| 3 | QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. | 7 | Validation of WRF model on simulating forcing data for Heihe River Basin显示文摘The research of coupling WRF(Weather Research and Forecasting Model)with a land surface model is enhanced to explore the interaction of the atmosphere and land surface;however,regional applicability of WRF model is questioned.In order to do the validation of WRF model on simulating forcing data for the Heihe River Basin,daily meteorological observation data from 15 sta-tions of CMA(China Meteorological Administration)and hourly meteorological observation data from seven sites of WATER(Watershed Airborne Telemetry Experimental Research)are used to compare with WRF simulations,with a time range of a whole year for 2008.Results show that the average MBE(Mean Bias Error)of daily 2-m surface temperature,surface pressure,2-m rela-tive humidity and 10-m wind speed were-0.19 °C,-4.49 hPa,4.08% and 0.92 m/s,the average RMSE(Root Mean Square Error)of them were 2.11 °C,5.37 hPa,9.55% and 1.73 m/s,and the average R(correlation coefficient)of them were 0.99,0.98,0.80 and 0.55,respectively.The average MBE of hourly 2-m surface temperature,surface pressure,2-m relative humidity,10-m wind speed,downward shortwave radiation and downward longwave were-0.16 °C,-6.62 hPa,-5.14%,0.26 m/s,33.0 W/m2 and-6.44 W/m2,the average RMSE of them were 2.62 °C,17.10 hPa,20.71%,2.46 m/s,152.9 W/m2 and 53.5 W/m2,and the average R of them were 0.96,0.97,0.70,0.26,0.91 and 0.60,respectively.Thus,the following conclusions were obtained:(1)regardless of daily or hourly validation,WRF model simulations of 2-m surface temperature,surface pressure and relative humidity are more reliable,especially for 2-m surface air temperature and surface pressure,the values of MBE were small and R were more than 0.96;(2)the WRF simulating downward shortwave radiation was relatively good,the average R between WRF simulation and hourly observation data was above 0.9,and the average R of downward longwave radiation was 0.6;(3)both wind speed and rainfall simulated from WRF model did not agree well with observation data. | XiaoDuo Pan Xin Li | 2011 | Research in Cold and Arid Regions2011,3,4: | 9 |

| 15 | Constructing a biomimetic robust bi-layered hydrophilic lubrication coating on surface of silicone elastomer显示文摘Silicone elastomers-based materials have been extensively involved in the field of biomedical devices,while their use is extremely restricted due to the poor surface lubricity and inherent hydrophobicity.This paper describes a novel strategy for generating a robust layered soft matter lubrication coating on the surface of the polydimethylsiloxane(PDMS)silicone elastomer,by entangling thick polyzwitterionic polyelectrolyte brush of poly(sulfobetaine methacrylate)(PSBMA)into the sub-surface of the initiator-embedded stiff hydrogel coating layer of P(AAm-co-AA-co-HEMA-Br)/Fe,to achieve a unified low friction and high load-bearing properties.Meanwhile,the stiff hydrogel layer with controllable thickness is covalently anchored on the surface of PDMS by adding iron powder to provide catalytic sites through surface catalytically initiated radical polymerization(SCIRP)method and provides high load-bearing capacity,while the topmost brush/hydrogel composite layer is highly effective for aqueous lubrication.Their synergy effects are capable of attaining low friction coefficient(COFs)under wide range of loaded condition in water environment with steel ball as sliding pair.Furthermore,the influence of mechanical modulus of the stiff hydrogel layer on the lubrication performance of layered coating is investigated,for which the COF is the lowest only when the modulus of the stiff hydrogel layer well matches the PDMS substrate.Surprisingly,the COF of the modified PDMS could remain low friction(COF<0.05)stably after encountering 50,000 sliding cycles under 10 N load.Finally,the surface wear characterizations prove the robustness of the layered lubricating coating.This work provides a new route for engineering lubricious silicon elastomer with low friction,high load-bearing capacity,and considerable durability. | Luyao GAO Xiaoduo ZHAO Shuanhong MA Zhengfeng MA Meirong CAI Yong-Min LIANG Feng ZHOU | 2022 | Friction2022,10,7: | 2 |
| 16 | Integration of high-throughput phenotyping,GWAS,and predictive models reveals the genetic architecture of plant height in maize显示文摘Plant height(PH)is an essential trait in maize(Zea mays)that is tightly associated with planting density,biomass,lodging resistance,and grain yield in the field.Dissecting the dynamics of maize plant architecture will be beneficial for ideotype-based maize breeding and prediction,as the genetic basis controlling PH in maize remains largely unknown.In this study,we developed an automated high-throughput phenotyping platform(HTP)to systematically and noninvasively quantify 77 image-based traits(i-traits)and 20 field traits(f-traits)for 228 maize inbred lines across all developmental stages.Time-resolved i-traits with novel digital phenotypes and complex correlations with agronomic traits were characterized to reveal the dynamics of maize growth.An i-trait-based genome-wide association study identified 4945 traitassociated SNPs,2603 genetic loci,and 1974 corresponding candidate genes.We found that rapid growth of maize plants occurs mainly at two developmental stages,stage 2(S2)to S3 and S5 to S6,accounting for the final PH indicators.By integrating the PH-association network with the transcriptome profiles of specific internodes,we revealed 13 hub genes that may play vital roles during rapid growth.The candidate genes and novel i-traits identified at multiple growth stages may be used as potential indicators for final PH in maize.One candidate gene,ZmVATE,was functionally validated and shown to regulate PH-related traits in maize using genetic mutation.Furthermore,machine learning was used to build predictive models for final PH based on i-traits,and their performancewas assessed across developmental stages.Moderate,strong,and very strong correlations between predictions and experimental datasets were achieved from the early S4(tenth-leaf)stage.Colletively,our study provides a valuable tool for dissecting the spatiotemporal formation of specific internodes and the genetic architecture of PH,as well as resources and predictive models that are useful for molecular design breeding and predicting maize varieties with ideal plant architectures. | Weixuan Wang Weijun Guo Liang Le Jia Yu Yue Wu Dongwei Li Yifan Wang Huan Wang Xiaoduo Lu Hong Qiao Xiaofeng Gu Jian Tian Chunyi Zhang Li Pu | 2023 | Molecular Plant2023,16,2: | 1 |
