| 2 | Diagnostic and prognostic value of minor elevated cardiac troponin levels for percutaneous coronary intervention-related myocardial injury:a prospective,single-center and double-blind study显示文摘Cardiac troponin-I(cTnI) and-T(cTnT) are sensitive and specific markers of myocardial injury.However,the role of increased cTnI and cTnT in percutaneous coronary intervention(PCI)-related myocardial injury remains controversial.In this prospective,single-center and double-blind study,we aimed to determine the diagnostic and prognostic value of cTnI as well as cTnT(cTns) in PCI-related myocardial injury in a Chinese population.A total of 1,008 patients with stable angina pectoris and non-ST-segment elevation acute coronary syndrome were recruited.The levels of cTnI and cTnT were examined before and after PCI.All patients were followed up for26±9 months to observe the incidence of major adverse cardiac events(MACEs).Our results showed that postPCI cTnI and/or cTnT levels were increased to more than the 99^(th) percentile upper reference limit(URL) in 133(13.2%) patients,among which 22(2.2%) were more than 5×99^(th) percentile URL.By univariate analysis,an elevation in cTns after PCI was not an independent predictor of increased MACEs,HR 1.35(P = 0.33,95%CI:0.74-2.46).In conclusion,our data demonstrate that the incidence of PCI-related myocardial injury is not common in a Chinese population and minor elevated cTns levels may not be a sensitive prognostic marker for MACEs. | Min Zhang Huiwei He Ze-Mu Wang Zhihui Xu Ningtian Zhou Zhengxian Tao Bo Chen Chunjian Li Tiebing Zhu Di Yang Liansheng Wang Zhijian Yang | 2014 | The Journal of Biomedical Research2014,28,2: | 12 |

| 8 | Abiogenic hydrocarbons in commercial gases from the Songliao Basin,China显示文摘This paper discusses the kinetic fractionation, composition and distribution characteristics of carbon and hydrogen isotopes for various alkane gases formed in different environments, by different mecha- nisms and from different sources in nature. It is demonstrated that the biodegradation or thermode- gradation of complex high-molecule sedimentary organic material can form microbial gas or thermogenic gas. The δ 13C1 value ranges from -110‰ to -50‰ for microbial gases but from -50‰ to -35‰ (even heavier) for thermogenic gases. Controlled by the kinetic isotope fractionation, both microbial and thermogenic gases have δ 13C and δ D values characterized by normal distribution, i.e. δ 13C1< δ 13C2< δ 13C3< δ 13C4 and δ DCH4< δ DC2H6< δ DC3H8<δ DC4H10, and by a positive correlation between the δ 13C and δ D values. Simple carbonbearing molecules (CH4, CO and CO2) can form abiogenic alkane gases via polymerization in the abiological chemical process in nature, with δ 13C1 heavier than -30‰. Moreover, controlled by the kinetic isotope fractionation, abiogenic alkane gases are characterized by a reverse distribution of δ 13C values and a normal trend of δ D values, namely δ 13C1> δ 13C2> δ 13C3> δ 13C4 and δ DCH4<δ DC2H6< δ DC3H8< δ DC4H10. The δ 13C values and δ D values are negatively correlated. Natural gases from 26 commercial gas wells distributed in the Xujiaweizi and Yingshan-Miaotaizi faulted depressions in the Songliao Basin, China, show δ13C1 values ranging from -30.5‰ to -16.7‰ with a very narrow δ D range between -203‰―-196‰. These gases are characterized by a reverse distribution of δ 13C values but a normal distribution of δ D values, and a negative correlation between their δ 13C and δ D values, indicating an abiological origin. The present study has revealed that abiogenic hydrocarbons not only exist in nature but also can make significant contribution to commercial gas reserviors. It is estimated that the reserve volume of alkane gases with abiogenic characteristics in these 26 gas wells in the Songliao Basin is over 500×108 m3. In the present study, we found that D. latiflorus leaf extract(DLE) reduced fasting blood glucose levels, body weight,and low-density lipoprotein cholesterol with low liver toxicity in db/db mice. In addition, gene expression profiling was performed and pathway enrichment analysis showed that DLE affected metabolic pathways.Importantly, DLE activated the AKT signaling pathway and reduced glucose production by downregulating glucose-6-phosphatase(G6 PC) and phosphoenolpyruvate carboxykinase 1(PCK1) expression. Moreover, network pharmacology analysis identified rutin as an active component in DLE through targeting insulin growth factor 1 receptor(IGF1 R), an upstream signaling transducer of AKT. | 17 | Testing the role of genetic variation of the MC4R gene in Chinese population in antipsychotic-induced metabolic disturbance显示文摘Antipsychotic-induced metabolic disturbance(AIMD) is a common adverse effect of antipsychotics with genetics partly underpinning variation in susceptibility among schizophrenia patients. Melanocortin4 receptor(MC4 R) gene, one of the candidate genes for AIMD, has been under-studied in the Chinese patients. We conducted a pharmacogenetic study in a large cohort of Chinese patients with schizophrenia. In this study, we investigated the genetic variation of MC4 R in Chinese population by genotyping two SNPs(rs489693 and rs17782313) in 1,991 Chinese patients and examined association of these variants with the metabolic effects that were often observed to be related to AIMD. Metabolic measures, including body mass index(BMI), waist circumference(WC), glucose, triglyceride, high-density lipoprotein(HDL), and low-density lipoprotein(LDL) levels were assessed at baseline and after 6-week antipsychotic treatment. We found that interaction of SNP×medication status(drug-na?ve/medicated) was significantly associated with BMI, WC, and HDL change %, respectively. Both SNPs were significantly associated with baseline BMI and WC in the medicated group. Moderate association of rs489693 with WC, Triglyceride, and HDL change % were observed in the whole sample. In the drug-na?ve group, we found recessive effects of rs489693 on BMI gain more than 7%, WC and Triglyceride change %, with AA incurring more metabolic adverse effects. In conclusion, the association between rs489693 and the metabolic measures is ubiquitous but moderate. Rs17782313 is less involved in AIMD. | 19 | Improvement of cardiac function by hepatocyte growth factor via intracoronary transfection in the swine myocardial infarction model显示文摘Objective:To study the amelioration effect of Adenovirus5-mediated human hepatocyte growth factor(Ad5-HGF)on postinfarction heart failure in the swine myocardial infarction model.Methods:Twelve SuZhong young swine were randomly divided into 2 groups with 6 swine in each group:Ad5-HGF-treated group and null-Ad5 group.Four weeks after ligation at left anterior descending coronary artery in swine hearts,Ad5-HGF was transferred to the swine myocardium.Simultaneously,Gated myocardial perfusion imaging was performed to evaluate cardiac perfusion and heart function.After three weeks,Gated myocardial perfusion imaging was performed again,then the hearts were harvested and sectioned to examine the expression of HGF through ELISA.Results:High expression of human HGF was observed in the myocardium of Ad5-HGF-treated group.From 4 weeks to 7 weeks after operation,Left ventricular ejection fraction was increased in Ad5-HGF-treated group.The improvement in LVEF was greater in Ad5-HGF-treated group than that in null-Ad5 group at 7 weeks after operation.Cardiac perfusion was significantly improved in the Ad5-HGF-treated group.Conclusion:High expression of human HGF was observed in the myocardium through intracoronary transfection,which suggests that HGF can ameliorate heart function in swine with postinfarction heart failure. | Wei Wang Zhijian Yang Dongchao Ma Shunlin Xu Yourong Zhang Yuqing Zhang Liansheng Wang Bo Chen Kejiang Cao Wenzhu Ma | 2006 | Journal of Nanjing Medical University2006,20,4: | 2 |
