated.Results showed that:the changing trends of color(L^(*),a^(*),and b^(*)values)monitored by the system were basically the same as the results detected by color difference meter,and the average errors of L^(*),a^(*),and b^(*)values were 0.93,0.52,and 0.73,respectively.The average relative error of moisture content between the system monitoring and manual static detection was 0.18%.The average error of vitamin C and reducing sugar content between the system prediction and manual detection were 50 mg/100 g in dry basis and 0.71g/100 g in dry basis,respectively.The current work can provide a useful reference for real-time monitoring of quality attributes of fruits and vegetables during the drying process.Yuxue Cao Xuedong Yao Yongzhen Zang Yubao Niu Hongwei Xiao Huan Liu Rongguang Zhu Xia Zheng Qiang Wang Xiangnan Zhang Shiyu Wei 2022International Journal of Agricultural and Biological Engineering2022,15,3:0
