| 3 | Protective Effect of Magnesium Isoglycyrrhizinate on Ethanol-Induced Testicular Injuries in Mice显示文摘Objective:Ethanol treatment induces an increase in oxidative stress. As licorice compounds are potent antioxidants, our aim was to examine whether magnesium isoglycyrrhizinate attenuated lipid peroxidation, the major end-point of oxidative damage resulting from ethanol administration. Methods: Four groups(18 animals in each group) of male Kunming mice were used. The first group served as control and received 0.4 ml normal saline daily for 18 days orally. The second group of mice was given 56% ethanol at 16 ml/kg body weight per day for 18 days orally. The third group was given the same dose of ethanol and administrated magnesium isoglycyrrhizinate (15 mg/kg.d, i.p.) for 18 days. The fourth group was given the same dose of ethanol and administrated with magnesium isoglycyrrhizinate (45 mg/kg.d, i.p.) for 18 days. Twenty four hours after 9 days or 18 days of treatment the mice were sacrificed using 10% chloral hydrate. Sperm counts and motility in the epididymis were assessed. The lipid peroxidation and antioxidants of testicular mitochondria were also determined. The pathological changes of testicle tissue of the mice were observed by light microscopy. Results: Magnesium isoglycyrrhizinate effectively prevented the ethanol-induced seminiferous epithelium disorganization and degeneration of Sertoli cells and germ cells. Sperm counts and motility of the magnesium isoglycyrrhizinate treated groups were higher than those of the alcohol treated group, but were lower than those of the control group. The drug exhibited an ability to counteract ethanol induced oxidative challenge as it effectively reduced testicular malondialdehyde (MDA) and increased the activities of superoxide dismutase and glutathione peroxidase. | 10 | Macrophage membrane-biomimetic adhesive polycaprolactone nanocamptothecin for improving cancer-targeting efficiency and impairing metastasis显示文摘The recent remarkable success and safety of mRNA lipid nanoparticle technology for producing severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccines has stimulated intensive efforts to expand nanoparticle strategies to treat various diseases.Numerous synthetic nanoparticles have been developed for pharmaceutical delivery and cancer treatment.However,only a limited number of nanotherapies have enter clinical trials or are clinically approved.Systemically administered nanotherapies are likely to be sequestered by host mononuclear phagocyte system(MPS),resulting in suboptimal pharmacokinetics and insufficient drug concentrations in tumors.Bioinspired drug-delivery formulations have emerged as an alternative approach to evade the MPS and show potential to improve drug therapeutic efficacy.Here we developed a biodegradable polymer-conjugated camptothecin prodrug encapsulated in the plasma membrane of lipopolysaccharide-stimulated macrophages.Polymer conjugation revived the parent camptothecin agent(e.g.,7-ethyl-10-hydroxy-camptothecin),enabling lipid nanoparticle encapsulation.Furthermore,macrophage membrane cloaking transformed the nonadhesive lipid nanoparticles into bioadhesive nanocamptothecin,increasing the cellular uptake and tumor-tropic effects of this biomimetic therapy.When tested in a preclinical murine model of breast cancer,macrophage-camouflaged nanocamptothecin exhibited a higher level of tumor accumulation than uncoated nanoparticles.Furthermore,intravenous administration of the therapy effectively suppressed tumor growth and the metastatic burden without causing systematic toxicity.Our study describes a combinatorial strategy that uses polymeric prodrug design and cell membrane cloaking to achieve therapeutics with high efficacy and low toxicity.This approach might also be generally applicable to formulate other therapeutic candidates that are not compatible or miscible with biomimetic delivery carriers. | Kangkang Ying Yifeng Zhu Jianqin Wan Chenyue Zhan Yuchen Wang Binbin Xie Peirong Xu Hongming Pan Hangxiang Wang | 2023 | Bioactive Materials2023,,2: | 1 |
| 11 | The Influence of Meridional Variation in North Pacific Sea Surface Temperature Anomalies on the Arctic Stratospheric Polar Vortex显示文摘This study examines the dependence of Arctic stratospheric polar vortex(SPV)variations on the meridional positions of the sea surface temperature(SST)anomalies associated with the first leading mode of North Pacific SST.The principal component 1(PC1)of the first leading mode is obtained by empirical orthogonal function decomposition.Reanalysis data,numerical experiments,and CMIP5 model outputs all suggest that the PC1 events(positive-minus-negative PC1 events),located relatively northward(i.e.,North PC1 events),more easily weaken the Arctic SPV compared to the PC1 events located relatively southward(i.e.,South PC1 events).The analysis indicates that the North PC1-related Aleutian low anomaly is located over the northern North Pacific and thus enhances the climatological trough,which strengthens the planetary-scale wave 1 at mid-to-high latitudes and thereby weakens the SPV.The weakened stratospheric circulation further extends into the troposphere and favors negative surface temperature anomalies over Eurasia.By contrast,the South PC1-related Aleutian low anomaly is located relatively southward,and its constructive interference with the climatological trough is less efficient at high latitudes.Thus,the South PC1 events could not induce an evident enhancement of the planetary-scale waves at high latitudes and thereby a weakening of the SPV on average.The Eurasian cooling associated with South PC1 events(positive-minus-negative PC1 events)is also not prominent.The results of this study suggest that the meridional positions of the PC1 events may be useful for predicting the Arctic SPV and Eurasian surface temperature variations. | Tao WANG Qiang FU Wenshou TIAN Hongwen LIU Yifeng PENG Fei XIE Hongying TIAN Jiali LUO | 2023 | Advances in Atmospheric Sciences2023,40,12: | 1 |

| 14 | Qilan preparation(芪蓝颗粒) inhibits proliferation and induces apoptosis by down-regulating microRNA-21 in humaqin Tca8113 tongue squamous cell carcinoma cells显示文摘OBJECTIVE: The aim of this study was to examine the antitumor effects of Qilan preparation(芪蓝颗粒) on oral squamous cell carcinoma(OSCC) in vitro and to investigate its underlying mechanisms of action. METHODS: Cell proliferation, cell cycle distribution and apoptosis were examined using cell counting kit-8(CCK8) and flow cytometry(FCM). The expression of PTEN and PDCD4 were determined by western blot. Changes in miR-21 levels were quantified using Taq Man stem-loop real-time PCR. After miR-21 was transiently transfected into Tca8113 cells using Lipofectamine?3000, cell proliferation, apoptosis and miR-21 and PDCD4 expression levels were measured. RESULTS: Qilan preparation inhibited Tca8113 cell growth in a dose-and time-dependent manner by inducing apoptosis and cell cycle arrest in S-phase, decreasing miR-21 levels and increasing PTEN and PDCD4 expression. | 20 | Modeling of the whole hydrocarbon-generating process of sapropelic source rock显示文摘Based on experimental data from hydrocarbon generation with a semi-open system, hydrocarbon generation kinetics modeling in gold tube of closed system, high temperature pyrolysis chromatography mass spectrometry experiment with open system and geological data, the characteristics of whole hydrocarbon-generating process, hydrocarbon expulsion efficiency and retained hydrocarbon quantity, origins of natural gas generated in high-over mature stage and cracking temperature of methane homologs were investigated in this study. The sapropelic source rock has a hydrocarbon expulsion efficiency of 30%-60% and 60%-80% in the major oil generation window(with Ro of 0.8%-1.3%) and high maturity stage(with Ro of 1.3%-2.0%) respectively; and the contribution ratio of kerogen degradation gas to oil cracking gas in total generated gas in high maturity stage is about 1:4. The degradation gas of kerogen accounts for 20%, the retained liquid hydrocarbon cracking gas accounts for 13.5%, and the amount of out-reservoir oil cracking gas(including aggregation type and dispersed oil cracking gas) accounts for 66.5%. The lower limit of gas cracking is determined preliminarily.
