| 1 | Olfactory ecto-mesenchymal stem cell-derived exosomes ameliorate murine Sjögren’s syndrome by modulating the function of myeloid-derived suppressor cells显示文摘Sjögren’s syndrome(SS)is a systemic autoimmune disease characterized by progressive inflammation and tissue damage in salivary glands and lacrimal glands.Our previous studies showed that myeloid-derived suppressor cells(MDSCs)exhibited impaired immunosuppressive function during disease progression in patients with SS and mice with experimental Sjögren’s syndrome(ESS),but it remains unclear whether restoring the function of MDSCs can effectively ameliorate the development of ESS.In this study,we found that murine olfactory ecto-mesenchymal stem cell-derived exosomes(OE-MSC-Exos)significantly enhanced the suppressive function of MDSCs by upregulating arginase expression and increasing ROS and NO levels.Moreover,treatment with OE-MSC-Exos via intravenous injection markedly attenuated disease progression and restored MDSC function in ESS mice.Mechanistically,OE-MSC-Exo-secreted IL-6 activated the Jak2/Stat3 pathway in MDSCs.In addition,the abundant S100A4 in OE-MSC-Exos acted as a key factor in mediating the endogenous production of IL-6 by MDSCs via TLR4 signaling,indicating an autocrine pathway of MDSC functional modulation by IL-6.Taken together,our results demonstrated that OE-MSC-Exos possess therapeutic potential to attenuate ESS progression by enhancing the immunosuppressive function of MDSCs,possibly constituting a new strategy for the treatment of Sjögren’s syndrome and other autoimmune diseases. | Ke Rui Yue Hong Qiugang Zhu Xiaofei Shi Fan Xiao Hailong Fu Qing Yin Yida Xing Xinfeng Wu Xiaodan Kong Huaxi Xu Jie Tian Shengjun Wang Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,2: | 12 |

| 3 | Bacteriostatic effect of lipopeptides from Bacillus subtilis N-2 on Pseudomonas putida using soybean meal by solid-state fermentation显示文摘Bacillus subtilis N-2 which was isolated from natto,produced lipopeptides using soybean meal as a substrate.This work aimed to purify,identify,and determine the antibacterial mechanism of lipopeptides produced by B.subtilis N-2.The fermented product obtained by solid-state fermentation was subjected to water extraction,acid precipitation,and methanol extraction.Fractions were separated and collected using a two-step ultrafiltration method and then identified by LC-MS/MS.Mass spectrometry characterization revealed the presence of four variants of iturin A that differed according to the p-amino fatty acid chain from C14 to C17 as well as the amino acid positions.A new lipopeptide(m/z 1070.3)was identified and its structure was different from the previously reported lipopeptides.The lipopeptides were shown to inhibit the growth of an isolate of Pseudomonas putida,a common pathogen in decaying fish,by changing membrane permeability.These results suggest that the lipopeptides from B.subtilis N-2 could be used as a biocontrol agent in aquaculture. | Meng Li Haijin Mou Qing Kong Tan Zhang Xiaodan Fu | 2020 | Marine Life Science & Technology2020,2,2: | 1 |
| 4 | Crosstalk between PC 12 cells and endothelial cells in an artificial neurovascular niche constructed by a dual-functionalized selfassembling peptide nanofiber hydrogel显示文摘The coordination between neurogenesis and angiogenesis plays an important role in nerve tissue development and regeneration.Recently,using bioactive materials to drive neurogenic and angiogenic responses has gained increasing attention.Understanding the neurovascular link between regulatory cues offers valuable insight into the mechanisms underlying nerve regeneration and the design of new bioactive materials.In this study,we utilized a dual-functionalized peptide nanofiber hydrogel presenting the brain-derived neurotrophic factor and vascular endothelial growth factor mimetic peptides RGIDKRHWNSQ(RGI)and KLTWQELYQLKYKGI(KLT)to construct an artificial neurovascular microenvironment.The dual-functionalized peptide nanofiber hydrogel enhanced the neurite outgrowth of pheochromocytoma(PC12)cells and tube-like structures formation of human umbilical vein endothelial cells(HUVECs)in vitro,and promoted rapid lesion infiltration of neural and vascular cells in a rat brain injury model.Using indirect co-culture models,we found that the dual-functionalized peptide hydrogel effectively mediated neurovascular crosstalk by regulating secretion of paracrine factors from PC12 cells and HUVECs.When the two cells types were directly co-cultured on the dua卜functionalized peptide hydrogel,the efficiency of cell-cell communication was enhanced,which further accelerated the differentiation and maturation of PC12 cells with an increased number of pseudopodia and spread morphology,and HUVECs tube-like structure formation.In summary,the dual-functionalized peptide nanofiber hydrogel successfully formed an artificial neurovascular niche to directly regulate the behaviors of neural and vascular cells and promote their neurovascular crosstalk through paracrine signaling and direct cell-cell contact. | Zhe Zhang Yi Chai He Zhao Shuhui Yang Wei Liu Zihui Yang Weilong Ye Chenlong Wang Xiaohan Gao Xiangdong Kong Xiaodan Sun Lingyun Zhao Tuoyu Chen Yuqi Zhang Jiaju Lu Xiumei Wang | 2022 | Nano Research2022,15,2: | 0 |

| 6 | Preparation of 3-D porous fibroin scaffolds by freeze drying with treatment of methanol solutions显示文摘In this study, silk scaffolds with appropriate porous structures were prepared by adjusting solution concentrations and providing treatment with methanol solutions in the way of freeze drying. The effects of the preparation conditions on the microstructures and properties of the scaffolds were discussed. Fibroin solutions with different concentrations of 4, 6, 8, 10 wt% were used respectively to prepare the scaffolds. The effects of the addition of 20 vol% methanol before or after freeze drying to the 4 wt% fibroin solution were investigated. As demonstrated by Scanning Electron Microscope (SEM), the fib-roin scaffolds prepared without methanol had porous microstructures composed of thin sheets, and the sizes of the pores decreased with the increase of the fibroin solution concentrations, while the scaffolds prepared in the presence of methanol showed porous microstructures formed by fine-particle aggregates. The porosities and mechanical properties of the prepared fibroin scaffolds under different conditions were tested. | 9 | Response of Salmonella enterica serovar Typhimurium to alginate oligosaccharides fermented with fecal inoculum:integrated transcriptomic and metabolomic analyses显示文摘Alginate oligosaccharides(AOS),extracted from marine brown algae,are a common functional feed additive;however,it remains unclear whether they modulate the gut microbiota and microbial metabolites.The response of Salmonella enterica serovar Typhimurium,a common poultry pathogen,to AOS fermented with chicken fecal inocula was investigated using metabolomic and transcriptomic analyses.Single-strain cultivation tests showed that AOS did not directly inhibit the growth of S.Typhimurium.However,when AOS were fermented by chicken fecal microbiota,the supernatant of fermented AOS(F-AOS)exhibited remarkable antibacterial activity against S.Typhimurium,decreasing the abundance ratio of S.Typhimurium in the fecal microbiota from 18.94 to 2.94%.Transcriptomic analyses showed that the 855 diferentially expressed genes induced by F-AOS were mainly enriched in porphyrin and chlorophyll metabolism,oxidative phosphorylation,and Salmonella infection-related pathways.RT-qPCR confrmed that F-AOS downregulated key genes involved in fagellar assembly and the type III secretory system of S.Typhimurium,indicating metabolites in F-AOS can infuence the growth and metabolism of S.Typhimurium.Metabolomic analyses showed that 205 microbial metabolites were signifcantly altered in F-AOS.Among them,the increase in indolelactic acid and 3-indolepropionic acid levels were further confrmed using HPLC.This study provides a new perspective for the application of AOS as a feed additive against pathogenic intestinal bacteria. | Jiaying Cheng Mengshi Xiao Xinmiao Ren Francesco Secundo Ying Yu Shihao Nan Weimiao Chen Changliang Zhu Qing Kong Youtao Huang Xiaodan Fu Haijin Mou | 2023 | Marine Life Science & Technology2023,5,2: | 0 |
| 10 | B1-cell-produced anti-phosphatidylserine antibodies contribute to lupus nephritis development via TLR-mediated Syk activation显示文摘Autoantibodies produced by B cells play a pivotal role in the pathogenesis of systemic lupus erythematosus (SLE). However, both the cellular source of antiphospholipid antibodies and their contributions to the development of lupus nephritis (LN) remain largely unclear. Here, we report a pathogenic role of anti-phosphatidylserine (PS) autoantibodies in the development of LN. Elevated serum PS-specific IgG levels were measured in model mice and SLE patients, especially in those with LN. PS-specific IgG accumulation was found in the kidney biopsies of LN patients. Both transfer of SLE PS-specific IgG and PS immunization triggered lupus-like glomerular immune complex deposition in recipient mice. ELISPOT analysis identified B1a cells as the main cell type that secretes PS-specific IgG in both lupus model mice and patients. Adoptive transfer of PS-specific B1a cells accelerated the PS-specific autoimmune response and renal damage in recipient lupus model mice, whereas depletion of B1a cells attenuated lupus progression. In culture, PS-specific B1a cells were significantly expanded upon treatment with chromatin components, while blockade of TLR signal cascades by DNase I digestion and inhibitory ODN 2088 or R406 treatment profoundly abrogated chromatin-induced PS-specific IgG secretion by lupus B1a cells. Thus, our study has demonstrated that the anti-PS autoantibodies produced by B1 cells contribute to lupus nephritis development.
