| 1 | Image denoising and deblurring: non-convex regularization, inverse diffusion and shock filter显示文摘A large number of applications in image processing and computer vision depend on image quality. In this paper, main concerns are image denoising and deblurring simultaneously in a restoration task by three types of methodologies: non-convex regularization, inverse diffusion and shock filter. We discuss their relations in the context of image deblurring: the inverse diffusion implied by the non-convex regularization, and the superior ability of deblurring edge of the shock filter to that of the inverse diffusion, both in 1D and 2D cases. Finally, we propose a region-based adaptive anisotropic diffusion with shock filter method, which shows advantages of deblurring edges, denoising and smoothing contours in experiments, compared with some related methods. | 2 | Deep ultraviolet light-emitting diodes based on a well-ordered AlGaN nanorod array显示文摘The nanorod structure is an alternative scheme to develop high-efficiency deep ultraviolet light-emitting diodes(DUV LEDs). In this paper, we first report the electrically injected 274-nm AlGaN nanorod array DUV LEDs fabricated by the nanosphere lithography and dry-etching technique. Nanorod DUV LED devices with good electrical properties are successfully realized. Compared to planar DUV LEDs, nanorod DUV LEDs present>2.5 times improvement in light output power and external quantum efficiency. The internal quantum efficiency of nanorod LEDs increases by 1.2 times due to the transformation of carriers from the exciton to the free electron–hole, possibly driven by the interface state effect of the nanorod sidewall surface. In addition, the nanorod array significantly facilitates photons escaping from the interior of LEDs along the vertical direction, contributing to improved light extraction efficiency. A three-dimensional finite-different time-domain simulation is performed to analyze further in detail the TE-and TM-polarized photon extraction mechanisms of the nanostructure. | 3 | Mutation analyses of integrated HBV genome in hepatitis B patients显示文摘Little has been learnt in the last 30 years about detection of HBV genome as well as its mutation analysis between hepatitis B fathers (HBF) and their children. In this study, we used nest polymerase chain reaction (PCR), fluorescence in situ hybridization (FISH), and DNA sequencing analysis, to examine the integrated HBV genome in paraffin-embedded testis tissues, which were taken as samples from HBF, and in peripheral blood mononuclear cells (PBMC) from 74 cases of HBFs and their children who were born after their fathers’ HBV infection (caHBF). We found that HBV DNA existed in testis tissues, mainly in the basilar parts of the seminiferous tubules, and also in PBMC of HBF. It was also documented that there were point mutations of poly-loci, insertions and deletions of nucleotides in integrated HBV genomes, and the types of gene mutations in the HBFs were similar to those in caHBF. | 4 | Effects of Culture Conditions on Growth and Docosahexaenoic Acid Production from Schizochytrium limacinum显示文摘The effects of temperature, initial pH, salinity of culture medium, and carbon and nitrogen sources on growth and doco- sahexaenoic acid (C22 : 6 n-3, DHA) production from Schizochytrium limacinum OUC88 were investigated in the present study. The results revealed that the optimal temperature, initial pH and salinity level of the medium for DHA production were 23℃, 7.0 and 18, respectively. Glucose was proved the best carbon source for the growth and DHA production from S. limacinum. Among the nitrogen sources tested, soybean cake hydrolysate, a cheap by-product, was found to be effective for the accumulation of DHA in S. lima- cinum cells. In addition, increasing the concentration of carbon sources in the medium caused a significant increase in cell biomass; however, accumulation of DHA in cells was mainly stimulated by the ratio of C/N in the medium. | 7 | Expression of the Phycoerythrin Gene of Gracilaria lemaneiformis (Rhodophyta) in E. coli and Evaluation of the Bioactivity of Recombinant PE显示文摘Phycoerythrin (PE) is one of the most important proteins involved in light capturing during photosynthesis in red algae. Its potential biological activities had gained wide concerns. In the present study, tumor cytotoxic and hydroxyl radical assay were preformed to detect the bioactivity of recombinant PE. Recombinant plasmids pGEX-PE and pBGL were transformed into E.coli BL21 to make two recombinant strains BEX (pGEX-PE) and BGL (pBGL). PE expressing in BEX (pGEX-PE) was validated by SDS-PAGE and Western blotting analysis. SDS-PAGE analysis indicated that the PE-GST fusion protein was mostly inclusion bod- ies. Specific expression of PE was confirmed by Western blotting analysis. The recombinant E. coli BEX (pGEX-PE) cells were col- lected and sonicated. The supernatants were reserved for the tumor cytotoxic experiments. The result of tumor cytotoxic assay indi- cated that the supernatants containing PE had the activity of inhibiting the growth of Hela cells and with the increase of protein con- centration, the inhibiting rate increased from 37.31% to 63.26%, which showed significant difference from the control. Hydroxyl radical scavenging effect was tested with supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates treated with sonication and heating. For the sonication samples, the scavenging rates of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were significantly higher than the negative control BL21(pGEX-4T) (P<0.02), and the scavenging rates increased slowly following the increase of the protein content. For the heating samples, except for the 0.2 mg mL-1 BGL (pBGL) products, the scavenging effects of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were stronger than that of negative control BL21(pGEX-4T). However, the effect intensity was not positively correlated with the increase of the protein concentration. Though a partially de- creased hydroxyl radical scavenging activity was led by heating, the biological activity was still retained and conspicuous. | 8 | Analysis of expressed sequence tags of a marine red alga,Gracilaria lemaneiformis显示文摘The expressed sequence tags (EST) has been proved to be a useful tool for discovering and identifying functional genes, especially in some species whose genetic information is unavailable. A total of 180 ESTs have been generated from a cDNA library of gametophytic Gracilaria lemaneiformis in this study. These clones are clustered into 151 groups, among which 8 groups are highly homologous to chloroplast genes and are abundant in the library. After searching for matches in the EST database of red alga, 22 groups are found to match with the registered ESTs of Rhodophyta and 6 with Gracilaria. | 12 | Heavy Metal Bioaccumulation and Toxicity with Special Reference to Microalgae显示文摘The bioaccumulation and toxicity of heavy metals were reviewed with special reference to microalgae, the key compo- nent of the food web in aquatic ecosystems. Heavy metals enter algal cells either by means of active transport or by endocytosis through chelating proteins and affect various physiological and biochemical processes of the algae. The toxicity primarily results from their binding to the sulphydryl groups in proteins or disrupting protein structure or displacing essential elements. Metals can break the oxidative balance of the algae, inducing antioxidant enzymes, such as superoxide dismutase (SOD), glutathione peroxidase (GPX) and ascorbate peroxidase (APX). The amount of oxidized proteins and lipids in the algal cells thus indicates the severity of the stress. Algal tolerance to heavy metal is highly dependent upon the defense response against the probable oxidative damages. Pro- duction of binding factors and proteins, exclusion of metals from cells by ion-selective transporters and excretion or compartmen- talization have been suggested with regard to reducing heavy metal toxicity. | 13 | Systematic study on the mechanical and electric behaviors of the nonbuckling interconnect design of stretchable electronics显示文摘Recently, we developed a nonbuckling interconnect design that provides an effective approach to simultaneously achieving high elastic stretchability, easiness for encapsulation, and high electric performance for stretchable electronics. This paper aims to systematically study its mechanical and electric behaviors, including comparisons of the nonbuckling and buckling interconnect designs on stretchability, effects of the thickness on electric performance, and modeling and experimental investigations on the finite deformation mechanics. It is found that the results on stretchability depend on the layouts. Long straight segments and small arc radii for nonbuckling interconnects yield an enhancement of stretchability, which is much better than that of buckling designs. On the other hand, shorter straight segments or thicker interconnects are better to lower the resistances of interconnects.Therefore, optimization of the designs needs to balance the requirements of both the mechanical and electric performances. The finite deformation of interconnects during stretching is analyzed. The established analytic model is well validated by both the finite element modeling and experimental investigations. | 14 | Optimization of the Purification Methods for Recovery of Recombinant Growth Hormone from Paralichthys olivaceus显示文摘This study aimed to optimize the purification of recombinant growth hormone from Paralichthys olivaceus. Recombinant flounder growth hormone (r-fGH) was expressed by Escherichia coli in form of inclusion body or as soluble protein under different inducing conditions. The inclusion body was renatured using two recovery methods, i.e., dilution and dialysis. Thereafter, the refolded protein was purified by Glutathione Sepharase 4B affinity chromatography and r-fGH was obtained by cleavage of thrombin. For soluble products, r-fGH was directly purified from the lysates by Glutathione Sepharase 4B affinity chromatography. ELISA-receptor assay demonstrated that despite its low receptor binding activity, the r-fGH purified from refolded inclusion body had a higher yield (2.605 mgL-1) than that from soluble protein (1.964 mgL-1). Of the tested recovery methods, addition of renaturing buffer (pH 8.5) into denatured inclusion body yielded the best recovery rate (17.9%). Results showed that each isolated promoter drived the expression of gfp in E.coli,and Promoter Ⅲ was the strongest one according to the GFP level. | 17 | Identification of a Putative Tetrasporophyte-Specific Gene in Gracilaria lemaneif ormis (Gracflariales, Rhodophyte)显示文摘A putative tetrasporophyte-specific gene,designated as SSH466(GenBank accession No. DQ019223) ,was one of the genes identified in this work using suppression subtractive hybridization(SSH) method in Gracilaria lemaneiformis. The full length of the gene was obtained using SMART RACE strategy. Sequence analysis revealed that the gene had 1 019 nucleotides,including an open reading frame of 498 nucleotides encoding 166 amino acid residues,158 nucleotides of 5' untranslated region and 363 nucleotides of 3' non-coding region. Protein motif and secondary structure prediction showed that there existed a transmembrane domain with a unique β-sheet. Thus,SSH466 protein might be a cross-membrane protein. Sequence homology search in the public GenBank databases did not reveal any significant match with SSH466. | 18 | Cloning, Sequencing and Phylogenetic Study of rbcL Gene from Cyanobacteria Arthrospira and Spirulina显示文摘Large subunit gene of rubisco (rbcL) of cyanobacteria Arthrospira platensis FACHB341, A. Platensis FACHB439, A. Maxima OUQDSM and Spirulina sp. FACHB440 is cloned, sequenced and characterized. Results show that GC content of the gene in strain Spirulina sp. FACHB440 is higher than that in the others. The alignments based on deduced amino acid sequences indicate that Spirulina sp. FACHB440 is different from that in other three samples of Arthrospira, though they have the same conserved functional sites (95, 98, 121, 124, 221, 257). The nucleotide sequence similarity among the three strains of the genus of Arthrospira (96.5~99.6%) is higher than that between Arthrospira and Spirulina (78.1~78.5%). By comparison of the corresponding sequence of other cyanobacteria, a phylogenetic tree with two clusters is constructed. A. Platensis FACHB341, A. Maxima OUQDSM and A. Platensis FACHB439 form the monophyletic linage, which is fully supported by bootstrap values (1000), while Spirulina sp. FACHB440 and Anabaena sp. | 20 | Freeze-drying-assisted Synthesis of Mesoporous CoMoO4 Nanosheets as Anode Electrode Material for Enhanced Lithium Batteries显示文摘A facile and green freeze-drying-assisted method was proposed to synthesize C0MoO4 mesoporous nano-sheets(MPNSs).The resulting product exhibits a Mgh specific capacity and good rate perfomance when evalimte an anode material for lithium-ion batteries(LIBs).The reversible specific capacity can be kept at 1105.2 mA·h·g^-1 after 100 cycles at a current density of 0.2 A/g.Even at the current densities of 1 and 4 A/gs the CoMoO4 MPNSs electrode can still retain the reversible capacities of 1148.7 and 540 mA·h·g^-1,respectively.Furthermore,the full cell(LiPePO4 catliode/CoMoO4 MPNSs anode)displays a stable discharge capacity of 146.7 mA·h·g^-1 at 0.1 C(1 C=170 mA/g)together with an initial coulombic efficiency of 98.2%.In addition,the CoMoO4 crystal structure is destroyed and reduced into Co^0 and Mo^0 in the first discharge process.In the subsequent cycles,the attractive Li storage properties come from the reversible conversions between Co/Co^2+and Mo/Mo^6+.The improved electroche-mical performance of CoMoO4 MPNSs is mainly attributed to their unique porous structures,which not only possess a good ion diffusion and electronic conduction pathway,but also provide many cavities to alleviate the volume changes during repeated cycling.This work offers a new perspective to the design of other porous electrode materials with a good energy storage performance. | WANG Wei WANG Tao FAN Xuecheng ZHANG Cuilin HU Jinxing CHEN Hui FANG Zhenxing YAN Jiefeng LIU Bing | 2019 | Chemical Research in Chinese Universities2019,35,2: | 1 |
