1IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development.Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu 2019Cellular & Molecular Immunology2019,16,12:13

3High-quality Arabidopsis thaliana Genome Assembly with Nanopore and HiFi Long Reads显示文摘Arabidopsis thaliana is an important and long-established model species for plant molecular biology,genetics,epigenetics,and genomics.However,the latest version of reference genome still contains a significant number of missing segments.Here,we reported a high-quality and almost complete Col-0 genome assembly with two gaps(named Col-XJTU)by combining the Oxford Nanopore Technologies ultra-long reads,Pacific Biosciences high-fidelity long reads,and Hi-C data.The total genome assembly size is 133,725,193 bp,introducing 14.6 Mb of novel sequences compared to the TAIR10.1 reference genome.All five chromosomes of the Col-XJTU assembly are highly accurate with consensus quality(QV)scores>60(ranging from 62 to 68),which are higher than those of the TAIR10.1 reference(ranging from 45 to 52).We completely resolved chromosome(Chr)3 and Chr5 in a telomere-to-telomere manner.Chr4 was completely resolved except the nucleolar organizing regions,which comprise long repetitive DNA fragments.The Chrl centromere(CEN1),reportedly around 9 Mb in length,is particularly challenging to assemble due to the presence of tens of thousands of CEN180 satellite repeats.Using the cutting-edge sequencing data and novel computational approaches,we assembled a 3.8-Mb-long CEN1 and a 3.5-Mb-long CEN2.We also investigated the structure and epigenetics of centromeres.Four clusters of CEN180 monomers were detected,and the centromere-specific histone H3-like protein(CENH3)exhibited a strong preference for CEN180 Cluster 3.Moreover,we observed hypomethylation patterns in CENH3-enriched regions.We believe that this high-quality genome assembly,Col-XJTU,would serve as a valuable reference to better understand the global pattern of centromeric polymorphisms,as well as the genetic and epigenetic features in plants.Bo Wang Xiaofei Yang Yanyan Jia Yu Xu Peng Jia Ningxin Dang Songbo Wang Tun Xu Xixi Zhao Shenghan Gao Quanbin Dong Kai Ye 2022Genomics, Proteomics & Bioinformatics2022,20,1:6
4Maize orthologs of rice GS5 and their transregulator are associated with kernel development显示文摘Genome information from model species such as rice can assist in the cloning of genes in a complex genome,such as maize.Here,we identified a maize ortholog of rice GS5 that contributes to kernel development in maize.The genomewide association analysis of the expression levels of ZmGS5,and 15 of its 26 paralogs,identified a trans-regulator on chromosome 7,which was a BAKi-like gene.This gene that we named as ZmBAK1-7 could regulate the expression of ZmGS5 and three of the paralogs.Candidate-gene association analyses revealed that these five genes were associated with maize kernel development-related traits.Linkage analyses also detected that ZmGSs and ZmBAK1-7 co-localized with mapped QTLs.A transgenic analysis of ZmGS5 in Arabidopsis thaliana L.showed a significant increase in seed weight and cell number,suggesting that ZmGS5 may have a conserved function among different plant species that affects seed development.Jie Liu Min Deng Huan Guo Sharif Raihan Jingyun Luo Yuancheng Xu Xiaofei Dong Jianbing Yan 2015Journal of Integrative Plant Biology2015,57,11:4

6Architecture of Co-layered double hydroxide nanocages/graphene composite electrode with high electrochemical performance for supercapacitor显示文摘A facile hydrolysis method was applied to fabricate high-performance Co-layered double hydroxide(LDH)nanocages/graphene composites for supercapacitors. The materials exhibit enhanced rate capability than the counterpart electrode free of graphene while maintaining a high specific capacitance. In addition,such Co-LDH nanocages/graphene composites display an excellent cycling stability; the capacitance retention of Co-LDH nanocages/graphene composite electrode remains 90.4% after 10000 cycles at a current density of 2 A g(-1). 7In situ nitrogen removal processes in intertidal wetlands of the Yangtze Estuary显示文摘Estuarine and intertidal wetlands are important sites for nitrogen transformation and elimination.However,the factors controlling nitrogen removal processes remain largely uncertain in the highly dynamic environments.In this study,continuous-flow experiment combined with 15 N isotope pairing technique was used to investigate in situ rates of denitrification and anaerobic ammonium oxidation(anammox)and their coupling with nitrification in intertidal wetlands of the Yangtze Estuary.The measured rates varied from below the detection limit to 152.39μmol N/(m^2·hr)for denitrification and from below the detection limit to 43.06μmol N/(m^2·hr)for anammox.The coupling links of nitrogen removal processes with nitrification were mainly dependent on nitrate,organic carbon,sulfide,dissolved oxygen and ferric iron in the estuarine and intertidal wetlands.Additionally,it was estimated that the actual nitrogen removal processes annually removed approximately 5%of the terrigenous inorganic nitrogen discharged into the Yangtze Estuary.This study gives new insights into nitrogen transformation and fate in the estuarine and intertidal wetlands.Cheng Liu Lijun Hou Min Liu Yanling Zheng Guoyu Yin Hongpo Dong Xia Liang Xiaofei Li Dengzhou Gao Zongxiao Zhang 2020Journal of Environmental Sciences2020,32,7:2
8Calcium ion pinned vanadium oxide cathode for high-capacity and long-life aqueous rechargeable zinc-ion batteries显示文摘Rechargeable aqueous zinc ion batteries (ZIBs),with the easy operation,cost effectiveness,and high safety,are emerging candidates for high-energy wearable/portable energy storage systems.Unfortunately,the unsatisfactory energy density and undesired long-term cycling performance of the cathode hinder the development of ZIBs.Here,we report the chemical preintercalation of a small amount of calcium ions into V2O5 as the cathode material.The cathode of Ca0.04V2O5·1.74H2O (CVO)was demonstrated to have a high specific capacity of 400 mA h g^-1at the current density of 0.05 A g^-1and 187 mA h g^-1at 10 A g^-1,along with impressive capacity retention (100%capacity retention at 10 A g^-1 for 3,000 cycles).Meanwhile,the CVO//Zn battery exhibits a high energy density of 308 Wh kg^-1and a power density of 467 W kg^-1at 0.5 A g^-1.The superior performance originates from the pinning effect of the calcium ions and the lubricating effect of the structural water.The energy storage mechanism of the CVO cathode was also investigated in detail.The new phase (Zn3(OH)2V2O7·2H2O) generated upon cycling participates in the electrochemical reaction and thus contributes to the excellent electrochemical performance.The small amount of Ca^2+ pre-inserted into the interlayer of V2O5 sheds light on constructing cathodes with high energy density for ZIBs.Min Du Feng Zhang Xiaofei Zhang Wentao Dong Yuanhua Sang Jianjun Wang Hong Liu Shuhua Wang 2020Science China Chemistry2020,63,12:2
9Ecological damage of submerged macrophyte Myriophyllum spicatum by cell extracts from microcystin(MC)-and non-MC-producing cyanobacteria,Microcystis显示文摘To explore how decomposed Microcystis-dominant cyanobacterial blooms affect submerged macrophytes,the submerged plant Myriophyllum spicatum was exposed to cell extracts from microcystin(MC)-and non-MC-producing Microcystis strains in a laboratory experiment.Results showed that both Mcracystis cell extracts exerted obvious damages to plant biomass,photosynthesis,primary and secondary metabolism measures,and resistance of plant antioxidant systems,with MC-producing Microcystis having stronger effects due to the presence of MCs.Cyanotoxins other than MCs responsible for the negative effects from both strains needs further identification.The Shannon diversity and Chao1 indices of epiphytic and planktonic bacteria were decreased by the cell extracts from both Microcystis strains.However,epiphytic and planktonic bacterial communities responded differently to cell extracts at the genus level.The dominant genera of planktonic bacteria including Enterobacter,Pseudomonas,and Novosphingobium from phylum Proteobacteria,Chryseobacterium from phylum Bacteroidetes,and Microbacterium from Actinobacteriota in the treatments with cell extracts were previously reported to have strains with algicidal and MC-degrading capabilities.B acterial genes associated with energy production and conversion,amino acid transport and metabolism,and inorganic ion transport and metabolism,were more abundant in both treatments than the control for planktonic bacteria,but less abundant for epiphytic bacteria.We speculate that planktonic bacterial communities have the potential to use and degrade substances derived from Microcystis cell extracts,which may be beneficial for M.spicatum to alleviate damages from Microcystis.Further research is needed to verify the structure and function dynamics of epiphytic and planktonic bacteria in the interaction between cyanobacteria and submerged macrophytes.Yunni GAO Hui YANG Xiaofei GAO Mei LI Man ZHANG Jing DONG Jingxiao ZHANG Longfei LI Xuejun LI Michele A BURFORD 2022Journal of Oceanology and Limnology2022,40,5:2
10CD14 macrophage and IL-10 levels in the peripheral blood of breast cancer patients and their diagnostic value显示文摘Objective To explore the correlation between macrophages and interleukin-10(IL-10 in the peripheral blood of breast cancer(BC)patients and the diagnostic value of joint detection.Methods BC patients(n=50)and healthy controls(n=40)were prospectively recruited.The percentage of circulating cluster of differentiation 14(CD 14)macrophage cells was analyzed by flow cytometry,and an enzyme-linked immunosorbent assay(ELISA)was used to detect IL-10 expression levels.Receiver operating characteristic(ROC)curves were used to verify the diagnostic value of the models based on the expression of CD14 macrophage cell populations and IL-10.In addition,the association between model expression and clinicopathological characteristics was investigated.Another 30 patients with BC and 30 with benign breast disease were selected to validate the IL-10 and CD14 macrophage joint detection model using the same method.Results CD14 macrophage and IL-10 expression levels in BC patients were higher than those in healthy controls(P<0.05).The ROC curve showed that the area under the curve(AUC)of CD14+macrophages combined with IL-10 was 0.830,the sensitivity was 72.0%,and the specificity was 87.5%.Its diagnostic efficiency was better than all other single and joint detections.Correlation analysis of clinicopathological features showed that IL-10 and CD14+macrophage joint detection was significantly correlated with tumor size,tumor-node-metastasis(TNM)stage,and lymph node,estrogen receptor(ER),and Ki-67 expression(P<0.05).The validation analysis results were consistent with the test results.Conclusion Peripheral blood macrophages can be an independent diagnostic marker for BC.Joint detection of CD14-macrophages and IL-10 suggests poor prognosis,which has unlimited potential to guide BC development and provides a new theory for studying tumor-associated macrophages in BC.Mengting Dong Jiayu Sheng Chunyang Li Patiguli·JIAPAER Xiaofei Li Minjia Yuan Xiaohong Xue Ke Jiang 2022Oncology and Translational Medicine2022,8,4:2

12The effect of multilayer structure on magnetic properties of FePt thin films显示文摘The Fe/Pt multilayer films with different structures were deposited by RF magnetron sputtering on glass substrates, and the L10-FePt films were obtained after the as- deposited samples were subjected to vacuum annealing at various temperatures. Re- sults show that the Fe/Pt multilayer structure can effectively reduce the ordering temperature of FePt film, and the in-plane coercivity of [Fe (5.2 nm)/Pt (5.2 nm)]7 multilayers can reach 161.2 kA/m after annealed at 350 ffC for 30 min. When Fe and Pt layer thickness is equal, the coercivity of the film is the largest. On the other hand, the different Fe-Pt crystalline phases such as Fe3Pt and FePt3 phases are formed after annealing when the thickness ratio of Fe/Pt deviates from 1 after annealing. 13UP-TreeRec: Building Dynamic User Profiles Tree for News Recommendation显示文摘Online news recommendation systems aim to address the information explosion of news and make personalized recommendations for users. The key problem of personalized news recommendation is to model users' interests and track their changes. A common way to deal with the user modeling problem is to build user profiles from observed behavior. However, the majority of existing methods make static representations of user profiles and little research has focused on effective user modeling that could dynamically capture user interests in news topics. To address this problem, in this paper, we propose UP-TreeRec, a news recommendation framework based on a user profile tree(UP-Tree), which is a novel framework combining content-based and collaborative filtering techniques. First, by exploiting a novel topic model namely UILDA, we obtain the representation vectors for news content in a topic space as the fundamental bridge to associate user interests with news topics. Next, we design a decision tree with a dynamically changeable structure to construct a user interest profile from the user's feedback. Furthermore, we present a clustering-based multidimensional similarity computation method to select the nearest neighbor of the UP-Tree efficiently. We also provide a Map-Reduce framework-based implemen-tation that enables scaling our solution to real-world news recommendation problems. 14Studies on Mo/HZSM-5 Complex Catalyst for Methane Aromatization显示文摘The influence of adding Fe, Cr, Co, and Ga into 3%Mo/HZSM-5 catalyst on methane aromatization, and the influence of additives ratio on methane conversion, selectivity to hydrocarbons and coke,as well as distribution of aromatics were investigated. The experimental results showed that the addition of Fe, Cr, Co and Ga promoted the dehydrogenation and dissociation of methane. The results of NH3-TPD indicated that the acidity of HZSM-5 was changed by adding Fe and Co components, consequently the catalytic properties of Mo/HZSM-5 were changed. It was also revealed that strong acid sites were the center of methane aromatization. 15Effect of Ni doping on the microstructure and magnetic properties of FePt films显示文摘FePtNi films with different Ni contents were deposited on glass substrates by RF magnetron sputtering, and the L10-FePtNi films were obtained after the asdeposited samples were subjected to vacuum annealing. The magnetic properties, structures, and orientations of the L10-FePt films with Ni doping were studied. The results show that with increasing Ni content, the (001) peak position of the FePtNi films shifts to a higher angle in comparison with FePt, which suggests that there is partial Ni substitution in the L10 lattice. The perpendicular coercivity decreases from 661 to 142 kA/m and magnetization decreases from 512 to 433 kA/m with increasing Ni content. 16High-density/efficient surface active sites on modified separators to boost Li-S batteries via atomic Co^(3+)-Se termination显示文摘Various and critical electrocatalytic processes are involved during the redox reactions in the Li-S batteries,which extremely depend on the surface structure and chemical state.Recently,single-atom concept unlocks a route to maximize the use of surface-active atoms,however,further increasing the density of active site is still strictly limited by the inherent structure that single-atoms are only highly-dispersed on substrate.Herein,we provide a viewpoint that an elaborate facet design with singlecrystalline structure engineering can harvest high-density surface active sites,which can significantly boost the electrocatalyst performance for excellent Li-S batteries.Specifically,the single-crystal CoSe_(2)(scCS)exhibits three-types of terminated(011)facet,efficiently obtaining the surface with a high-rich Co^(3+)–Se bond termination,in contrast with lots of surface grain boundaries and dangling bonds in polycrystalline CoSe_(2).Surprisingly,the surface active sites concentration can reach more than 69%.As anticipated,it can provide high-density and high-efficient active sites,enormously suppressing the shuttle effect and improving the reaction kinetics via accelerating the conversion and deposition of polysulfides and Li2S.This surface lattice strategy with element terminated mode is a promising approach for designing electrocatalyst effect-based energy system,not merely for Li-S batteries.Shujie Liu Xiaofei Liu Manfang Chen Dong Wang Xin Ge Wei Zhang Xiyang Wang Chunhui Wang Tingting Qin Haozhe Qin Liang Qiao Dan Zhang Xing Ou Weitao Zheng 2022Nano Research2022,15,8:1

20KIF15 expression characteristics: Relevance toneo-adjuvant chemotherapy efficacy in breast cancer显示文摘Objective The relationship between the expression of kinesin family member 15 (KIF15) andclinicopathological features in breast cancer (BC) remains controversial. In this study, we aimed to explorethe influence of KIF15 expression on the efficacy of neoadjuvant chemotherapy (NAC) and evaluate itsclinical value in predicting prognosis for BC patients.Methods Immunohistochemistry was used to detect KIF15 expression in 93 BC patients undergoingNAC to analyze the relationship between KIF15 expression and clinical efficacy and analytical parameters.Results Of the 93 BC patients enrolled, 24.73% who underwent NAC had higher KIF15 expression levels,showing positive correlations with ER, HER-2, Ki67, and lymph node metastasis (P < 0.05). The clinicalbenefit of NAC was 70.97%, and the major histological response (MHR) rate was 61.29%. The effectivetherapeutic rate in patients with high KIF15 expression was 95.65%, while the MHR rate was 65.22%.Various molecular BC subtypes with varied clinical and pathological responses exhibited correlation toa large extent. Of all the BC patients studied, 84% of the triple-negative breast cancer (TNBC) patientswere evaluated as clinically effective, and 52% of the TNBC patients were evaluated as pathologicallyeffective, and these values were significantly higher than those of the other molecular types (P < 0.05).The expression of KIF15 in 25 TNBC patients showed positive correlations with lymph node metastasis.Conclusion Overexpression of KIF15 was shown to increase BC sensitivity to chemotherapy anddemonstrated better outcomes.Mengting Dong Chunyang Li Patiguli·Jiapaer Xiaofei Li Xiaohong Xue Ke Jiang Jiayu Sheng 2021Oncology and Translational Medicine2021,7,2:1
