2Risk assessment models of integrative medicine indicators for malignant transformation of chronic atrophic gastritis:A registry study protocol显示文摘Objective:Traditional Chinese medicine(TCM)and modern medicine have both been used in arresting malignant transformation of chronic atrophic gastritis(CAG)in China with good therapeutic effect.However,no studies have been undertaken to assess the risk of malignant transformation in CAG patients using both modern medicine and TCM features.Our study aimed to develop risk assessment models for malignant transformation of CAG combining indicators of both TCM and modern medicine.These models will facilitate early warning and control of malignant transformation of CAG from the perspective of evidence-based integrative medicine.Methods:In the proposed registry study,a total of 1000 eligible CAG patients will be recruited from four hospitals in China.A 10-year follow-up study will be conducted both on-site and off-site to track the events of malignant transformation.Frequency analysis and chi-squared tests will be used to perform the comparative analysis on the prevalence of malignant transformation events and indicators in TCM or modern medicine in different groups.A multivariate Cox proportional hazard model will be used to perform correlation analyses of malignant transformation events and factors of TCM or modern medicine.Conclusion:The proposed study has been designed with a large sample size and long follow-up period,in which wide-ranging modern medicine and TCM indicators can be gathered over the whole process of malignant transformation of CAG.Based on this study,risk assessment models for malignant transformation ofCAGmaybe constructed fromthe perspective of integrative medicine.This may provide clinicians and patients with an optimized early warning system as well as prevention strategies for malignant transformation of CAG.Yue Liu Wanjuan Wang Yuan Li Fusheng Liu Li Zhang Tao Tang Mingxuan Zang Yin Zhang Xia Ding 2019Journal of Traditional Chinese Medical Sciences2019,6,3:3
3Mitotic motor CENP-E cooperates with PRC1 in temporal control of central spindle assembly显示文摘Error-free cell division depends on the accurate assembly of the spindle midzone from dynamic spindle microtubules to ensure chromatid segregation during metaphase-anaphase transition.However,the mechanism underlying the key transition from the mitotic spindle to central spindle before anaphase onset remains elusive.Given the prevalence of chromosome instability phenotype in gastric tumorigenesis,we developed a strategy to model context-dependent cell division using a combination of light sheet microscope and 3D gastric organoids.Light sheet microscopic image analyses of 3D organoids showed that CENP-E inhibited cells undergoing aberrant metaphase-anaphase transition and exhibiting chromosome segregation errors during mitosis.Highresolution real-time imaging analyses of 2D cell culture revealed that CENP-E inhibited cells undergoing central spindle splitting and chromosome instability phenotype.Using biotinylated syntelin as an affinity matrix,we found that CENP-E forms a complex with PRC1 in mitotic cells.Chemical inhibition of CENP-E in metaphase by syntelin prevented accurate central spindle assembly by perturbing temporal assembly of PRC1 to the midzone.Thus,CENP-E-mediated PRC1 assembly to the central spindle constitutes a temporal switch to organize dynamic kinetochore microtubules into stable midzone arrays.These findings reveal a previously uncharacterized role of CENP-E in temporal control of central spindle assembly.Since CENP-E is absent from yeast,we reasoned that metazoans evolved an elaborate central spindle organization machinery to ensure accurate sister chromatid segregation during anaphase and cytokinesis.Xu Liu Leilei Xu Junying Li Phil Y.Yao Wanjuan Wang Hazrat Ismail Haowei Wang Bryce Liao Zhihong Yang Tarsha Ward Ke Ruan Jianchun Zhang Quan Wu Ping He Xia Ding Dongmei Wang Chuanhai Fu Zhen Dou Feng Yan Wenwen Wang Xing Liu Xuebiao Yao 2020Journal of Molecular Cell Biology2020,12,8:3
4Acetytation of ACAP4 regutates CCL18-elicited breast cancer cell migration and invasion显示文摘Tumor metastasis represents the main causes of cancer-related death.Our recent study showed that chemokine CCL18 secreted from tumor-associated macrophages regulates breast tumor metastasis,but the underlying mechanisms remain less clear.Here, we show that ARF6 GTPase-activating protein ACAP4 regulates CCL18-elicited breast cancer cell migration via the acetyltransferase PCAF-mediated acetylation.CCL18 stimulation elicited breast cancer cell migration and invasion via PCAF-dependent acetylation.ACAP4 physically interacts with PCAF and is a cognate substrate of PCAF during CCL18 stimulation.The acetylation site of ACAP4 by PCAF was mapped to Lys311 by mass spectrometric analyses.Importantly,dynamic acetylation of ACAP4 is essential for CCL18-induced breast cancer cell migration and invasion,as overexpression of the persistent acetylation-mimicking or nonacetylatable ACAP4 mutant blocked CCL18-elicited cell migration and invasion.Mechanistically,the acetylation of ACAP4 at Lys311 reduced the lipid-binding activity of ACAP4 to ensure a robust and dynamic cycling of ARF6-ACAP4 complex with plasma membrane in response to CCL18 stimulation.Thus,these results present a previously undefined mechanism by which CCL18-elicited acetylation of the PH domain controls dynamic interaction between ACAP4 and plasma membrane during breast cancer cell migration and invasion.Xiaoyu Song Wei Liu Xiao Yuan Jiying Jiang Wanjuan Wang McKay Mullen Xuannv Zhao Yin Zhang Fusheng Liu Shihao Du Adeel Rehman Ruijun Tian Jian Li Andra Frost Zhenwei Song Hadiyah-Nicole Green Calmour Henry Xing Liu Xia Ding Dongmei Wang Xuebiao Yao 2018Journal of Molecular Cell Biology2018,10,6:3
5A population randomization-based multi-objective genetic algorithm for gesture adaptation in human-robot interaction显示文摘In recent years,vision-based gesture adaptation has attracted great attention from many experts in the field of human-robot interaction,and many methods have been proposed and successfully applied,such as particle swarm optimization and genetic algorithm.However,the reduction of the error and energy consumption of a robot while paying attention to more subtle attitude changes is very important and challenging.In view of these problems,we propose a population randomization-based multi-objective genetic algorithm.The gesture signal is processed with a slight change by imitating the biological evolution mechanisms.In the proposed algorithm,a random out-of-order matrix is added in the process of population evolution synthesis to prevent the premature grouping convergence of the new population.The weights of the objective function and the elite retention strategy are adopted,and the most adaptable individuals in each generation are inherited directly in the next generation without any recombination or mutation.To verify the effectiveness of the algorithm,preliminary application experiments are performed on the gesture adaptation of a robotic arm.The results are compared with the original signal,and the comparison shows that by using the proposed method,the energy consumption is reduced,and the end error is decreased to less than 3 mm while ensuring the tracking effect of the robotic arm.These obtained results meet the communication requirements for human-robot interactions such as handshakes.Moreover,the proposed method has better performance,uses less energy,and has a smaller tracking error than the particle swarm optimization,the single-objective genetic algorithm,and the traditional multi-objective genetic algorithm.A preliminary application experiment indicates that the robotic arm can adapt to human gestures in real time.Luefeng CHEN Wanjuan SU Min LI Min WU Witold PEDRYCZ Kaoru HIROTA 2021Science China(Information Sciences)2021,64,1:2

7Characterization of grass carp FosB, Fosl2, JunD transcription factors in response to GCRV infection显示文摘Activating protein-1(AP-1)composed of Jun and Fos family proteins,which are frequently activated by pathogens to induce cellular inflammation and immune responses.In this study,by using RNA sequencing(RNA-seq)technology,we found that FosB,Fosl2,and JunD expression was co-induced 12 and 20 h in grass carp(Ctenopharyngodon idella)kidney cell line(CIK cells)after grass carp reovirus(GCRV)infection.Full-length cDNA of the three genes were cloned and phylogenetic analysis indicated that the corresponding proteins shared high homology with those in higher vertebrates.The tissue distribution analysis of the three genes showed they were ubiquitously expressed in various tissues of healthy grass carp.In infection assay,FosB,Fosl2,and JunD were up-regulated and co-expressed 8 h after GCRV infection in CIK cells.Protein interaction and co-expression network analysis revealed that FosB,Fosl2,and JunD showed similar functions and were co-expressed in mammals and fish.Taken together,these results indicated that the co-expression of the AP-1 family proteins in grass carp uniformly responds to GCRV infection.These findings provided insights into the further study of AP-1 transcription factors function during grass carp reovirus infection.Fei Yu Wanjuan Li Longlong Wang Shunzheng Que Liqun Lu 2022Aquaculture and Fisheries2022,7,3:0
8mRNA expression of CYP4 in marine polychaete Marphysa sanguinea in response to benzo[a]pyrene显示文摘Rapid amplification of cDNA ends (RACE) and real-time polymerase chain reaction (RT-PCR) were carried out to analyze the CYP4 gene expression in polychaete Marphysa sanguinea exposed to benzo[a]pyrene (BaP) in this study. The full length of MsCYP4 cDNA was 2 470 bp, and it encoded 512 amino acids. The deduced amino acid sequence showed 47% identity with CYP4F from frog Xenopus tropicalis and shared high homology with other known CYP4 sequences. To analyse the role of CYP4 in protecting M. sanguinea from BaP exposure, three BaP groups were established: 0.5, 5 and 50 μg/L. Polychaetes were sampled after 3, 7 and 12 d. At 0.5 μg/L, the effect of BaP on MsCYP4 gene expression increased with time prolonged. MsCYP4 gene expression curve showed Ushaped trend with time in 5 and 50 μg/L BaP groups. 9Travelling-Wave Solutions and Interfaces for Non-Newtonian Diffusion Equations with Strong Absorption显示文摘In this paper, we first find finite travelling-wave solutions, and then investigate the short time development of interfaces for non-Newtonian diffusion equations with strong absorption. We show that the initial behavior of the interface depends on the concentration of the mass of u(x, 0) near x = 0.
