1miR-503-3p promotes epithelialemesenchymal transition in breast cancer by directly targeting SMAD2 and E-cadherin显示文摘Although progress in clinical and basic research has significantly increased our understanding of breast cancer, little is known about the molecular mechanism underlying breast cancer metastasis. Identification of effective therapeutic targets to prevent breast cancer metastasis is urgently needed. The function of mi R-503-3p has been investigated in other cancers, but its role in breast cancer remains undefined.Here, we found that mi R-503-3p was overexpressed in breast cancer tissue and plasma compared with adjacent normal breast tissue and with plasma from healthy individuals. Moreover, we identified mi R-503-3p to be an oncogene of breast cancer cell proliferation, migration and invasion. Upregulation of mi R-503-3p in breast cancer cells inhibited expression of epithelialemesenchymal transition(EMT)-related protein SMAD2 and the epithelial marker protein E-cadherin by directly binding to their m RNA30 untranslated region, whereas increased expression of mesenchymal marker proteins, including vimentin and N-cadherin. 2Multi-stage Dynamic Post-disaster Recovery Strategy for Distribution Networks Considering Integrated Energy and Transportation Networks显示文摘To improve the resilience of distribution networks(DNs),a multi-stage dynamic recovery strategy is proposed in this paper,which is designed for post-disaster DN considering an integrated energy system(IES)and transportation network(TN).First,the emergency response quickly increases the output of gas turbines(GTs)in the natural gas network(NGN),and responsively reconfigures the DN in microgrids,to maximize the amount of loads to be restored.The single-commodity flow model is adopted to construct spanning tree constraints.Then,in the second stage of energy storage recovery,mobile energy storage systems(MESSs)are deployed to cover the shortages of power demands,i.e.,to further restore the loads after evaluating the load recovery situation.The Floyd algorithm based dynamic traffic assignment(DTA)is selected to obtain the optimal path of the MESSs.In the third stage,the outputs of various post-disaster recovery measures are adjusted to achieve an economically optimized operation.Case studies demonstrate the effectiveness of the proposed dynamic post-disaster recovery strategy.Xinyi Jiang Jian Chen Ming Chen Zhen Wei 2021CSEE Journal of Power and Energy Systems2021,7,2:6
3In vitro and in vivo activity of D-serine in combination with β-lactam antibiotics against methicillin-resistant Staphylococcus aureus显示文摘As D-amino acids play important roles in the physiological metabolism of bacteria,combination of D-amino acids with antibiotics may provide synergistic antibacterial activity. The aim of the study was to evaluate in vitro and in vivo activity of D-serine alone and in combination withβ-lactams against methicillin-resistant Staphylococcus aureus(MRSA) strains, and to explore the possible sensitization mechanisms. The activity of D-serine, β-lactams alone and in combinations was evaluated both in vitro by standard MICs, time–kill curves and checkerboard assays, and in vivo by murine systemic infection model as well as neutropenic thigh infection model. An in vitro synergistic effect was demonstrated with the combination of D-serine and β-lactams against MRSA standard and clinical strains.Importantly, the combinations enhanced the therapeutic efficacy in the animal models as compared toβ-lactam alone groups. Initial mechanism study suggested possible revision of D-alanine-D-alanine residue to D-alanine-D-serine in peptidoglycan by adding of D-alanine in the medium, which may cause decreased affinity to PBPs during transpeptidation. In conclusion, D-serine had synergistic activity in combination with β-lactams against MRSA strains both in vitro and in vivo. 6Diamond/β-SiC film as adhesion-enhanced interlayer for top diamond coatings on cemented tungsten carbide substrate显示文摘In present work, diamond/β-SiC composite interlayers were deposited on cemented tungsten carbide(WC-6%Co) substrates by microwave plasma enhanced chemical vapor deposition(MPCVD) using H_2,CH_4 and tetramethylsilane(TMS) gas mixtures. The microstructure, chemical bonding, element distribution and crystalline quality of the composite interlayers were systematically characterized by means of field-emission scanning electron microscopy(FE-SEM), X-ray diffraction(XRD), X-ray photoelectron spectrometer(XPS), electron probe microanalysis(EPMA), Raman spectroscopy and transmission electron microscropy(TEM). The influences of varying TMS flow rates on the diamond/β-SiC composite interlayers were investigated. Through changing the TMS flow rates in the reaction gas, the volume fraction of β-SiC in the composite interlayers were tunable in the range of 12.0%–68.1%. XPS and EPMA analysis reveal that the composite interlayers are composed of C, Si element with little cobalt distribution. The better crystallinity of the diamond in the composite is characterized based on the Raman spectroscopy, which are helpful to deposit top diamond coatings with high quality. Then, the adhesion of top diamond coatings were estimated using Rockwell C indentation analysis, revealing that the adhesion of top diamond coatings on the WC-6%Co substrates can be improved by the interlayers with the diamond/β-SiC composite structures. Comprehensive TEM interfacial analysis exhibits that the cobalt diffusion is weak from WC-6%Co substrate to the composite interlayer. The homogeneous microcrystalline diamond coatings with the most excellent adhesion can be fabricated on the substrates with the composite interlayer with the β-SiC/diamond ratio of about 45%. 7Silicon prepared by electro-reduction in molten salts as new energy materials显示文摘Silicon has a large impact on the energy supply and economy in the modern world. In industry, high purity silicon is firstly prepared by carbothermic reduction of silica with the produced raw silicon being further refined by a modified Siemens method. This process suffers from the disadvantages of high cost and contaminant release and emission. As an alternative, the molten salt electrolysis approach, particularly the FFC Cambridge Process(FFC: Fray-Farthing-Chen), could realize high purity silicon products with morphology-controllable nanostructures at low or mild temperatures(generally 650–900 ℃). In this article, we review the development, reaction mechanisms, and electrolysis conditions of silicon production by the FFC Cambridge Process. 8Optimal Cooperative Sensing Order Using Inactive Secondary Users in Cognitive Radio Networks显示文摘A novel cooperative spectrum sensing order which utilizes inactive Secondary users(SUs) efficiently based on maximum throughput has been proposed in Cognitive radio networks(CRNs). In order to predict the states of Primary users(PUs), we build the PU's traffic pattern as a Continuous-time Markov chain(CTMC) process. CRNs obtain the maximum throughput while SUs sense the licensed channels with the optimal order. The numerical simulation results show that the proposed order based on spectrum sensing scheme can achieve larger channel utilization and lower sensing overhead as compared with the spectrum sensing scheme without using the optimal order sensing. 9A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences.We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://bencao.bmicc.cn),annotated the sequences by sequence-based clustering,and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools,our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas,part of the predicted human target genes were experimentally validated,suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines.We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions.Moreover,the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction,demonstrated potent medical effects.We propose a Bencao(herbal)Index,including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine.The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine,and may provide potential remedies for the theory and practice of one medicine.Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang 2023Science China(Life Sciences)2023,66,10:2
10SINR Driven Joint Network Selection Policy in the Heterogeneous Internet of Things显示文摘In the heterogeneous Internet of things(Io T), the Signal to interference plus noise ratio(SINR)and delay constraint are two important factors that influence the throughput of Io T and the performance of users.Until recently, most network selection policy researches were based on either the Shannon theory or the signal strength, while the combined influence of the delay constraint and the SINR, which has a significant impact on resource utilization, is hardly considered. We therefore propose an SINR driven joint network selection policy, which incorporates the delay constraint and the signal strength into the SINR. 13Two-step Optimal Allocation of Stationary and Mobile Energy Storage Systems in Resilient Distribution Networks显示文摘Energy storage systems (ESSs) are acknowledged to be a promising option to cope with issues in high penetration of renewable energy and guarantee a highly reliable power supply. In this paper, a two-step optimal allocation model is proposed to obtain the optimal allocation (location and size) of stationary ESSs (SESSs) and mobile ESSs (MESSs) in the resilient distribution networks (DNs). In the first step, a mixed-integer linear programming (MILP) problem is formulated to obtain the preselected location of ESSs with consideration of different scenarios under normal operation conditions. In the second step, a two-stage robust optimization model is established to get the optimal allocation results of ESSs under failure operation conditions which are solved by column-and-constraint generation (C&CG) algorithm. A hybrid ESS allocation strategy based on the subjective and objective weight analysis is proposed to give the final allocation scheme of SESSs and MESSs. 14Stress-controlled LCF experiments and ratcheting behaviour simulation of a nickel-based single crystal superalloy with [001] orientation显示文摘Uniaxial ratcheting behaviour and low cycle fatigue(LCF)failure mechanism of nickel-based single crystal superalloy DD6 with[001]orientation are investigated through the stresscontrolled LCF tests with stress ratio of-1.Then the deformation behaviour during the wholelifetime from the beginning of the experiment to the fracture of the specimen,as well as the fractographic/metallographic morphology,are compared with the strain-controlled LCF experimental results.Through the scanning electron microscope(SEM)observations,it is shown that the failure characteristics under stress-controlled LCF loading are similar with those under strain-controlled loading.Nevertheless,unlike strain-controlled LCF loading,even under fully reversed cycle loading for stress-controlled LCF,DD6 shows significant ratcheting behaviour due to the tensioncompression asymmetry.In addition,the LCF lifetimes under stress control are significantly shorter than the LCF lifetimes under strain control,and the culprit might be the detrimental effect of ratcheting strain on LCF lifetime.Based on these phenomena,an improved crystal plasticity constitutive model on the basis of slip-based Walker constitutive model is developed through modifying the kinematic hardening rule in order to overcome the inaccurate prediction of decelerating stageand stable stage of ratcheting behaviour.Furthermore,combining the continuum damage mechanics,a damage-coupled crystal plasticity constitutive model is proposed to reflect the damage behaviour of DD6 and the accelerating stage of ratcheting behaviour.The simulation results for the stress-controlled LCF deformation behaviour including the whole-lifetime ratcheting behaviour show good agreement with the experimental data.Bin ZHANG Rongqiao WANG Dianyin HU Kanghe JIANG Jianxing MAO Fulei JING Xinyi HAO 2021Chinese Journal of Aeronautics2021,34,8:2

16Polymyxin resistance caused by large-scale genomic inversion due to IS26 intramolecular translocation in Klebsiella pneumoniae显示文摘Polymyxin B and polymyxin E(colistin)are presently considered the last line of defense against human infections caused by multidrug-resistant Gram-negative organisms such as carbapenemase-producer Enterobacterales,Acinetobacter baumannii,and Klebsiella pneumoniae.Yet resistance to this last-line drugs is a major public health threat and is rapidly increasing.Polymyxin S2(S2)is a polymyxin B analogue previously synthesized in our institute with obviously high antibacterial activity and lower toxicity than polymyxin B and colistin.To predict the possible resistant mechanism of S2for wide clinical application,we experimentally induced bacterial resistant mutants and studied the preliminary resistance mechanisms.Mut-S,a resistant mutant of K.pneumoniae ATCC BAA-2146(Kpn2146)induced by S2,was analyzed by whole genome sequencing,transcriptomics,mass spectrometry and complementation experiment.Surprisingly,large-scale genomic inversion(LSGI)of approximately 1.1 Mbp in the chromosome caused by IS26mediated intramolecular transposition was found in Mut-S,which led to mgrB truncation,lipid A modification and hence S2resistance.The resistance can be complemented by plasmid carrying intact mgrB.The same mechanism was also found in polymyxin B and colistin induced drug-resistant mutants of Kpn2146(Mut-B and Mut-E,respectively).This is the first report of polymyxin resistance caused by IS26 intramolecular transposition mediated mgrB truncation in chromosome in K.pneumoniae.The findings broaden our scope of knowledge for polymyxin resistance and enriched our understanding of how bacteria can manage to survive in the presence of antibiotics.Haibin Li Lang Sun Han Qiao Zongti Sun Penghe Wang Chunyang Xie Xinxin Hu Tongying Nie Xinyi Yang Guoqing Li Youwen Zhang Xiukun Wang Zhuorong Li Jiandong Jiang Congran Li Xuefu You 2023Acta Pharmaceutica Sinica B2023,13,9:1
17Nanopolyphenol rejuvenates microglial surveillance of multiple misfolded proteins through metabolic reprogramming显示文摘Microglial surveillance plays an essential role in clearing misfolded proteins such as amyloid-beta,tau,andα-synuclein aggregates in neurodegenerative diseases.However,due to the complex structure and ambiguous pathogenic species of the misfolded proteins,a universal approach to remove the misfolded proteins remains unavailable.Here,we found that a polyphenol,α-mangostin,reprogrammed metabolism in the disease-associated microglia through shifting glycolysis to oxidative phosphorylation,which holistically rejuvenated microglial surveillance capacity to enhance microglial phagocytosis and autophagy-mediated degradation of multiple misfolded proteins.Nanoformulation ofα-mangostin efficiently deliveredα-mangostin to microglia,relieved the reactive status and rejuvenated the misfolded-proteins clearance capacity of microglia,which thus impressively relieved the neuropathological changes in both Alzheimer’s disease and Parkinson’s disease model mice.These findings provide direct evidences for the concept of rejuvenating microglial surveillance of multiple misfolded proteins through metabolic reprogramming,and demonstrate nanoformulatedα-mangostin as a potential and universal therapy against neurodegenerative diseases.Dayuan Wang Xiao Gu Xinyi Ma Jun Chen Qizhi Zhang Zhihua Yu Juan Li Meng Hu Xiaofang Tan Yuyun Tang Jianrong Xu Minjun Xu Qingxiang Song Huahua Song Gan Jiang Zaiming Tang Xiaoling Gao Hongzhuan Chen 2023Acta Pharmaceutica Sinica B2023,13,2:1

20Growth behavior of CVD diamond films with enhanced electron field emission properties over a wide range of experimental parameters显示文摘In this study, diamond films were synthesized on silicon substrates by microwave plasma enhanced chemical vapor deposition(CVD) over a wide range of experimental parameters. The effects of the microwave power,CH_4/H_2 ratio and gas pressure on the morphology, growth rate, composition, and quality of diamond films were investigated by means of scanning electron microscope(SEM), X-ray diffraction(XRD), Raman spectroscopy and X-ray photoelectron spectroscopy(XPS). A rise of microwave power can lead to an increasing pyrolysis of hydrogen and methane, so that the microcrystalline diamond film could be synthesized at low CH_4/H_2 levels. Gas pressure has similar effect in changing the morphology of diamond films, and high gas pressure also results in dramatically increased grain size. However,diamond film is deteriorated at high CH_4/H_2 ratio due to the abundant graphite content including in the films. Under an extreme condition of high microwave power of 10 kW and high CH_4 concentration, a hybrid film composed of diamond/graphite was successfully formed in the absence of N_2 or Ar,which is different from other reports. This composite structure has an excellent measured sheet resistance of 10-100 Ω/Sqr. which allows it to be utilized as field electron emitter.
