| 1 | Clinicopathological and prognostic analysis of 429 patients with intrahepatic cholangiocarcinoma显示文摘AIM:To understand the clinicopathological characteristics and treatment selections and improve survival and provide valuable information for patients with intrahepatic cholangiocarcinoma(ICC). METHODS:We retrospectively evaluated 5311 liver cancer patients who received resection between October 1999 and December 2003.Of these,429(8.1%)patients were diagnosed with ICC,and their clinicopathological, surgical,and survival characteristics were analyzed. RESULTS:Upper abdominal discomfort or pain(65.0%), no symptoms(12.1%),and hypodynamia(8.2%)were the major causes for medical attention.Laboratory tests showed 198(46.4%)patients were HBsAg positive, 90(21.3%)hadα-fetoprotein>20μg/L,50(11.9%) carcinoembryonic antigen>10μg/L,and 242(57.5%) carbohydrate antigen 19-9(CA19-9)>37 U/mL.Survival data was available for 329(76.7%)patients and their mean survival time was 12.4 mo.The overall survival of the patients with R0,R1 resection and punching exploration were 18.3,6.6 and 5.6 mo,respectively. Additionally,CA19-9>37 U/mL was associated with lymph node metastases,but inversely associated withcirrhosis.Multivariate analysis indicated that radical resection,lymph node metastases,macroscopic tumor thrombi and size,and CA19-9 were associated with prognosis. | 2 | Enhancement of acellular cartilage matrix scaffold by Wharton’s jelly mesenchymal stem cell-derived exosomes to promote osteochondral regeneration显示文摘Articular cartilage defect repair is a problem that has long plagued clinicians.Although mesenchymal stem cells(MSCs)have the potential to regenerate articular cartilage,they also have many limitations.Recent studies have found that MSC-derived exosomes(MSC-Exos)play an important role in tissue regeneration.The purpose of this study was to verify whether MSC-Exos can enhance the reparative effect of the acellular cartilage extracellular matrix(ACECM)scaffold and to explore the underlying mechanism.The results of in vitro experiments show that human umbilical cord Wharton’s jelly MSC-Exos(hWJMSC-Exos)can promote the migration and proliferation of bone marrow-derived MSCs(BMSCs)and the proliferation of chondrocytes.We also found that hWJMSC-Exos can promote the polarization of macrophages toward the M2 phenotype.The results of a rabbit knee osteochondral defect repair model confirmed that hWJMSC-Exos can enhance the effect of the ACECM scaffold and promote osteochondral regeneration.We demonstrated that hWJMSC-Exos can regulate the microenvironment of the articular cavity using a rat knee joint osteochondral defect model.This effect was mainly manifested in promoting the polarization of macrophages toward the M2 phenotype and inhibiting the inflammatory response,which may be a promoting factor for osteochondral regeneration.In addition,microRNA(miRNA)sequencing confirmed that hWJMSC-Exos contain many miRNAs that can promote the regeneration of hyaline cartilage.We further clarified the role of hWJMSC-Exos in osteochondral regeneration through target gene prediction and pathway enrichment analysis.In summary,this study confirms that hWJMSC-Exos can enhance the effect of the ACECM scaffold and promote osteochondral regeneration. | Shuangpeng Jiang Guangzhao Tian Zhen Yang Xiang Gao Fuxin Wang Juntan Li Zhuang Tian Bo Huang Fu Wei Xinyu Sang Liuqi Shao Jian Zhou Zhenyong Wang Shuyun Liu Xiang Sui Quanyi Guo Weimin Guo Xu Li | 2021 | Bioactive Materials2021,6,9: | 11 |
| 3 | Molecular Cloning and Expression of Squalene Epoxidase from a Medicinal Plant, Bupleurum chinense显示文摘Objective In plant, squalene epoxidase(SE) catalyzes the first oxygenation step in the biosynthetic pathway of triterpenoid and phytosterol, representing one of the rate-limiting enzymes in this pathway. Bupleurum chinense is an important medicinal herb with its major active constituents such as triterpenoid saponins and saikosaponins. In order to obtain the series of enzymatic genes involved in saikosaponin biosynthesis, a c DNA of SE, designated Bc SE1, was cloned from B. chinense. Methods The Bc SE1 gene was cloned by homology-based PCR and 5'/3' RACE methods from the adventitious roots of B. chinense. The physical and chemical parameters of Bc SE1 protein were predicted by protparam. In order to discover hints in amino acid sequences on the dominant functions in the biosynthesis of saponin or phytosterol, sequences of SE from other plants were downloaded from NCBI for sequences alignment and phylogenetic analysis. Bc SE1 was cloned into a yeast mutant KLN1(MATa, erg1::URA3, leu2, ura3, and trp1) to verify the enzyme activity of Bc SE1. Additionally, the tissue-specific expression and methyl jasmonate(Me JA) inducibility of Bc SE1 were investigated using quantitative real-time PCR. Results The predicted protein of Bc SE1 is highly similar to SEs from other plants sharing amino acid sequence identities of up to 88%. The Bc SE1 can functionally complement with yeast SE gene(ERG1) when expressed in the KLN1 mutant(MATa, erg1::URA3, leu2, ura3, and trp1). Using as controls with β-amyrin synthase(β-AS) which is presumed to catalyze the first committed step in saikosaponin biosynthesis and a cycloartenol synthase(CAS) relating to the phytosterol biosynthesis, the transcript of Bc SE1 was significantly elevated by Me JA in adventitious roots of B. chinense and the transcript of Bc SE1 was most abundant in the fruits and flowers of plants, followed by that in the leaves and roots, and least in stems. | 5 | Si-and alkali-rich melt inclusions in minerals of mantle peridotites from eastern China:Implication for lithospheric evolution显示文摘Minerals of spinel- and garnet-facies mantle xenoliths entrained in Cenozoic basalts from eastern China (North China, Northeastern China and Southeastern China coastal area) contains lots of melt inclusions. Studies on these melt inclusions show that the glass inclusions are rich in SiO2 (60%―68%) and alkalis (K2O+Na2O=5%―11%, especially for K2O) as well as volatiles such as H2O and CO2 (2%―7%), which belong to dacites and andesites of the high-K calcic alkali series rocks with few shoshonites. High Al and Ca diopside in melt inclusion is the product of melt crystallization at high temperature and pressure, rather than the product of devitrification. Results show that these K-rich (in general K2O>3%) intermediate-acidic silicate melt inclusions have characteristics of continent without a genetical link to host basalts and their phenocrystic minerals. Thus, these trapped melt inclusions represent melts of Mesozoic lithospheric mantle-crust interaction and imply that the continental litho-spheric mantle beneath eastern China had undergone fragmentation and recreation processes during the Mesozoic and Cenozoic periods. This result undoubtly provides important implication for the evo-lution of sub-continental lithosphere beneath eastern China. | 6 | Perchlorate removal using granular activated carbon supported iron compounds: Synthesis, characterization and reactivity显示文摘Synthesis and use of the iron compounds supported on granular activated carbon (ICs/GAC) have shown significant environmental implications for perchlorate (ClO 4 ) removal. ICs/GAC was synthesized via hydrolyzing FeSO 4 ·7H 2 O on GAC, reduced by NaBH 4 solution in polyethylene glycol 6000 and ethanol solution, dried in vacuum condition and exposed to air. Synthesized ICs/GAC was characterized using transmission electron micrograph (TEM), Brunauer-Emmett-Teller, X-ray photoelectron spectroscopy (XPS). ICs/GAC was determined to be containing a large amount of FeOHSO 4 , Fe 2 O 3 and a small amount of zero-valent iron (ZVI) nanoparticles according to TEM and XPS measurements. Batch static kinetic tests showed that 97% of ClO 4 was removed within 10 hr at 90°C and 86% of ClO 4 was removed within 12 hr at 25°C, at ICs/GAC dosage of 20 g/L. The experimental results also showed that FeOHSO 4 and Fe 2 O 3 nanoparticles have the function of perchlorate adsorption and play important roles in ClO 4 removal. | 7 | Dietary saturated fatty acid and polyunsaturated fatty acid oppositely affect hepatic NOD-like receptor protein 3 inflammasome through regulating nuclear factor-kappa B activation显示文摘AIM: To investigate the effect of different dietary fatty acids on hepatic inflammasome activation.METHODS: Wild-type C57BL/6 mice were fed either a high-fat diet or polyunsaturated fatty acid(PUFA)-enriched diet. Primary hepatocytes were treated with either saturated fatty acids(SFAs) or PUFAs as well as combined with lipopolysaccharide(LPS). The expression of NOD-like receptor protein 3(NLRP3) inflammasome, peroxisome proliferator-activated receptor-γ and nuclear factor-kappa B(NF-κB) was determined by real-time PCR and Western blot. The activity of Caspase-1 and interleukine-1β production were measured.RESULTS: high-fat diet-induced hepatic steatosis was sufficient to induce and activate hepatic NLRP3 inflammasome. SFA palmitic acid(PA) directly activated NLRP3 inflammasome and increased sensitization to LPS-induced inflammasome activation in hepatocytes. In contrast, PUFA docosahexaenoic acid(Dh A) had thepotential to inhibit NLRP3 inflammasome expression in hepatocytes and partly abolished LPS-induced NLRP3 inflammasome activation. Furthermore, a highfat diet increased but PUFA-enriched diet decreased sensitization to LPS-induced hepatic NLRP3 inflammasome activation in vivo. Moreover, PA increased but Dh A decreased phosphorylated NF-κB p65 protein expression in hepatocytes.CONCLUSION:Hepatic NLRP 3 inflammasome activation played an important role in the development of non-alcoholic fatty liver disease. | 8 | A study on pathogenicity of hepatitis G virus显示文摘AIM To study the pathogenicity of hepatitis G virus (HGV) and observe the genesis and pathological process of hepatitis G.METHODS HGV-RNA in serum was detected by RT-PCR assay. The immunohistochemical assays of liver tissue were performed with HGV monocoloned antibody (McAb)expressed from the region of HGV NS5 nucleic acid sequence. The clinical and pathological data of 52 patients with hepatitis G were discussed. In animal experiment,the Chinese Rhesus monkeys were infected with the serum of a patient with HGV infection. And the dynamic changes in serology and liver histology of animals were observed.RESULTS One hundred and fifty-four patients with HGVRNA positive were selected from 1552 patients with various kinds of hepatitis. Of 154 patients with HGV infection, 52 were infected with HGV only, which accounted for 33.8% (52/154) and 102 with positive HGVRNA were super-infected with other hepatitis viruses,which accounted for 66.2% (102/154). The clinical and pathological observation showed that the acute and chronic hepatitis could be induced by HGV. | 17 | Cloning and Expression Analysis on 3-Hydroxy-3-methylglutaryl-coenzyme A Reductase from Aquilaria sinensis显示文摘Objective To clone the full-length cDNA of 3-hydroxy-3-methylglutaryl-coenzyme A reductase(HMGR)from Aquilaria sinensis(AsHMGR1)and to analyze its expression profile in different tissues and in response to different treatments.HMGR is the first rate-limiting enzyme for sesquiterpene synthesis in the mevalonate pathway.Methods RT-PCR and RACE were used to clone the full-length cDNA of HMGR from A.sinensis based on the conserved HMGR gene fragments.The bioinformatic analysis was performed on its nucleic acid and protein sequence.The expression profile of AsHMGR1 in different tissues and in response to different treatments was analyzed by quantitative RT-PCR.Results The full-length AsHMGR1 cDNA was 2026 bp,containing a 1719 bp open reading frame which encoded a protein of 572 amino acids.Amino acid sequence homology alignment and phylogenetic analysis demonstrated that AsHMGR1 belonged to the HMGR gene family.The detection of tissue expression patterns showed that AsHMGR1 was mainly expressed in the stem,followed by roots and branches.AsHMGR1 could be stimulated by methyl jasmonate and H2O2to varying degrees in a time-dependent manner.Conclusion These data will provide a foundation for further investigation on AsHMGR1 functions and regulatory mechanisms in sesquiterpene synthesis in A.sinensis. | XU Yan-hong YANG Xin WEI Jian-he ZHANG Xing-li ZHANG Zheng GAO Zhi-hui SUI Chun | 2013 | Chinese Herbal Medicines2013,5,3: | 6 |
| 18 | Synthesis of graphene materials by electrochemical exfoliation: Recent progress and future potential显示文摘Synthesis of structurally controlled graphene materials is critical for realizing their practical applications.The electrochemical exfoliation of graphite has emerged as a simple method to produce graphene materials.This review examines research progress in the last 5 years,from 2015 to 2019.Graphene material synthesis methods generally have a trade‐off between increasing production yield and achieving better material property control.The synthesis conditions for synthesizing pristine graphene,graphene oxide(GO),and graphene composites are significantly different.Thus,in this review,we first discuss synthesis methods for graphene materials with high C/O ratios from four aspects:graphite electrodes,equipment engineering,electrolytes,and additional reduction methods.Next,we survey synthesis methods for GO and examine how the pretreatment of the graphite electrodes,electrolytes,and operation parameters,such as applied voltages,electrolyte temperatures,and mechanical forces,affect the quality of GO.Further,we summarize electrochemical exfoliation methods used to dope graphene materials,introduce covalent functional groups,incorporate various nanoparticles,and assembly of graphene architectures.For all synthesis methods,we compare the properties of resulting graphene materials such as C/O ratios,lateral size,layer numbers,and quality characterized by Raman spectroscopy.Lastly,we propose our perspectives on further research.We hope this review stimulates more studies to realize the on‐demand production of graphene materials with desired properties using electrochemical exfoliation methods. | Fei Liu Chaojun Wang Xiao Sui Muhammad Adil Riaz Meiying Xu Li Wei Yuan Chen | 2019 | Carbon Energy2019,1,2: | 6 |
| 19 | Study on the resolution of multi-aircraft flight conflicts based on an IDQN显示文摘With the rapid growth of flight flow,the workload of controllers is increasing daily,and handling flight conflicts is the main workload.Therefore,it is necessary to provide more efficient conflict resolution decision-making support for controllers.Due to the limitations of existing methods,they have not been widely used.In this paper,a Deep Reinforcement Learning(DRL)algorithm is proposed to resolve multi-aircraft flight conflict with high solving efficiency.First,the characteristics of multi-aircraft flight conflict problem are analyzed and the problem is modeled based on Markov decision process.Thus,the Independent Deep Q Network(IDQN)algorithm is used to solve the model.Simultaneously,a’downward-compatible’framework that supports dynamic expansion of the number of conflicting aircraft is designed.The model ultimately shows convergence through adequate training.Finally,the test conflict scenarios and indicators were used to verify the validity.In 700 scenarios,85.71%of conflicts were successfully resolved,and 71.51%of aircraft can reach destinations within 150 s around original arrival times.By contrast,conflict resolution algorithm based on DRL has great advantages in solution speed.The method proposed offers the possibility of decision-making support for controllers and reduce workload of controllers in future high-density airspace environment. | Dong SUI Weiping XU Kai ZHANG | 2022 | Chinese Journal of Aeronautics2022,35,2: | 6 |
