| 1 | A method for the production and expedient screening of CRISPR/Cas9-mediated non-transgenic mutant plants显示文摘Developing CRISPR/Cas9-mediated non-transgenic mutants in asexually propagated perennial crop plants is challenging but highly desirable.Here,we report a highly useful method using an Agrobacterium-mediated transient CRISPR/Cas9 gene expression system to create non-transgenic mutant plants without the need for sexual segregation.We have also developed a rapid,cost-effective,and high-throughput mutant screening protocol based on Illumina sequencing followed by high-resolution melting(HRM)analysis.Using tetraploid tobacco as a model species and the phytoene desaturase(PDS)gene as a target,we successfully created and expediently identified mutant plants,which were verified as tetra-allelic mutants.We produced pds mutant shoots at a rate of 47.5%from tobacco leaf explants,without the use of antibiotic selection.Among these pds plants,17.2%were confirmed to be non-transgenic,for an overall non-transgenic mutation rate of 8.2%.Our method is reliable and effective in creating non-transgenic mutant plants without the need to segregate out transgenes through sexual reproduction.This method should be applicable to many economically important,heterozygous,perennial crop species that are more difficult to regenerate. | Longzheng Chen Wei Li Lorenzo Katin-Grazzini Jing Ding Xianbin Gu Yanjun Li Tingting Gu Ren Wang Xinchun Lin Ziniu Deng Richard J.McAvoy Frederick G.Gmitter Jr. Zhanao Deng Yunde Zhao Yi Li | 2018 | Horticulture Research2018,5,1: | 12 |
| 2 | Effect of the Cutter Parameters and Machining Parameters on the Interference in Gear Slicing显示文摘Current researches have not yet found the effect law of the cutter parameters and machining parameters on the interference in gear slicing,the interference between the cutter and machined gear often happens because the appropriate cutter parameters and machining parameters cannot be set,which reduces the gear machining accuracy.The relative position between the major flank face and edge-sweeping surface,distribution law of the interference area in forming process of edge-sweeping surface,and effect law of relative positions among edge-sweeping surfaces on the interference are studied by graphical analysis.The effect law of the cutter parameters and machining parameters on the interference is found.The effect law shows that the interference in gear slicing can be controlled when the relief angle measured on the top edge and feed of every rotation are chosen respectively larger than 9°and smaller than 0.15 mm/r.An internal helical gear is sliced with the spur slice cutter and the cutter parameters and machining parameters are set based on above the effect law.The machined gear is measured in Gear Measuring Center and the detection result shows that the comprehensive accuracy reaches GB/T Class 7,where some reach GB/T Class 6.The result can meet the gear machining accuracy requirement and shows that the effect law found is valid.The problem of the interference in gear slicing is solved and the gear machining accuracy can be improved. | CHEN Xinchun LI Jia LOU Benchao SHI Jiang YANG Qijun | 2013 | Chinese Journal of Mechanical Engineering2013,26,6: | 10 |

| 9 | Influence of tribofilm on superlubricity of highly-hydrogenated amorphous carbon films in inert gaseous environments显示文摘In this study,we mainly focus on the structural morphology and inter-atomic bonding state of tribofilms resulting from a highly-hydrogenated amorphous carbon(a-C:H) film in order to ascertain the underlying mechanisms for its superlubric behavior(i.e.,less than 0.01 friction coefficient).Specifically,we achieved superlubricity(i.e.,friction coefficients of down to 0.003) with this film in dry nitrogen and argon atmospheres especially when the tribo-pair is made of an a-C:H coated Si disk sliding against an a-C:H coated steel ball,while the a-C:H coated disk against uncoated ball does not provide superlubricity.We also found that the state of superlubricity is more stable in argon than in nitrogen and the formation of a smooth and uniformly-thick carbonaceous tribofilm appears to be one of the key factors for the realization of such superlubricity.Besides,the interfacial morphology of sliding test pairs and the atomic-scale bond structure of the carbon-based tribofilms also play an important role in the observed superlubric behavior of a-C:H films.Using Raman spectroscopy and high resolution transmission electron microscopy,we have compared the structural differences of the tribofilms produced on bare and a-C:H coated steel balls.For the a-C:H coated ball as mating material which provided superlow friction in argon,structural morphology of the tribofilm was similar or comparable to that of the original a-C:H coating;while for the bare steel ball,the sp^2-bonded C fraction in the tribofilm increased and a fingerprint-like nanocrystalline structure was detected by high resolution transmission electron microscopy(HRTEM).We also calculated the shear stresses for different tribofilms,and established a relationship between the magnitude of the shear stresses and the extent of sp^3-sp^2 phase transformation. | LIU ShuWei ZHANG ChenHui OSMAN Eryilmaz CHEN XinChun MA TianBao HU YanZhong LUO JianBin ALI Erdemir | 2016 | Science China(Technological Sciences)2016,59,12: | 2 |
| 10 | Modified ADM1 for modeling free ammonia inhibition in anaerobic acidogenic fermentation with high-solid sludge显示文摘Anaerobic acidogenic fermentation with high-solid sludge is a promising method for volatile fatty acid(VFA) production to realize resource recovery. In this study, to model inhibition by free ammonia in high-solid sludge fermentation, the anaerobic digestion model No. 1(ADM1) was modified to simulate the VFA generation in batch, semicontinuous and full scale sludge. The ADM1 was operated on the platform AQUASIM 2.0.Three kinds of inhibition forms, e.g., simple inhibition, Monod and non-inhibition forms,were integrated into the ADM1 and tested with the real experimental data for batch and semi-continuous fermentation, respectively. The improved particle swarm optimization technique was used for kinetic parameter estimation using the software MATLAB 7.0. In the modified ADM1, the Ksof acetate is 0.025, the km,acis 12.51, and the KI_NH3is 0.02,respectively. The results showed that the simple inhibition model could simulate the VFA generation accurately while the Monod model was the better inhibition kinetics form in semi-continuous fermentation at p H 10.0. | 13 | Effects of Long-Term Fertilization Strategies on Soil Productivity and Soybean Rhizobial Diversity in a Chinese Mollisol显示文摘Rhizobial diversity is affected by interactions between soil features, fertilization strategy, and cropping system. However, interactions among the rhizobial community, chemical-organic manure fertilization, and plant production have not been well documented in Mollisols from long-term experiments. Aimed at maintaining and recovering the productivity of Chinese Mollisols, a long-term fertilization experiment had been carried out for 29 years under a wheat-maize-soybean rotation system, involving the application of recycled organic manure (ROM), chemical fertilizers (N, P, and/or K), or ROM plus N, P, and/or K. In the present study, the effects of different treatments were evaluated by determining soil physicochemical features, soybean production, and soybean rhizobial diversity. The results showed that application of ROM plus NPK maintained or increased soil fertility, which was accompanied by higher production and higher diversity of rhizobia, as compared with the other treatments. The negative association of Bradyrhizobium japonicum with N fertilizer, positive association of B. diazoefficiens with soil pH, and alleviation of N-inhibition on the diversity of Bradyrhizobium by the addition of ROM were recorded as new findings. | 20 | Covert communication with beamforming over MISO channels in the finite blocklength regime显示文摘This paper considers beamforming techniques for covert communication over multiple input single output(MISO) channels with finite blocklength. We first show that the optimal input distribution for covert communication over complex Gaussian channels is circular symmetric complex Gaussian. By reviewing our previous results on the throughput of Gaussian random coding over additive white Gaussian noise(AWGN) channels, the achievability and converse bounds on the attainable throughput over MISO channels are analyzed. Then, the optimal beamforming strategies and their relationship with the transmitting power are thoroughly investigated in a variety of situations in terms of several types of channel state information available at both ends of the system. We reveal the fact that the maximal allowable transmit power is not constrained by covertness requirement when there is full channel information of the adversary, while there is an upper bound of the transmit power which is based on beamforming and an outage-based covertness criterion if there is only partial channel information of the adversary.
