1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

3Effects of annealing treatment on the microstructure and electrochemical properties of low-Co hydrogen storage alloys containing Cu and Fe显示文摘The effects of annealing treatment on the microstructure and electrochemical properties of low-Co LaNi 3.55 Mn 0.35 Co 0.20 Al 0.20 Cu 0.75 Fe 0.10 hydrogen storage alloys were investigated. X-ray diffraction (XRD) analysis indicated that annealing treatment remarkably reduced the lattice strain and defects, and increased the unit-cell volume. The optical microscope analysis showed that the as-cast alloy had a crass dendrite microstructure with noticeable composition segregation, which gradually disappeared with increasing annealing temperature, and the micro-structure changed to an equiaxed structure after annealing the alloy at 1233 K. The electrochemical tests indicated that the annealed alloys demonstrated much better cycling stability compared with the as-cast one. The capacity retention at the 100th cycle increased from 90.0% (as-cast) to 94.7% (1273 K). The annealing treatment also improved the discharge capacity. 4Virus-induced Phytoene Desaturase(PDS) Gene Silencing Using Tobacco Rattle Virus in Lilium × formolongi显示文摘Gene function analysis is challenging for Lilium due to the lack of an efficient method for stable genetic transformation. Virus-induced gene silencing(VIGS) is an attractive tool for determining gene function in plants. This study reported that Tobacco rattle virus(TRV)-based VIGS of a PHYTOENE DESATURASE(PDS) ortholog(LhPDS) can be achieved in Lilium × formolongi seedlings, with a survival rate of 92%, using the inoculation method of rubbing plus injection. Compared with untreated and mock-treated seedlings, the photobleached leaf phenotype of silenced plants significantly correlates with down-regulation of endogenous LhPDS(P ≤ 0.05). In addition, the silencing phenomenon can be observed in the growing points of plants, indicating that systemic viral infection was achieved using the protocol. The results indicate that TRV-based VIGS can be used to characterize gene function in Lilium × formolongi. 5An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 6Bone marrow mesenchymal stem cells with Nogo-66 receptor gene silencing for repair of spinal cord injury显示文摘We hypothesized that RNA interference to silence Nogo-66 receptor gene expression in bone marrow mesenchymal stem cells before transplantation might further improve neurological function in rats with spinal cord transection injury. After 2 weeks, the number of neurons and BrdU-positive cells in the Nogo-66 receptor gene silencing group was higher than in the bone marrow mesenchymal stem cell group, and significantly greater compared with the model group. After 4 weeks, behavioral performance was significantly enhanced in the model group. After 8 weeks, the number of horseradish peroxidase-labeled nerve fibers was higher in the Nogo-66 receptor gene silencing group than in the bone marrow mesenchymal stem cell group, and significantly higher than in the model group. 9Induced electric fields and plasmonic interactions between a metallic nanotube and a thin metallic film显示文摘We have numerically simulated the induced electric fields and the plasmonic interactions of a metallic nanotube near a thin metallic film. Our study shows that the energies and intensities of the plasmon resonances depend strongly on the aspect ratio (the ratio of the inner to outer radius) of the nanotube as well as the separation between the center of the nanotube and the upper surface of the metallic film and the thickness of the film. The enhancement of the induced electric field of this system reaches as high as 10 4 orders of magnitude and its field distribution is characterized by waveguide-mode resonance. 10Towards smart optical focusing:deep learningempowered dynamic wavefront shaping through nonstationary scattering media显示文摘Optical focusing through scattering media is of great significance yet challenging in lots of scenarios,including biomedical imaging,optical communication,cybersecurity,three-dimensional displays,etc.Wavefront shaping is a promising approach to solve this problem,but most implementations thus far have only dealt with static media,which,however,deviates from realistic applications.Herein,we put forward a deep learning-empowered adaptive framework,which is specifically implemented by a proposed Timely-Focusing-Optical-Transformation-Net(TFOTNet),and it effectively tackles the grand challenge of real-time light focusing and refocusing through time-variant media without complicated computation.The introduction of recursive fine-tuning allows timely focusing recovery,and the adaptive adjustment of hyperparameters of TFOTNet on the basis of medium changing speed efficiently handles the spatiotemporal non-stationarity of the medium.Simulation and experimental results demonstrate that the adaptive recursive algorithm with the proposed network significantly improves light focusing and tracking performance over traditional methods,permitting rapid recovery of an optical focus from degradation.It is believed that the proposed deep learning-empowered framework delivers a promising platform towards smart optical focusing implementations requiring dynamic wavefront control.YUNQI Luo SUxIA YAN HUANHAO LI PUXIANG LAI YUANJIN ZHENG 2021Photonics Research2021,9,8:3
11Estimating the minimum in-stream flow requirements via wetted perimeter method based on curvature and slope techniques显示文摘Under the assumptions of triangular cross section channel and uniform stable flow, an analytical solution of the minimum ecological in-stream flow requirement (MEIFR) is deduced. Based on the analytical solution, the uncertainty of the wetted perimeter method is analyzed by comparing the two techniques for the determination of the critical point on the relationship curve between wetted perimeter, P and discharge, Q. It is clearly shown that the results of MEIFR based on curvature technique (corresponding to the maximum curvature) and slope technique (slope being 1) are significantly different. On the P-Q curve, the slope of the critical point with the maximum curvature is 0.39 and the MEIFR varied prominently with the change of the slope threshold. This indicates that if a certain value of the slope threshold is not available for slope technique, curvature technique may be a better choice. By applying the analytical solution of MEIFR in the losing rivers of the Western Route South-to-North Water Transfer Project in China, the MEIFR value via curvature technique is 2.5%-23.7% of the multi-year average annual discharge, while that for slope technique is 11%-105.7%. 12Multiple active components synergistically driven heteroatom-doped porous carbon as high-performance counter electrode in dye-sensitized solar cells显示文摘A facile template-free in situ self-activation approach for the multiple active components synergistically driven porous carbon was presented via a feasible annealing process.The biomass-derived carbon without additional activation reagents was fabricated using K-rich pomelo peel(PP)as the carbon source,which possesses a high electric conductivity where abundant functional hetero-metal atoms are doped into the carbon framework that playing the role of catalytic graphitization.The K^+that exists within the biomass can induce self-activation during pyrolysis apart from the activating gases during the pyrolysis process.The resulting electrocatalyst of PP-850(PP was pyrolyzed at 850°C in an N_2atmosphere)with abundant heteroatoms possesses a higher power conversion efficiency(PCE)of 7.81%as the counter electrode(CE)of dye-sensitized solar cells(DSCs)compared with the CEs calcinated at other temperatures and a similar PCE with Pt counterpart(8.24%)based on the liquid I_3^-/I^-electrolyte.The better electrocatalytic performance is attributed to the synergistic effect between self-activation and the co-doping of nitrogen,sulfur and phosphorus all together in a carbon matrix.Due to the feasibility of large-scale production,rich heteroatom doping,the PP-derived carbon,which simplifies the procedure and decreases the cost,has a potential application for an alternative electrocatalyst for high-performance photovoltaic devices.Hongyu Jing Danyang Wu Suxia Liang Xuedan Song Yonglin An Ce Hao Yantao Shi 2019Journal of Energy Chemistry2019,28,4:2
13Simulation Research on Stress Intensity Factors of Different Crack Aspect Ratios on Hollow Axles显示文摘Because of the wicked service environment of the high speed train,it is possible that the hollow axle of the train may encounter the foreign object damage and form a sharp notch.Under the fatigue loading a crack can initiate from the notch and propagate to failure.It is noted that the stress intensity factor is the control parameter of the crack propagating,for the purpose of getting the more exact propagation characteristics,the stress intensity factor is studied mainly.The service loads of hollow axles are defined,and the stress distribution of hollow axles is obtained according to the load spectrum.The semi-ellipse crack configuration is defined with three parameters:the aspect ratio,the relative depth and the relative location along the crack front.Quarter point 20-node isoparametric degenerate singular elements are used for the region near the crack tip.The finite element model of crack extension of hollow axle is created,and the crack front is dispersed which can realize orthogonal extension.Based on this the stress intensity factors of crack front were calculated,and the distribution rules of the stress intensity factors of different initial crack shapes are obtained.The conclusions are compared with that of the analytic method and they agree with each other very well,and the calculating results show that there is a close relationship between the stress intensity factor and the initial crack shape.For a round crack the stress intensity factor at the surface point increases faster than the one at the center point with the crack propagation.However,for a narrow crack,the results are in contrast with that of a round one.So,all the cracks with different shapes propagate toward to a similar shape,and they grow at this shape to end.The study may contribute to the crack propagate characteristics research.ZHOU Suxia XIE Jilong 2009Chinese Journal of Mechanical Engineering2009,22,5:2
14Effects of the local environment on plasmonic coupling of metallic nanotube arrays显示文摘We have theoretically investigated the effect of the dielectric core and the dielectric embedding medium separately on the transmission spectra and plasmonic properties of coupled metallic nanotube arrays.It is found that the plasmonic properties of the nanotube arrays are strongly influenced by the presence of the dielectric which induces additional screening charges.We show that instead of one single photonic bandgap for the hollow nanotube arrays placed in air,an additional photonic bandgap arises from the presence of dielectric media in the transmission spectra.Based on the localized nature of the electric field distributions,we also clearly show the presence of the local plasmonic resonant modes that originate from multipolar plasmon polaritons in the cross section of these nanotube arrays,and that a large amount of opposing surface charges are built up in the gap between adjacent nanotubes.LI HongJian FU ShaoLi XIE SuXia XU HaiQing ZHOU Xin WU JinJun 2011Science China(Physics,Mechanics & Astronomy)2011,54,1:2

16Repurposing econazole as a pharmacological autophagy inhibitor to treat pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma(PDAC)is characterized by the highest mortality among carcinomas.The pathogenesis of PDAC requires elevated autophagy,inhibition of which using hydroxychloroquine has shown promise.However,current realization is impeded by its suboptimal use and unpredictable toxicity.Attempts to identify novel autophagy-modulating agents from already approved drugs offer a rapid and accessible approach.Here,using a patient-derived organoid model,we performed a comparative analysis of therapeutic responses among various antimalarial/fungal/parasitic/viral agents,through which econazole(ECON),an antifungal compound,emerged as the top candidate.Further testing in cell-line and xenograft models of PDAC validated this activity,which occurred as a direct consequence of dysfunctional autophagy.More specifically,ECON boosted autophagy initiation but blocked lysosome biogenesis.RNA sequencing analysis revealed that this autophagic induction was largely attributed to the altered expression of activation transcription factor 3(ATF3).Increased nuclear import of ATF3 and its transcriptional repression of inhibitor of differentiation-1(ID-1)led to inactivation of the AKT/mammalian target of rapamycin(m TOR)pathway,thus giving rise to autophagosome accumulation in PDAC cells.The magnitude of the increase in autophagosomes was sufficient to elicit ER stress-mediated apoptosis.Furthermore,ECON,as an autophagy inhibitor,exhibited synergistic effects with trametinib on PDAC.This study provides direct preclinical and experimental evidence for the therapeutic efficacy of ECON in PDAC treatment and reveals a mechanism whereby ECON inhibits PDAC growth.Ningna Weng Siyuan Qin Jiayang Liu Xing Huang Jingwen Jiang Li Zhou Zhe Zhang Na Xie Kui Wang Ping Jin Maochao Luo Liyuan Peng Edouard C.Nice Ajay Goel Suxia Han Canhua Huang Qing Zhu 2022Acta Pharmaceutica Sinica B2022,12,7:2

19Relationship between Expression of CEA,E-cadherin and Liver Metastasis in Colorectal Cancer显示文摘OBJECTIVE To investigate the expression of E-cadherin andCEA in serum in colorectal carcinoma and their relationship withliver metastasis.METHODS CEA level was measured post-operativelyby radioimmunoassay of 60 patients with colorectal cancer.Immunohistochemical analysis was used to evaluate theexpression of E-cadherin.RESULTS In liver metastasis group,24 patients (24/26,92.3%)were high level of CEA,but only 9 patients in non-liver metastasisgroup.The difference is significant (P=0.004).Expression ofE-cadherin significantly correlated with differentiation,but wasnot associated with Tstage or N stage.Liver metastatic rate innegative expression was higher than that in positive expression.And the survival analysis showed that time of liver metastasis wassignificant different in two groups (P<0.05).CONCLUSION The expression of CEA in serum can be usedto predict liver mestatasis of colorectal cancer after operation.E-cadherin,associated with tumor differentiation,is also a hopefulindicator for the prediction of liver metastasis in patients withcolorectal cancer.Ning Li Xiaobing Chen Xiangbin Wan Suxia Luo Lili Han Mengqiang Zhou Dengwen Yin Li Zhang 2008Chinese Journal of Clinical Oncology2008,5,6:2
20Secondary structural analysis of the mRNA regions encoding the hemagglutinin cleavage site basic amino acids of the avian influenza virus H5N1 subtype samples显示文摘Here we report the codon bias and the mRNA secondary structural features of the hemagglutinin(HA)cleavage site basic amino acid regions of avian influenza virus H5N1 subtypes.We have developed a dynamic extended folding strategy to predict RNA secondary structure with RNAstructure 4.1 program in an iterative extension process.Statistical analysis of the sequences showed that the HA cleavage site basic amino acids favor the adenine-rich codons,and the corresponding mRNA fragments are mainly in the folding states of single-stranded loops.Our sequential and structural analyses showed that to prevent and control these highly pathogenic viruses,that is,to inhibit the gene expression of avian influenza virus H5N1 subtypes,we should consider the single-stranded loop regions of the HA cleavage site-coding sequences as the targets of RNA interference.ZHANG SuXia WANG Xin CHEN XueFeng CAO Huai ZHANG Wen LIU CiQuan 2008Chinese Science Bulletin2008,53,3:2
