| 1 | Research on unloading nonlinear mechanical characteristics of jointed rock masses显示文摘Geological environments of rock mass projects are always very complicated, and further investigations on rock mechanical characteristics are needed. There are considerable distinctions in rock mechanical characteristics under unloading and loading conditions. A series of tests are conducted to study the stress-strain relationship of rock masses under loading and unloading conditions. Also, the anisotropy, the size effect, and the rheological property of unloading rock mass are investigated. The tests presented in the paper include model test and granite rheological test, which are conducted considering geological condition, rock mass structure, in-situ stress field of the permanent shiplock of the Three Gorges Project. The main differences between loading and unloading rock masses are stress paths, yield criteria, deformation and strength parameters, etc.. Different structural plane directions affect unloading rock mass evidently. With increasing size, the tensile strength, the compressive strength, the deformation modulus, the Poisson's ratio and the anisotropy of rock mass all decrease. For sandstone samples with parallel bedding planes, the cohesion c increases but the internal friction angle ? decreases under unloading condition when compared with the values under loading condition. While for samples with vertical bedding planes, the trend is adverse. The rheological property of rocks has close relationship with the tensile stresses of rock masses. When the sandstone samples are tested under high stress condition, their rheological properties are very obvious with the unloading of confining pressure, and three typical rheological stages are shown. | 3 | Bandgap prediction by deep learning in configurationally hybridized graphene and boron nitride显示文摘It is well-known that the atomic-scale and nano-scale configuration of dopants can play a crucial role in determining the electronic properties of materials.However,predicting such effects is challenging due to the large range of atomic configurations that are possible.Here,we present a case study of how deep learning algorithms can enable bandgap prediction in hybridized boron–nitrogen graphene with arbitrary supercell configurations.A material descriptor that enables correlation of structure and bandgap was developed for convolutional neural networks.Bandgaps calculated by ab initio calculations,and corresponding structures,were used as training datasets.The trained networks were then used to predict bandgaps of systems with various configurations.For 4×4 and 5×5 supercells they accurately predict bandgaps,with a R^(2) of >90% and root-mean-square error of~0.1 eV.The transfer learning was performed by leveraging data generated from small supercells to improve the prediction accuracy for 6×6 supercells.This work will pave a route to future investigation of configurationally hybridized graphene and other 2D materials.Moreover,given the ubiquitous existence of configurations in materials,this work may stimulate interest in applying deep learning algorithms for the configurational design of materials across different length scales. | Yuan Dong Chuhan Wu Chi Zhang Yingda Liu Jianlin Cheng Jian Lin | 2019 | npj Computational Materials2019,,1: | 5 |
| 4 | Magnitude of modulation of gene expression in aneuploid maize depends on the extent of genomic imbalance显示文摘Aneuploidy has profound effects on an organism,typically more so than polyploidy,and the basis of this contrast is not fully understood.A dosage series of the maize long arm of chromosome 1(1L)was used to compa re relative global gene expression in diffe rent types and degrees of aneuploidy to gain insights into how the magnitude of genomic imbalance as well as hypoploidy affects global gene expression.While previously available methods require a selective examination of specific genes,RNA sequencing provides a whole-genome view of gene expression in aneuploids.Most studies of global aneuploidy effects have concentrated on individual types of aneuploids because multiple dose aneuploidies of the same genomic region are difficult to produce in most model genetic organisms.The genetic toolkit of maize allows the examination of multiple ploidies and 1-4 doses of chromosome arms.Thus,a detailed examination of expression changes both on the varied chromosome arms and elsewhere in the genome is possible,in both hypoploids and hyperploids,compared with euploid controls.Previous studies observed the inverse trans effect,in which genes not varied in DNA dosage were expressed in a negative relationship to the varied chromosomal region.This response was also the major type of changes found globally in this study.Many genes varied in dosage showed proportional expression changes,though some were seen to be partly or fully dosage compensated.It was also found that the effects of aneuploidy were progressive,with more severe aneuploids producing effects of greater magnitude. | Adam F·Johnson Jie Hou Hua Yang Xiaowen Shi Chen Chen Md Soliman Islam Tieming Ji Jianlin Cheng James A·Birchler | 2020 | Journal of Genetics and Genomics2020,47,2: | 3 |

| 9 | Active compounds of Caodoukou(Semen Alpinia Katsumadai)inhibit the migration,invasion and metastasis of human pancreatic cancer cells by targeting phosphoinosmde-3-kinase/protein kinase B/mammalian target of rapamycin pathway显示文摘OBJECTIVE:To detect the effects of active compounds of Caodoukou(Semen Alpinia Katsumadai)(ACAK)on the proliferation,migration and invasion of pancreatic cancer,and explain the possible molecular mechanism of ACAK interacting with these processes.Methods:Cell counting kit-8 method,cell scratch repair experiment,Transwell migration and invasion experiment,immunohistochemistry,western blot assay and real-time polymerase chain reaction experiment were used to evaluate the effect of ACAK on the proliferation,migration and invasion of pancreatic cancer cells.The levels of active molecules involved in the phosphoinosmde-3-kinase(PI3K)/Akt/the mammalian target of rapamycin(m TOR)signal transduction were detected by Western blot assay.In addition,the function of ACAK in vivo was evaluated by xenotransplantation tumor model in nude mice.Results:The inhibitory effect of ACAK on the proliferation of pancreatic cancer cells showed certain time-dose dependence.The results of scratch repair test,Transwell test,Western blotting and real time polymerase chain reaction assay showed that ACAK could inhibit the migration and invasion of pancreatic cancer cells in vitro.In addition,the regulatory effect of ACAK on epithelialmesenchymal transition(EMT)is partly attributed to PI3K/Akt/mT OR signaling pathway.The experimental results in vivo showed that ACAK regulated the development of pancreatic cancer.Conclusions:ACAK can partly inhibit the activity of EMT and matrix metallopeptidases by down-regulating the downstream proteins of PI3K/Akt/mTOR signal pathway,thus inhibiting the ability of migration and invasion of pancreatic cancer. | YANG Xiaohui WANG Jian CHENG Li ZHANG Yuxi HUANG Jianlin LIU Minghua | 2023 | Journal of Traditional Chinese Medicine2023,43,5: | 0 |
| 10 | Genomic imbalance modulates transposable element expression in maize显示文摘Genomic imbalance refers to the more severe phenotypic consequences of changing part of a chromosome compared with the whole genome set.Previous genome imbalance studies in maize have identified prevalent inverse modulation of genes on the unvaried chromosomes(trans)with both the addition or subtraction of chromosome arms.Transposable elements(TEs)comprise a substantial fraction of the genome,and their reaction to genomic imbalance is therefore of interest.Here,we analyzed TE expression using RNA-seq data of aneuploidy and ploidy series and found that most aneuploidies showed an inverse modulation of TEs,but reductions in monosomy and increases in disomy and trisomy were also common.By contrast,the ploidy series showed little TE modulation.The modulation of TEs and genes in the same experimental group were compared,and TEs showed greater modulation than genes,especially in disomy.Class Ⅰ and Ⅱ TEs were differentially modulated in most aneuploidies,and some superfamilies in each TE class also showed differential modulation.Finally,the significantly upregulated TEs in three disomies(TB-7Lb,TB9Lc,and TB-10L19)did not increase the proportion of adjacent gene expression when compared with non-differentially expressed TEs,indicating that modulations of TEs do not compound the effect on genes.These results suggest that the prevalent inverse TE modulation in aneuploidy results from stoichiometric upset of the regulatory machinery used by TEs,similar to the response of core genes to genomic imbalance. | Hua Yang Xiaowen Shi Chen Chen Jie Hou Tieming Ji Jianlin Cheng James A.Birchler | 2023 | Plant Communications2023,4,2: | 0 |
| 11 | 降低Pin1可以抑制大肠癌细胞SW620的侵袭转移能力(英文)显示文摘Objective:The aim of our study was to investigate the effect of Pin1 on the expression of MMP-2 and MMP-9 in human colorectal carcinoma SW620 cells. Methods: We constructed a eukaryotic expression vector of RNA interfering (shRNA) for Pin1 gene (pGenesil-1-Pin1), and then observed its expression in SW620 cells by Western blotting. The cells motility were tested by wound healing assay and Boyden chamber assay. The protein levels and activity of MMP-2 and MMP-9 were tested by Western blotting and Gelatin zymography in SW620 cells after transfected with pGenesil-1-PIN1. Results: pGenesil-1-PIN1 was successfully constructed, which was confirmed by sequencing. Silencing the Pin1 by RNAi significantly decreased the cells motility from 96.4±3.9 per field (×10 objective) to 52.7±4.4 per field (P<0.05, Student's t-test) for SW620 cells transfected with pGenesil-1-PIN1 (SW620/p-shRNA) in Boyden chamber assay, and reduced the MMP-2 and MMP-9 expressions and activity in SW620 cells. The protein relative levels of MMP-2 were 0.32±0.04 for SW620/p-shRNA, and 0.76±0.03 for SW620/p-Con; MMP-9 were 0.41±0.09 for SW620/p-shRNA, and 0.94±0.07 for SW620/p-Con (p<0.05). | 12 | A back analysis scheme for refined soil stratification based on integrating borehole and CPT data显示文摘Utilizing both borehole and Cone Penetration Testing(CPT)data in soil stratification helps to get more convincing soil stratification results.However,the soil classification results revealed by borehole(Unified Soil Classification System,USCS)and CPT tests(soil behavior type,SBT)are commonly not con-sistent.This study proposes a feasible solution to integrate the borehole and CPT data with the tree-based method.The tree-based method is naturally suitable for soil stratification tasks as it aims to divide the subsurface space into several clusters based on the similarities of the soil types.A novel boundary dic-tionary method is proposed to enhance the model performance on complex soil layer conditions.A prob-abilistic mapping matrix between the USCS-SBT system is built based on a collected municipal database with collocated borehole and CPT data.The optimal soil stratification results can be selected based on considering multiple borehole information and pruning the structure of trees.The structure of the trees can be optimized in a back analysis perspective with the Sequential Model-Based Global Optimization(SMBO)algorithm which aims to maximize the possibility of observing the borehole information based on the USCS-SBT probabilistic mapping matrix.The uncertainties of the optimal soil stratification results can be estimated based on a weighted Gini index method.The performance of the proposed method is validated based on a real case in New Zealand with a cross-validation method.The results indicate that the proposed method is robust and effective. | Jiawei Xie Cheng Zeng Jinsong Huang Yuting Zhang Jianlin Lu | 2024 | Geoscience Frontiers2024,15,1: | 0 |
| 13 | Observation-based sources evolution of non-methane hydrocarbons (NMHCs) in a megacity of China显示文摘Both concentrations and emissions of many air pollutants have been decreasing due to implement of control measures in China,in contrast to the fact that an increase in emissions of non-methane hydrocarbons(NMHCs)has been reported.This study employed seven years continuous NMHCsmeasurements and the related activities data of Shanghai,a megacity in China,to explore evolution of emissions and effectiveness of air pollution control measures.The mixing ratio of NMHCs showed no statistical interannual changes,of which their compositions exhibited marked changes.This resulted in a decreasing trend of ozone formation potential by 3.8%/year(p<0.05,the same below),which should be beneficial to ozone pollution mitigation as its production in Shanghai is in the NMHCs-limited regime.Observed alkanes,aromatics and acetylene changed by+3.7%/year,-5.9%/year and-7.4%/year,respectively,and alkenes showed no apparent trend.NMHCs sources were apportioned by a positive matrix factorization model.Accordingly,vehicular emissions(-5.9%/year)and petrochemical industry emissions(-7.1%/year)decreased significantly,but the decrease slowed down;significant reduction in solvent usage(-9.0%/year)appeared after 2010;however,emissions of natural gas(+12.6%/year)and fuel evaporation(with an increasing fraction)became more important.The inconsistency between observations and inventories was found in interannual trend and speciation as well as source contributions,emphasizing the need for further validation in NMHCs emission inventory.Our study confirms the effectiveness of measures targeting mobile and centralized emissions from industrial sources and reveals a need focusing on fugitive emissions,which provided new insights into future air policies in polluted region. | Yarong Peng Hongli Wang Qian Wang Shengao Jing Jingyu An Yaqin Gao Cheng Huang Rusha Yan Haixia Dai Tiantao Cheng Qiang Zhang Meng Li Jianlin Hu Zhihao Shi Li Li Shengrong Lou Shikang Tao Qinyao Hu Jun Lu Changhong Chen | 2023 | Journal of Environmental Sciences2023,,2: | 0 |
