1MiR-30a Positively Regulates the Inflammatory Response of Microglia in Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS) is a classical inflammatory demyelinating disease of the central nervous system(CNS). Microglia are the main resident immune cells in the CNS and are closely associated with the pathogenesis of MS.In the present study, we found that mi R-30 a was highly expressed in jellyfish-like microglia in chronic active lesions of MS patients, as well as in the microglia of mice with experimental autoimmune encephalomyelitis(EAE) at the chronic phase. In vitro, the conditioned supernatant of mouse microglia overexpressing miR-30 a promoted the apoptosis of oligodendrocyte precursor cells(OPCs), and inhibited OPC differentiation. In vivo, overexpressing miR-30 a in transplanted microglia exacerbated the progression of EAE.Overexpression and knock-down experiments in primary cultured mouse microglia showed that mi R-30 a increased the expression of IL-1 b and i NOS, which are pro-inflammatory, while inhibiting the expression of Ym-1 and CD206.Mechanistically, mi R-30 a inhibited the expression of Ppargc1 b, which is the co-activator of peroxisome proliferator-activated receptor gamma, resulting in pro-inflammatory effects. 2Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities.Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao 2021Cancer Communications2021,41,3:10

5Significant downward trend of SO_2 observed from 2005 to 2010 at a background station in the Yangtze Delta region,China显示文摘SO2 is an important gas in atmosphere with great environmental and climate implications.SO2 emission in China has been receiving great attention as the economy grows and the amount of coal consumption has increased in the past few decades.SO2 has been observed from July 2005 to June 2010 at Linan WMO GAW regional station(30.3 °N,119.73 °E,138 m a.s.l.) located in the Yangtze Delta region in eastern China.These observation data are analyzed to understand the trend of regional SO2 background concentration.Strict quality controls are conducted to ensure the temporal comparability of the data.Significant downward trend with ?2.4 ppb/yr(P < 0.0001) of surface SO2 is observed from 2005 to 2010,especially after 2008.The average concentration of SO2 from July 2005 to June 2008 is 14.2±3.1 ppb,which is slightly higher than the mean values of 13.5±5.1 ppb during 1999?2000 and is two folds of the average value(7.1±3.1 ppb) from July 2008 to June 2010.More than 50% of the SO2 has been cut down after 2008 in the Yangtze Delta region due to the implementation of stricter emission control measures.The peak SO2 concentration appears around 10 o'clock in the morning after 2009 while appears at night before 2009.These diurnal variations of SO2 might indicate that after 2009,more SO2 is from the vertical exchange process than from the local accumulation.QI HuangXiong LIN WeiLi XU XiaoBin YU XiangMing MA QianLi 2012Science China Chemistry2012,55,7:7
6Effectiveness of ZiYin Xiehuo granules and Zishen Qinggan granules on partial precocious puberty in girls: a multicenter, randomized, single-blind, controlled trial显示文摘OBJECTIVE: To evaluate the effect of ZiYin Xiehuo granules(ZYXH) and Zishen Qinggan granules(ZSQG) on partial precocious puberty(PPP).METHODS: This was a multicenter, randomized, single-blind, positive-controlled trial. A total of 143 pa-tients were assigned to either the ZYXH group or the ZSQG group using a random number table. The ZYXH group received ZYXH three times daily for 6 months and the ZSQG group received ZSQG three times daily for 6 months. Mammary nucleus diameter; the results of uterus, ovarian, and maximum follicle measures; and Chinese medicine symptom pattern scores were compared at baseline and after 3 months and 6 months of treatment.RESULTS: After 3 months' treatment, there were no significant differences between the two groups in mammary nucleus index changes(left 3.4 ± 3.1 vs3.5 ± 3.1, P = 0.790; right 3.0 ± 2.9 vs 3.6 ± 3.0, P =0.719). The uterine volume in the ZYXH group was smaller than that in the ZSQG group(2.1 ± 1.6 vs2.6 ± 2.2, P = 0.006). There were no significant between-group differences in ovarian volume and maximum follicular diameter on either side(ovarian volume: left 1.2 ± 0.7 vs 1.3 ± 0.6, P = 0.8; right1.2 ± 0.7 vs 1.4 ± 1.1, P = 0.984; maximum follicular diameter: left 3.9 ± 1.7 vs 3.5 ± 2.2, P = 0.158; right3.5 ± 1.7 vs 3.9 ± 2.1, P = 0.314).CONCLUSION: ZYXH granules and ZSQG granules both affected the size of the mammary nucleus in girls with PPP, and improved Chinese medicine symptom patterns. 7Hot deformation behavior and workability characteristic of a fine-grained Mg–8Sn–2Zn–2Al alloy with processing map显示文摘The hot deformation behavior of a fine-grained Mg–8 Sn–2 Zn–2 Al(TZA822, in wt%) alloy was investigated in the temperature range of 150–350°C and the strain rate of 0.01–10 s^-1 employing thermomechanical simulator. In most of the cases, the material showed typical dynamic recrystallization(DRX) features i.e., a signal peak value followed by a gradual decrease or to reach a steady state. The work hardening rate was found to increase with decreasing temperature and increasing strain rate, while strain rates had great effects on work hardening behavior. Meanwhile, the constitutive analysis indicated that cross-slip of dislocations was likely to be the dominant deformation mechanism. In addition, the processing map at the strain of 0.1–0.7 showed two stability domains with high power dissipation efficiencies and the optimum hot working parameters for the studied alloy was determined to be 350°C/0.01 s^-1 and 350°C/10 s^-1, at which continuous DRX(CDRX) and discontinuous DRX(DDRX) as main softening mechanism. 9Cryo-self-assembled silk fibroin sponge as a biodegradable platform for enzyme-responsive delivery of exosomes显示文摘Although advances in protein assembly preparation have provided a new platform for drug delivery during tissue engineering,achieving long-term controlled exosome delivery remains a significant challenge.Diffusion-dominated exosome release using protein hydrogels results in burst release of exosomes.Here,a fibroin-based cryo-sponge was developed to provide controlled exosome release.Fibroin chains can self-assemble into silk I structures under ice-cold conditions when annealed above the glass transition temperature.Exosome release is enzyme-responsive,with rates primarily determined by enzymatic degradation of the scaffolds.In vivo experiments have demonstrated that exosomes remain in undigested sponge material for two months,superior to their retention in fibrin glue,a commonly used biomaterial in clinical practice.Fibroin cryo-sponges were implanted subcutaneously in nude mice.The exosome-containing sponge group exhibited better neovascularization and tissue ingrowth effects,demonstrating the efficacy of this exosome-encapsulating strategy by realizing sustained release and maintaining exosome bioactivity.These silk fibroin cryo-sponges containing exosomes provide a new platform for future studies of exosome therapy.Muyang Sun Qi Li Huilei Yu Jin Cheng Nier Wu Weili Shi Fengyuan Zhao Zhenxing Shao Qingyang Meng Haifeng Chen Xiaoqing Hu Yingfang Ao 2022Bioactive Materials2022,7,2:4

11Solidification structure of eutectic Al-Si alloy under a high magnetic field-aid-electromagnetic vibration显示文摘An electromagnetic vibration was generated by simultaneously imposing a strong static magnetic field (up to 10 T) and an alternative electricity current to the metal. Its effects on the solidification structure of eutectic Al-Si alloy have been investigated experimentally. It is found that the eutectic structure has been refined by solely imposing high magnetic field while it is coarsened under the electromagnetic vibration. Furthermore, polyhedral Si grains and non-dendritic α-Al appeared when the electromagnetic vibration strength was strong enough. The refining of eutectic structure is attributed to the decrease of diffusion coeffcient caused by the strong magnetic field. The coarseness of eutectic structure may be attributed to the convection caused by electromagnetic vibration. 12Methodology for credibility assessment of historical global LUCC datasets显示文摘Land use-induced land cover change(LUCC)is an important anthropogenic driving force of global change that has influenced,and is still influencing,many aspects of regional and global environments.Accurate historical global land use/cover datasets are essential for a better understanding of the impacts of LUCC on global change.However,there are not only evident inconsistencies in current historical global land use/cover datasets,but inaccuracies in the data in these global dataset revealed by historical record-based reconstructed regional data throughout the world.A focus in historical LUCC and global change research relates to how the accuracy of historical global land cover datasets can be improved.A methodology for assessing the credibility of existing historical global land cover datasets that addresses temporal as well as spatial changes in the amount and distribution of land cover is therefore needed.Theoretically,the credibility of a global land cover dataset could be assessed by comparing similarities or differences in the data according to actual land cover data(the'true value').However,it is extremely difficult to obtain historical evidence for assessing the credibility of historical global land cover datasets,which cannot be verified through field sampling like contemporary global land cover datasets.We proposed a methodological framework for assessing the credibility of global land cover datasets.Considering the types and characteristics of the available evidence used for assessments,we outlined four methodological approaches:(1)accuracy assessment based on regional quantitative reconstructed land cover data,(2)rationality assessment based on regional historical facts,(3)rationality assessment based on expertise,and(4)likelihood assessment based on the consistency of multiple datasets.These methods were illustrated through five case studies of credibility assessments of historical cropland cover data.This framework can also be applied in assessments of other land cover types,such as forest and grassland.Xiuqi FANG Wanyi ZHAO Chengpeng ZHANG Diyang ZHANG Xueqiong WEI Weili QIU Yu YE 2020Science China Earth Sciences2020,63,7:4
13Mechanisms of Traditional Chinese Medicine Bushenantai granules(补肾安胎颗粒) in promoting angiogenesis at the maternal-fetal interface of recurrent spontaneous abortion mice显示文摘OBJECTIVE:To assess the effects of Bushenantai(BSAT)granule(补肾安胎颗粒)on angiogenesis-related factors[E2,P,and vascular endothelial growth factor(VEGF)]at the maternal-fetal interface of recurrent spontaneous abortion(RSA)mice,and to evaluate the role of BSAT in promoting angiogenesis at the maternal-fetal interface by influencing the expression of sex hormones,and VEGF.METHODS:A mouse model with normal pregnancy and another with Clark’s classic RSA were established.The RSA mice were randomly assigned to six groups:normal,model,progesterone,high-doseBSAT granule(BSAT-H),medium-dose-BSAT granule(BSAT-M),and low-dose-BSAT granule(BSAT-L)(n=10 for each group).The embryo loss rate and the histopathological changes in the decidual tissues were measured.Serum levels of estrogen(E2),progesterone(P),and VEGF were detected by enzyme-linked immunosorbent assay.The m RNA and protein expressions of estradiol receptor(ER),progesterone receptor(PR),VEGF,and vascular endothelial growth factor receptor 2(VEGFR2)in the decidual tissues were identified by immunohistochemistry,Western blotting,and quantitative reverse transcription polymerase chain reaction.RESULTS:The embryo loss rate in all groups that received BSAT treatment was reduced,while the number of blood vessels at decidual tissues was increased.The serum levels of E2,P and VEGF were elevated,and the m RNA and protein expressions of ER,PR,VEGF,and VEGFR2 in the decidual tissues were enhanced.CONCLUSION:BSAT can improve angiogenesis at the maternal-fetal interface and reduce the embryo loss rate,which may be associated with its ability to increase the serum levels of estrogen,progesterone,and VEGF,in addition to up-regulation of m RNA and protein expression of ER,PR,VEGF,and VEGFR2 in the decidual tissues.LI Yuanqi LI Weili YU Xinhui WU Hua WANG Yuanzhong JIN Ya HAO Lele LIU Mingmin SONG Xiaoge 2021Journal of Traditional Chinese Medicine2021,41,4:3

15Tenofovir vs.entecavir on recurrence of hepatitis B virus-related hepatocellular carcinoma beyond Milan criteria after hepatectomy显示文摘Background:Hepatectomy for hepatocellular carcinoma(HCC)beyond the Milan criteria is shown to be beneficial.However,a high rate of post-operative HCC recurrence hinders the long-term survival of the patients.This study aimed to investigate and compare the impacts of tenofovir(TDF)and entecavir(ETV)on the recurrence of hepatitis B viral(HBV)-related HCC beyond the Milan criteria.Methods:Data pertaining to 1532 patients who underwent hepatectomy and received antiviral therapy between January 2014 and January 2019 were collected from five centers.Recurrence-free survival(RFS)analysis was performed using the Kaplan-Meier method.Cox proportional hazards regression analysis was performed to determine prognostic factors for HCC recurrence.Results:The analysis incorporates 595 HBV-related HCC patients.The overall 5-year RFS was 21.3%.Among them,533 and 62 patients received ETV and TDF treatment,respectively.The 1-,3-,and 5-year RFS rates were 46.3%,27.4%,and 19.6%,respectively,in the ETV group compared with 65.1%,41.8%,and 37.2%,respectively,in the TDF group(P<0.001).Multivariate analysis showed that TDF treatment(hazard ratio[HR]:0.604,P=0.005),cirrhosis(HR:1.557,P=0.004),tumor size(HR:1.037,P=0.008),microvascular invasion(MVI)(HR:1.403,P=0.002),portal vein tumor thrombus(PVTT)(HR:1.358,P=0.012),capsular invasion(HR:1.228,P=0.040),and creatinine levels(CREA)(HR:0.993,P=0.031)were statistically significant prognostic factors associated with RFS.Conclusions:Patients with HCC beyond the Milan criteria exhibited a high rate of HCC recurrence after hepatectomy.Compared to the ETV therapy,TDF administration significantly lowered the risk of HCC recurrence.Junyi Shen Weili Qi Junlong Dai Shusheng Leng Kangyi Jiang Yu Zhang Shun Ran Chuan Li Tianfu Wen 2022Chinese Medical Journal2022,,3:3
16Photothermally Responsive Conjugated Polymeric Singlet Oxygen Carrier for Phase Change-Controlled and Sustainable Phototherapy for Hypoxic Tumor显示文摘Hypoxia significantly compromises the therapeutic performance of photodynamic therapy(PDT)owing to the oxygen level which plays a key role in the production of singlet oxygen(^(1)O_(2)).Herein,the photothermally responsive phase change materials(PCM)are used to encapsulate 1,4-dimethylnaphthalene-functionalized platinum(II)-acetylide conjugated polymer(CP1)with intense nearinfrared(NIR)absorption to prepare new ^(1)O_(2) nanocarriers(CP1-NCs).The 1,4-dimethylnaphthalene moieties in CP1-NCs can trap the ^(1)O_(2) produced from CP1 under irradiation and form a stable endoperoxide.Then,the endoperoxide undergoes cycloreversion to controllably release ^(1)O_(2) via the NIR light-triggered photothermal effect of CP1 and controllable phase change of PCM,which can be used for oxygen-independent PDT for hypoxic tumor.Furthermore,the in vivo luminescence imagingguided synergistic PDT and photothermal therapy showed better efficiency in tumor ablation.The smart design shows the potent promise of CP1-NCs in PCM-controlled and sustainable phototherapy under tumor hypoxic microenvironment,providing new insights for constructing oxygen-independent precise cancer phototherapeutic platform.Guo Li Ruyi Zhou Weili Zhao Bo Yu Jie Zhou Shujuan Liu Wei Huang Qiang Zhao 2020Research2020,,1:3
17Modulating the optical and electrical properties of MAPbBr_(3) single crystals via voltage regulation engineering and application in memristors显示文摘Defect density is one of the most significant characteristics of perovskite single crystals(PSCs)that determines their optical and electrical properties,but few strategies are available to tune this property.Here,we demonstrate that voltage regulation is an efficient method to tune defect density,as well as the optical and electrical properties of PSCs.A three-step carrier transport model of MAPbBr_(3) PSCs is proposed to explore the defect regulation mechanism and carrier transport dynamics via an applied bias.Dynamic and steady-state photoluminescence measurements subsequently show that the surface defect density,average carrier lifetime,and photoluminescence intensity can be efficiently tuned by the applied bias.In particular,when the regulation voltage is 20 V(electrical poling intensity is 0.167 Vμm^(−1)),the surface defect density of MAPbBr_(3) PSCs is reduced by 24.27%,the carrier lifetime is prolonged by 32.04%,and the PL intensity is increased by 112.96%.Furthermore,a voltage-regulated MAPbBr_(3) PSC memristor device shows an adjustable multiresistance,weak ion migration effect and greatly enhanced device stability.Voltage regulation is a promising engineering technique for developing advanced perovskite optoelectronic devices.Jun Xing Chen Zhao Yuting Zou Wenchi Kong Zhi Yu Yuwei Shan Qingfeng Dong Ding Zhou Weili Yu Chunlei Guo 2020Light(Science & Applications)2020,9,1:3
18Nondestructive effect of the cusp magnetic field on the dendritic microstructure during the directional solidification of Nickel-based single crystal superalloy显示文摘The mechanical-property improvement of directionally-solidified Nickel-based single crystal(SC)superalloy with the single-direction magnetic fields is limited by their destructiveness on the dendritic microstructure.Here,the work present breaks through the bottleneck.It shows that the application of the cusp magnetic field(CMF)ensures that the dendrites are not destroyed.This feature embodies that the primary dendrite trunks arrange regularly and orderly,as well the secondary dendrite arms grow symmetrically.By contrast,both the unidirectional transverse and longitudinal magnetic field destroy the dendrite morphology,and there are a number of stray grains near the totally-re melted interface.The nondestructive effect is achieved mainly by the combined action of the thermoelectromagnetic force on the dendrites and thermoelectromagnetic convection in the melt during directional solidification.The investigation should contribute a new route for dramatically and effectively improving the crystal quality and mechanical properties of the directionally-solidified alloys.Xiaotan Yuan Tao Zhou Weili Ren Jianchao Peng Tianxiang Zheng Long Hou Jianbo Yu Zhongming Ren Peter K.Liaw Yunbo Zhong 2021Journal of Materials Science & Technology2021,,3:2
19Genome Assembly and Pathway Analysis of Edible Mushroom Agrocybe cylindracea显示文摘Agrocybe cylindracea,an edible mushroom,is widely cultivated for its abundance of nutrients and flavor,and many of its metabolites are reported to have beneficial roles,such as medicinal effects on tumors and chronical illnesses.However,the lack of genomic information has hindered further molecular studies on this fungus.Here,we present a genome assembly of A.cylindracea together with comparative genomics and pathway analyses of Agaricales species.The draft,generated from both next-generation sequencing(NGS)and single-molecule real-time(SMRT)sequencing platforms to overcome high genetic heterozygosity,is composed of a56.5 Mb sequence and 15,384 predicted genes.This mushroom possesses a complex reproductive system,including tetrapolar heterothallic and secondary homothallic mechanisms,and harbors several hydrolases and peptidases for gradual and effective degradation of various carbon sources.Our pathway analysis reveals complex processes involved in the biosynthesis of polysaccharides and other active substances,including B vitamins,unsaturated fatty acids,and N-acetylglucosamine.RNA-seq data show that A.cylindracea stipes tend to synthesize carbohydrate for carbon sequestration and energy storage,whereas pilei are more active in carbon utilization and unsaturated fatty acid biosynthesis.These results reflect diverse functions of the two anatomical structures of the fruiting body.Our comprehensive genomic and transcriptomic data,as well as preliminary comparative analyses,provide insights into the molecular details of the medicinal effects in terms of active compounds and nutrient components.Yuan Liang Dengxue Lu Sen Wang Yuhui Zhao Shenghan Gao Rongbing Han Jun Yu Weili Zheng Jianing Geng Songnian Hu 2020Genomics, Proteomics & Bioinformatics2020,18,3:2
