2Chromosome analysis of esophageal squamous cell carcinoma cell line KYSE 410-4 by repetitive multicolor fluorescence in situ hybridization显示文摘Chromosome aberrations are distinctive features of human malignant tumors. Analysis of chromosomal changes can illuminate the molecular mechanisms underlying the development and progression of cancer. To establish the technique of multicolor fluorescence in situ hybridization (M-FISH) for identifying chromosome aberrations in esophageal carcinoma cell line KYSE 410-4, four pools of 6-color whole-chromosome painting probes have been designed and hybridized on the same metaphase spread by four rounds of repetitive FISH. Repetitive 6-color M-FISH was successfully established and the cytogenetic abnormalities in KYSE 410-4 cells were characterized. Chromosome gains occurred at 2q, 3, 8, 17p, and X. An isochromosome 3q was visualized in the cell line, which might be one intermedi- ate mechanism leading to 3p losses and/or 3q gains. Furthermore, 16 structural arrangements were detected, including four derivative chromosomes. The rearrangement of the centromeric regions accounted for approximately 44% of all rearrangements. The results added a more complete and accurate information of the genetic alterations to the classical cytogenetic description of KYSE 410-4 and provided a detailed cytogenetic background data for appropriate use of the cell line. 3Selective oxidation of cyclopentene with H2O2 by using H3PW12O40 and TBAB as a phase transfer catalyst显示文摘The selective oxidation of cyclopentene by aqueous H2O2 using H3PW12O40 and tetrabutyl ammonium bromide(TBAB) as a phase transfer catalyst has been investigated. The results show that the presence of TBAB significantly improved the oxidation selectivity of cyclopentene. The effects of the reaction conditions on the conversion of cyclopentene were investigated in detail. The optimal reaction conditions are as follows: the H3PW12O40 to TBAB molar ratio, 1:1–1:3;H3PW12O40 to cyclopentene molar ratio,0.54:100–0.64:100;and molar ratio of H2O2 to cyclopentene, 1.6:1. The conversion reached to 59.8% in 4h at 35.0 ℃, while the selectivity of glutaraldehyde was 38.0% and the selectivity of 1,2-cyclopentanediol was 55.6%. 12Functional characterization of novel NPRL3 mutations identified in three families with focal epilepsy显示文摘Focal epilepsy accounts for 60% of all forms of epilepsy, but the pathogenic mechanism is not well understood. In this study,three novel mutations in NPRL3(nitrogen permease regulator-like 3), c.937_945del, c.1514dup C and 6,706-bp genomic DNA(g DNA) deletion, were identified in three families with focal epilepsy by linkage analysis, whole exome sequencing(WES) and Sanger sequencing. NPRL3 protein is a component of the GATOR1 complex, a major inhibitor of m TOR signaling. These mutations led to truncation of the NPRL3 protein and hampered the binding between NPRL3 and DEPDC5, which is another component of the GATOR1 complex. Consequently, the mutant proteins enhanced m TOR signaling in cultured cells, possibly due to impaired inhibition of m TORC1 by GATOR1. Knockdown of nprl3 in Drosophila resulted in epilepsy-like behavior and abnormal synaptic development. 14Global characterization of OsPIP aquaporins reveals that the H_(2)O_(2)transporter OsPIP2;6 increases resistance to rice blast显示文摘Plasma membrane intrinsic proteins(PIPs)are conserved plant aquaporins that transport small molecules across the plasma membrane to trigger instant stress responses and maintain cellular homeostasis under biotic and abiotic stress.To elucidate their roles in plant immunity to pathogen attack,we characterized the expression patterns,subcellular localizations,and H_(2)O_(2)-transport ability of 11 OsPIPs in rice(Oryza sativa),and identified OsPIP2;6 as necessary for rice disease resistance.OsPIP2;6 resides on the plasma membrane and facilitates cytoplasmic import of the immune signaling molecule H_(2)O_(2).Knockout of OsPIP2;6 increases rice susceptibility to Magnaporthe oryzae,indicating a positive function in plant immunity.OsPIP2;6 interacts with OsPIP2;2,which has been reported to increase rice resistance to pathogens via H_(2)O_(2)transport.Our findings suggest that OsPIP2;6 cooperates with OsPIP2;2 as a defense signal transporter complex during plant–pathogen interaction.Gousi Li Jingluan Han Chen Yi Hao Luo Yuzhu Wang Fengpin Wang Xiaoyu Wang Letian Chen Yaling Zhang 2024The Crop Journal2024,12,1:0
15Formation of multifaceted nano-groove structure on rutile TiO_(2) photoanode for efficient electron-hole separation and water splitting显示文摘Photoelectrochemical(PEC)water-splitting using solar energy holds great promise for the renewable energy future,and a key challenge in the development of industry viable PEC devices is the unavailability of high-efficient photoanodes.Herein,we designed a TiO_(2) model photocatalyst with nano-groove pattern and different surface orientation using low-energy Ar+irradiation and photoetching of TiO_(2),and significantly improved the intrinsic activity for PEC water oxidation.High-resolution transmission electron microscopy directly manifests that the grooves consist of highly stepped surface with<110>steps and well-crystallized.Transient absorption spectroscopy reveals the groove surface that allows for increased recovery lifetime,which ensures promoted electron-hole separation efficiency.Surface photovoltage directly shows the carrier separation and transportation behaviors,verified by selective photodeposition,demonstrating the groove surface on TiO_(2) contributes to electron-hole separation.This work proposes an efficient and scalable photoanode strategy,which potentially can open new opportunities for achieving efficient PEC water oxidation performance.Xiaoyi Zhan Yaling Luo Ziyu Wang Yao Xiang Zheng Peng Yong Han Hui Zhang Ruotian Chen Qin Zhou Hongru Peng Hao Huang Weimin Liu Xin Ou Guijun Ma Fengtao Fan Fan Yang Can Li Zhi Liu 2022Journal of Energy Chemistry2022,31,2:0
16Heterostructure of Ta_(3)N_(5) nanorods and CaTaO_(2)N nanosheets fabricated using a precursor template to boost water splitting under visible light显示文摘Fabrication of heterostructure composed of one-dimensional(1D)and 2D semiconductors has inspired extensive interest in promoting photogenerated charge separation as well as performances of solar fuel production,but it is still challenging for(oxy)nitride photocatalysts due to their uncontrollable ammonia thermal preparative process.In this work,we report a synthesis on heterostructure of Ta_(3)N_(5)nanorods and CaTaO_(2)N(CTON)nanosheets(denoted as Ta_(3)N_(5)/CTON)by directly nitriding a 2D Dion-Jacobson(DJ)type of perovskite KCa_(2)Ta_(3)O_(10)(KCTO)precursor under the assistance of K_(2)CO_(3)flux.It is demonstrated that the 2D morphology of KCTO can be well inherited to get 2D CTON,and the Ta-rich(nonstoichiometric ratio of Ca:Ta compared to CTON)feature of the KCTO as well as the easy evaporation of K species results in the formation of 1D Ta_(3)N_(5)nanorods.Meanwhile,the formation of intermediate species K_(2)Ca_(2)Ta_(3)O_(9)N owning similar crystal lattice as Ta_(3)N_(5)was detected and deduced to be responsible for the generation of Ta_(3)N_(5)nanorods and observation of intimate interface between CTON and Ta_(3)N_(5).Benefitting from the formation of special 1D/2D type-II heterostructure,obviously promoted charge separation as well as photocatalytic water splitting performance can be obtained.Extended discussion demonstrates the generality of the hard-template preparative strategy developed here.To our knowledge,this should be the first fabrication of 1D/2D heterostructure for the(oxy)nitride semiconductors,and the developed hard-template strategy may provide an alternative way of fabricating heterostructures of other semiconductors prepared at high temperature.Yanpei Luo Hao Li Yaling Luo Zheng Li Yu Qi Fuxiang Zhang Can Li 2022Journal of Energy Chemistry2022,31,4:0

19Vitamin B6 prevents Isocarbophos-induced posterior cerebral artery injury in offspring rats through up-regulating S1P receptor expression显示文摘We have previously reported that the long-term exposure of Isocarbophos,a kind of organophosphorus compounds,induces vascular dementia(VD)in rats.Studies have also shown that organophosphorus compounds have adverse effects on offsprings.Vitamin B6 is a coenzyme mainly involved in the regulation of metabolism and has been demonstrated to ameliorate VD.Sphingosine-1-phosphate(S1P),a biologically active lipid,plays a vital role in the cardiovascular system.However,whether S1P is involved in the therapeutic effects of Vitamin B6 on posterior cerebral artery injury has yet to be further answered.In the present study,we aimed to explore the potential influence of Vitamin B6 on Isocarbophos-induced posterior cerebral artery injury in offspring rats and the role of the S1P receptor in this process.We found that Vitamin B6 significantly improves the vasoconstriction function of the posterior cerebral artery in rats induced by Isocarbophos by the blood gas analysis and endothelium-dependent relaxation function assay.We further demonstrated that Vitamin B6 alleviates the Isocarbophos-induced elevation of ICAM-1,VCAM-1,IL-1,and IL-6 by using the enzyme-linked immunosorbent assay kits.By performing immunofluorescence and the western blot assay,we revealed that Vitamin B6 prevents the down-regulation of S1P in posterior cerebral artery injury.It is worth noting that Fingolimod,the S1P inhibitor,significantly inhibits the Vitamin B6-induced up-regulation of S1P in posterior cerebral artery injury.Collectively,our data indicate that Vitamin B6 may be a novel drug for the treatment of posterior cerebral artery injury and that S1P may be a drug target for its treatment.Yanhua Liu Kunli Yang Ling Wang Jinfang Yang Yang Wang Hu Luo Peng Li Yaling Yin 2021Acta Biochimica et Biophysica Sinica2021,53,12:0
