2Multifunctional MgF_2/Polydopamine Coating on Mg Alloy for Vascular Stent Application显示文摘Mg alloy is of great potential in the application of vascular stent due to its degradation in physical environment and proper mechanical property.However its mechanical integrity does not meet the clinical requirement due to relatively fast degradation.Besides,in order to accelerate the reendothelialization of Mg-based stents,it needs surface modification to improve the attachment,growth and adhesion of endothelial cells(ECs).To solve the main obstacles,an anti-corrosion and quick endothelialization coating was prepared on novel Mg-Zn-Y-Nd alloy via a simple two-step immersion method in the present study,first in hydrofluoric acid(HF) then in dopamine tris-Hydrochloric acid(tris-HC1) solution.The coating was uniform and thin,which consisted of two layers—the upper was polydopamine(PDA) layer and the lower was MgF_2 layer.The alloy with the coating demonstrated dramatic corrosion resistance enhancement in vitro by immersion test and electrochemical test.Moreover the HFPDA-treated Mg alloy exhibited great performance of cell adhesion and proliferation.The coating created a favorable environment for ECs to have a competitive advantage over vascular smooth muscle cells(VSMCs),which was preferable for re-endothelialization.The results suggest that HF-PDA-treated Mg-Zn-Y-Nd alloy has great potential in the application of vascular stent and the surface coating method is of great application value in biodegradable Mg alloy stent due to its simplicity and effectiveness.Xiaoli Liu Zhen Zhen Jing Liu Tingfei Xi Yudong Zheng Shaokang Guan Yufeng Zheng Yan Cheng 2015Journal of Materials Science & Technology2015,31,7:8

4A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 5Quantification of microcystin-producing and non-microcystin producing Microcystis populations during the 2009 and 2010 blooms in Lake Taihu using quantitative real-time PCR显示文摘Lake Taihu,a large,shallow hypertrophic freshwater lake in eastern China,has experienced lake-wide toxic cyanobacterial blooms annually during summer season in the past decades.Spatial changes in the abundance of hepatotoxin microcystin-producing and nonmicrocystin producing Microcystis populations were investigated in the lake in August of 2009 and 2010.To monitor the densities of the total Microcystis population and the potential microcystin-producing subpopulation,we used a quantitative real-time PCR assay targeting the phycocyanin intergenic spacer(PC-IGS) and the microcystin synthetase gene(mcyD),respectively.On the basis of quantification by real-time PCR analysis,the abundance of potential toxic Microcystis genotypes and the ratio of the mcyD subpopulation to the total Microcystis varied significantly,from 4.08×104 to 5.22×107 copies/mL,from 5.7% to 65.8%,respectively.Correlation analysis showed a strong positive relationship between chlorophyll-a,toxic Microcystis and total Microcystis;the abundance of toxic Microcystis correlated positively with total phosphorus and ortho-phosphate concentrations,but negatively with TN:TP ratio and nitrate concentrations.Meanwhile the proportion of potential toxic genotypes within Microcystis population showed positive correlation with total phosphorus and ortho-phosphate concentrations.Our data suggest that increased phosphorus loading may be a significant factor promoting the occurrence of toxic Microcystis bloom in Lake Taihu.Daming Li Fanxiang Kong Xiaoli Shi Linlin Ye Yang Yu Zhen Yang 2012Journal of Environmental Sciences2012,24,2:5
6Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 7Effects of elevated CO_2 on dynamics of microcystin-producing and non-microcystin-producing strains during Microcystis blooms显示文摘In an attempt to elucidate the effects of different CO2concentrations(270, 380, and 750 μL/L) on the competition of microcystin-producing(MC-producing) and non-MC-producing Microcystis strains during dense cyanobacteria blooms, an in situ simulation experiment was conducted in the Meiliang Bay of Lake Taihu in the summer of 2012. The abundance of total Microcystis and MC-producing Microcystis genotypes was quantified based on the 16 S r DNA and mcy D gene using real-time PCR. The results showed that atmospheric CO2 elevation would significantly decrease the p H value and increase the dissolved inorganic carbon(DIC) concentration.Changes in CO2 concentration did not show significant influence on the abundance of total Microcystis population. However, CO2 concentrations may be an important factor in determining the subpopulation structure of Microcystis. The enhancement of CO2 concentrations could largely increase the competitive ability of non-MC-producing over MC-producing Microcystis, resulting in a higher proportion of non-MC-producing subpopulation in treatments using high CO2 concentrations. Concurrently, MC concentration in water declined when CO2 concentrations were elevated. 8A model to assess fundamental and realized carrying capacities of island ecosystem:A case study in the southern Miaodao Archipelago of China显示文摘Research on carrying capacity, aiming at maintaining the limited ability of the Earth's life supporting system to sustain human development, requires a comprehensive and ecosystem-based approach to monitor and assess the localized sustainability of coupled social and ecological systems. A definition termed the ecosystem-based carrying capacity of island(EBCCI) was developed in this paper of which the indices of fundamental and realized carrying capacities of island(FCCI and RCCI) were highlighted to emphasize the inter-dependencies between social systems and ecological systems. In order to avoid the difficulties and uncertainties of direct assessment, the related assessment model was established on the basis of synthetic evaluation of inherent and external factors affecting the EBCCI. The southern Miaodao Archipelago(SMA) located in the intersection of the Bohai Sea and the Yellow Sea, China, was taken as a typical example to acquire integrated recognition of the island ecosystem and its carrying capacity so as to discuss its sustainable development. The index scores of FCCI and RCCI in the SMA were 0.818 5 and 0.712 9 out of 1.0, respectively, with acceptable uncertainties. The results showed a relatively well capacity to sustain progress and relatively well realization of the carrying capacity of island ecosystem, owing to a well capacity of ecologically regulating, general performance of both ecologically supporting and resource provisioning, and a relatively high level of social supporting system. The study implied that it was critical to optimize the inter-dependencies and to sustain the relative balance between social systems and ecological systems so as to improve the RCCI and further facilitate the sustainability of SMA. 9Foam Forming: An Effective Method to Prepare Polyimide Fiber-based Paper显示文摘Well-dispersed fiber suspension is the precondition of good paper formation. Compared with cellulosic fibers, synthetic fibers are prone to flocculate because of their long length and hydrophobic nature, resulting in poor paper formation. To solve this problem, dispersants and extremely low forming consistency are typically adopted during the traditional wet-forming process, which cause a large amount of water consumption and treatment cost. Therefore, increasing forming consistency without compromising paper formation remains a challenge for papermakers. In this work, foam forming was adopted to disperse polyimide fibers (PI) with high forming consistency. The results showed that the formation index of handsheets increased when the bubble size and distribution became small and narrow. Compared with traditional wet-forming process with the same consistency (0.4%), the formation index of handsheets by foam forming increased by approximately 100% when C8 alkyl glucoside (APG08) concentration reached 16 g/L. Notably, forming consistency could increase by eight times while keeping the same level of paper formation. 10p190A inactivating mutations cause aberrant RhoA activation and promote malignant transformation via the Hippo-YAP pathway in endometrial cancer显示文摘The Rho family of GTPases is strictly regulated by a large family of GTPase-activating proteins(GAPs)that stimulate the relatively weak intrinsic GTP-hydrolyzing activity of Rho GTPases.p190A is a potent and widely expressed GAP that acts on RhoA GTPases.p190A is frequently mutated in endometrial cancer,but the contribution of p190A mutations to endometrial tumorigenesis remains unclear.Here we identified that p190A is an upstream regulator of the Hippo-YAP signaling pathway,which is a critical regulator of cell proliferation,apoptosis,and cell fate.p190A knockout in endometrial cancer cells promoted cell proliferation,migration,and epithelial–mesenchymal transition(EMT),which were partially dependent on YAP activation.Wild-type p190A,but not endometrial cancer-associated mutants,suppressed the nuclear localization,transcriptional activity,and malignant transformation function of YAP.Moreover,the nuclear localization of YAP was enhanced in p190A-mutated endometrial cancer.These findings reveal novel molecular mechanisms underlying Hippo-YAP pathway-driven endometrial tumorigenesis and elucidate the potential for therapy targeting the Hippo-YAP pathway in p190A-mutated endometrial cancer.Xiaoli Wen Jing Wan Qizhi He Mengfei Wang Shuangdi Li Mei Jiang Zhen Qian Binya Liu Wen Lu Kai Wang Kun Gao Xiaoping Wan 2020Signal Transduction and Targeted Therapy2020,5,1:3
11Adaptive immune responses to SARS-CoV-2 infection in severe versus mild individuals显示文摘The global Coronavirus disease 2019(COVID-19)pandemic caused by SARS-CoV-2 has affected more than eight million people.There is an urgent need to investigate how the adaptive immunity is established in COVID-19 patients.In this study,we proled adaptive immune cells of PBMCs from recovered COVID-19 patients with varying disease severity using single-cell RNA and TCR/BCR V(D)J sequencing.The sequencing data revealed SARS-CoV-2-specic shufing of adaptive immune repertories and COVID-19-induced remodeling of peripheral lymphocytes.Characterization of variations in the peripheral T and B cells from the COVID-19 patients revealed a positive correlation of humoral immune response and T-cell immune memory with disease severity.Sequencing and functional data revealed SARS-CoV-2-specic T-cell immune memory in the convalescent COVID-19 patients.Furthermore,we also identied novel antigens that are responsive in the convalescent patients.Altogether,our study reveals adaptive immune repertories underlying pathogenesis and recovery in severe versus mild COVID-19 patients,providing valuable information for potential vaccine and therapeutic development against SARS-CoV-2 infection.Fan Zhang Rui Gan Ziqi Zhen Xiaoli Hu Xiang Li Fengxia Zhou Ying Liu Chuangeng Chen Shuangyu Xie Bailing Zhang Xiaoke Wu Zhiwei Huang 2020Signal Transduction and Targeted Therapy2020,5,1:3
12Expression patterns and functional divergence of homologous genes accompanied by polyploidization in cotton(Gossypium hirsutum L.)显示文摘Naturally allotetraploid cotton has been widely used as an ideal model to investigate gene expression remodeling as a consequence of polyploidization.However,the global gene pattern variation during early fiber development was unknown.In this study,through RNA-seq technology,we comprehensively investigated the expression patterns of homologous genes between allotetraploid cotton(G.hirsutum)and its diploid progenitors(G.arboreum and G.raimondii)at the fiber early development stage.In tetraploid cotton,genes showed expression level dominance(ELD)bias toward the A genome.This phenomenon was explained by the up-/downregulation of the homologs from the nondominant progenitor(D genome).Gene ontology(GO)enrichment results indicated that the ELD-A genes might be a prominent cause responsible for fiber property change through regulating the fatty acid biosynthesis/metabolism and microtubule procession,and the ELD-D genes might be involved in transcription regulation and stress inducement.In addition,the number and proportion of completely A-and D-subfunctionalized gene were similar at different fiber development stages.However,for neofunctionalization,the number and proportion of reactivated D-derived genes were greater than those of A at 3 and 5 DPA.Eventually,we found that some homologous genes belonging to several specific pathways might create novel asymmetric transcripts between two subgenomes during polyploidization and domestication process,further making the fiber property meet the human demands.Our study identified determinate pathways and their involved genes between allotetraploid cotton and their progenitors at early fiber development stages,providing new insights into the mechanism of cotton fiber evolution.Zhen Peng Hua Cheng Gaofei Sun Zhaoe Pan Xiao Wang Xiaoli Geng Shoupu He Xiongming Du 2020Science China(Life Sciences)2020,63,10:2
