| 3 | CD24 aggravates acute liver injury in autoimmune hepatitis by promoting IFN-γ production by CD4^(+) T cells显示文摘The T-cell-mediated immune response is implicated in many clinical hepatic injuries, such as autoimmune hepatitis and acute virus hepatitis. CD24 is widely expressed by different immune cells and plays an important role in the pathogenesis of many autoimmune diseases. However, the role of CD24 in T-cell-mediated liver injury has not been elucidated until now. Here we showed that CD24 deficiency protects mice from concanavalin A (ConA)-induced fulminant liver injury by reducing serum interferon-γ (IFN-γ) levels. CD24 expression by hepatic T cells was markedly increased following ConA challenge. Moreover, decreased IFN-γ production by hepatic CD4^(+) T cells in CD24-deficient mice was detected, which was correlated with downregulated phosphorylation of STAT1 in hepatic tissue. In vitro experiments also supported the conclusion that CD24 deficiency impaired IFN-γ production by CD4^(+) T cells following ConA, CD3/CD28 and phorbol myristate acetate/ionomycin stimulation. | 4 | Effects of sintered metal distributor on fluidization quality of the air dense medium fluidized bed显示文摘Dry coal beneficiation using an air dense medium fluidized bed (ADMFB) requires the formation of a sta- ble and uniform bed from the dense medium. Others have shown that the structure and geometric parameters of the air distributor have a significant influence on the experimentally observed fluidization quality. In this study we used a sintered metal distributor (SMD) in the ADMFB separator and study its effect on the fluidization quality. The results show that for the same open area ratio (OAR), a smaller aper- ture in the SMD will provide improved fluidization quality. If aperture size is held constant bigger open area ratios result in improved fluidization quality. And, the fluidization quality also improves when the pressure drop across the SMD increases. | 5 | Effects of acrous gramineus and its component, alpha-asarone, on apoptosis of hippocampal neurons after seizure in immature rats显示文摘BACKGROUND: α-asarone and acrous gramineus have been shown to play a necessary function in enhancing the reactivity and convulsant threshold to electric stimulation of immature rats. They have also been shown to effectively suppress epileptic seizures induced by pentylenetetrazol in young rats. However, the mechanisms for these roles have been still unclear. OBJECTIVE: To observe the effects in immature rats of acrous gramineus and α-asarone on apoptosis of hippocampal neurons after epileptic seizure at the protein level, and to analyze the mechanism for these effects. DESIGN: A randomized controlled animal experiment. SETTINGS: Department of Pediatrics, First Hospital of Jilin University; Department of Histology and Embryology, Norman Bethune Medical School of Jilin University; Department of Internal Medicine, Children's Hospital of Changchun City; Department of Neurology, First Clinical Hospital affiliated to Harbin Medical University. MATERIALS: Fifty 3-week old Wistar rats, 34-40 g, irrespective of gender, were provided by Gaoxin Research Center of Medical Animal Experiment, Changchun. The animals were treated according to the animal ethical standards. The following chemicals were used for this study: acrous gramineus powders or infusion (Batch No. 0307113, Tianjiang Medicine Company Limited, Jiangyin), α-asarone tablets (Batch No. 030219, Tianwei Pharmaceutical Factory, Shenyang), and phenobarbital sodium tablets (Batch No. 020608, Xinya Medicine Company Limited, Shanghai). The animals were divided into five groups randomly. First, ten rats were chosen as the normal controls. The remaining rats were treated with i.p. injections of pentylenetetrazol to stimulate an epileptic model. METHODS: The experiments were performed at the Neurological Laboratory of the First Hospital of Jilin University between October and December 2004. The rats were treated with i.p. injections of pentylenetetrazol (60 mg/kg) to establish an epileptic model. According to Racine' s standard, animals that reached stage 4 and 5 were chosen and randomly divided into 4 groups: model group, phenobarbital sodium, acrous gramineus, and α-asarone group. The normal control group was treated with an i.p. injection of physiological saline (0.5 mL). After modeling, the model groups were intragastrically administrated 0.5 mL saline. The phenobarbital sodium, acrous gramineus, and α -asarone groups were intragastrically administrated 18 mg/kg/d phenobarbital sodium, 2 350 mg/kg/d acrous gramineus and 29 mg/kg/d α -asarone, respectively. The course of treatment was twice a day for 7 days. The normal group received intragastric administration of 0.5 mL saline at the same time. The rats were sacrificed and brain sections were prepared for light microscopy and electron microscopy. MAIN OUTCOME MEASURES: ① Pathological changes of CA1 and CA3 hippocampal region neurons were observed by light microscopy and electronic microscopy. ② Neuronal apoptosis in the CA1 and CA3 region was measured by TUNEL staining. ③ Bcl-2 and Bax expression in CA1 and CA3 region neurons was detected by immunohistochemistry and a ratio of Bcl-2/Bax was calculated. RESULTS: All 50 immature rats were included in the final analysis. ① Pathological changes of CA1 and CA3 region hippocampal neurons: there were different pathological changes in all groups other than the normal control group. The number of damaged neurons in the model group was highest. The phenobarbital sodium, acrous gramineus, and α-asarone group exhibited different degrees of improvement. ② Neuronal apoptosis in the CA1 and CA3 regions: there were less TUNEL-positive cells in the CA1 and CA3 regions in the normal control group. One week after PTZ-induced seizure, numerous TUNEL-positive cells were detected in the CA1 and CA3 regions in the remaining four groups. There was a significant difference between the normal control group and the remaining four groups (t = 12.089-19.162, P < 0.01). The number of TUNEL-positive cells was less in the phenobarbital sodium, acrous gramineus, and α-asarone groupscompared to the model group (t = 4.707-6.268, P < 0.01). ③Bcl-2 and Bax expression of neurons in the CA1 and CA3 regions: The number of Bcl-2- and Bax-positive cells was less in the normal control group. The Bax-positive cells exhibited a normal shape and had large round nuclei that were predominant. One week after PTZ-induced epilepsy, the number of Bcl-2- and Bax-positive cells in the CA1 and CA2 regions was significantly increased in the remaining four groups compared to the normal control group (t = 11.606-27.042, P < 0.01). The Bax-positive cells exhibited a reduced size and nuclear pyknosis was predominant. However, there was no significant difference among the four groups (P > 0.05). The number of Bcl-2-positive cells in the phenobarbital sodium, acrous gramineus, and α -asarone groups were significantly increased compared to the model group (t = 4.051-6.404, P < 0.01). However, the number of Bax-positive cells was not significantly different among the four groups. The ratio of Bcl-2 to Bax expression was approximately 6.0 in the normal controls, 0.7 in the model group, and 1.0 in the remaining three groups. | 6 | Effects of acrous gramimeus and its main component alpha-asarone on the reactivity and convulsive threshold of immature rats to electric stimulation显示文摘BACKGROUND: The traditional Chinese medicine acrous gramimeus is the dry rhizome of Acrous gramimeus Soland, a kind of Araceae familial perennial herb, which has a sedation action, anticonvulsant and antiepileptic effect. Its effective component has not been known yet, and α-asarone, the major component of the volatile oil extracted from acrous gramineus, has been supposed to play a necessary role in it. OBJECTIVE: To explore the effects of acrous gramimeu and α-asarone on the reactivity and convulsive threshold to electric stimulation in immature rats, furthermore, attempt to definitize the anticonvulsant effect of αasarone. DESIGN: A randomized controlled study. SETTINGS: Department of Pediatrics, First Hospital of Jilin University; Department of Histology and Embryology, School of Basic Medical Sciences of Jilin University; Department of Neurology, First Clinical Hospital affiliated to Harbin Medical University; Department of Internal Medicine, Children’s Hospital of Changchun City. MATERIALS: Seventy 3-week immature Wistar rats (either males or females) of 34-40 g were used. Acrous gramimeu (1 g/bag, the content of α-asarone was 0.046 26%-0.070 16%) with the batch number of 0307113 was provided by Tianjiang Medicine Company Limited, Jiangyin City. α-asarone tablet (60 mg per tablet) with the batch number of 030219 was provided by Tianwei Pharmaceutical Factory, Shenyang City. α-asarone injectable preparation (2 mL per piece) with the batch number of 030105 was provided by Shuanghe Medicine Limited Company, Beijing City. METHODS: The experiments were carried out in the Neurological Laboratory of the First Hospital of Jilin University between August and October in 2004. ① The 70 rats were randomly divided into intragastric subset and intraperitoneal subset. The intragastric subset included four groups of control, phenobarbital sodium, acrous gramimeu and α-asarone; the intraperitoneal subset included three groups of control, phenobarbital sodium and α-asarone. There were 10 rats per group. ② In the intragastric subset, different group was treated with saline (1 mL for each time, phenobarbital sodium (18 mg/kg per day), acrous gramineu (2 350 mg/kg per day) and α-asarone (29 mg/kg per day) respectively twice every day for 5 days. In the intraperitoneal subset, different group was treated with saline (0.5 mL), phenobarbital sodium (29 mg/kg) and α-asarone (2.9 mg/kg) respectively. ③ Before and after administration for 5 days in the intragastric subset as well as before and after administration for about 1 hour in the intraperitoneal subset respectively, the rats were given electric stimulation with the NIHOM KOMDEM multifunctional electrophysiological recorder, and the reactivity and convulsive threshold to electric stimulation of the rats were recorded. MAIN OUTCOME MEASURES: The reactivity and convulsive threshold to electric stimulation in immature rats were compared. RESULTS: All the rats were involved in the analysis of results. ① Results for intragastric administration: Before intragastric administration, there were no obvious differences in the reactivity and convulsive threshold to electric stimulation among the groups (P > 0.05). After intragastric administration for 5 days, the reactivity and convulsive threshold to the electric stimulation had no obvious changes in the control group, but those were significantly higher than before administration in the drug administration groups (t=3.317-7.401, P < 0.01), which were also obviously higher than those in the control group (t=3.027-8.941, P < 0.01), and those in the acrous gramimeu group and α-asarone group were not markedly different from those in the phenobarbital sodium group. ② Results for intraperitoneal injection: Before intraperitoneal injection, the reactivity and convulsive threshold to the electric stimulation had no obvious differences among the groups. After the intraperitoneal injection for 1 hour, the reactivity and convulsive threshold to the electric stimulation had no obvious change in the control group, but those were significantly higher than before administration in the drug administration groups (P < 0.01), which were also obviously higher than those in the control group (t=6.211-7.237, P < 0.01; t=4.085-5.633, P < 0.05), and there was no marked difference between α-asarone group and phenobarbital sodium group (P > 0.05). | 9 | Several miRNAs derived from serum extracellular vesicles are potential biomarkers for early diagnosis and progression of Parkinson’s disease显示文摘Background:Blood-based test for predicting disease progression and early diagnosis of Parkinson’s disease(PD)is an unmet need in the clinic.The profiles of microRNAs(miRNAs)are regarded as potential diagnostic biomarkers for human diseases,whereas miRNAs in the periphery are susceptible to the influence of various components.MiRNAs enriched in serum extracellular vesicles(EVs)have demonstrated disease-specific advantages in diagnosis due to their high abundance,stability and resistance to degradation.This study was aimed to screen differentially expressed EV-derived miRNAs between healthy controls and PD patients to aid in diagnosis of PD.Methods:A total of 31 healthy controls and 72 patients with a diagnosis of PD at different Hoehn and Yahr stages in Tangdu Hospital were included.In total,185 differentially expressed miRNAs were obtained through RNA sequencing of serum EVs as well as edgeR and t-test analyses.Subsequently,the weighted gene co-expression network analysis(WGCNA)was utilized to identify the commonly expressed miRNAs in all stages of PD by constructing connections between modules,and specifically expressed miRNAs in each stage of PD by functional enrichment analysis.After aligning these miRNAs with PD-related miRNAs in Human miRNA Disease Database,the screened miRNAs were further validated by receiver operating characteristic(ROC)curves and quantitative real-time polymerase chain reaction(qRT-PCR)using peripheral blood EVs from 40 more participants.Results:WGCNA showed that 4 miRNAs were commonly associated with all stages of PD and 13 miRNAs were specifically associated with different stages of PD.Of the 17 obtained miRNAs,7 were validated by ROC curve analysis and 7 were verified in 40 more participants by qRT-PCR.Six miRNAs were verified by both methods,which included 2 miRNAs that were commonly expressed in all stages of PD and 4 miRNAs that were specifically expressed in different stages of PD.Conclusions:The 6 serum EV-derived miRNAs,hsa-miR-374a-5p,hsa-miR-374b-5p,hsa-miR-199a-3p,hsa-miR-28-5p,hsa-miR-22-5p and hsa-miR-151a-5p,may potentially be used as biomarkers for PD progression and for early diagnosis of PD in populations. | Shulei He Lu Huang Ci Shao Tiejian Nie Li Xia Bozhou Cui Fangfang Lu Lin Zhu Bolin Chen Qian Yang | 2021 | Translational Neurodegeneration2021,10,3: | 1 |
| 10 | Occurrence characteristics of mercury in high-mercury coal and distribution rule of mercury in products of an air dense medium fluidized bed显示文摘Mercury emitted from direct combustion of high-mercury coal does great harm to the environment. To make good use of high-mercury coal, it is necessary to study the occurrence characteristics of mercury in coal and high-efficient methods of mercury removal. In this paper, high-mercury coal of Guizhou Province of China was taken as an object to study the nature of mercury in coal and the rule of mercury removal by dry preparation method. Mercury mainly distributes in the medium and high density fractions, and has a good affinity with silicon, aluminum, iron, titanium, potassium, calcium, zinc, sodium and magnesium. It exists in minerals formed by these elements and presents significant correlation with ash in coal. After the high-mercury coal is separated by air dense medium fluidized bed, mercury content of clean coal is reduced to 1/10 of raw coal, while mercury content of gangue is increased to 3 times that of raw coal. This indicates that mercury in raw coal is rejected greatly by ADMFB and enriched in highdensity fractions. | 11 | Microstructure,segregation and precipitate evolution in directionally solidified GH4742 superalloy显示文摘The evolution of microstructure,elemental segregation,and precipitation in GH4742 superalloy under a wide range of cooling rates was investigated using zonal melting liquid metal cooling(ZMLMC) experiments.Comparing various nickel-based superalloys,the primary dendrite spacing is significantly linearly correlated with G^(-1/2)V^(-1/4) at high cooling rates,where G and V are temperature gradient and drawing rate,respectively.As the cooling rate decreases,the primary dendrite spacing increases in a dispersive manner.The secondary dendrite arm spacing is significantly correlated with(GV)^(-0.4) for all cooling rate ranges.The degree of elemental segregation increases and then decreases as the cooling rate increases,which is due to the competition between solute counter-diffusion and dendrite tip subcooling.With increasing the solidification rate,the size of γ′,carbides,and non-metallic inclusions gradually decreases.The morphology of the γ′ precipitate changes from plume-like to cubic to spherical.The morphology of carbide changes from block to fine-strip then to Chinese-script.The morphology of carbide is controlled by both dendrite interstitial shape and element diffusion.The inclusions are mainly composite inclusions,which usually show the growth of Ti(C,N) with oxide as the heterogeneous nucleation center and carbide on the outer surface of the carbonitride.As the cooling rate increases,the number density of composite inclusions first increases and then decreases,which is closely related to the elemental segregation behavior. | Shulei Yang Shufeng Yang Wei Liu Jingshe Li Jinguo Gao Yi Wang | 2023 | International Journal of Minerals,Metallurgy and Materials2023,30,5: | 0 |

data at center-of-mass energies ranging from 4.13 to 4.44 GeV.These data samples are primarily used for the study of excited charmonium and charmoniumlike states.By analyzing the di-muon process e^(+)e^(-)→(γISR=FSR)μ^(+)μ^(-),we measure the center-of-mass energies of the data samples with a precision of 0.6 MeV.Through a run-by-run study,we find that the center-of-mass energies were stable | 15 | Effects of increased human tumor necrosis factor-like molecule 1A expression in peripheral blood of children with acute Guillain-Barre syndrome on interferon-gamma secretion显示文摘BACKGROUND:Human tumor necrosis factor-like molecule 1A(hTL1A) is a strong T helper cell type 1(Th1) co-stimulator.Guillain-Barre syndrome(GBS) is an autoimmune disorder of the nervous system,which is mediated by Th1 cells. OBJECTIVE:To determine hTL1A expression in peripheral blood T lymphocytes of acute GBS children and the effects of hTL1 A on secretion of interferon-γ. DESIGN,TIME AND SETTING:A randomized,controlled,neuroimmunological in vitro study was performed at the Central Laboratory of First Hospital of Jilin University,China from November 2005 to November 2007. MATERIALS:Venous blood samples were obtained from 6 healthy donors,aged 6-12 years(all routine blood examination items were normal),and 6 additional children with acute GBS,aged 6-12 years.The GBS children fell ill within 1 week and were not treated with hormones or immunoglobulin. Purified recombinant human soluble tumor necrosis factor-like molecule 1A(rhsTL1A,1 mg/mL, relative molecular mass 22 000,6×His tag,soluble form) was supplied by the Central Laboratory of First Hospital of Jilin University,China. METHODS:Peripheral blood mononuclear cells were isolated from healthy donors using the standard Ficoll gradient centrifugation and were incubated in 96-well culture plates.The cells were assigned to the following groups:control(2μg/mL phytohemagglutinin),2μg/mL phytohemagglutinin + 25,100 and 400 ng/mL rhsTL1A.T cell proliferation was quantified using the tritiated thymidine(~3H-TdR) method.Serum interferon-γlevels in acute GBS children were detected by enzyme-linked immunosorbent assay(ELISA).The ratio of hTL1 A-positive T cells to CD3-positive T cells in peripheral blood of acute GBS children was determined using flow cytometry. Following in vitro pre-activation of peripheral blood mononuclear cells by 2μg/mL phytohemagglutinin,the peripheral blood mononuclear cells were treated with 400 ng/mL exogenous rhsTL1A.Finally,peripheral blood mononuclear cell-secreted interferon-γlevels were measured by ELISA. MAIN OUTCOME MEASURES:The following parameters were measured:rhsTL1A stimulation index to stimulate proliferation of T cells;the serum interferon-γlevels in acute GBS children;the ratio of hTL1A-positive cells to CD3-positive cells;the levels of interferon-γsecreted by peripheral blood mononuclear cells in acute GBS children,as well as rhsTL1A-stimulated interferon-γlevels. RESULTS:T cell proliferation assay revealed that the stimulation index in each rhsTL1A group was greater than the control group.The stimulation index of the 400 ng/mL rhsTL1A group was the greatest.Serum interferon-γlevels in acute GBS children were significantly greater than the control group(P<0.05).The ratio of hTL1 A^+ CD3^+ T cells to CD3^+ T cells in acute GBS children was significantly greater than the control group(P<0.01).Phytohemagglutinin stimulated peripheral blood mononuclear cells to a greater extent than 400 ng/mL rhsTL1A in the acute GBS group,and the secreted interferon-γlevels were significantly increased(P<0.05). | 17 | Seasonal compensation implied no weakening of the land carbon sink in the Northern Hemisphere under the 2015/2016 El Niño显示文摘The recurrent extreme El Niño events are commonly linked to reduced vegetation growth and the land carbon sink over many but discrete regions of the Northern Hemisphere(NH).However,we reported here a pervasive and continuous vegetation greening and no weakened land carbon sink in the maturation phase of the 2015/2016 El Niño event over the NH(mainly in the extra-tropics),based on multiple evidences from remote sensing observations,global ecosystem model simulations and atmospheric CO_(2)inversions.We discovered a significant compensation effect of the enhanced vegetation growth in spring on subsequent summer/autumn vegetation growth that sustained vegetation greening and led to a slight increase in the land carbon sink over the spring and summer of 2015(average increases of 23.34%and 0.63%in net ecosystem exchange from two independent datasets relative to a 5-years average before the El Niño event,respectively)and spring of 2016(6.82%),especially in the extra-tropics of the NH,where the water supply during the pre-growing-season(November of the previous year to March of the current year)had a positive anomaly.This seasonal compensation effect was much stronger than that in 1997 and 1998 and significantly alleviated the adverse impacts of the 2015/2016 El Niño event on vegetation growth during its maturation phase.The legacy effect of water supply during the pre-growing-season on subsequent vegetation growth lasted up to approximately six months.Our findings highlight the role of seasonal compensation effects on mediating the land carbon sink in response to episodic extreme El Niño events. | Fangzhong SHI Xiuchen WU Xiaoyan LI Philippe CIAIS Hongyan LIU Chao YUE Yuting YANG Shulei ZHANG Shushi PENG Yi YIN Benjamin POULTER Deliang CHEN | 2024 | Science China Earth Sciences2024,67,1: | 0 |
| 18 | Biosynthesis and Immunological Evaluation of a Dual-Antigen Nanoconjugate Vaccine Against Brucella melitensis显示文摘Brucellosis,caused by Brucella,is one of the most common zoonosis.However,there is still no vaccine for human use.Although some live attenuated vaccines have been approved for animals,the protection effect is not ideal.In this study,we developed a dual-antigen nanoconjugate vaccine containing both polysaccharide and protein antigens against Brucella.First,the antigenic polysaccharide was covalently coupled to the outer membrane protein Omp19 using protein glycan coupling technology,and then it was successfully loaded on a nano-carrier through the SpyTag/SpyCatcher system.After confirming the efficient immune activation and safety performance of the dual-antigen nanoconjugate vaccine,the potent serum antibody response against the two antigens and remarkable protective effect in non-lethal and lethal Brucella infection models were further demonstrated through different routes of administration.These results indicated that the dual-antigen nanoconjugate vaccine enhanced both T helper 1 cell(Th1)and Th2 immune responses and protected mice from Brucella infection.Furthermore,we found that this protective effect was maintained for at least 18 weeks.To our knowledge,this is the first Brucella vaccine bearing diverse antigens,including a protein and polysaccharide,on a single nanoparticle.Thus,we also present an attractive technology for co-delivery of different types of antigens using a strategy applicable to other vaccines against infectious diseases. | Jing Huang Yufei Wang Kangfeng Wang Shulei Li Peng Sun Yan Guo Jiankai Liu Ruifu Yang Ming Zeng Chao Pan Hengliang Wang Li Zhu | 2023 | Engineering2023,,10: | 0 |
| 19 | Toward high-performance lithium-oxygen batteries with cobalt-based transition metal oxide catalysts:Advanced strategies and mechanical insights显示文摘Aprotic lithium-oxygen(Li-O_(2))batteries represent a promising next-generation energy storage system due to their extremely high theoretical specific capacity compared with all known batteries.Their practical realization is impeded,however,by the sluggish kinetics for the most part,resulting in high overpotential and poor cycling performance.Due to the high catalytic activity and favorable stability of Co-based transition metal oxides,they are regarded as the most likely candidate catalysts,facilitating researchers to solve the sluggish kinetics issue.Herein,this review first presents recent advanced design strategies for Co-based transition metal oxides in Li-O_(2)batteries.Then,the fundamental insights related to the catalytic processes of Co-based transition metal oxides in traditional and novel Li-O_(2)electrochemistry systems are summarized.Finally,we conclude with the current limitations and future development directions of Co-based transition metal oxides,which will contribute to the rational design of catalysts and the practical applications of Li-O_(2)batteries. | Zhenjie Liu Zhiwei Zhao Wang Zhang Yang Huang Ying Liu Dianlun Wu Lei Wang Shulei Chou | 2022 | InfoMat2022,4,4: | 0 |
